Developing the radiation-based sterile insect technique (SIT) for controlling Aedes aegypti: identification of a sterilizing dose.

BACKGROUND: The sterile insect technique (SIT) is emerging as a tool to supplement traditional pesticide-based control of Aedes aegypti, a prominent mosquito vector of microbes that has increased the global burden of human morbidity and mortality over the past 50 years. SIT relies on rearing, sterilizing and releasing large numbers of male mosquitoes that will mate with fertile wild females, thus reducing production of offspring from the target population. In this study, we investigated the effects of ionizing radiation (gamma) on male and female survival, longevity, mating behavior, and steri

Controlled insect-sting challenge in 55 patients: correlation between activation of plasminogen and the development of anaphylactic shock.

The pathogenesis of anaphylactic shock is not completely understood. Mast cell degranulation products may stimulate endothelial cells, leading to activation of fibrinolytic and coagulation systems. We investigated the activation of these systems in insect-sting anaphylaxis. Fifty-five patients with a previous insect-sting anaphylactic reaction and 8 volunteers were challenged with an in-hospital sting. Plasma levels of von Willebrand factor (vWF), coagulation, and fibrinolytic parameters were assessed. After the sting challenge, 20 patients developed anaphylactic symptoms, 7 of whom developed

Design and stable flight of a 21 g insect-like tailless flapping wing micro air vehicle with angular rates feedback control.

An insect-like tailless flapping wing micro air vehicle (FW-MAV) without feedback control eventually becomes unstable after takeoff. Flying an insect-like tailless FW-MAV is more challenging than flying a bird-like tailed FW-MAV, due to the difference in control principles. This work introduces the design and controlled flight of an insect-like tailless FW-MAV, named KUBeetle. A combination of four-bar linkage and pulley-string mechanisms was used to develop a lightweight flapping mechanism that could achieve a high flapping amplitude of approximately 190°. A new approach to insect-pest control--combination of neurotoxins interacting with voltage sensitive sodium channels to increase selectivity and specificity.

Voltage-sensitive sodium channels are responsible for the generation of electrical signals in most excitable tissues and serve as specific targets for many neurotoxins. At least seven distinct classes of neurotoxins have been designated on the basis of physiological activity and competitive binding studies. Although the characterization of the neurotoxin receptor sites was predominantly performed using vertebrate excitable preparations, insect neuronal membranes were shown to possess similar receptor sites. We have demonstrated that the two mutually competing anti-insect excitatory and depress

Nanoformulation of poly(ethylene glycol) polymerized organic insect repellent by PIT emulsification method and its application for Japanese encephalitis vector control.

The utilization of increased dosage of insect repellents to overcome mosquito resistance has raised environmental concerns globally. In accord to this, we have formulated an efficacious, water-dispersive, nanometric formulation of a poor water-soluble insect repellent, diethylphenylacetamide (DEPA) by poly(ethylene glycol) (PEG) polymerization followed by PIT emulsification method. The critical micelle concentration of PEG in the spontaneously emulsified conventional DEPA droplets was determined, based on the droplets physical stability. Subjecting them to PIT emulsification yielded monodisper

The role of the United States military in the development of vector control products, including insect repellents, insecticides, and bed nets.

Arthropod-borne diseases such as malaria, dengue, scrub typhus, and leishmaniasis continue to pose a significant threat to U.S. military forces deployed in support of operational and humanitarian missions. These diseases are transmitted by a variety of arthropods, including mosquitoes, ticks, chiggers, sand flies, and biting midges. In addition to disease threats, biting arthropods can cause dermatitis, allergic reactions, and sleep loss; therefore, monitoring of vector impact and integrated use of personal protective measures (PPM) and methods to reduce the vector populations are needed to pr

Boosting the sterile insect technique with pyriproxyfen increases tsetse flies Glossina palpalis gambiensis sterilization in controlled conditions.

Tsetse flies (Diptera: Glossinidae) are the main vectors of animal and human trypanosomoses in Africa. The Sterile Insect Technique (SIT) has proven effective in controlling tsetse flies when applied to isolated populations but necessitates the production of large numbers of sterile males. A new approach, called boosted SIT, combining SIT with the contamination of wild females by sterile males coated with biocides has been proposed for large-scale control of vector populations. The aim of the study was to evaluate this new approach using pyriproxyfen on the riverine species Glossina palpalis g

Preparation for targeted sterile insect technique to control invasive Aedes aegypti (Diptera: Culicidae) in southern California: dose-dependent response, survivorship, and competitiveness.

Aedes aegypti is fast spreading across California, with over 300 cities within 22 central and southern counties being infested since its introduction in 2013. Due to its cryptic breeding habitats, control efforts have not been successful so far. This calls for innovative tools such as sterile insect technique (SIT) to reinforce the existing integrated pest management (IPM). Here, we assessed fitness, survivorship, and dose response of X-ray irradiated male Ae. aegypti in California. Locally acquired Ae. aegypti eggs were hatched and reared in temperature-controlled laboratory setting at the We

Viral diversity in predatory insects used for biological control: Insights from a multi-country meta-viromic analysis.

Predatory insects such as ladybirds, green lacewings, and an aphid midge have been commercialized and deployed worldwide to deal with the challenge of pest control. However, limited information is available on their associated viruses. To start filling this gap, this study employed an integrative bioinformatics approach with 21 publicly available RNA-seq libraries, from six countries, of eight natural enemy species used to control aphids. A total of 41 putative novel viruses were identified and classified into 16 families and 1 genus. In addition, 13 known viruses were detected, including aphi

Effectiveness of the sterile insect technique in controlling Aedes albopictus as part of an integrated control measure: evidence from a first small-scale field trial in Switzerland.

BACKGROUND: The invasive Asian tiger mosquito (Aedes albopictus) poses growing health risks across Europe. In Switzerland, a preliminary field trial was conducted to assess the feasibility of integrating the sterile insect technique (SIT) into existing integrated vector management (IVM), which includes breeding site removal and application of biological larvicides. SIT involves repeated releases of irradiated sterile males, which mate with wild females, producing non-viable eggs and leading to population decline. METHODS: Following a preliminary release test in 2022, a small-scale SIT trial t

Feasibility of using the radiation-based sterile insect technique (SIT) to control the olive fruit fly, Bactrocera oleae Gmelin (Diptera: Tephritidae) in Iran.

Bactrocera oleae is one of the most hazardous pests threatening olive orchards in Iran. SIT is an environment-friendly system of pest control based on releasing sterile males able to compete with wild males to mate with wild females. To determine sterile doses of radiation, pupae were irradiated to the doses of 0-160 Gy. Doses of 90-100 Gy were found optimal providing the necessary sterilization without severely impairing the competitiveness of the irradiated males in mating.

Evaluation of different irradiation doses for Aedes aegypti (Linnaeus) to optimize the Sterile Insect technique for arbovirus control in Burkina Faso (West Africa).

The sterile insect technique (SIT) relies on the release of sterile males that mate with wild females, thereby reducing the reproductive capacity of the target population. Its effectiveness depends on identifying an irradiation dose that induces high sterility while preserving male biological quality. This study evaluated the effects of gamma irradiation doses of 20, 30, 50, 70, and 90 Gy on adult emergence, flight ability, survival, fecundity, and fertility in a laboratory strain of Aedes aegypti established from a population collected in Burkina Faso. The results show that irradiation did no

Sustainable cellulose acetate beads impregnated with eugenol, thymol, β-citronellol, and their mixtures via supercritical carbon dioxide for microbial and pest insect control.

Post-harvest losses of legumes caused by microbial contamination and pest insects represent a major challenge for food security and sustainable storage systems. In this study, biodegradable and bio-based cellulose acetate beads (CAB) were developed as functional materials for bean protection using an environmentally benign supercritical impregnation process. Eugenol, thymol, β-citronellol, and their mixtures were incorporated into CAB, achieving loadings of up to 7.5 wt% within a 3 h process. Impregnation resulted in a pronounced reduction in average pore size, from approximately 0.5 to 0.2 μm

Isolation and Quantification of L-Tryptophan from Protaetia brevitarsis seulensis Larvae as a Marker for the Quality Control of an Edible Insect Extract.

Protaetia brevitarsis seulensis (Kolbe, 1886) larvae have traditionally been used in East Asian medicine and have recently attracted attention as functional food ingredients because of their pharmacological potential. However, chemical investigations remain limited, and no marker compounds have been established for quality control. This study aimed to isolate and identify a primary constituent from the 70% ethanol extract of P. brevitarsis (PBE) and to develop an analytical method for its quantification. Among the solvent-partitioned fractions, the n-butanol fraction (PBE-B) exhibited a major

Efficacy of four insect repellents against mosquito bites: a double-blind randomized placebo-controlled field study in Senegal.

Insect-borne diseases represent a worldwide threat. In addition to fight against vectors (insecticides) and disease prevention (vaccination against yellow fever, chemoprophylaxis against malaria), insect repellents applied on the skin could help reduce the heavy burden related to these diseases. In a field study performed in Senegal, we compared the efficacy of one skin application between 3 and 4 p.m. of four spray repellents [icaridine 20%, para-menthane-diol (PMD) 20% and 50% and DEET 50%] against placebo, among 100 healthy male and female volunteers experienced with mosquito capture. Impact of Wolbachia-mediated incompatible and sterile insect techniques on dengue incidence: a large-scale cluster randomised controlled trial.

BACKGROUND: Wild-type female Aedes aegypti which mate with Wolbachia-infected male Aedes aegypti (Wolbachia-Aedes) produce non-viable eggs due to cytoplasmic incompatibility. Sustained releases of Wolbachia-infected male Aedes aegypti can suppress wild-type mosquito populations, but robust evidence of its efficacy in reducing dengue incidence is needed. METHODS: We conducted a large-scale cluster-randomised controlled trial involving releases of male wAlbB-infected Ae. aegypti mosquitoes (wAlbB-SG) for dengue control in Singapore. Mosquitoes were irradiated to sterilise potential residual fem

Construction of a genetic sexing strain for Aedes albopictus: a promising tool for the development of sterilizing insect control strategies targeting the tiger mosquito.

BACKGROUND: Aedes albopictus is an invasive mosquito species of global medical concern as its distribution has recently expanded to Africa, the Americas and Europe. In the absence of prophylaxis protecting human populations from emerging arboviruses transmitted by this mosquito species, the most straightforward control measures rely on the suppression or manipulation of vector natural populations. A number of environmental-friendly methods using mass releases of sterilizing males are currently under development. However, these strategies are still lacking an efficient sexing method required fo
