Mosquito repellency mediated by skin commensal Bacillus species via volatile metabolite reprogramming.

Individual variation in human attractiveness to mosquitoes critically influences vector-borne disease transmission, and this phenomenon arises from multiple underlying factors. This study integrated human skin microbiome profiling, non-human primate in situ intervention, and volatilomics to uncover a microbial-mediated mechanism underlying host-specific mosquito repellency. Bacillus species were identified as a microbial signature associated with reduced mosquito attraction in humans. Targeted Intervention of B. licheniformis and B. paralicheniformis on primate skin was sufficient to significa

A single mutation in the mosquito (Aedes aegypti) olfactory receptor 8 causes loss of function to 1-octen-3-ol.

The host-seeking behavior of mosquitoes have long been established to be primarily odor-mediated. In this process, olfactory receptors (Ors) play a critical role. 1-Octen-3-ol is a common volatile compound that is attractive to hematophagous arthropods such as mosquitos. The olfactory receptor 8 (AaOr8) on the tip of the stylet and maxillary palp of Aedes aegypti is tuned to 1-octen-3-ol, which is required for mosquitoes to quickly find blood vessels from a vertebrate host. However, little is known about the interaction of AaOr8 with 1-octen-3-ol which was studied in vivo and in silico in this

Linkages of Weather and Climate With Ixodes scapularis and Ixodes pacificus (Acari: Ixodidae), Enzootic Transmission of Borrelia burgdorferi, and Lyme Disease in North America.

Lyme disease has increased both in incidence and geographic extent in the United States and Canada over the past two decades. One of the underlying causes is changes during the same time period in the distribution and abundance of the primary vectors: Ixodes scapularis Say and Ixodes pacificus Cooley and Kohls in eastern and western North America, respectively. Aside from short periods of time when they are feeding on hosts, these ticks exist in the environment where temperature and relative humidity directly affect their development, survival, and host-seeking behavior. Other important factor

Escherichia coli Mono-Association Modulates Ionotropic Receptor-Dependent Behaviors in Drosophila melanogaster.

Chemosensory systems are crucial for insect survival, enabling host-seeking, food acquisition, and oviposition site selection. While insect-associated microbes are known to influence host development and immunity, their role in modulating chemosensory behavior remains poorly understood. Here, we show that mono-association with Escherichia coli alters sensory-driven behaviors in both larval and adult axenic Drosophila melanogaster. In larvae, E. coli mono-association altered phototaxis and mechanosensory responses across genotypes, while changes in tunneling and thermosensory behaviors were red

Effect of body size on host seeking and blood meal utilization in Anopheles gambiae sensu stricto (Diptera: Culicidae): the disadvantage of being small.

The survival, metabolic reserves, and host-seeking response of unfed Anopheles gambiae Giles sensu stricto females of different size classes were investigated from 1 to 6 d after emergence. In addition, the effect of blood meal size and frequency of feeding on the accumulation of metabolic reserves and reproductive development were investigated during the 1st gonotrophic cycle. Large females survived longer and contained significantly more protein, glycogen, and lipid at emergence than did small females. Over 6 d, the host-seeking response of large mosquitoes was greater than that of small one

Environmental Factors Affecting Survival of Immature Ixodes scapularis and Implications for Geographical Distribution of Lyme Disease: The Climate/Behavior Hypothesis.

Recent reports suggest that host-seeking nymphs in southern populations of Ixodes scapularis remain below the leaf litter surface, while northern nymphs seek hosts on leaves and twigs above the litter surface. This behavioral difference potentially results in decreased tick contact with humans in the south, and fewer cases of Lyme disease. We studied whether north-south differences in tick survival patterns might contribute to this phenomenon. Four month old larvae resulting from a cross between Wisconsin males and South Carolina females died faster under southern than under northern condition

Transcriptomic analysis of eggs, rediae and cercariae reveal stage-specific adaptations in the rumen fluke Calicophoron daubneyi.

Rumen flukes, particularly the trematode Calicophoron daubneyi, are emerging parasites of livestock in Europe, yet transcriptomic insights into their environmental and intermediate host stages remain limited. Here, we present a comprehensive transcriptomic analysis of eggs at three distinct developmental stages (freshly excreted, early developmental and eye-spot stages), as well as rediae and cercariae, of C. daubneyi. High-quality RNA-sequencing (RNA-seq) datasets revealed both shared and stage-specific transcriptional profiles with each developmental stage exhibiting its own distinct express

Dog hair volatiles attract Rhipicephalus sanguineus sensu lato (Acari: Ixodidae) females.

Rhipicephalus sanguineus sensu lato (s.l.) (Latreille) feeds on domestic dogs worldwide. This tick species uses dog volatiles during host-seeking behavior. In this study, we identified volatile compounds from dog hairs involved in the host location of R. sanguineus s.l. The R. sanguineus s.l. females, but not males, were attracted to hair samples and Super Q extracts from Schnauzer dogs in Y-tube olfactometer bioassays. A total of 54 compounds from dog hair extracts were identified by gas chromatography coupled to mass spectrometry, including hydrocarbons, aldehydes, alcohols, ketones, and car

Invasive brown algae (Sargassum spp.) as a potential source of biocontrol against Aedes aegypti.

Influxes of sargassos are responsible for economic and environmental disasters in areas where they bloom, especially in regions whose main income relies on tourism and with limited capacity for sanitation and public health response. A promising way of valorization would be to convert this incredible biomass into tools to fight the deadly vector mosquito Aedes aegypti. In the present study, we generated hydrolates and aqueous extracts from three main Sargassum morphotypes identified in Guadeloupe (French West Indies): Sargassum natans VIII, Sargassum natans I and Sargassum fluitans. Host movement and time of year influence tick parasitism in Pantanal birds.

Ticks are among the best studied parasitic groups as they spread important pathogens of medical and veterinary importance worldwide. Migratory birds can play an important role in transporting ticks infected with pathogens across wide geographic regions. It is therefore important to understand which factors promote tick parasitism rates across their avian hosts and the associated potential for disease spread. Here, we identified the host attributes of infestation probability of ticks from the genus Amblyomma in 955 birds from Pantanal, Brazil. Infestation rates exhibited considerable variation

Chemical Ecology and Management of Dengue Vectors.

Dengue, caused by the dengue virus, is the most widespread arboviral infectious disease of public health significance globally. This review explores the communicative function of olfactory cues that mediate host-seeking, egg-laying, plant-feeding, and mating behaviors in Aedes aegypti and Aedes albopictus, two mosquito vectors that drive dengue virus transmission. Aedes aegypti has adapted to live in close association with humans, preferentially feeding on them and laying eggs in human-fabricated water containers and natural habitats. In contrast, Ae. albopictus is considered opportunistic in

Impact of disabled circadian clock on yellow fever mosquito Aedes aegypti fitness and behaviors.

Like other insects, Aedes aegypti displays strong daily patterns in host seeking and mating. Much of these behaviors are believed to be under the control of a circadian clock, an endogenous timekeeping mechanism relying on transcriptional/translational negative feedback loops that drive rhythmic physiology and behavior. To examine the connection between the circadian clock and various Ae. aegypti behaviors, we knocked out the core clock gene cycle using CRISPR/Cas9. We found that the rhythmic pattern and intensity of mRNA expression of seven circadian genes, including AeCyc-/-, were altered ac

Ammonium transporter AcAmt mutagenesis uncovers reproductive and physiological defects without impacting olfactory responses to ammonia in the malaria vector mosquito Anopheles coluzzii.

Anopheline mosquitoes are the sole vectors of malaria and rely on olfactory cues for host seeking in which ammonia derived from human sweat plays an essential role. To investigate the function of the Anopheles coluzzii ammonium transporter (AcAmt) in the mosquito olfactory system, we generated an AcAmt null mutant line using CRISPR/Cas9. AcAmt-/- mutants displayed a series of novel phenotypes compared with wild-type mosquitoes including significantly lower insemination rates during mating and increased mortality during eclosion. Furthermore, AcAmt-/- males showed significantly lower sugar cons

Established populations of Ixodes silvanus (Acari: Ixodidae) in the Chaco Serrano, Argentina.

The aim of the study was to report the occurrence of Ixodes silvanus outside its distribution range, in the Chaco Serrano vegetation unit of the Chaco Phytogeographic Province, central Argentina. Ticks were collected from vegetation and on birds between November 2021 and December 2023. A possible seasonal activity was observed in the immature stages of I. silvanus, with larvae in autumn-winter and nymphs in spring-summer, consistent with a behavioral diapause. A total of 937 ticks were identified as I. silvanus and confirmed molecularly, 741 host-seeking and 196 parasitizing 49 passerine birds

Olfactory circuits and behaviors of nematodes.

Over one billion people worldwide are infected with parasitic nematodes. Many parasitic nematodes actively search for hosts to infect using volatile chemical cues, so understanding the olfactory signals that drive host seeking may elucidate new pathways for preventing infections. The free-living nematode Caenorhabditis elegans is a powerful model for parasitic nematodes: because sensory neuroanatomy is conserved across nematode species, an understanding of the microcircuits that mediate olfaction in C. elegans may inform studies of olfaction in parasitic nematodes. Here we review circuit mecha

Methods of sampling malaria vectors and their reliability in estimating entomological indices in Africa.

In efforts to intensify malaria control through vector control and hasten the progress towards elimination, the impact of control interventions needs to be evaluated. This requires sampling vector population using appropriate trapping methods. The aim of this article is to critically review methods of sampling malaria vectors and their reliability in estimating entomological indicators of malaria transmission in Africa. The standard methods are human landing catch (HLC), pyrethrum spray catch, and pit shelter for sampling host-seeking, indoor resting, and outdoor resting malaria vectors, respe

[The effect of ivermectin on the transmission of Onchocerca volvulus in southern Mexico].

We assessed the transmission potential and the mean infected and infective biting density of S. ochraceum in an onchocerciasis endemic community in the southern Mexican state of Chiapas. In addition, the impact of first treatment of ivermectin during the peak of natural infection of host-seeking S. ochraceum populations was also evaluated. Monthly entomological sampling was carried out during the year before treatment to assess the seasonal biting behavior of parous host-seeking S. ochraceum females, and either infected (with whatever stage of O. volvulus) or infective females (with only third

Sexually Dimorphic Visual Processing in the Yellow-Fever Mosquito, Aedes aegypti.

Flying insects process motion across the visual scene to regulate their ground speed, correct course deviations, and respond to behaviorally relevant stimuli such as avoiding predators or finding hosts. The yellow-fever mosquito Aedes aegypti provides a useful model for studying these visually guided behaviors, as both sexes use vision to track moving features. Approaching a moving target presents mosquitoes with a demanding visual problem: they must extract target motion from the changing visual scene while simultaneously controlling their own flight and responding to potentially dangerous mo

Early life stress and susceptibility to addiction in adolescence.

Early life stress (ELS) is a risk factor for developing a host of psychiatric disorders. Adolescence is a particularly vulnerable period for the onset of these disorders and substance use disorders (SUDs). Here we discuss ELS and its effects in adolescence, especially SUDs, and their correlates with molecular changes to signaling systems in reward and stress neurocircuits. Using a maternal separation (MS) model of neonatal ELS, we studied a range of behaviors that comprise a "drug-seeking" phenotype. We then investigated potential mechanisms underlying the development of this phenotype. Cortic

Density and behavior of capybara (Hydrochoerus hydrochaeris) ticks (Acari: Ixodidae) Amblyomma sculptum and Amblyomma dubitatum with notes on Rickettsia bellii infection: Assessing human exposure risk.

In several urban and peri‑urban areas of Brazil, populations of Amblyomma sculptum and Amblyomma dubitatum ticks are maintained by capybaras (Hydrochoerus hydrochaeris). In some of these areas, this host and these tick species are associated with Brazilian spotted fever (BSF), a lethal human disease caused by the bacterium Rickettsia rickettsii. In this work, we evaluated the risk of human exposure to these tick species using four collection techniques to discern host-seeking behavior. The study was carried out in 10 urban sites inhabited by capybaras in Uberlândia, a BSF-free municipality in
