The human fungal pathogen Candida glabrata appears to utilise unique stealth, evasion and persistence strategies in subverting the onslaught of host immune response during systemic infection. However, macrophages actively deprive the intracellular fungal pathogen of glucose, and therefore alternative carbon sources probably support the growth and survival of engulfed C. glabrata. The present study aimed to investigate the role of the glyoxylate cycle gene ICL1 in alternative carbon utilisation and its importance for the virulence of C. glabrata. The data showed that disruption of ICL1 rendered

The taxonomic status was determined of an aerobic, facultatively methylotrophic strain, JZL-4(T), isolated from activated sludge. The cells were gram-negative, asporogenous, colourless, motile, short rods. The strain utilized methanol, methylamine, formate and a variety of polycarbon compounds, but not methane, dichloromethane or CO(2)/H(2), as carbon and energy sources. C(1) compounds were assimilated via the isocitrate lyase-negative serine pathway. Optimal growth occurred at 30 °C, pH 6.5-7.5 and 0.5 % (w/v) NaCl. The major cellular fatty acids were C(18 : 1)ω7c and C(18 : 0). The major pho

Mycobacterium tuberculosis (Mtb) represents one of the most persistent bacterial threats to human health and new drugs are needed to limit its impact. Conditional knockdown mutants can help validate new drug targets, but the analysis of individual mutants is laborious and time consuming. Here, we describe quantitative DNA tags (qTags) and their use to simultaneously analyze conditional Mtb knockdown mutants that allowed silencing the glyoxylate and methylcitrate cycles (via depletion of isocitrate lyase, ICL), the serine protease Rv3671c, and the core subunits of the mycobacterial proteasome,

Information regarding the presence of the glyoxylate cycle in chick liver was sought. This metabolic pathway has long been thought to be absent from vertebrate tissues. Previous studies in other tissues have shown that, when present, this pathway is sensitive to vitamin-D. Thus, the effect of long-term vitamin-D deficiency and subsequent vitamin-D replacement on liver structure was studied by light microscopy. In addition, specific biochemical assays for the presence of glyoxylate cycle enzymes were performed. Light microscopy of lipid extracted tissues, light microscopic histochemistry, and q

BACKGROUND: Tuberculous meningitis (TBM) is caused by the dissemination of Mycobacterium tuberculosis (MTB) from the primary site of infection to the central nervous system. However, the bacterial factors associated with the pathogenesis of TBM remain unclear. This study employed transcriptomic and proteomic methods to comprehensively analyze the changes in genes and proteins and their associated pathways in MTB strains isolated from cerebrospinal fluid (CSF) of TBM and sputum of pulmonary TB (PTB) cases.
METHODOLOGY: Five MTB strains were subjected to OMICs (transcriptomic and proteomic) ana

The ABC transporter Cdr1 protein of Candida albicans, which plays a major role in antifungal resistance, has two transmembrane domains (TMDs) and two nucleotide-binding domains (NBDs). The 12 transmembrane helices of TMDs that are interconnected by extracellular and intracellular loops (ICLs) mainly harbor substrate recognition sites where drugs bind while cytoplasmic NBDs hydrolyze ATP which powers drug efflux. The coupling of ATP hydrolysis to drug transport requires proper communication between NBDs and TMDs typically accomplished by ICLs. This study examines the role of cytoplasmic ICLs of

Itaconate has garnered significant attention in recent years due to its immunomodulatory and antimicrobial functions. During inflammation and pathogenic infections, itaconate is formed through the decarboxylation of cis-aconitate in the mitochondrial tricarboxylic acid cycle and accumulates in large quantities to counteract excessive inflammation and pathogenic infections. However, many pathogenic bacteria have also evolved pathways to directly degrade itaconate or indirectly resist its stimulatory effects. We first review the research history and metabolic pathways of itaconate. Then we focus

In May 2024, outbreaks of massive mortality happened in the American bullfrog (Aquarana catesbeiana) farm in Honghu, Hubei Province China. A pathogenic strain of Elizabethkingia miricola, named as 2024EM05, was isolated and identified from diseased bullfrogs based on biochemical tests and molecular identification. Subsequently, the growth characteristic, pathogenicity and antibiotic sensitivity of the E. miricola strain, as well as the immune response of A. catesbeiana following E. miricola infection were both investigated. Virulence genes encoding polar flagella, capsule, urease accessory pro

Histoplasma capsulatum is a thermally dimorphic fungus distributed worldwide, but with the highest incidence in the Americas within specific geographic areas, such as the Mississippi River Valley and regions in Latin America. This fungus is the etiologic agent of histoplasmosis, an important life-threatening systemic mycosis. Dimorphism is an important feature for fungal survival in different environments and is related to the virulence of H. capsulatum, and essential to the establishment of infection. Proteomic profiles have made important contributions to the knowledge of metabolism and path

Earlier studies demonstrated that forestomach herbivores are less insulin sensitive than monogastric omnivores. The present study was carried out to determine if different distribution patterns of the glucose transporters GLUT1 and GLUT4 may contribute to these different insulin sensitivities. Western blotting was used to measure GLUT1 and GLUT4 protein contents in oxidative (masseter, diaphragm) and glycolytic (longissimus lumborum, semitendinosus) skeletal muscle membranes of monogastric omnivores (rats and pigs), and of forestomach herbivores (cows, adult goats, goat kids, and camels). Cordyceps militaris is an edible entomopathogenic fungus with substantial nutritional and medicinal value. However, the molecular mechanisms underlying the vegetative mycelia-to-primordia (PR) transition and bioactive metabolite synthesis are still poorly defined. Deletion of the acyl-CoA dehydrogenase gene (ACAD) caused malformed, stunted PR-like structures together with excessive aerial mycelial growth, severely disrupting PR initiation and fruiting-body elongation in C. militaris. Transcriptomic, untargeted metabolomic and multi-omics analyses were performed on wild-type (WT) and ACAD-knock

BACKGROUND: The role of vitamin D in obesity and diabetes is debated. Obese and/or diabetic patients have elevated levels of free fatty acids, increased susceptibility to gastrointestinal symptoms and are suggested to have altered vitamin D balance. The enteric nervous system is pivotal in regulating gastrointestinal activity and high fat diet (HFD) has been shown to cause loss of enteric neurons in ileum and colon. This study investigates the effect of vitamin D on HFD- and palmitic acid-induced enteric neuronal loss in vivo and in vitro.
METHODS: Mice were fed either a normal diet (ND) or H

Fourteen novel Schiff base compounds (AS-1∼AS-14) containing 5-amino-1H-1,2,4-triazole-3-carboxylic acid and substituted benzaldehyde were successfully synthesized, and their structures were verified by melting point, elemental analysis (EA) and spectroscopic techniques (Fourier Transform Infra-Red (FT-IR) and Nuclear Magnetic Resonance (NMR)). In vitro hyphal measurements were used to investigate the antifungal activities of the synthesised compounds against Wheat gibberellic, Maize rough dwarf and Glomerella cingulate. The preliminary studies indicated that all compounds had good inhibitory

BACKGROUND: Metabolic adaptation and maladaptation are hallmarks of the failing heart and may be a target for therapeutic interventions. For example, sustained glucose oxidation during cardiac stress is associated with increased activity and abundance of ACL (ATP-dependent citrate lyase, Acly), which produces acetyl-coenzyme A (CoA) from citrate and CoA and supports de novo lipid synthesis. However, our understanding of how ACL supports cardiac metabolic adaptation and its potential to modulate disease pathophysiology has not yet been investigated.
METHODS: We used human heart tissue samples

Identification of novel drug candidates against Candidozyma auris (C. auris), an emerging fungal pathogen associated with global healthcare outbreaks, is of considerable importance. A major clinical challenge in C. auris infection is its ability to evade host immune responses through distinct processes, including reduced exposure of the immunostimulatory cell-wall component β-1,3-glucan and metabolic reprogramming through the glyoxylate cycle (GC), which supports survival under low-carbon conditions. In the present study, we evaluated the antifungal efficacy of two novel carbazate derivatives,

Dehydroepiandrosterone (DHEA) treatment is effective in preventing or delaying the onset of various genetic and induced disorders of mice and rats. Associated with the beneficial therapeutic effects exerted by action of this steroid is the development of hepatomegaly. To determine whether the changes associated with hepatomegaly also involve alterations in activities of tissue enzymes, we evaluated the effects of DHEA (0.45% in food, w/w) on hepatic protein kinases, phosphatases, and lipogenic enzymes in mice of various strains. The rates of fatty acid and cholesterol syntheses also were evalu

The taxonomic position of an actinomycete, strain 15-116AT, isolated from a blood culture of a 31-year-old male from the United States was determined using polyphasic taxonomic analysis. Genome-based phylogeny showed that the strain was closely related to Streptomyces composti, Streptomyces thermocarboxydovorans, and Streptomyces thermospinosisporus species. Digital DNA-DNA (dDDH) relatedness and average nucleotide identity (ANI) between the draft genome sequence of the strain and those of its closest phylogenomic neighbors listed above were between 35.9% and 37.3% (< 70%) and 88.5%, 88.1%, an
