A two-stage study was carried out to test the mechanism of arginase in ammonia detoxification of yellow catfish. At stage 1, fish was injected lethal half concentration ammonium acetate and 0.9% sodium chloride respectively every 12 h in six replicates for 72 h. The result found that no significant different in serum ammonia contents of fish in ammonium acetate group at hours 12, 24, 36, 48, 60 and 72. At stage 2, ammonium acetate group was split in two, one continued to injected with ammonium acetate (NH3 group) and the other with ammonium acetate and valine (an inhibitor of arginase; Val gro

Siegesbeckia orientalis L. is a promising species for cadmium (Cd) phytoextraction with large biomass and fast growth rate, while little information about their intracellular mechanisms involved in Cd tolerance and detoxification has been explored. A soil pot experiment with total target Cd concentrations of 0, 10, 50, 100, and 150 mg kg-1 were designed to investigate the subcellular distribution, chemical forms and thiol synthesis characteristics of Cd in S. orientalis. More than 90% of Cd was bound to the soluble fractions (48.4-76.5%) and cell walls (19.9-46.3%). Increasing soil Cd concentr

Detoxification (detox) plays a major role in pesticide action and resistance. The mechanisms involved are sometimes part of the discovery and development process in seeking new biochemical targets and metabolic pathways. Genetically modified and chemical-safener-modified crops are a marked exception and often involve herbicide detox by design to achieve the required crop tolerance. This perspective evaluates the role of detox by design or chance and target-site-based selectivity in insecticide, herbicide, and fungicide action and human health and environmental effects.

The urinary excretion of thiocyanate by hens after dosage with cyanide (30 mumol) has been studied in a series of acute experiments involving 6 hr urine collection periods. More than half of the dose could be recovered as thiocyanate when cyanide was given by intravenous infusion and the rate of excretion closely paralleled plasma thiocyanate concentration. Little cyanide was excreted directly. The excretion of thiosulphate fell by an amount that suggested that availability of sulphane sulphur might limit the extent of conversion. However, neither thiosulphate nor sulphur amino acids enhanced

Eight volunteers were exposed to trichloroethylene vapour (1,042 μg./l.) for five hours; 51 to 64% of the inhaled trichloroethylene was retained. The concentration of trichloroethanol and trichloroacetic acid in the urine was studied daily for a three-week period; on the third day both metabolites were determined in faeces, sweat, and saliva. The concentration of trichloroacetic acid in plasma and red blood cells was studied on alternate days. Of the trichloroethylene retained, 38·0 to 49·7% was excreted in the urine as trichloroethanol and 27·4 to 35·7% as trichloroacetic acid. Of both metabo

Pleural mesothelioma (PM) is one of the deadliest cancers, with limited therapeutic options due to its therapeutically intractable genome, which is characterized by the functional inactivation of tumor suppressor genes (TSGs) and high tumor heterogeneity, including diverse metabolic adaptations. However, the molecular mechanisms underlying these metabolic alterations remain poorly understood, particularly how TSG inactivation rewires tumor metabolism to drive tumorigenesis and create metabolic dependencies. Through integrated multi-omics analysis, we identify for the first time that NF2 loss o

Ornithine decarboxylase (ODC) can be induced up to 100-fold over basal levels four hours after addition of glutamine to the medium of HeLa cells growing in suspension culture. As demonstrated in several other cell types, ODC is inactivated very rapidly in HeLa cells, and the rate of inactivation is seen to vary with a half life of 9-15 minutes in uninduced cells and rises to ca. 60 minutes at the peak of induction. Quantitatively, the change in rate of inactivation cannot completely account for the observed rise in activity, thus synthesis or activation of ODC must also be involved in the indu

Rahnella species are environmentally versatile bacteria widely distributed in terrestrial and aquatic ecosystems; however, the genomic basis of formaldehyde detoxification and stress adaptation in cryoconite-associated Rahnella remains poorly understood. In this study, we performed phylogenomic, comparative genomic, pan-genomic, stress-response, and functional analyses of Rahnella sp. PAMC25617 isolated from polar cryoconite to investigate glutathione (GSH)-dependent formaldehyde detoxification and its evolutionary conservation. The insect larval fat body is the primary organ for detoxification of environmental pesticides and metabolic waste. During lepidopteran metamorphosis, the larval fat bodies undergo degradation and reprogramming for development, leaving the process of detoxification largely unknown at this stage. Using integrative approaches in the silkworm, Bombyx mori, we demonstrate that the pupal rectal sac, a greatly enlarged hindgut structure, serves as a central detoxification hub during the non-feeding metamorphic period. We show that it actively sequesters metabolic waste (uric acid) and, remarkably, c

Polyhalogenated carbazoles (PHCZs) are widely recognized for their persistence and widespread presence in aquatic environments. Although PHCZ accumulation in aquatic organisms induces various forms of biological toxicity, the effects on liver metabolism are not yet fully understood. This study investigated the biological concentration factor (BCF), liver histopathology, metabolomics, and responses of key metabolic genes in the liver tissues of male zebrafish exposed to 20 and 100 μg/L of 3,6-dichlorocarbazole (3,6-DCCZ) for 21 days. The bioconcentration factor of 3,6-DCCZ was found to reach 10

Diverse forms of mercury (Hg) have various effects on animals and humans because of a variety of routes of administration. Inorganic mercury (iHg) binds to thiol groups of proteins and enzymes in one's body or is methylated by microorganisms. Organic form of Hg, contrary to the iHg, is more stable but may be demethylated to Hg2+ in the tissue of intestinal flora. Selenium (Se) also occurs in a variety of chemical forms in one's body but both of these elements behave very differently from one another. Mercury binding to selenide or Se-containing ligands is a primary molecular mechanism that red

The ability to detoxify defensive compounds of competitors provides key ecological advantages that can influence community-level processes. Although common in plants and bacteria, this type of detoxification interaction is extremely rare in animals. Here, using laboratory behavioral assays and analyses of videotaped interactions in South America, we report widespread venom detoxification among ants in the subfamily Formicinae. Across both data sets, nine formicine species, representing all major clades, used a stereotyped grooming behavior to self-apply formic acid (acidopore grooming) in resp

Secondary metabolic products of molds, called mycotoxins, negatively affect animal health and production. They constitute a significant problem in veterinary and medical sciences, and their presence has been confirmed in feed all over the world. Applying appropriate agricultural practices and ensuring proper storage conditions significantly reduces the contamination of agricultural products with mycotoxins. However, this does not guarantee that raw materials are completely free from contamination. Many detoxification methods are currently used, but their insufficient effectiveness and negative

Aflatoxin B1 (AFB1) is one of the most harmful mycotoxins, raising serious global health and economic problems. Searching for biological approaches for effective and safe AFB1 degradation is imminent. In our study, Microbacterium proteolyticum B204 isolated from bovine faeces degraded 77% of AFB1 after 24 h, becoming the first reported bacteria from the Microbacterium family to possess AFB1 degradation characteristics. Temperature variation showed little effect on its degradation ratio, demonstrating high thermostability of 75% and 79% after boiling and sterilization, respectively. We suppose

Fluoride toxicity to microorganisms was a predominant factor contributing to the failure of a commercial scale bioleach heap. An integrated control strategy for fluoride complexation without jarosite generation by stepwise adding ferric ions was first proposed to enable the bioleaching of the chalcopyrite associated with fluoride-bearing gangue mineral by Acidithiobacillus ferrooxidans. Chemical speciation calculation revealed that with the presence of Fe3+, the concentration of the main lethal fluoride to microorganism, HF, decreased dramatically. The pure culture study showed that the detrim

Mercury (Hg) is a global pollutant that poses a serious threat to human and the environment. Rice was found as an important source for human exposure to Hg in some areas. In this study, the transportation and transformation of IHg and MeHg in rice plants exposed to IHg or MeHg were investigated. The IHg and MeHg concentrations in rice roots and shoots collected every five days were analyzed by HPLC-ICP-MS and SR-XANES. When exposed to MeHg, the percent of IHg in rice roots and shoots increased while MeHg decreased significantly, suggesting prominent demethylation of MeHg occurred. However no n

Glyoxals are reactive alpha-oxoaldehydes that are formed endogenously from sugars, the levels of which are increased in various pathological conditions associated with hyperglycaemia and thiamine deficiency. However, the molecular cytotoxic mechanisms of glyoxal are not known. Results presented here and in the other studies cited provide a glimpse into the cytotoxicity mechanisms involved and their pathological implications. We found that glyoxal (10 microM) markedly increased the susceptibility of hepatocyte glutathione (GSH) to oxidation by hydrogen peroxide (H(2)O(2)) and markedly increased

Kinetic studies of the serum albumin-catalyzed reaction of cyanide with colloidal elemental sulfur have been carried out to elucidate the formal mechanism of this reaction. The results indicate that one of the several sulfur-binding sites on the protein molecule is involved in a single displacement catalytic cycle in the presence of cyanide. At high velocities the rate of thiocyanate production is limited by an albumin-independent second order process which produces the form of sulfur that serves as substrate. The catalyzed reaction is also subject to substrate inhibition at high colloidal sul

A case of cyanide poisoning from laetrile ingestion is presented as an illustration of the recognition and treatment of cyanide intoxication. The pharmacology of laetrile, of cyanide, and of antidotes to cyanide intoxication are discussed as they relate to the acute management and successful treatment of this patient after this highly lethal ingestion.
