Development of a Physiologically Based Pharmacokinetic Model for Inorganic Mercury Salts in Rodents and Humans and Its Application in the Derivation of Health-Based Toxicity Metrics.

Inorganic mercury salts (iHg) are mercury compounds that lack carbon-hydrogen bonds. Exposure to iHg is associated with toxic effects, primarily shown in animal studies, yet the lack of cross-species toxicokinetic models has limited our ability to assess human health risks. This study aimed to develop and validate the first physiologically based pharmacokinetic (PBPK) model for iHg, incorporating species-specific physiological and chemical parameters to better characterize iHg kinetics in mice, rats, and humans. We included a hierarchical Markov chain Monte Carlo (MCMC) approach to address the

Thiotemplated Polyketide Chain Fusion and Reductive Cyclization Build the Reactive Butenolide Core of Malleicyprol.

Malleicyprol, a virulence factor of notorious animal and human pathogens of the Burkholderia pseudomallei (BP) group, features a molecular cyclopropanol warhead linked to a reactive butenolide core. Biosynthetic considerations suggested that this heterocycle was formed by the merger of two individual polyketide chains, but the precise mechanism has remained elusive. By combining chemical synthesis, complete in vitro reconstitution of the biotransformation, and mutational analysis, we show that two individually generated polyketide chains are joined by a noncanonical condensation domain of the

Exploring the Dynamics of Canine-Assisted Interactions: A Wearable Approach to Understanding Interspecies Well-Being.

Canine-assisted interactions (CAIs) have been explored to offer therapeutic benefits to human participants in various contexts, from addressing cancer-related fatigue to treating post-traumatic stress disorder. Despite their widespread adoption, there are still unresolved questions regarding the outcomes for both humans and animals involved in these interactions. Previous attempts to address these questions have suffered from core methodological weaknesses, especially due to absence of tools for an efficient objective evaluation and lack of focus on the canine perspective. In this article, we

Neuropeptide oxytocin enhances μ opioid receptor signaling as a positive allosteric modulator.

Oxytocin (OT) is a 9-amine neuropeptide that plays an essential role in mammalian labor, lactation, maternal bonding, and social affiliation. OT has been reported to exert an analgesic effect in both humans and animals, and the results of certain animal experiments have shown that the analgesic effect of OT is partially blocked by opioid receptor antagonists. To investigate the relationship between OT and μ opioid receptor (MOR), we evaluated how OT affects MOR in vitro by performing an electrical impedance-based receptor biosensor assay (CellKey™ assay), an intracellular cAMP assay, and a com

Cure of SCID mice bearing human B-lymphoma xenografts by an anti-CD74 antibody-anthracycline drug conjugate.

PURPOSE: The purpose of this research was to test the therapeutic efficacy of an anthracycline-antibody conjugate for the treatment of human B-cell lymphoma in a preclinical animal model. EXPERIMENTAL DESIGN: Doxorubicin (dox) conjugates of the murine and humanized versions of the anti B-cell antibody LL1, targeting CD74, were prepared, along with a nonspecific control dox-antibody conjugate, targeting carcinoembryonic antigen. Antibody conjugates carried approximately 8-10 drug molecules attached site-specifically at thiols of reduced interchain disulfide bonds. Conjugates were tested, initi

Genetic and peripheral markers of the oxytocin system and parental care jointly support the cross-generational transmission of bonding across three generations.

BACKGROUND: Human and animal research indicates that oxytocin (OT) plays a key role in the cross-generational transmission of parental bonding, and human studies suggest that allelic variations on the oxytocin receptor gene (OXTR) and circulating OT levels interact with patterns of parental care to shape children's social-affiliative competencies. Yet, no study to date has tested the joint contribution of OT and parental care across three generations. METHODS: The study included 345 participants comprising 115 family lines of grandmothers, mothers, and their infants. Salivary OT and allelic v

ER O-glycosylation in synovial fibroblasts drives cartilage degradation.

How arthritic synovial fibroblasts (SFs) activate cartilage ECM degradation remains unclear. GALNT enzymes initiate O-glycosylation in the Golgi; when relocated to the ER, their activity stimulates ECM degradation. Here, we show that in human rheumatoid and osteoarthritic synovial SFs, GALNTs are relocated to the ER. In an RA mouse model, GALNTs relocation occurs shortly before arthritis symptoms and abates as the animal recovers. An ER GALNTs inhibitor prevents cartilage ECM degradation in vitro and expression of this chimeric protein in SFs results in the protection of cartilage. One of the

A europium (III) functionalized hydrogen-bonded organic framework for sensitively ratiometric fluorescent sensing of tetracycline.

As a kind of antibiotic, tetracycline (TC) might remain in animal blood and milk products during use, which poses a risk to humans after consumption. Therefore, a ratiometric fluorescence probe was proposed for the detection of TC, which was based on an Eu3+ functionalized hydrogen-bonded organic framework (HOF). Since there are a large number of N and O atoms in the skeleton of HOF, more Eu3+ could be loaded onto HOF by forming coordinate bonds, while preserving the fluorescence of luminol monomer in HOF. In the presence of TC, the fluorescence of luminol monomer was attenuated at 425 nm due

Transmission and epidemiology of zoonotic protozoal diseases of companion animals.

Over 77 million dogs and 93 million cats share our households in the United States. Multiple studies have demonstrated the importance of pets in their owners' physical and mental health. Given the large number of companion animals in the United States and the proximity and bond of these animals with their owners, understanding and preventing the diseases that these companions bring with them are of paramount importance. Zoonotic protozoal parasites, including toxoplasmosis, Chagas' disease, babesiosis, giardiasis, and leishmaniasis, can cause insidious infections, with asymptomatic animals bei

Opioid Regulation of Social Homeostasis: Connecting Loneliness to Addiction.

Loneliness heightens the risk of substance use disorder, and a desire to escape this negative feeling motivates drug use. Opioid drugs in particular are believed to target neurobiological circuits involved in social bonding, increasing vulnerability to opioid addiction when social connectedness is lacking. In this narrative review, we consider how current understanding of μ opioid modulation of reward and threat processing across domains sheds light on the mechanisms that link loneliness and substance use. We discuss evidence for state- and context-dependent μ opioid modulation of social affec

Unmet Pet Caregiving Needs are Associated with Depression and Anxiety Symptoms Among People Living with HIV.

People living with HIV (PLWH) experience disproportionately high rates of depression and anxiety. Human-animal interactions (HAI), particularly through pet ownership, may provide emotional support and companionship; however, the human-animal bond can be threatened by structural and socioeconomic factors that impede one's ability to meet pets' needs. The current study examined how comfort from pets and unmet pet caregiving needs were associated with depression and anxiety symptoms among pet-owning PLWH. Data were derived from the Florida Cohort (2021-2023), a survey study of PLWH in Florida. Pa

Pet Ownership is Associated with Durable Viral Suppression but Presents Healthcare Access Challenges for People with HIV.

Human-animal interaction (HAI; e.g., pet ownership) may contribute to enhanced health and well-being among individuals managing chronic conditions like HIV; however, responsibilities associated with pet ownership may also prevent owners from accessing timely healthcare. This study investigates the relationship between pet ownership, pet-related barriers to healthcare (PRBH), and comfort derived from pets with durable HIV viral suppression among people with HIV (PWH) in Florida. We tested three hypotheses using survey data from the Florida Cohort Study linked with Florida Department of Health H

Colonization dynamics of Klebsiella pneumoniae in the pet animals and human owners in a single household.

Klebsiella pneumoniae resides in the gastrointestinal (GI) microbiota of humans and animals. To characterize the population dynamics of GI-colonizing K. pneumoniae, we examined the clonality of K. pneumoniae isolates, which were longitudinally collected from the fecal samplings of a healthy married couple and their pet animals during Sep. 2015 to Oct. 2016. As revealed by XbaI-PFGE analysis, the K. pneumoniae populations detected in the male owner and in one of the dogs, consisted of clonally diverse K. pneumoniae isolates; whereas, a dominant clone persisted in the GI tract of the female owne

Secretory mouse quiescin sulfhydryl oxidase 1 aggregates defected human and mouse spermatozoa in vitro and in vivo.

A flavin-dependent enzyme quiescin Q6 sulfhydryl oxidase 1 (QSOX1) catalyzes the oxidation of thiol groups into disulfide bonds. QSOX1 is prominently expressed in the seminal plasma. However, its role in male reproduction is elusive. Here, we purified the secreted form of QSOX1, i.e., QSOX1c, from mouse seminal vesicle secretions and revealed for the first time its function involved in sperm physiology. Exogenous addition of QSOX1c time-dependently promoted the in vitro aggregation of thiol-rich, oxidative stressed, and apoptotic mouse and human sperm cells. Also, in vivo aggregated sperm cell

Interaction of plant polyphenols with salivary proteins.

Tannins are polyphenols that occur widespread in plant-based food. They are considered to be part of the plant defense system against environmental stressors. Tannins have a number of effects on animals, including growth-rate depression and inhibition of digestive enzymes. Tannins also have an effect on humans: They are, for example, the cause of byssinosis, a condition that is due to exposure to airborne tannin. Their biological effect is related to the great efficiency by which tannins precipitate proteins, an interaction that occurs by hydrophobic forces and hydrogen bonding. Two groups of

Effects of cocaine on maternal behavior and neurochemistry.

Drug addiction is a chronic relapsing disorder that involves drug seeking and abuse despite the negative social and health consequences. While the potential effects of cocaine on child development have been extensively studied over the last 30 years, few researchers have focused on the effects of cocaine on maternal behavior, which includes offspring care and maternal aggression towards an unfamiliar individual. In humans, maternal cocaine use can lead to child neglect, abuse, and disrupt the mother-child bond. While it has been argued the developmental effects of maternal cocaine use on child

Sensitive determination of the peptide AP301--a motif of TNF-α--from human plasma using HPLC-MS/MS.

The AP301 peptide mimics the lectin-like domain of TNF-α. The synthetic peptide AP301 (molecular weight 1923.1amu) is composed of 17 amino acids and contains an intramolecular disulfide bond between the N-terminal and the C-terminal cysteine. AP301 interacts with the endothelial sodium channel (ENaC) and activates pulmonary liquid clearance both in vitro and in animal studies. Currently, AP301 is subject to clinical investigations for the treatment of pulmonary oedema. With HPLC-MS/MS on reversed phase chromatography a determination limit of 1ng AP301/mL human plasma can be achieved. The MS-io
