Maximizing TLR9 Activation in Cancer Immunotherapy with Dual-Adjuvanted Spherical Nucleic Acids.

Nucleic-acid-based immune adjuvants have been extensively investigated for the design of cancer vaccines. However, nucleic acids often require the assistance of a carrier system to improve cellular uptake. Yet, such systems are prone to carrier-associated adaptive immunity, leading to difficulties in a multidose treatment regimen. Here, we demonstrate that a spherical nucleic acid (SNA)-based self-adjuvanting system consisting of phosphodiester oligonucleotides and vitamin E can function as a potent anticancer vaccine without a carrier. The two functional modules work synergistically, serving

Interleukin 8 activity influences the efficacy of adenoviral oncolytic immunotherapy in cancer patients.

After the landmark approval of T-VEC, oncolytic viruses are finding their way to the clinics. However, response rates have still room for improvement, and unfortunately there are currently no available markers to predict responses for oncolytic immunotherapy. Interleukin 8 (IL-8) production is upregulated in many cancers and it also connects to several pathways that have been shown to impair the efficacy of adenoviral immunotherapy. We studied the role of IL-8 in 103 cancer patients treated with oncolytic adenoviruses. We found high baseline serum IL-8 concentration to be independently associa

Activated cytotoxic T-lymphocyte immunotherapy is effective for advanced oral and maxillofacial cancers.

Conventional cancer treatments are surgery, radiotherapy, and chemotherapy, but treatment efficiency is insufficient and cancer recurrence is common. Immunotherapy has been added as an important cancer treatment component, but no reports on its efficacy in oral and maxillofacial cancers exist. We evaluated the clinical efficacy of adoptive immunotherapy using ex vivo-activated cytotoxic T lymphocytes (CTL) in the treatment of 7 patients with advanced oral and maxillofacial cancers with stage IV disease at diagnosis. The mean follow-up period was 26.2 months. Phenotype of the lymphocyte assay r

Tumor-Activated Carrier-Free Dual-Prodrug Nanoassemblies for Synergistic Chemo-Immunotherapy.

Chemo-immunotherapy is limited by the poor bioavailability, nonspecific distribution, and systemic toxicity of free drugs. Here, we develop a reactive oxygen species (ROS)-responsive prodrug strategy by conjugating paclitaxel (PTX) and the TLR7/8 agonist resiquimod (R848) with a phenylboronic ester (PBE) moiety to generate two ROS-activatable prodrugs, PTX-PBE and R848-PBE. These prodrugs spontaneously coassemble into carrier-free nanoparticles (PR NPs) with ultrahigh loading efficiency and tunable drug ratios. PBE modification improves systemic stability while enabling ROS-triggered drug rele

TLR9-activating cholesterol azetidine derivative-assisted therapeutic vaccines for cancer immunotherapy.

Therapeutic cancer vaccines show great promise for de novo induction of antigen-specific T cell responses against tumors. However, weak coordination between innate immune activation and antigen delivery remains a major obstacle to vaccine efficacy. Here, we present a cholesterol azetidine derivative-assisted polymeric carrier, Aze-Chol NP, which stimulates innate immunity and primes tumor-specific CD8+ T cell responses. Mechanistically, Aze-Chol NP induces maturation and activation of dendritic cells (DCs) through the Toll-like receptor 9 (TLR9) pathway while simultaneously delivering protein

Lower airway microbiome transcriptional and metabolic activities influence responsiveness to immunotherapy in NSCLC.

Atrial fibrillation (AF) is an abnormal heart rhythm related to an increased risk of heart failure, dementia, and stroke. The distinction between valvular and non-valvular AF remains a debate. In this study, proteomics and metabolomics were integrated to describe the dysregulated metabolites and proteins of AF patients relative to sinus rhythm (SR) patients. Totally 47 up-regulated and 41 down-regulated proteins in valvular AF, and 59 up-regulated and 149 down-regulated proteins in non-valvular AF were recognized in comparison to SR patients. Moreover, 58 up-regulated and 49 significantly down

Assembled DNA Nanostructure to Precisely Induced cGAS-STING Activation for Cancer Immunotherapy.

Triple-negative breast cancer (TNBC) has emerged as a major challenge in cancer therapy due to its aggressive nature and lack of effective targeted treatments. Activating the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway to stimulate innate immunity is considered a promising strategy for TNBC treatment. However, current STING agonists are limited by several severe drawbacks that hinder their further application. Herein, a cross-shaped DNA skeleton was rationally engineered by leveraging programmable DNA assembly and AS1411 aptamer-mediated tumor targeting to achie

Lipid metabolism in γδ T-cell activation: Implications for immunotherapy in ovarian cancer.

Ovarian cancer (OC) remains a leading cause of cancer-related mortality among women worldwide, largely due to asymptomatic progression, late-stage diagnosis, therapeutic resistance, and profound immunosuppression within the tumor microenvironment (TME). This is particularly relevant in advanced disease, where metastatic spread to the omentum creates a lipid-rich niche that promotes tumor growth and weakens anti-tumor immunity. This review examines lipid metabolic reprogramming and γδ T-cell function as a targetable immunometabolic axis in OC, with selected discussion of iNKT and CD8+ T-cells a

STING-activating layered double hydroxide nano-adjuvants for enhanced cancer immunotherapy.

Cancer vaccines represent a promising therapeutic strategy in oncology, yet their effectiveness is often hampered by suboptimal antigen targeting, insufficient induction of cellular immunity, and the immunosuppressive tumor microenvironment. Advanced delivery systems and potent adjuvants are needed to address these challenges, though a restricted range of adjuvants for human vaccines that are approved, and even fewer are capable of stimulating robust cellular immune response. In this work, we engineered a unique self-adjuvanted platform (MLDHs) by integrating STING agonists manganese into a la

A Bimodal MRI-Traceable Nanozyme with Immune-Activated Capability for Immunotherapy of Lung Metastases.

The therapeutic efficacy of cancer is greatly hampered by hypoxia, overexpressed glutathione (GSH) in the tumor microenvironment (TME), and the high propensity for tumor metastasis. Developing advanced diagnostic and therapeutic strategies capable of destroying tumors holds great promise. Herein, multifunctional manganese-based nanoparticles (FMBI NPs) with multiple nanozyme activities and GSH-depleting capabilities were designed to enhance cancer therapy by remodeling the TME. The release of Mn ions from FMBI NPs under TME conditions can modulate the tumor stroma, activate the cyclic GMP-AMP

Delivery of Macrophage Activating Nanoparticles and Checkpoint Inhibitors Improves Melanoma Immunotherapy in Mice.

The immune system clears tumors from forming in the body by first activating the innate immune system, which subsequently triggers the adaptive immune system to mount robust responses against cancer cells. This strategy involves initially activating phagocytic immune cells, such as dendritic cells and macrophages, followed by the reinvigoration of adaptive immune cells, like T cells, to develop effective immune responses against tumors. Herein, the one-two punch strategy is utilized of first activating the innate immune system with biomaterial-based adjuvants composed of succinate polymers and

A dual PROTAC nanocarrier amplifies DNA damage and STING activation for cancer immunotherapy.

The cytoplasmic accumulation of damaged DNA to activate the STING pathway has emerged as a promising strategy to enhance tumor immunogenicity and improve the efficacy of immune checkpoint blockade (ICB) therapy. Herein, a stimuli-responsive and dual PROTAC-embedded immunoactivator (denoted as Sd@Lip) is developed to potentiate ICB through amplified DNA damage and robust STING activation. Sd@Lip comprises an acid-sensitive liposomal nanocarrier co-encapsulating two targeted degraders of dBET1 (a BRD4 degrader) and SK-575 (a PARP1 degrader), which enhances drug solubility, stability, and enable

Fabrication of RIG-I-Activating Nanoparticles for Intratumoral Immunotherapy via Flash Nanoprecipitation.

Intratumoral immunotherapy is a promising strategy for stimulating local and systemic antitumor immunity while eliminating or reducing immune-related adverse events often attendant to systemic administration. Activation of the cytosolic pattern recognition receptor retinoic acid-inducible gene I (RIG-I) at tumor sites stimulates innate immunity that can potentiate a T cell-dependent adaptive antitumor immune response. However, the activity and efficacy of 5'-triphosphate RNA (3pRNA) agonists of RIG-I are hindered by poor in vivo stability, rapid degradation, limited cellular uptake, and ineffi

Tumor microenvironment-activated Zn//MnO2 battery for sustained and local electrochemical immunotherapy.

Metal ions are widely adopted to potentiate oxidative damage-induced immunogenic cell death (ICD) and regulate immune pathways. However, current ion delivery platforms lack precise administration over distribution and retention time within the tumor microenvironment, leading to high systemic toxicity and suboptimal therapeutic efficacy. Here, local electrochemical immunotherapy is developed by implanting a tumor microenvironment-activated Zn//MnO2 battery, which uses body fluid as an electrolyte and endogenous glutathione as a redox mediator to promote sustained generation and localized retent

Ultrasound-activated nanovesicles for adenosine exhaustion and immune checkpoint blockade in cancer immunotherapy.

Conventional antitumor therapies induce immunogenic cell death (ICD), releasing large amounts of ATP that are rapidly converted into immunosuppressive adenosine within the tumor microenvironment (TME). This accumulation of adenosine promotes tumor immune evasion by inhibiting effector immune cells and upregulating inhibitory immune checkpoints. To overcome this challenge, we designed ultrasound (US)-activated nanovesicle system ADA/Ce6@tLipo which is composed of T cell membranes displaying multiple immune checkpoint molecules and a liposome encapsulating chlorin e6 (Ce6) and adenosine deaminas

Breaking the cycle: immune complexes, complement activation, and novel immunotherapies in lupus nephritis.

Lupus nephritis (LN), a severe manifestation of systemic lupus erythematosus (SLE), is driven by immune complex deposition and complement activation, resulting in glomerular inflammation and podocyte injury. Beyond being passive targets, podocytes actively modulate renal immunity through cytokine secretion and antigen presentation. Recent advances in urinary biomarkers such as NGAL, TWEAK, and MCP-1 and composite indices like the Renal Activity Index for Lupus (RAIL) offer dynamic and noninvasive monitoring of disease activity. Immunotherapy has transitioned from nonspecific immunosuppression

STING-activating nanoparticles normalize the vascular-immune interface to potentiate cancer immunotherapy.

The tumor-associated vasculature imposes major structural and biochemical barriers to the infiltration of effector T cells and effective tumor control. Correlations between stimulator of interferon genes (STING) pathway activation and spontaneous T cell infiltration in human cancers led us to evaluate the effect of STING-activating nanoparticles (STANs), which are a polymersome-based platform for the delivery of a cyclic dinucleotide STING agonist, on the tumor vasculature and attendant effects on T cell infiltration and antitumor function. In multiple mouse tumor models, intravenous administr

Augmented Passive Immunotherapy with P4 Peptide Improves Phagocyte Activity in Severe Sepsis.

INTRODUCTION: Antimicrobial resistance threatens to undermine treatment of severe infection; new therapeutic strategies are urgently needed. Preclinical work shows that augmented passive immunotherapy with P4 peptide increases phagocytic activity and shows promise as a novel therapeutic strategy. Our aim was to determine ex vivo P4 activity in a target population of patients admitted to critical care with severe infection. METHODS: We prospectively recruited UK critical care unit patients with severe sepsis and observed clinical course (≥3 months postdischarge). Blood samples were taken in ea

DAP12-based activating chimeric antigen receptor for NK cell tumor immunotherapy.

NK cells are emerging as new effectors for immunotherapy of cancer. In particular, the genetic engraftment of chimeric Ag receptors (CARs) in NK cells is a promising strategy to redirect NK cells to otherwise NK cell-resistant tumor cells. On the basis of DNAX-activation protein 12 (DAP12), a signaling adaptor molecule involved in signal transduction of activating NK cell receptors, we generated a new type of CAR targeting the prostate stem cell Ag (PSCA). We demonstrate in this article that this CAR, designated anti-PSCA-DAP12, consisting of DAP12 fused to the anti-PSCA single-chain Ab fragme
