Plate tectonic history and ocean oxygenation shaping biogeography of hydrothermal bacterial community.

The influence of plate tectonics on microbial distributions remains poorly understood. Here, we demonstrate that the global biogeography of hydrothermal vent-endemic chemoautotrophic microbiota is structured by the tectonic history of the global major oceans. These microbiota, particularly anaerobic ones, are significantly more abundant in early-origin Pacific, Arctic, and Mediterranean oceans, whereas they are notably scarce in late-formed Atlantic and Indian Oceans. This pattern was attributed to the timing of ocean formation and its interplay with global redox evolution. Fully oxygenated co

Hydrothermal origin of metabolic phosphorylation.

Phosphate is central to modern bioenergetics and to all theories for the origin of life. How phosphate entered metabolism is unknown, though microbial physiology and geochemical environments can provide important clues. Some bacteria obtain electrons and energy from phosphite (HPO3 2-), a reduced form of phosphate (HPO4 2-), that naturally occurs in serpentinizing (H2-producing) hydrothermal systems. Here we show that the insoluble, solid-state catalyst native palladium, which is naturally deposited in serpentinizing hydrothermal systems, catalyzes the oxidation of phosphite to phosphate and H

Diverse biosynthetic pathways in Arctic hydrothermal biofilms.

Biosynthetic gene clusters (BGCs) which encode diverse secondary metabolites are ubiquitous across microbiomes and support critical ecological functions. They also serve as attractive targets for new drug discovery. Here we combined genome-resolved metagenomics with long-read Nanopore RNA sequencing, yielding 1016 bacterial and 124 archaeal medium-to-high quality MAGs from previously unchartered microbial communities in Arctic hydrothermal vent biofilms. We identified 2965 BGCs from 870 metagenome-assembled genomes (MAGs) comprising a distinctive and rich diversity of BGCs, with ribosomally sy

Electrosynthetic bacterial growth under conditions simulating electric discharge in deep-sea hydrothermal fields.

Microbial electrosynthesis is a metabolic process in which extracellular electrons are utilized as the primary energy source for carbon fixation. While microbial electrosynthesis has been proposed as a novel concept for ecological primary production, our understanding of how such microorganisms are distributed in natural environments remains limited. In this study, we constructed a laboratory-scale electrochemical cultivation system that simulates electric discharge conditions in deep-sea hydrothermal fields. Microscopic counts revealed increased cell numbers in the electrochemical culture, an

Shiny metal catalysts at the origin of metabolism.

Metabolism is the reaction network that converts inanimate matter into living cells. There is no life without it. The network of approximately 400 enzymatic reactions that synthesize amino acids, nucleosides, and cofactors from compounds in the environment is conserved across life. Before enzymes, how did metabolism arise? The subsurface chemical environment of Earth's crust reveals clues. Serpentinizing (H2 producing) hydrothermal vents contain naturally deposited, shiny metals in their reduced (native) state and phosphite, a reduced form of phosphate. In the laboratory, native metal catalyst

Theoretical Study of Formation of Methanol under Hydrothermal Conditions.

We have examined the formation process of methanol by the reduction of formaldehyde under hydrothermal conditions. A formaldehyde absorbs a hydrogen molecule and turns to a methanol. Water molecules near a formaldehyde help to transfer protons to proceed the reduction process efficiently. The energy barrier for the reduction of a formaldehyde becomes 32.8 kcal/mol when a water cluster with five water molecules catalyzes the reduction. The ionic product becomes the largest under hydrothermal conditions. We introduce the acid-base catalytic effect due to hydronium and hydroxide on the reduction

Arsenic and Mercury Distribution in an Aquatic Food Chain: Importance of Femtoplankton and Picoplankton Filtration Fractions.

Arsenic (As) and mercury (Hg) were examined in the Yellowstone Lake food chain, focusing on two lake locations separated by approximately 20 km and differing in lake floor hydrothermal vent activity. Sampling spanned from femtoplankton to the main fish species, Yellowstone cutthroat trout and the apex predator lake trout. Mercury bioaccumulated in muscle and liver of both trout species, biomagnifying with age, whereas As decreased in older fish, which indicates differential exposure routes for these metal(loid)s. Mercury and As concentrations were higher in all food chain filter fractions (0.1

Halophilic Archaea determined from geothermal steam vent aerosols.

Hydrothermal vents, known as 'fumaroles', are ubiquitous features of geothermal areas. Although their geology has been extensively characterized, little is known about the subsurface microbial ecology of fumaroles largely because of the difficulty in collecting sufficient numbers of cells from boiling steam water for DNA extraction and culture isolation. Here we describe the first collection, molecular analysis and isolation of microbes from fumarole steam waters in Russia (Kamchatka) and the USA (Hawaii, New Mexico, California and Wyoming). Surprisingly, the steam vent waters from all the fum

Protein biochemistry and engineering drive the development of a carbonic anhydrase-based carbon dioxide sequestration strategy.

The sequestration of carbon dioxide using carbonic anhydrase (CA) is one of the most effective methods for mitigating global warming. The burning of fossil fuels releases large quantities of flue gas; because of its high temperature and of the alkaline conditions required for CaCO3 precipitation in the mineralization process, thermo-alkali-stable CAs are needed. In this context, Manyumwa et al. conducted a biochemical characterization of three CAs derived from thermophilic bacteria. They then employed a rational design approach to enhance the specific activity and stability of the enzyme from

Unconventional Stiffening Behavior of Amphiphilic Triblock Copolymers in Aqueous Environments for Thermally Activated Artificial Muscles.

Biological systems achieve multifunctionality through dynamic stiffness modulation, yet replicating this adaptability in synthetic actuators remains challenging. Paradigm-shifting thermally triggered shape-memory polymers (SMPs) are reported that defies conventional thermal softening behavior. By designing amphiphilic triblock copolymers with various block chain lengths, water-induced microphase separation is exploited to achieve anomalous enhancement in stiffness-specially, a remarkable 170-fold modulus increase in aqueous environments at 25 °C, along with a sustained 1.8-fold amplification e

Key role of hydrogen in regulating hydrogenases and the reductive TCA cycle in a thermophilic, autotrophic sulfur-reducing bacterium.

UNLABELLED: Many thermophilic autotrophs in deep-sea hydrothermal vents are anaerobes that require H2 for growth. However, our understanding of how non-methanogenic thermophilic hydrogenotrophs adapt to low H2 conditions is nascent. In this study, the thermophilic autotrophic thiosulfate-reducing bacterium Desulfurobacterium thermolithotrophum HR11 was grown at 72°C in a chemostat at 2 and 43 µM aqueous H2 concentrations to quantify changes in growth and H2S production kinetics and in its proteome in response to H2 availability. At 2 μM H2, cultures showed a decrease in specific growth rate, m

Possible Roles of Hypotaurine and Thiotaurine in the Vesicomyid Clam Phreagena okutanii.

AbstractVesicomyid clams, which inhabit deep-sea hydrothermal vents and hydrocarbon seeps, are nutritionally dependent on symbiotic, chemoautotrophic bacteria that produce organic matter by using hydrogen sulfide. Vesicomyid clams absorb hydrogen sulfide from the foot and transport it in their hemolymph to symbionts in the gill. However, mechanisms to cope with hydrogen sulfide toxicity are not fully understood. Previous studies on vent-specific invertebrates, including bathymodiolin mussels, suggest that hypotaurine, a precursor of taurine, mitigates hydrogen sulfide toxicity by binding it to

Gene-centered metagenome analysis of Vulcano Island soil (Aeolian archipelago, Italy) reveals diverse microbial key players in methane, hydrogen and sulfur cycles.

The Aeolian archipelago is known worldwide for its volcanic activity and hydrothermal emissions, of mainly carbon dioxide and hydrogen sulfide. Hydrogen, methane, and carbon monoxide are minor components of these emissions which together can feed large quantities of bacteria and archaea that do contribute to the removal of these notorious greenhouse gases. Here we analyzed the metagenome of samples taken from the Levante bay on Vulcano Island, Italy. Using a gene-centric approach, the hydrothermal vent community appeared to be dominated by Proteobacteria, and Sulfurimonas was the most abundant

Continuous radon monitoring during seven years of volcanic unrest at Campi Flegrei caldera (Italy).

This is a seven-year study (1/7/2011-31/12/2017) of radon monitoring at two sites of Campi Flegrei caldera (Neaples, Southern Italy) that in the last 70 years experienced repeated phases of volcanic unrest. The sites are equipped with devices for radon detection, based on the spectrometry analysis of the α-particles of radon daughters. A hybrid method, as combination of three known methods, is applied for the identification of residuals (anomalies) and trends of the time series of Radon. The results are compared with the following indicators of current caldera unrest: the tremor caused by the

Comparative Metagenomics Highlight a Widespread Pathway Involved in Catabolism of Phosphonates in Marine and Terrestrial Serpentinizing Ecosystems.

Serpentinizing hydrothermal systems result from water circulating into the subsurface and interacting with mantle-derived rocks notably near mid-ocean ridges or continental ophiolites. Serpentinization and associated reactions produce alkaline fluids enriched in molecular hydrogen, methane, and small organic molecules that are assumed to feed microbial inhabitants. In this study, we explored the relationships linking serpentinization to associated microbial communities by comparative metagenomics of serpentinite-hosted systems, basalt-hosted vents, and hot springs. The shallow Prony bay hydrot

Soil processes modify the composition of volatile organic compounds (VOCs) from CO2- and CH4-dominated geogenic and landfill gases: A comprehensive study.

Degradation mechanisms affecting non-methane volatile organic compounds (VOCs) during gas uprising from different hypogenic sources to the surface were investigated through extensive sampling surveys in areas encompassing a high enthalpy hydrothermal system associated with active volcanism, a CH4-rich sedimentary basin and a municipal waste landfill. For a comprehensive framework, published data from medium-to-high enthalpy hydrothermal systems were also included. The investigated systems were characterised by peculiar VOC suites that reflected the conditions of the genetic environments in whi

Evolutionarily divergent DUF4465 domains have a common vitamin B12-binding function.

The DUF4465 family (DUF, domain of unknown function) contains more than 1000 members distributed across eight bacterial clades with species from diverse microenvironments including various gut microbiomes, hydrothermal vents, and soil. In the gut commensal Bacteroides thetaiotaomicron (B. theta), DUF4465 containing proteins act as high-affinity B12-binding proteins that scavenge this cofactor to ensure bacterial survival. Such B12 capture is essential for bacteria that have lost the ability to synthesize B12 de novo. This raises the question of whether B12-binding is ubiquitous across this fam
