Novel Disk Diffusion Assay on Magnesium Oxalate Agar To Evaluate the Susceptibility of Yersinia pestis to Type III Secretion System Inhibitors.

Current methods for screening small molecules that inhibit the plasmid pCD1-encoded Yersinia pestis type III secretion system (T3SS) include lengthy growth curves followed by multistep luminescence assays or Western blot assays to detect secretion, or lack thereof, of effector proteins. The goal of this research was to develop a novel disk diffusion assay on magnesium oxalate (MOX) agar as a simple way to evaluate the susceptibility of Y. pestis to type III secretion system inhibitors. MOX agar produces distinct Y. pestis growth characteristics based on the bacteria's ability or inability to s

First report of Theileria orientalis Ikeda genotype associated with disease in cattle, Georgia, USA.

Theileria orientalis genotype Ikeda is a tickborne parasite that infects red and white blood cells and causes bovine infectious anemia. The primary tick vector for T. orientalis Ikeda is the longhorned tick (Haemaphysalis longicornis). This tick was first detected in the United States in 2017 and in the state of Georgia in 2021, however, to date, no clinical cases of theileriosis in cattle have been reported in the state of Georgia. A Highland calf was clinically diagnosed with anemia, marked lethargy, tachycardia, tachypnea, decreased rumen contraction rate, dry and flaking skin, and thin and

Ehrlichia canis in dogs experimentally infected, treated, and then immune suppressed during the acute or subclinical phases.

BACKGROUND: Concerns for recrudescence of Ehrlichia canis infection arise when immunosuppressive drugs are used to treat immune-mediated diseases in dogs previously infected with E. canis. OBJECTIVES: Determine whether administration of prednisolone and cyclosporine would reactivate E. canis infection in dogs previously treated with doxycycline during the acute or subclinical phases. ANIMALS: Seven beagles previously experimentally infected with E. canis and administered doxycycline for 4 weeks were included. Three of the 7 dogs were incidentally concurrently infected with Anaplasma platys a

Venous Air Embolism in a Horse After Jugular Venous Catheterization: Echocardiographic Diagnosis.

A Quarter Horse mare was presented with a history of imidocarb intoxication. During the initial evaluation, the patient presented an initial episode of syncope, in addition to a Grade III/IV diastolic murmur. On the basis of the observed condition, the animal underwent echocardiographic evaluation, in which artifacts were detected and subsequently characterized as air bubbles, restricted to the right cardiac chambers. A diagnosis of venous air embolism resulting from right jugular venocatheterization was made. This is the first reported case of venous air embolism secondary to jugular venocath

Theileria haneyi: An Emerging Equids Hemoparasite with Milder Virulence but Major Diagnostic and Therapeutic Implications.

Theileria haneyi, a recently discovered tick-borne hemoparasite infecting equids globally, has significant implications for equine health. Although it is closely related to T. equi (sharing 23% genomic divergence), it establishes an asymptomatic carrier state in persistently infected horses, creating a silent transmission reservoir. Its discovery and unique genetics justify its classification as a new taxon. A critical diagnostic challenge is that the lack of the ema-1 gene in T. haneyi prevents its detection by the standard T. equi cELISA, emphasizing the need for species-specific tools. Alth

Identification of the piroplasms isolated from horses with clinical piroplasmosis in Poland.

The study was aimed at determining the cause of the diseases in three horses exhibiting symptoms of fever, ataxia, mucus membrane paleness, haematuria and thrombocytopenia. The PCR technique revealed the presence in the blood of 18S RNA Babesia/Theileria spp. genetic material. DNA amplification using primers RLB F2 and RLB R2 produced 430 bp size products. The sequences of these PCR products demonstrated a 95.6-97.5% similarity with the sequence of the fragment of 18S RNA Babesia equi, gene number DQ287951 in the GenBank. The treatment utilizing the subcutaneous application of the imidocarb re

Clinical and serological observations on experimental infections with Babesia canis and its diagnosis using the IFAT.

Naive Beagle dogs (n = 5) were experimentally inoculated with two stocks of Babesia canis. Dogs were examined regularly for parasitaemia and for antibodies, using the indirect immunofluorescent antibody test (IFAT). This test proved to be useful for the diagnosis of babesiosis. After primo-infection dogs seem to develop a certain degree of immunity, although this immunity is neither absolute nor of long duration. Treatment of infected dogs with imidocarb (6 mg/kg) cleared the infection, but did not prevent the production of IFAT-antibodies. An infected dog treated with long acting oxytetracycl

Association between decreased blood pressure and azotaemia in canine babesiosis.

Acute tubular necrosis (ATN) was described in canine babesiosis. Hypotension is considered as one of the factors which influence the development of hypoxic renal damage. In this study hypotension defined as mean arterial pressure (MAP) < 80 mmHg was detected in 7 out of 48 dogs (14.6%) infected with Babesia canis. Lower systolic arterial pressure (SAP), diastolic arterial pressure (DAP) and MAP were detected in azotaemic dogs infected with B. canis. Statistically significant negative correlations between blood pressures (SAP, DAP and MAP) and serum creatinine and urea concentrations showed the

Canine babesiosis - a disease rarely considered in the context of male infertility.

BACKGROUND: Little is known about the impact of babesiosis on semen quality and fertility in dogs. CASE PRESENTATION: Four cases of infertility in male dogs after infection with Babesia canis are described. In all dogs sperm quality was low. Two dogs were castrated pharmacologically or surgically. In two dogs fertility was restored after supplementation with selenium and Vitamin E. As possible causes of spermatogenesis disorders due to the treatment of infection with Babesia canis with imidocarb, fever and disturbed testicular microcirculation are discussed. CONCLUSIONS: These cases indicate

A review of Babesiosis caused by Babesia ovis in small ruminants.

Babesiosis is a significant tick-borne disease that impacts both domestic and wild animals, as well as humans, with its prevalence being particularly high in tropical and subtropical areas. Among small ruminants, Babesia ovis is a leading cause of babesiosis, especially in areas where Rhipicephalus bursa ticks are prevalent. The transmission of the disease occurs through tick bites, imposing a considerable economic strain on livestock industries in regions where it is prevalent. Clinical manifestations of the disease include hemolytic anemia, fever, jaundice, lethargy, anorexia, weight loss, a

Chemotherapy of Babesia divergens in the gerbil, Meriones unguiculatus.

It was found that surprisingly low doses of four babesicides were effective against Babesia divergens in gerbils and it was concluded that this was due to the involvement of host resistance, which may be of a non-specific nature. The efficacy of the drugs relative to each other was the same in gerbils as in cattle and this host-parasite system is evidently more suitable for the screening of babesicides than are other rodent babesia systems. The prophylactic dose of imidocarb dipropionate required to provide a similar degree of protection in gerbils as in cattle was found to be much higher and

First case of clinical canine hepatozoonosis in Ukraine.

Canine hepatozoonosis is caused by the tick-borne protozoans Hepatozoon canis and Hepatozoon americanum (Apicomplexa, Hepatozoidae). While H. americanum is widespread in the south-central and south-eastern United States, H. canis is documented in Europe, Africa, Asia, and South America, and is transmitted to dogs by ingesting infected ticks, primarily Rhipicephalus sanguineus. In Europe, this vector is mainly distributed in the Mediterranean region but endemic areas within Central Europe have also been reported, possibly facilitated by climate change. A mixed-breed male puppy, ∼3-7 months-old,

Fatal acute babesiosis associated with Babesia venatorum infection (Babesia sp. EU1) in a captive reindeer calf in Switzerland.

Babesia venatorum was isolated from a captive reindeer calf in Switzerland. The clinical signs consistent with acute babesiosis included hemolytic anemia and hemoglobinuria. The diagnosis was made based on visualization of intraerythrocytic parasites in the stained blood smears and confirmed by PCR analysis of the 18S rRNA gene, with subsequent species identification within Babesia confirmed by sequencing. The reindeer calf was initially treated with supportive care and an antiprotozoal drug (imidocarb dipropionate) but died a few days after hospitalization. Babesia venatorum is also known as

Severe anemia caused by babesiosis in a maned wolf (Chrysocyon brachyurus).

An 8-yr-old, captive, spayed, female maned wolf (Chrysocyon brachyurus) developed progressive lethargy and weakness over a 24-hr period. Clinical signs included vomiting, recumbency, horizontal nystagmus, possible blindness, pale icteric mucus membranes, and port-wine colored urine. A complete blood cell count revealed severe anemia (packed cell volume [PCV], 6%) and intraerythrocytic piroplasms consistent with a Babesia species. Polymerase chain reaction testing later confirmed babesiosis. The wolf was treated with imidocarb dipropionate, antibiotics, and fluid therapy. A whole-blood transfus

Pharmacotherapeutics of drugs used in treatment of anaplasmosis and babesiosis.

Treatment of anaplasmosis and babesiosis in cattle is practical and effective when given early in the course of infection, before the onset of severe anemia or neurologic disorders. The tetracyclines (usually oxytetracycline or chlortetracycline) are the only effective specific compounds approved for use against anaplasmosis in the United States. Oxytetracycline, at the rate of 6.6 to 11 mg/kg of body weight given one to three times IM or IV, effectively moderates the course of infection. Chlortetracycline administered orally in dosages as small a 1.1 mg/kg will prevent infection and in dosage

Methods to improve the health of cattle in the tropics: immunisation and chemoprophylaxis against haemoparasitic infections.

A study of methods to improve the health of native cattle in tropical areas of Colombia showed an advantage using immunisation techniques against haemoparasitic infections in comparison with other control methods. The control of anaplasmosis and babesiosis by immunisation of cattle with fully virulent Anaplasma marginale, Babesia argentina and B. bigemina is feasible in tropical cattle when the postimmunisation reaction is controlled by appropriate drug therapy. Chemoprophylaxis was found less effective in controlling haemoparasitic diseases; however, treated was found less effective in contro
