Extracellular vesicles (EVs) are key mediators of intercellular communication across biological kingdoms, with central roles in immune regulation and disease processes. Despite shared structural features, EVs derived from bacteria, plants, and mammalian cells differ substantially in their biogenesis, molecular composition, and immunological functions. EV formation pathways generate vesicles with distinct cargo profiles, including pathogen-associated molecular patterns (PAMPs) in bacterial EVs, regulatory small RNAs in plant-derived vesicles, and cytokines, microRNAs, and antigen-presenting com

Despite our constant exposure to potential dangers, our ability to withstand infection and tissue damage relies on a network of coordinated cellular security systems. One of these security features is our innate immune system, which senses danger signals from invading pathogens as well as host-derived stress signals while remaining largely unresponsive to normal host molecules and the flora of the host microbiota. This intertwining network of cellular guardians expresses pattern recognition receptors to recognise specifically pathogen-associated molecular patterns and danger-associated molecul

Exogeneous pathogen-associated molecular patterns and endogenous danger signals bind to pattern recognition receptors and activate innate immunity cells, leading to proinflammatory cytokine production and activation of acquired immue cells. These are important factors in the pathogenesis of autoimmune-mediated neuroimmunological disorders such as multiple sclerosis. Furthermore, recent advances in the study of innate immunity revealed that innate immunity is a major players in the pathogenesis of some neuroimmunological diseases such as Behçet's disease and herpes simplex virus encephalitis.

Katherine C. Barnett studies cell death and damage in viral infections, focusing on how these processes activate innate immunity and shape viral pathogenesis. In this mSphere of Influence article, she highlights how the work of Aguirre et al. (Nat Microbiol 2:17037, 2017, https://doi.org/10.1038/nmicrobiol.2017.37) changed her perception of how RNA viruses interact with pattern recognition receptors and how later the work of Wang et al. (Nature 616:152-158, 2023, https://doi.org/10.1038/s41586-023-05851-w) shaped her concept of cell death in viral infections. Framing these findings in differen

Adaptive immunity towards tuberculosis (TB) has been extensively studied for many years. In addition, in recent years the profound contribution of innate immunity to host defence against this disease has become evident. The discovery of pattern recognition receptors, which allow innate immunity to tailor its response to different infectious agents, has challenged the view that this arm of immunity is nonspecific. Evidence is now accumulating that innate immunity can remember a previous exposure to a microorganism and respond differently during a second exposure. Although the specificity and me

Exposed to diverse pathogenic and non-pathogenic insults, the airway epithelium must balance effective host defense while minimizing unnecessary inflammation and tissue damage. In studies of influenza A virus infection including spatial transcriptomics, Nguyen et al. illustrate how tissue-level organization of viral sensing may tune the intensity of innate immune responses.

The gradual assembly and oligomerization of cell surface receptors are critical for regulating immune activation and turnover. Yet, how dynamic, spatiotemporally regulated assembly enables rapid and sustained signaling in plant innate immunity remains unclear, owing to the inherent complexity of their dynamic associations. Using a synthetic engineering approach, we precisely controlled the oligomerization states of the Arabidopsis FLS2 receptor. Single-particle imaging with quantitative calibration revealed that FLS2 transits from a predominantly monomeric resting state to transient homo-assoc

Red blood cells (RBCs) were historically regarded as immunologically inert; however, current research has revealed their active participation in the innate immune response through the expression of various surface immune receptors. Notably, the nucleic-acid-sensing receptor toll-like receptor 9 (TLR9) enables RBCs to sequester and scavenge cell-free DNA under basal conditions and to deliver DNA during inflammatory states. Consequently, the presence of TLR9 on RBCs endows them with distinct immune functions. Novel techniques were developed to study the immunological function of red blood cells.

Astrocytes, long considered supportive cells of the central nervous system (CNS), have critical roles in innate immunity. This Review explores immune signaling pathways in astrocytes, including pattern recognition through Toll-like receptors, nucleic acid sensors and inflammasomes. These pathways enable the detection of danger signals and initiate protective responses and endogenous innate immune functions. Downstream signaling pathways, including the interferon, NF-κB and STAT3 pathways, mediate astrocyte reactivity and drive cytokine secretion, antiviral responses, phagocytosis and many othe

The complement system as a part of innate immunity is considered to provide rapid tough incomplete antimicrobial activity. However, besides providing a first-line defence, innate immunity plays additional important roles: it initiates and improves the slower, but more specific, acquired immune response. The recognition and destruction of noxious substances, as well as initiation of the acquired immune response, are accompanied by potentially hazardous inflammation. The inflammatory process has thus to be tightly regulated. In this overview, innate immunity and its interactions with acquired im

Fungal cell wall polysaccharides - such as β-glucans, chitin, and mannans - are key immunogenic structures recognized by Toll-like receptors and C-type lectin receptors on innate immune cells, initiating phagocytosis, cytokine production, and antigen presentation, and shaping downstream adaptive immunity. CD4+ T cells differentiate into Th1, Th2, Th17, and Treg subsets to maintain immune balance, whereas B cells contribute to fungal clearance and memory through species-specific antibodies. The boundary between protective and pathogenic immunity varies with fungal species, infection stage, and

Innate immunity is the first defense against viral infections, limiting viral replication and initiating adaptive immunity. Viral pathogens are recognized by pattern recognition receptors (PRRs), including Toll-like receptors (TLRs), (RIG-I-like receptors) RLRs, and the (cyclic GMP-AMP synthase) cGAS-STING pathway, which trigger interferon production and antiviral responses. This review compares innate immune responses to major RNA viruses (influenza, SARS-CoV-2, HIV) and DNA viruses (HSV, HBV, CMV). While these viruses activate similar pathways, they differ in interferon dynamics, inflammator

Inflammation plays a key role in the formation, progression, and rupture of intracranial aneurysms, leading to aneurysmal subarachnoid hemorrhage. Pathogen PPRs (pattern recognition receptors) that are activated by pathogen-derived molecules are key mediators of the innate immune response against microbes. As PPRs are activated by molecules resembling microbes, that is, pathogen-associated molecular patterns or damage-associated molecular patterns that can be of nonmicrobial origin, exposure to pathogen-associated molecular patterns and damage-associated molecular patterns may promote an exces

Respiratory infections caused by viral pathogens, including influenza A virus, respiratory syncytial virus, and SARS-CoV-2, as well as bacterial pathogens, remain major contributors to global morbidity and mortality. The outcomes of these infections are largely determined by how the host innate immune system detects pathogen-derived nucleic acids and regulates subsequent inflammatory responses. Pattern recognition receptors (PRRs) located in endosomal and cytosolic compartments, including toll-like receptors, RIG-I-like receptors, absent in melanoma 2, and Z-DNA-binding protein 1, as well as t

Over 10 species of Rickettsia cause serious human disease across the globe, including deadly spotted fever and typhus. These arthropod-borne microbes and their close relatives are the only bacteria that obligately reside in the eukaryotic cytosol, making them distinctive tools to study innate immunity. Yet, the molecular details on how Rickettsia species interact with innate immunity have remained largely enigmatic. Recent advances are revealing that Rickettsia species are not only distinct from other bacterial pathogens that reside in the cytosol, but in some cases, they are highly distinct f

Influenza virus and coronavirus are two important respiratory viruses, which often cause serious respiratory diseases in humans and animals after infection. In recent years, highly pathogenic avian influenza virus (HPAIV) and SARS-CoV-2 have become major pathogens causing respiratory diseases in humans. Thus, an in-depth understanding of the relationship between viral infection and host innate immunity is particularly important to the stipulation of effective control strategies. As the first line of defense against pathogens infection, innate immunity not only acts as a natural physiological b

Viruses are obligate parasites which are able to infect cells of all living organisms. Multiple antiviral defense mechanisms have appeared early in evolution of the immune system. Higher vertebrates have the most complex antiviral immunity which is based on both innate and adoptive immune responses. However, majority of living organisms, including plants and invertebrates, rely exclusively on innate immune mechanisms for protection against viral infections. There are some striking similarities in several components of the innate immune recognition between mammals, plants and insects, rendering
