Investigation of dietary exogenous protease and humic substance on growth, disease resistance to Flavobacterium covae and immune responses in juvenile channel catfish (Ictalurus punctatus).

Many bacterial pathogens impact the US catfish industry, and disease control can be challenging for producers. Columnaris disease in channel catfish, Ictalurus punctatus, is primarily caused by Flavobacterium covae (formerly F. columnare). Immunostimulants may enhance nonspecific immune responses and offer an alternative to antibiotic treatments in catfish. Furthermore, dietary protein sources and inclusions are also essential to fish health and nutrition and may enhance overall fish performance in pond culture. The current project evaluated two immunostimulants: a protease complex (PC) and a

Pathologic Changes Associated With Respiratory Compromise And Morbidity Due To Massive Interlamellar Henneguya Exilis Infection In Channel × Blue Hybrid Catfish.

There are multiple Henneguya spp. (Myxozoa: Myxobolidae) endemic to North American catfish aquaculture that affect the gills of channel catfish and their hybrids. These parasites are morphologically similar, and confusion exists regarding the predilection sites and pathologic changes associated with different species. In the spring of 2018, channel (Ictalurus punctatus) female × blue (Ictalurus furcatus) male hybrid catfish from 2 separate commercial operations in northwest Mississippi were submitted for diagnostic assessment in response to observed morbidity and reduced feeding activity. Fish

Expression profiling analysis of immune-related genes in channel catfish (Ictalurus punctatus) skin mucus following Flavobacterium columnare challenge.

Fish are covered by a watery gel-mucus, mainly secreted by the goblet cells, serving as the physical and biochemical barrier between the external environment and the interior milieu, playing more important roles in fish that without scale. Despite the important roles of mucus in fish immunity, the knowledge of detailed molecular events happened during infection process is still limited. While most studies were focused on characterizing the protein and enzyme activities in the mucus following challenge, no studies have examined the gene expression profiles in fish mucus. In this regard, herein

Taste bud development in the channel catfish.

Taste bud formation in channel catfish is first seen to occur in stage 39 embryos, when taste bud primordia (stage 1), consisting of three to five cells, including a single calretinin-positive cell, can be recognized within the oropharyngeal cavity and maxillary barbels. Within a short time (stage 40), stage 2 taste bud primordia are apparent and include two or three calretinin-positive cells. The number of calretinin-positive cells continues to increase (stage 3), and the primordia begin to erupt as mature taste buds (stage 4) by embryonic stage 48. This same pattern of taste bud development

The riddle of hepatic neoplasia in brown bullheads from relatively unpolluted waters in New York State.

Since 1985, pathologists at Cornell University have investigated the causes of lesions in freshwater fish throughout New York waters in order to clarify possible impairment of fish health by environmental contaminants. Fishermen and biologists alerted us to several relatively protected reservoirs and ponds in which we have found no evidence of elevated levels of anthropogenic environmental contaminants but in which up to 100% of brown bullheads exhibited skin neoplasia. Complete necropsies and histologic study revealed that over 30% of mature brown bullheads from some of these sites had benign

Extraction and determination by gas chromatography of S,S,S-tri-n-butyl phosphorotrithioate (DEF) in fish and water.

A simple, low-cost, fast method for the extraction and cleanup of DEF (S,S,S-tri-n-butyl phosphorotrithioate) from fish tissues and water samples was developed. The method combines extraction and cleanup in one step. The basis of the method is passing water samples or aqueous tissue homogenates containing DEF through a C-18 disposable cartridge. DEF is eluted from the cartridge by acetone or ethyl acetate. The eluates are analyzed by gas chromatography using a thermionic-specific detector. The method detects levels as low as 100 parts per trillion (ppt) in water samples; recovery efficiency fr

The role of angiotensin II to support arterial blood pressure following serial haemorrhage in the freshwater channel catfish (Ictalurus punctatus).

Reductions in blood volume and changes to environmental salinity are known regulators of the renin-angiotensin system (RAS) in marine and euryhaline species of fish. However, the mechanisms activating the RAS in freshwater stenohaline species remain poorly understood. The purpose of this study was to determine if reduction in blood volume stimulates the activation of the RAS in channel catfish (Ictalurus punctatus). To achieve this goal, this study sought to determine if angiotensin II (ANG II) functions to support arterial blood pressure following a moderate haemorrhage. Channel catfish (n =

The capsid architecture of channel catfish virus, an evolutionarily distant herpesvirus, is largely conserved in the absence of discernible sequence homology with herpes simplex virus.

Although herpesviruses have a wide host range and their genomes vary substantially in size, the nucleocapsid appears to be a conservative element of viral design. The capsid shell is icosahedrally symmetric (T = 16), and 125 nm in diameter and 15nm thick in the case of herpes simplex virus 1 (HSV-1). Channel catfish virus (CCV) has the gross morphology of a herpesvirus, although no relationship to other herpesviruses is evident from the sequences of its proteins. To examine CCV capsid architecture more closely, we have determined its structure by cryoelectron microscopy and three-dimensional i

Biochemical studies of taste sensation. XIII. Enantiomeric specificity of alanine taste receptor sites in catfish, Ictalurus punctatus.

Specific binding of amino acid taste stimuli is known to occur to a sedimentable fraction (P2) from catfish (Ictalurus punctatus) taste epithelium or to purified plasma membranes from that fraction. L-Alanine, a potent taste stimulus for the catfish, binds in a reversible and saturable manner to these preparations. The extent to which the enantiomeric stimuli, L- and D-alanine, interact with the same or different receptor/transduction processes is investigated here both electrophysiologically and biochemically. With an electrophysiological assay, L-alanine was the more potent stimulus across a

Edwardsiella ictaluri type III and type VI secretion system mutant strains as candidates for live attenuated vaccines.

OBJECTIVE: Edwardsiella ictaluri, the causative agent of enteric septicemia of catfish, causes substantial economic losses to Channel Catfish Ictalurus punctatus producers in the United States. This study evaluates three E. ictaluri strains, each carrying a markerless deletion mutation in a type III or type VI secretion system gene, as candidates for a live attenuated vaccine against enteric septicemia of catfish in Channel Catfish. Replication in Channel Catfish cells, in vivo invasion and persistence, virulence, and the ability to provide single-dose protection against a wild-type E. ictalur

Molecular characterization, phylogenetic analysis and adjuvant effect of channel catfish interleukin-1βs against Streptococcus iniae.

Channel catfish is one of the most extensively cultured species worldwide, which is widely used as a classical model for comparative immunology. Interleukin-1β (IL1β) is an immunoregulatory cytokine with the potential to enhance the immune response induced by vaccines in many animals. To characterize the molecular characterization and identify the immunoadjuvant role of channel catfish IL1β, molecular cloning, phylogenetic analysis, and expression of two IL1β genes were performed, the bioactivity of their recombinant proteins (rIL1β1 and rIL1β2) were detected in vitro and their adjuvant effect

Synergies between vaccination and dietary arginine and glutamine supplementation improve the immune response of channel catfish against Edwardsiella ictaluri.

Channel catfish was used to investigate the enhancement of vaccine efficacy following dietary supplementation with arginine (ARG, 4% of diet), glutamine (GLN, 2% of diet), or a combination of both. After vaccination against Edwardsiella ictaluri, humoral and cellular immune responses, along with lymphoid organ responses were evaluated. E. ictaluri-specific antibody titers in plasma were higher (P < 0.05) in fish fed the supplemented diets compared to those fed the basal diet as early as 7 d post-vaccination (dpv). B-cell proportion in head-kidney was higher (P < 0.05) at 14 dpv in vaccinated f

Biomarker assessment of the effects of coal strip-mine contamination on channel catfish.

A suite of biomarkers was used to evaluate acute (1-day) to semichronic (3-month) heavy metal-induced toxicity in channel catfish, lctalurus punctatus, caged at an abandoned strip mine and a noncontaminated reference site. Assays performed include indicators of metabolic, hematological, osmoregulatory, and genotoxic stress. Two cage designs were used to evaluate the importance of exposure routes: one excluding contact with the sediments and the other allowing contact with water and sediments. Significant DNA strand breakage was observed in catfish exposed to both exposure regimens, but evidenc

Beneficial effects of Ginseng polysaccharide improve the growth performance, intestinal immunity, intestinal microbiota, and resistance to Yersinia ruckeri in channel catfish (Ictalurus punctatus).

This study evaluated the effects of ginseng polysaccharide (GP) supplementation on the growth performance, immunity, intestinal microbiota, and antibacterial activity of channel catfish (Ictalurus punctatus). Over a 56-day feeding trial, 750 catfish (60.03 ± 0.5 g) were fed diets containing GP at five concentrations: 0 mg/kg (CON), 200 mg/kg (GP200), 400 mg/kg (GP400), 800 mg/kg (GP800), and 1600 mg/kg (GP1600). GP supplementation at 200-800 mg/kg significantly enhanced growth performance, digestive enzyme activities (amylase, lipase, and trypsin), and reduced the feed conversion ratio (P < 0.

Plasma and tissue transferrin and ferritin, and gene expression of ferritin, transferrin, and transferrin receptors I and II in channel catfish Ictalurus punctatus fed diets with different concentrations of inorganic or organic iron.

Ferritin, transferrin, and transferrin receptors I and II play a vital role in iron metabolism, health, and indication of iron deficiency anaemia in fish. To evaluate the use of high-iron diets to prevent or reverse channel catfish (Ictalurus punctatus) anaemia of unknown causes, we investigated the expression of these iron-regulatory genes and proteins in channel catfish fed plant-based diets. Catfish fingerlings were fed five diets supplemented with 0 (basal), 125, and 250 mg/kg of either inorganic iron or organic iron for 2 weeks. Cadmium effects on Ichthyophthirius: evidence for metal-sequestration in fish tissues following administration of recombinant vaccines.

We are developing Tetrahymena thermophila as a delivery system for recombinant vaccines against parasitic protozoa, including the common fish parasite, Ichthyophthirius multifiliis. T. thermophila cell lines expressing I. multifiliis genes under the control of a cadmium-inducible metallothionein gene promoter conferred strong protection against a lethal parasite challenge when administered parenterally to naive fish. Nevertheless, given that heavy metals can be toxic to parasites, a question arose as to whether protection resulted from Cd residues carried over with the vaccine, rather than acq

Autochthonous probiotic Lactococcus lactis MA5 improves recovery from acute hypoxia stress and resistance to Edwardsiella ictaluri in hybrid catfish (Ictalurus punctatus × I. furcatus).

Dietary supplementation with probiotics is considered an effective strategy to enhance aquaculture production efficiency and disease resistance. Recent research has highlighted the value of using autochthonous probiotics in aquaculture. Herein, an autochthonous probiotic strain (Lactococcus lactis MA5) isolated from pond-raised hybrid catfish (Ictalurus punctatus × I. furcatus) in a previous study was investigated for its ability to improve host recovery from acute hypoxia and resistance to bacterial infection. Hybrid catfish were fed a control diet, or diets containing either 104 or 106CFU/g

Sterilization of Channel Catfish (Ictalurus punctatus) via Overexpression of bax Gene Regulated by a Tet-off System in the Primordial Germ Cells.

Transgenic technologies have been used for genetic improvement of catfish performance with notable success. However, these developments are useless from a commercialization standpoint without extremely efficient confinement. Transgenic sterilization has the potential to accomplish 100% reproductive confinement and avoid genetic exchange between transgenic or domestic genotypes and wild populations. The present study reports a novel sterilization method for channel catfish by overexpressing the pro-apoptosis gene bax, specifically in the primordial germ cells, to inhibit their proliferation. Th
