Babesia rodhaini: passive protection of mice with immune serum.

Immune serum delayed the onset of parasitemia in both intact and splenectomized mice, but it neither prevented the development of Babesia rodhaini infection nor protected the mice from death even with further supplementation of immune serum during the infection. The protective antibodies in the serum are more effective in their action on free B. rodhaini parasites than on infected erythrocytes; the parasites (free or inside the red cells) being direct targets for the antibodies. Passive administration of the immune serum seemed to inhibit the active development of the host's own immune respons

Passive immunity against pseudomonas sepsis during granulocytopenia.

Specific passive immunity against Pseudomonas aeruginosa sepsis was assessed in granulocytopenic dogs. Dogs were infused with either normal or antipseudomonas immune plasma 24 h before pseudomonas challenge. They were challenged intravenously with 10(7) serotype 6 P. aeruginosa during granulocytopenia. Treatment was evaluated by observation of survival periods, febrile responses, type 6 pseudomonas antibody titers, and quantitative cultures of blood and tissues. The results demonstrated that passively immunized dogs did not survive infection. Both normal-plasma and immune-plasma recipients had

Passive immunization through polyclonal antibodies against IsdE showed protective activity in an experimental S.aureus challenge model.

Staphylococcus aureus, particularly methicillin-resistant strains (MRSA), continues to pose a major therapeutic challenge and highlights the need for alternative or supportive treatment strategies. In this study, the recombinant iron-regulated surface determinant E (IsdE) protein was expressed, purified, and used to generate rabbit polyclonal IgG antibodies.Antibody specificity was confirmed by ELISA and Western blotting, and functional activity was assessed using an opsonophagocytic assay. To evaluate in vivo efficacy, BALB/c mice were challenged with a 5 × 10⁸ CFU dose of MRSA and treated wit

Mucosal passive immunization with monoclonal antibodies targeting Candidalysin and Hyr proteins attenuates vaginal candidiasis in mice.

Vulvovaginal candidiasis (VVC) is a prevalent fungal infection primarily caused by the opportunistic pathogen Candida albicans. Among women affected by VVC, up to 8% experience more than four episodes per year, regarded as recurrent vulvovaginal candidiasis (RVVC). Current treatments for VVC are effective for isolated episodes but are insufficient for preventing recurrences, highlighting the need to develop novel treatments for the management of RVVC. In this study, we explore passive immunization as a potential alternative strategy to prevent VVC. The Candidalysin toxin (Cdlys) and the pariet

Passive Immunization with a Single Monoclonal Neutralizing Antibody Protects against Cutaneous and Mucosal Mouse Papillomavirus Infections.

We have established a mouse papillomavirus (MmuPV1) model that induces both cutaneous and mucosal infections and cancers. In the current study, we use this model to test our hypothesis that passive immunization using a single neutralizing monoclonal antibody can protect both cutaneous and mucosal sites at different time points after viral inoculation. We conducted a series of experiments involving the administration of either a neutralizing monoclonal antibody, MPV.A4, or control monoclonal antibodies to both outbred and inbred athymic mice. Three clinically relevant mucosal sites (lower genit

A subcutaneous cellular implant for passive immunization against amyloid-β reduces brain amyloid and tau pathologies.

Passive immunization against misfolded toxic proteins is a promising approach to treat neurodegenerative disorders. For effective immunotherapy against Alzheimer's disease, recent clinical data indicate that monoclonal antibodies directed against the amyloid-β peptide should be administered before the onset of symptoms associated with irreversible brain damage. It is therefore critical to develop technologies for continuous antibody delivery applicable to disease prevention. Here, we addressed this question using a bioactive cellular implant to deliver recombinant anti-amyloid-β antibodies in

Passive immunization with melanin-binding monoclonal antibodies prolongs survival of mice with lethal Cryptococcus neoformans infection.

Passive immunization with monoclonal antibodies (MAbs) to melanin prolonged the survival of and reduced the fungal burden in Cryptococcus neoformans-infected mice in comparison to controls. MAbs to melanin reduced the growth rate of in vitro-melanized C. neoformans cells, suggesting a new mechanism of antibody-mediated protection.

Passive immunization with monoclonal antibodies against herpes simplex virus glycoproteins protects mice against herpetic ocular disease.

The effects of passive immunization with specific monoclonal antibodies against herpes simplex virus glycoproteins gB, gC, gD, and gE on the course of herpetic keratitis, survival and the establishment of latency in an outbred mouse model are described. A total of nine monoclonal antibodies were tested in these experiments. Passive immunization at 24 or 48 hours post-inoculation had little effect on the severity of the initial epithelial infection of the cornea, but blocked dissemination of the virus to the central nervous system and periocular tissues and prevented development of blepharitis,

[Immune complex formation during the passive immunization of mice infected with the tick-borne encephalitis virus].

The results of investigations, indicating that the development of the infectious process in experimental encephalitis is accompanied by the formation of immune complexes circulating in the blood and localized in the brain tissue, are presented. The intravenous injections of a homologous serum preparation into intact animals induces the appearance of a rather low level of circulating immune complexes, which precedes the elimination of antibodies. The intravenous injection of a specific serum preparation two days after the infection of the animals with tick-borne encephalitis virus is accompanie

[Therapy with high-dose intravenous gamma globulin in the newborn infant with thrombocytopenia from passive immunization].

High doses of gammaglobulin intravenously have been found useful in controlling thrombocytopenia of a neonate born to a mother who developed acute idiopathic thrombocytopenic purpura during pregnancy. Administration of 0.4 gm/kg/day of intact gammaglobulin intravenously for five days was followed by dramatic rise of platelet count without any side-effect. After 1 month the platelet count was still normal. Passively-acquired immune thrombocytopenia of the newborn consequently is another indication for intravenous gammaglobulin therapy.

[Dynamics of the antismallpox antibodies detectable in the passive hemagglutination reaction with various animal immunization schemes].

Dynamics of accumulation and preservation of antibodies detectable in the PHAT (PHAT-AT) was studied on rabbits and guinea pigs with the use of various doses of the living inactivated virus and their combination in comparison with the virus-neutralizing antibodies, antihemagglutinins and precipitins. Accumulation of the virus-neutralizing antibodies did not coincide in time with the curve of the PHAT-AT accumulation; the titres of the virus-neutralizing antibodies were higher than the PHAT-AT titres. At the same time the percentage of seroconversions determined by PHAT was equal to 100 and the

Active immunization with Brucella abortus S19 phage lysate elicits serum IgG that protects guinea pigs against virulent B. abortus and protects mice by passive immunization.

Brucellosis is an economically important zoonosis of worldwide significance. Earlier (Jain et al., 2015) we reported methodology for generation of phage lysate preparations against Brucella abortus S19 using brucellaphage 'ϕLd'. In this study, using a fixed dose (Two mouse PD100) of lysates, the prophylactic efficacies of both plain and alum gel adjuvanted lysates were evaluated in guinea pig by direct virulent challenge and passive mouse protection test (PMPT). Strong humoral and cell mediated immune responses in guinea pigs and protection comparable to S19 vaccine was observed with low dose

Maternal Immunization Using a Protein Subunit Vaccine Mediates Passive Immunity against Zaire ebolavirus in a Murine Model.

The Ebola virus has caused outbreaks in Central and West Africa, with high rates of morbidity and mortality. Clinical trials of recombinant virally vectored vaccines did not explicitly include pregnant or nursing women, resulting in a gap in knowledge of vaccine-elicited maternal antibody and its potential transfer. The role of maternal antibody in Ebola virus disease and vaccination remains understudied. Here, we demonstrate that a protein subunit vaccine can elicit robust humoral responses in pregnant mice, which are transferred to pups in breastmilk. These findings indicate that an intramus

Immunization with Streptococcus suis bacterin plus recombinant Sao protein in sows conveys passive immunity to their piglets.

BACKGROUND: Streptococcus suis (S. suis) causes arthritis, meningitis, septicemia, and sudden death in pigs and is also an zoonotic agent for humans. The present study demonstrated that immunization with recombinant Sao-L (surface antigen one-L, rSao-L) protein from a strain of S. suis serotype 2 in pigs was able to increase cross-serotype protection against S. suis serotype 1 and 2 challenge. Since weaning piglets are more susceptible to S. suis infections due to the stresses associated with weaning, prepartum immunization in sows may convey passive immunity to piglets and provide protection.

Passive immunization with anti-ActA and anti-listeriolysin O antibodies protects against Listeria monocytogenes infection in mice.

Listeria monocytogenes is an intracellular pathogen that causes listeriosis. Due to its intracellular niche, L. monocytogenes has evolved to limit immune recognition and response to infection. Antibodies that are slightly induced by listerial infection are completely unable to protect re-infection of L. monocytogenes. Thus, a role of antibody on the protective effect against L. monocytogenes infection has been neglected for a long time. In the present study, we reported that passive immunization with an excessive amount of antibodies against ActA and listeriolysin O (LLO) attenuates severity o

Maternal antibodies by passive immunization with formalin inactivated respiratory syncytial virus confer protection without vaccine-enhanced disease.

Maternal immunization of mice with formalin inactivated respiratory syncytial virus (FI-RSV) resulted in the passive transfer of RSV antibodies but not cellular components to the offspring. The offspring born to FI-RSV immunized mothers showed serum RSV neutralizing activity, effectively controlled lung viral loads without vaccine-enhanced disease, did not induce pulmonary eosinophilia, and cytokine producing cells after live RSV infection. Therefore, this study provides evidence that maternal immunization provides an in vivo model in investigating the roles of antibodies independent of cellul

Passive immunization with a monoclonal IgM antibody specific for Strongyloides ratti HSP60 protects mice against challenge infection.

It is estimated that 30-100 million people are infected with the pathogenic nematode Strongyloides stercoralis worldwide but parasite control is still based on anti-helminthic treatment. To develop protective vaccination strategies, we use the murine model of Strongyloides ratti infection. We have shown recently that vaccination with alum-precipitated, but not with native or CFA-emulsified S. ratti heat shock protein 60 (srHSP60) conferred protection to challenge infection. Here we describe the generation of a monoclonal IgM specific for srHSP60. Anti-srHSP60 detected human and srHSP60 and sta
