SPME-based air sampling method for inhalation exposure assessment studies: case study on perchlorethylene exposure in dry cleaning.

Exposure to perchlorethylene, especially for dry cleaning workers and for people living near dry cleaning shops, could lead to several diseases and disorders. This study examines the value of solid-phase microextraction (SPME) for sampling perchlorethylene in the atmosphere of dry cleaning shops. Carboxen/polydimethylsiloxane (CAR/PDMS) in 0.5-cm retracted mode was selected. There were no significant differences between sampling rates at different temperatures (range of 20 to 30 °C) and air velocities (2 to 50 cm/s). On the opposite, relative humidity (RH) had a significant effect on sampling

Inhalation exposure to mycotoxins in occupational settings: an underrecognized and unregulated health hazard.

Mycotoxins represent secondary metabolic products generated by fungal genera, including Aspergillus, Penicillium, and Fusarium, and are known to trigger hepatotoxic, nephrotoxic, carcinogenic, and immunotoxic effects in humans. While well-defined regulatory thresholds exist to manage dietary mycotoxin intake for major mycotoxins such as aflatoxins, ochratoxins, fumonisins, deoxynivalenol, and zearalenone, the health consequences stemming from inhalation among the agricultural and food-processing workforce are still poorly understood. Importantly, there are no health-based occupational exposure

Inhalation Exposure to 2,4-Dichlorophenoxyacetic Acid Causes Tracheal Damage-A Study in Rats.

Exposure to 2,4-dichlorophenoxyacetic acid (2,4-D) occurs mainly by inhalation. Studies indicate that respiratory allergic reactions are induced by this pesticide. This study analyzed the effects of chronic inhalation exposure to 2,4-D in the trachea of rats. We exposed the animals during six months to three different concentrations of 2,4-D used for crop spraying. Animals exposed to low concentrations had an increase in the mast cells count, showing that this herbicide can cause allergic reactions in the airways. The tracheal epithelium thickness, the nuclear area and number of nucleolar orga

Oxidative stress mediates cardiac electrophysiological injury in inhalation exposure to flavored vaping products.

BACKGROUND: Electronic nicotine delivery systems (ENDS) heat "E-liquids" to generate "E-vapor", an aerosolized mixture containing nicotine and flavors. Flavored ENDS are popular among teens who vape, however, the possible cardiac electrophysiological harm of inhalation exposure to flavored ENDS are not fully understood. OBJECTIVE: To test if inhalation exposure to flavoring carbonyls in E-liquids compromises mitochondrial integrity, increases oxidative stress, and leads to cardiac electrophysiological toxicity. METHODS: Gas chromatography mass spectrometry (GC/MS), flow cytometry, oxygen con

Cardiovascular dysfunction induced by combined exposure to nicotine inhalation and high-fat diet.

Smoking and high-fat diet (HFD) consumption are two modifiable risk factors for cardiovascular (CV) diseases, and individuals who are overweight or obese due to unhealthy diet are more likely to use tobacco products. In this study, we aim to investigate the combined effects of nicotine (the addictive component of all tobacco products) and HFD on CV health, which are poorly understood. C57BL/6N male mice were placed on either HFD (60 kcal% fat) or regular diet (22 kcal% fat) and exposed to air or nicotine vapor for 10-12 wk. CV function was monitored by echocardiography and radiotelemetry, with

The risk factors and threshold level of subchronic inhalation exposure of reclaimed water.

Inhalation of reclaimed water is known to cause lung inflammation, and free endotoxins have been shown to be a major risk factor for acute exposure. Subchronic exposure has also been shown to induce inflammatory responses with visible tissue damage. However, subchronic risk factors have yet to be identified, and a threshold for the protection of occupational populations during urban reuse is necessary. In this study, potential risk factors in reclaimed water were examined by subchronic exposure with fractionated reclaimed water, and the health risk threshold was tested with a series of diluted

Contact and respiratory sensitizers can be identified by cytokine profiles following inhalation exposure.

There are currently no validated animal models that can identify low molecular weight (LMW) respiratory sensitizers. The Local Lymph Node Assay (LLNA) is a validated animal model developed to detect contact sensitizers using skin exposure, but all LMW respiratory sensitizers tested so far were also positive in this assay. Discrimination between contact and respiratory sensitizers can be achieved by the assessment of cytokine profiles. In a LLNA using the inhalation route, both contact and respiratory sensitizers enhanced proliferation in the draining lymph nodes. The question was if their cyto

Dermal and inhalation exposure to methylene bisphenyl isocyanate (MDI) in iron foundry workers.

Diisocyanates are a group of chemically reactive agents, which are used in the production of coatings, adhesives, polyurethane foams, and parts for the automotive industry and as curing agents for cores in the foundry industry. Dermal and inhalation exposure to methylene bisphenyl isocyanate (MDI) is associated with respiratory sensitization and occupational asthma. However, limited research has been performed on the quantitative evaluation of dermal and inhalation exposure to MDI in occupationally exposed workers. The objective of this research was to quantify dermal and inhalation exposure l

Effect of short-term inhalation exposure to 1,3-butadiene on murine immune functions.

Interest in 1,3-butadiene (BD) as a potential immunomodulator was prompted by reports of an increased incidence of neoplasia in humans exposed to BD during the manufacture of styrene-butadiene synthetic rubber, and by a recent study which demonstrated a high incidence of thymic lymphomas in B6C3F1 mice. B6C3F1 mice were exposed to 1250 ppm BD by inhalation 6 hr per day, 5 days per week, for 6 or 12 weeks. Immune function assays were selected to evaluate specific humoral and cell-mediated immunity and spontaneous cytotoxicity; lymphoid organ histopathology was also evaluated. A slight decrease

Acute tubular necrosis after inhalation exposure to methylene chloride. Report of a case.

Nephrotoxic reaction occurred after inhalation exposure to methylene chloride. This exposure is associated with acute renal failure, myoglobinuria, hypocomplementemia, and liver enzyme elevations. Pathologic specimens, both light and electron microscopic, demonstrate renal tubular damage. We conclude that methylene chloride may have potential as both a hepatotoxic and nephrotoxic agent when inhaled at high concentrations over an extended period of time.

Effect of carbon monoxide inhalation exposure in mice on drug metabolism in vivo.

Experiments were undertaken to evaluate the action of carbon monoxide (CO) on the mixed-function oxidase (MFO) system in vivo. Mice were exposed to 500 ppm CO for 8 h per day in an inhalation chamber under dynamic airflow conditions. Hexobarbital (150 mg/kg, i.p.), zoxazolamine (150 mg/kg, i.p.) or ethanol (2 mg/g, i.p.) was given to each group of mice during CO exposure and disappearance of the drug from blood or brain was determined while CO exposure continued. The experiments were repeated with different groups of animals which were exposed to CO for 3 or 5 days. Hexobarbital and ethanol me

Safety of Mechanically Fibrillated Cellulose Nanofibers (CNFs) by Inhalation Exposure Based on TG412.

An investigation into the acute toxicity of mechanically fibrillated cellulose nanofibers (fib-CNFs), with a fiber length ranging from 500 to 600 nm, was conducted in accordance with the OECD TG412 guidelines. In this study, rats were exposed to fib-CNFs via nasal inhalation for 6 h daily over a 28-day period. The highest exposure concentration was set at 35 mg/m3, with intermediate and low concentrations at 7.0 mg/m3 and 1.5 mg/m3, respectively. No significant differences were observed in body weight, hematological parameters, or biochemical profiles between the fib-CNF-exposed groups and the

Maternal Electronic Cigarette Inhalation Exposure During Gestation: Impacts on Prolactin and Xanthine Oxidase.

Pregnancy requires metabolic and endocrine changes that must occur to support fetal growth and development. Aberrations in these necessary modifications can impact maternal health and fetal growth. Exposure to toxicants during pregnancy can negatively affect fetal health and development, but studies on electronic cigarette (e-cig) exposure is limited. We hypothesized that maternal e-cig exposure during gestation leads to hormonal and redox imbalance and negatively impacts fetal development. Pregnant Sprague-Dawley rats were exposed to e-cig aerosols (1227 ± 131 mg/m3) or HEPA-filtered air for

Toxicity evaluation in rats following 28 days of inhalation exposure to xylitol aerosol.

Xylitol has reported to decrease gingival inflammation and nasopharyngeal pneumonia, which indicated that xylitol may have potential application in respiratory diseases. Although some studies have reported the inhalation toxicity of xylitol, however, the longest period tested was only for 14 days. The inhalation toxicity of xylitol is insufficient. This work investigated the potential subacute toxicity of xylitol according to the OECD TG 412. Rats were randomly divided into a control group and different dosage groups (2 g/m3, 3 g/m3, 5 g/m3), and exposed for 6 hours/day, 5 days/week for 28 day

The evaluation of inhalation studies for exposure quality: A case study with formaldehyde.

The inherent complexity of generating and monitoring a test article in an inhalation chamber can make inhalation toxicity testing challenging. Poor study design, human error, and electrical and mechanical problems can adversely affect an inhalation exposure and undermine a study's results. We have developed a process for evaluating seven key elements of exposure quality in inhalation chamber studies: 1) test article characterization, 2) generation method, 3) chamber sampling and analytical method, 4) chamber concentrations, 5) particle size characteristics, 6) chamber type, and 7) controls. In silico modeling of spore inhalation reveals fungal persistence following low dose exposure.

The human lung is constantly exposed to spores of the environmental mould Aspergillus fumigatus, a major opportunistic pathogen. The spectrum of resultant disease is the outcome of complex host-pathogen interactions, an integrated, quantitative understanding of which lies beyond the ethical and technical reach permitted by animal studies. Here we construct a mathematical model of spore inhalation and clearance by concerted actions of macrophages and neutrophils, and use it to derive a mechanistic understanding of pathogen clearance by the healthy, immunocompetent host. In particular, we invest
