Modeling resource allocation strategies for insecticide-treated bed nets to achieve malaria eradication.

Large reductions in the global malaria burden have been achieved, but plateauing funding poses a challenge for progressing towards the ultimate goal of malaria eradication. Using previously published mathematical models of Plasmodium falciparum and Plasmodium vivax transmission incorporating insecticide-treated nets (ITNs) as an illustrative intervention, we sought to identify the global funding allocation that maximized impact under defined objectives and across a range of global funding budgets. The optimal strategy for case reduction mirrored an allocation framework that prioritizes funding

Vector composition, abundance, biting patterns and malaria transmission intensity in Madang, Papua New Guinea: assessment after 7 years of an LLIN-based malaria control programme.

BACKGROUND: A malaria control programme based on distribution of long-lasting insecticidal bed nets (LLINs) and artemisinin combination therapy began in Papua New Guinea in 2009. After implementation of the programme, substantial reductions in vector abundance and malaria transmission intensity occurred. The research reported here investigated whether these reductions remained after seven years of sustained effort. METHODS: All-night (18:00 to 06:00) mosquito collections were conducted using human landing catches and barrier screen methods in four villages of Madang Province between September

Physical integrity and residual bio-efficacy of PBO-pyrethroid synergist-treated and pyrethroid-only LLINs after 1.5 years of field use in Western Kenya.

BACKGROUND: Long-lasting insecticidal nets (LLINs) are vital for malaria control in sub-Saharan Africa, but their durability is challenged by fabric decay and pyrethroid resistance. This study assessed the physical integrity and bioefficacy of piperonyl butoxide-LLINs (PBO-LLINs) and pyrethroid-only LLINs (pyrethroid-LLINs) after 1.5 years of use in western Kenya, where resistance is widespread. METHODS: A survey on net integrity and insecticide efficacy was conducted in randomly selected households (101-107 per group per visit) from three villages per net type group in Muhoroni Sub-County, K

Managing insecticide resistance in malaria vectors in Africa: case studies from Cameroon, Côte d'Ivoire and Tanzania.

BACKGROUND: Malaria remains a significant public health challenge in sub-Saharan Africa (SSA), where vector control strategies, particularly insecticide-treated nets (ITNs) and indoor residual spraying (IRS), have played a critical role in reducing transmission. However, the emergence and spread of insecticide resistance among malaria vectors threatens to undermine these gains. In response, many countries in the region have developed national insecticide resistance management (IRM) plans. This study evaluates the adequacy and implementation of these plans in Cameroon, Côte d'Ivoire, and mainla

Cascades of effectiveness of next-generation insecticide-treated nets against malaria, from entomological trials to real-life conditions.

As insecticide resistance spreads in Africa, next-generation insecticide-treated nets (ITNs) are increasingly being deployed to protect vulnerable populations against malaria. While these nets provide greater entomological efficacy against resistant mosquitoes, their effectiveness against malaria transmission also depends on other factors, such as durability, access, usage, and activity patterns of hosts and vectors. Here, we quantify the impact of two next-generation ITNs, namely Interceptor®G2 (chlorfenapyr-pyrethroid) and Olyset® Plus (piperonyl butoxide-pyrethroid), in a cascade from entom

Efficacy of PermaNet® Dual compared to Interceptor® G2 and PermaNet 3.0 in experimental huts in Siaya County, western Kenya.

BACKGROUND: Pyrethroid-chlorfenapyr nets have shown significant epidemiological impact over pyrethroid-only and pyrethroid plus piperonyl-butoxide (PBO) in Africa. A non-inferiority evaluation of PermaNet® Dual, a new chlorfenapyr plus deltamethrin net, compared to Interceptor® G2, was conducted in experimental huts in Siaya, Kenya against free-flying pyrethroid-resistant Anopheles funestus. METHODS: This study was an experimental hut trial, following a 7 by 7 Latin Square design. Seven treatments and seven sleepers were deployed in the experimental huts daily and rotated weekly and daily, re

Anopheles gambiae Re-Emergence and Resurgent Malaria Transmission in Eastern Rwanda, 2010-2020.

Rwanda achieved unprecedented malaria control gains from 2000 to 2010, but cases increased 20-fold between 2011 and 2017. Vector control challenges and environmental changes were noted as potential explanations, but no studies have investigated causes of the resurgence or identified which vector species drove transmission. We conducted a retrospective study in four sites in eastern Rwanda that conducted monthly entomological surveillance and outpatient malaria care. We compared sporozoite rates, human blood index (HBI), and relative abundance of the primary vectors, Anopheles gambiae and Anoph

An experimental hut study evaluating the impact of pyrethroid-only and PBO nets alone and in combination with pirimiphos-methyl-based IRS in Ethiopia.

BACKGROUND: Pyrethroid resistance observed in populations of malaria vectors is widespread in Ethiopia and could potentially compromise the effectiveness of insecticide-based malaria vector control interventions. In this study, the impact of combining indoor residual spraying (IRS) and insecticide-treated nets (ITNs) on mosquito behaviour and mortality was evaluated using experimental huts. METHODS: A Latin Square Design was employed using six experimental huts to collect entomological data. Human volunteers slept in huts with different types of nets (pyrethroid-only net, PBO net, and untreat

Efficacy of Olyset long-lasting bednets to control transmission of cutaneous leishmaniasis in Iran.

A large-scale intervention field trial of the effect of Olyset long-lasting insecticide-treated bednets on transmission of cutaneous leishmaniasis was carried out in 2 cities in the Islamic Republic of Iran from October 2003 to July 2005. We enrolled 8620 individuals in 3000 households in 6 pairs of sectors in each city. Epidemiological and entomological surveys were carried out pre- and post-intervention. In both cities a statistically significant reduction was found in the incidence of new cases in intervention sectors who received bednets compared with control areas. Entomological surveys s

Elevated Temperature Decreases the Insecticidal Efficacy of Yahe LN Insecticide-Treated Nets: Implications for Transport and Storage.

A recent durability study has revealed that Yahe LN® insecticide-treated nets (ITNs; Fujian Yamei Industry & Trade Co., LTD, Fuzhou, China) distributed in Papua New Guinea (PNG) in 2021 did not retain their insecticidal efficacy after several months post-distribution. Insecticide-treated nets are frequently shipped and stored in containers where they are exposed to elevated temperatures. When used in the tropics, ITNs may continue to be exposed to high ambient temperatures. The aim for the present study is to better understand the potential impact of elevated temperature during transport and s

A randomized controlled trial combining house screening and insecticide-treated nets reduces malaria transmission in northwestern Ethiopia.

House screening (HS) of doors, eaves, and windows using wire-mesh has demonstrated potential in the integrated vector management of malaria. However, limited epidemiological data are available to guide its implementation across different ecological settings. In a 16-month randomized controlled trial (follow-up period) conducted across three agroecological areas (dry mountain, plateau highland, and semi-arid) in Jabi Tehnan district, northwestern Ethiopia, treatment houses were equipped with HS combined with insecticide-treated nets (ITNs), while control houses received ITNs only. Characteristics of malaria vector populations and transmission before a randomised controlled trial assessing the efficacy of next-generation insecticide-treated nets in Côte d'Ivoire.

BACKGROUND: The World Health Organization (WHO) recommends mass distribution of insecticide-treated nets (ITNs) to prevent malaria transmission. Unfortunately, resistance to pyrethroids affects the efficacy of standard ITNs. To overcome this resistance and continue to protect the population, the WHO has recommended new types of ITNs that combine a pyrethroid insecticide with either a synergist (PBO) or a second insecticide, such as chlorfenapyr. This study examines the baseline characteristics of malaria vectors prior to the distribution of three types of insecticide-treated nets as part of a

The non-inferiority of piperonyl-butoxide Yorkool® G3 insecticide-treated nets compared to Olyset®Plus measured by Anopheles arabiensis mortality in experimental huts in Tanzania.

BACKGROUND: Non-inferiority trials are recommended by the World Health Organization (WHO) to demonstrate that health products show comparable efficacy to that of existing standard of care. As part of the WHO Global Malaria Programme (GMP) process of assessment of malaria vector control products, a second-in-class insecticide-treated net (ITN) must be shown to be non-inferior to a first-in-class product based on mosquito mortality. The public health impact of the first-in-class pyrethroid-piperonyl butoxide (PBO) ITN, Olyset® Plus, has been demonstrated in epidemiological trials in areas with i

Evaluation of improved coloured targets to control riverine tsetse in East Africa: A Bayesian approach.

BACKGROUND: Riverine tsetse (Glossina spp.) transmit Trypanosoma brucei gambiense which causes Gambian Human African Trypanosomiasis. Tiny Targets were developed for cost-effective riverine tsetse control, and comprise panels of insecticide-treated blue polyester fabric and black net that attract and kill tsetse. Versus typical blue polyesters, two putatively more attractive fabrics have been developed: Vestergaard ZeroFly blue, and violet. Violet was most attractive to savannah tsetse using large targets, but neither fabric has been tested for riverine tsetse using Tiny Targets. METHODS: We

Targeting indoor residual spraying for malaria using epidemiological data: a case study of the Zambia experience.

BACKGROUND: In Zambia and other sub-Saharan African countries affected by ongoing malaria transmission, indoor residual spraying (IRS) for malaria prevention has typically been implemented over large areas, e.g., district-wide, and targeted to peri-urban areas. However, there is a recent shift in some countries, including Zambia, towards the adoption of a more strategic and targeted IRS approach, in coordination with increased emphasis on universal coverage of long-lasting insecticidal nets (LLINs) and effective insecticide resistance management. A true targeted approach would deliver IRS to s

Long-lasting insecticidal nets are synergistic with mass drug administration for interruption of lymphatic filariasis transmission in Nigeria.

In central Nigeria Anopheles mosquitoes transmit malaria and lymphatic filariasis (LF). The strategy used for interrupting LF transmission in this area is annual mass drug administration (MDA) with albendazole and ivermectin, but after 8 years of MDA, entomological evaluations in sentinel villages showed continued low-grade mosquito infection rates of 0.32%. After long-lasting insecticidal net (LLIN) distribution by the national malaria program in late 2010, however, we were no longer able to detect infected vectors over a 24-month period. This is evidence that LLINs are synergistic with MDA i
