Improving the binding of metal complexes to DNA to boost cancer cell cytotoxicity requires fine tuning of their structural and chemical properties. Copper has been used as a metal center in compounds containing intercalating ligands due to its ability to catalytically generate reactive oxygen species (ROS), such as hydroxyl radicals (OH˙). We envision the synergy of DNA binding and ROS generation in proximity to target DNA as a powerful chemotherapy treatment. Here, we explore the use of [Cu(2CP-Bz-SMe)]2+ (2CP-Bz-SMe = 1,3-bis(1,10-phenanthrolin-2-yloxy)-N-(4-(methylthio)benzylidene)propan-2-

The interaction of calf thymus DNA (ct DNA) with anastrozole, which is acknowledged as an antineoplastic drug, has been enquired into in the absence and presence of histone H1, through the means of absorbance, fluorescence, circular dichroism spectroscopy, viscosity, thermal melting, and molecular modeling techniques. In addition, the effects of anastrozole on MCF 7 cell line have been thoroughly investigated. Fluorescence spectroscopy results have indicated that quenching mechanism of ct DNA-anastrozole are known as static quenching procedures, since the Stern-Volmer quenching constant (KSV)

The interaction of Cu(NO3)2·3H2O with the sulfonyl o-pyridine carboxamidoxime N'-(4-nitrophenylsulfonyloxy)picolinimidamide (L) resulted in the mononuclear complex [Cu(L1)2](L2)2 (1), where L1 = pyridine-2-carboxamidine ligand and (L2)- = 4-nitrobenzenesulfonate anion derived from the homolytic cleavage of the NO bond of L. The complex was characterized by diverse techniques including single-crystal X-ray crystallography. From the antimicrobial tests performed, complex 1 seems to be active against gram-negative bacterial strains. The complex binds tightly and reversibly to serum albumins and t

Circular dichroism (CD) is an useful technique for monitoring DNA conformation changes resulting from changes in environmental conditions, such as temperature, ionic strength, and pH, and also for the study of the interaction between DNA and ligands (including small molecules and proteins). CD spectroscopy of DNA arises from the asymmetric backbone sugars and by the helical structures often adopted by nucleic acids. By the interpretation of induced circular dichroism (ICD) of ligand signals resulting from the coupling of electric transition moments of the ligand, DNA bases within the asymmetri

Gilvocarcin V (GV), a coumarin, is a nucleic acid photosensitizer that is phototoxic to bacteria and mammalian cells at picomolar levels in the presence of near-UV radiation (UVA). We evaluated the effectiveness of GV plus UVA for inactivation of several viruses, including herpes simplex virus, type 1 (HSV) and the bacterial viruses phi X174, T7, PRD1 and phi 6. Some inactivation of the bacterial viruses was observed with UVA radiation alone (4-50% survival at 26 kJ/m2). Additional photosensitized inactivation was observed only with T7 and phi 6 at 2.0 microM GV. On the other hand, HSV was pho

Role of heterocyclic compounds with nitrogen substitution in therapeutic frontiers is well established. The efforts made in this study are directed to dissect the biological significance of benzimidazole-substituted zinc phthalocyanine derivative. Its capacity to act as an anticancer agent against the 2 B-lymphoma cell lines (low-grade and high-grade malignancy) was found out by recording florescence using Alamar blue dye. Further cytotoxic effect at the DNA level was analyzed by performing agarose gel electrophoresis. Molecular docking studies made mechanistic details crystal clear by showing

In view of their DNA intercalation activities as anticancer agents, novel fifteen [1,2,4]triazolo[4,3-c]quinazoline and bis([1,2,4]triazolo)[4,3-a:4',3'-c]quinazoline derivatives have been designed, synthesized and evaluated against HepG2 and HCT-116. The molecular design was performed to investigate the binding mode of the proposed compounds with DNA active site. The data obtained from biological testing highly correlated with that obtained from molecular modeling studies. HCT-116 was found to be more sensitive cell lines to the influence of the new derivatives. In particular, compounds 16, 1

With the urgent need for novel agents to combat emerging fungal resistance to existing drugs, activity in the exploration of small molecule DNA binders has increased. Recently, selected cationic heterocyclic compounds belonging to a broad class of molecules known to bind to the minor groove of DNA were revealed to have potent antifungal activity. These molecules are different from the conventional DNA-interacting drugs such as topoisomerase inhibitors, DNA alkylators or intercalators. Selected compounds are fungicidal towards a variety of pathogenic yeasts and molds, and a selected lead compou

In pursuit of novel anticancer agents with enhanced potency and selectivity, a new series of quinazoline-based derivatives was rationally designed, synthesized, and biologically evaluated as potential DNA intercalators and topoisomerase II (Topo II) inhibitors. Building upon a previously identified lead compound (6), structural modifications were introduced to reinforce key pharmacophoric features essential for DNA intercalation and enzyme inhibition. Two distinct series of compounds were developed, incorporating either N-phenylacetamide (VII-series) or N-(4-cinnamoylphenyl)acetamide (VIII-ser

Doxorubicin is a chemotherapeutic drug for cancer that intercalates into nucleosome-free regions at promoters. Doxorubicin was reported to result in loss of histone H3 trimethylated lysine 4 (H3K4me3). To further explore doxorubicin's mechanism of action, we determined the genomic location of the binding sites of doxorubicin in leukemic cells. The effect of doxorubicin intercalation into the chromatin of leukemic cells on histone modifications was also determined. We show that doxorubicin binding sites were present in the nucleosome-free regions associated with regulatory regions (promoters, e

The low binding affinity of unmodified triplex-forming oligonucleotides (TFO) is the main drawback to their promising utilization in gene therapy. In the present study, we have synthesized DNA intercalator 5-(pyren-1-ylethynyl)indole Y, known as twisted intercalating nucleic acid (TINA), by a Cu-mediated Sonogashira palladium-catalyzed coupling reaction of 1-ethynylpyrene with 5-iodoindole at a high temperature under anaerobic conditions. Coupling with indole C-5 was far more preferable in obtaining stable TINA-indole than enamine site C-3, as neither hydration of the triple bond to ketones no

Exposure to oxygen has been implicated as an important mechanism of mutations, cancer and aging. Most data supporting this notion have been obtained in vitro, but the elaborate defense systems against oxygen stress in aerobic organisms make it difficult to extrapolate in vitro data to in vivo conditions. In the present investigation the somatic mutation and recombination test (SMART) in Drosophila with the wing spot system (Graf et al., 1984) has been used as an in vivo system to study the effect of oxygen radicals generated by bleomycin (BLM). BLM causes a dose-related increase of wing spots

Ametantrone (HAQ) and mitoxantrone (DHAQ) are structurally similar antitumor drugs of the anthracenedione class. The cytostatic, cytotoxic, and antitumor activities of these drugs are different, with DHAQ being 10-100 times more potent, per molar basis. Both drugs are strong intercalators and intercalative modes of binding are suspected as relevant to their pharmacological activity. No significant differences, however, that could explain the differences in pharmacological activity are observed in their intercalative properties with respect to base specificity and binding affinity. A correlatio

BackgroundBoth human and veterinary cancer chemotherapy are undergoing a paradigm shift from a "one size fits all" approach to more personalized, patient-oriented treatment strategies. Personalized chemotherapy is dependent on the identification and validation of biomarkers that can predict treatment outcome and/or risk of toxicity. Many cytotoxic chemotherapy agents, including doxorubicin, base their mechanism of action by interaction with DNA and disruption of normal cellular processes. We developed a high-resolution/accurate-mass liquid chromatography-mass spectrometry DNA screening approac

This study explores the anticancer potential of rigid planar compounds capable of intercalating into tumor cell DNA, thereby inducing DNA damage and subsequent cell death. A novel series of compounds (c1-c20) were synthesized using 2H-1,4-benzoxazin-3(4H)-one as the planar core structure, with 1,2,3-triazole groups introduced at the 7-position. Biological evaluation across multiple human tumor cell lines revealed that several c-series compounds exhibited notable inhibitory activity against Huh-7 liver cancer cells, with IC50 values of 28.48 μM (c5), 32.60 μM (c14), 31.87 μM (c16), and 19.05 μM

Copper [Cu(II)] has been shown to enhance DNA damage in several biological systems. Binding of copper to DNA may be a key step in producing these lesions. The results of this study indicate that the DNA double helix contains at least two kinds of binding sites for copper. One site is present once every four nucleotides, has high affinity, and shows a cooperative effect. The other is an intercalating site for copper that is present in every base pair. This site is saturable, has a dissociation constant (Kd) for Cu(II) of 41 microM. In single-stranded DNA, we found an average copper binding site

Development of robust oligonucleotide-based probe technologies, capable of recognizing specific regions of double-stranded DNA (dsDNA) targets, continues to attract considerable attention due to the promise of tools for modulation of gene expression, diagnostic agents, and new modalities against genetic diseases. Our laboratory pursues the development of various strand-invading probes. These include Invader probes, i.e., double-stranded oligonucleotide probes with one or more +1 interstrand zipper arrangements of intercalator-functionalized nucleotides like 2'-O-(pyren-1-yl)methyl-RNA monomers
