Cross-reaction between Formosan termite (Coptotermes formosanus) proteins and cockroach allergens.

Cockroach allergens can lead to serious allergy and asthma symptoms. Termites are evolutionarily related to cockroaches, cohabitate in human dwellings, and represent an increasing pest problem in the United States. The Formosan subterranean termite (Coptotermes formosanus) is one of the most common species in the southern United States. Several assays were used to determine if C. formosanus termite proteins cross-react with cockroach allergens. Expressed sequence tag and genomic sequencing results were searched for homology to cockroach allergens using BLAST 2.2.21 software. Whole termite extr

Study on the role of RPL23 gene in active immunity of termite Reticulitermes chinensis against Metarhizium anisopliae.

Ribosomal proteins are considered to be involved in the immunity of different animals against pathogens. The protein level of RPL23 increased after fungal infection in termites, but how it influence active immunity in termites is unknown. The role of RPL23 gene was studied to evaluate its impact on active immunity of termite Reticulitermes chinensis against entomopathogenic fungus (EPF) Metarhizium anisopliae. The RPL23 gene fragment (414 bp) was cloned and phylogenetic analysis revealed that it's very close to termite Coptotermes formosanus. Expression of RPL23 gene was significantly higher i

Development of Multiplex Nested PCR for Simultaneous Detection of Ectoparasitic Fungi Laboulbeniopsis termitarius and Antennopsis gallica on Reticulitermes speratus (Blattodea: Rhinotermitidae).

Laboulbeniopsis termitarius (Thaxt) and Antennopsis gallica (Buchli and Heim) are two of the most common ectoparasitic fungi found on the body surface of termites. While visual observation under a dissecting microscope is a common method used to screen for such fungi, it generally requires a large number of termites and is thus very time consuming. In this study, we develop a fast, efficient protocol to detect fungal infection on the termite Reticulitermes speratus (Kolbe). Species-specific primers were designed based on sequence data and amplified using a number of universal fungus primer pai

Chemical Interaction among Termite-Associated Microbes.

Bacteria and fungi in shared environments compete with one another for common substrates, and this competition typically involves microbially-produced small molecules. An investigation of one shared environmental niche, the carton material of the Formosan subterranean termite Coptotermes formosanus, identified the participants on one of these molecular exchanges. Molecular characterization of several termite-associated actinobacteria strains identified eleven known antimicrobial metabolites that may aid in protecting the C. formosanus colony from pathogenic fungal infections. One particular ac

Mercury species in the nests and bodies of soil-feeding termites, Silvestritermes spp. (Termitidae, Syntermitinae), in French Guiana.

Mercury pollution is currently a major public health concern, given the adverse effects of mercury on wildlife and humans. Soil plays an essential role in speciation of mercury and its global cycling, while being a habitat for a wide range of terrestrial fauna. Soil fauna, primarily soil-feeding taxa that are in intimate contact with soil pollutants are key contributors in the cycling of soil mercury and might provide relevant indications about soil pollution. We studied the enrichment of various mercury species in the nests and bodies of soil-feeding termites Silvestritermes spp. in French Gu

Single-Cell DNA barcoding using sequences from the small subunit rRNA and internal transcribed spacer region identifies new species of Trichonympha and Trichomitopsis from the hindgut of the termite Zootermopsis angusticollis.

To aid in their digestion of wood, lower termites are known to harbour a diverse community of prokaryotes as well as parabasalid and oxymonad protist symbionts. One of the best-studied lower termite gut communities is that of Zootermopsis angusticollis which has been known for almost 100 years to possess 3 species of Trichonympha (T. campanula, T. collaris, and T. sphaerica), 1 species of Trichomitopsis (T. termopsidis), as well as smaller flagellates. We have re-assessed this community by sequencing the small subunit (SSU) rRNA gene and the internal transcribed spacer (ITS) region from a larg

Elucidating the redox cycle of environmental phosphorus using ion chromatography.

Historically, it was assumed that reactive, inorganic phosphorus present in pristine environments was solely in the form of orthophosphate. However, this assumption contradicts theories of biogenesis and the observed metabolic behavior of select microorganisms. This paper discusses the role of ion chromatography (IC) in elucidating the oxidation-reduction cycle of environmental phosphorus. These methods employ suppressed-IC, coupled with tandem conductivity and electrospray mass spectrometry detectors to identify and quantify phosphorus oxyanions in natural water, synthetic cosmochemical, and

A novel beta-1,4 glucanase produced by symbiotic Bacillus sp. CF96 isolated from termite (Anacanthotermes).

A novel beta-1,4-glucanase was purified and characterized from symbiotic Bacillus sp. CF96 of termite. The SDS-PAGE and zymogram analyses revealed a molecular mass of 35.6 kDa. Optimal activity was at 50 °C and pH 5.5, while the enzyme was active over a wide range of temperature 20-80 °C and pH 4-10 and interestingly more than 60% of the maximum activity remained up to pH 9. The enzyme activity increased in the presence of hexane, chloroform and methanol (20% v/v). while, the enzyme activity was inhibited by metal ions such as Mn2+, Hg2+, Cu2+, Zn2+, Mg2+, Fe2+. The isolated enzyme was able to

Natalenamides A⁻C, Cyclic Tripeptides from the Termite-Associated Actinomadura sp. RB99.

In recent years, investigations into the biochemistry of insect-associated bacteria have increased. When combined with analytical dereplication processes, these studies provide a powerful strategy to identify structurally and/or biologically novel compounds. Non-ribosomally synthesized cyclic peptides have a broad bioactivity spectrum with high medicinal potential. Here, we report the discovery of three new cyclic tripeptides: natalenamides A⁻C (compounds 1⁻3). These compounds were identified from the culture broth of the fungus-growing termite-associated Actinomadura sp. RB99 using a liquid c

Integrative taxonomy and metabolite profiling reveal functional divergence among wild edible Termitomyces from Indonesia.

Termitomyces species are termite-associated fungi that play essential roles in lignocellulose degradation and nutrient cycling and represent a promising source of bioactive metabolites. However, comparative understanding of metabolite diversity and its potential contribution to functional variation among species remains limited. This study investigated three wild edible Termitomyces species (Termitomyces intermedius, Termitomyces microcarpus, and Termitomyces eurrhizus) from Indonesia using an integrative approach combining morphological characterization, ITS rDNA phylogeny, proximate analysis

Biochemical and Structural Characterization of a Highly Glucose-Tolerant β-Glucosidase from the Termite Reticulitermes perilucifugus.

The enzymatic hydrolysis of lignocellulose is often hindered by the glucose-mediated inhibition of β-glucosidases, a major bottleneck in industrial cellulose degradation. Identifying novel glucose-tolerant β-glucosidases is essential for enhancing saccharification efficiency. In this study, we cloned and heterologously expressed a novel β-glucosidase, RpBgl8, from the termite Reticulitermes perilucifugus in Escherichia coli. Sequence and structural analyses classified RpBgl8 as a glycoside hydrolase family 1 enzyme. The purified enzyme exhibited optimal activity at 45 °C and pH 7.0, with broad

Selective isolation of agents of chromoblastomycosis from insect-associated environmental sources.

Chromoblastomycosis is a neglected disease characterized by cutaneous, subcutaneous or disseminated lesions. It is considered an occupational infectious disease that affects mostly rural workers exposed to contaminated soil and vegetal matter. Lesions mostly arise after a traumatic inoculation of herpotrichiellaceous fungi from the Chaetothyriales order. However, the environmental niche of the agents of the disease remains obscure. Its association with insects has been predicted in a few studies. Therefore, the present work aimed to analyze if social insects, specifically ants, bees, and termi

Application of termite nest for adsorption of Cr(VI).

This work proposes the use of tree termite nest as an adsorbent for the reduction/removal of Cr(VI) present in aqueous solution. In laboratory experiments, adsorption of Cr(VI) was sensitive to pH in the range investigated (2-5), with maximum adsorption capacity achieved at pH 2 (3.70 ± 0.04 mg g(-1), representing 93.2% removal of Cr). The termite nest was characterized by off-line pyrolysis GC/MS (py-GC/MS), infrared spectroscopy (FTIR), and electron paramagnetic resonance spectroscopy (EPR). Pyrolysis of the adsorbent produced a complex mixture of aromatic compounds, including the guaiacyl a

Hexamerin-based regulation of juvenile hormone-dependent gene expression underlies phenotypic plasticity in a social insect.

Worker termites of the genus Reticulitermes are temporally-arrested juvenile forms that can terminally differentiate into adultsoldier- or reproductive-caste phenotypes. Soldier-caste differentiation is a developmental transition that is induced by high juvenile hormone (JH) titers. Recently, a status quo hexamerin mechanism was identified, which reduces JH efficacy and maximizes colony fitness via the maintenance of high worker-caste proportions. Our goal in these studies was to investigate more thoroughly the influences of the hexamerins on JH-dependent gene expression in termite workers. Ou

[Ultrastructural basis of interactions between prokaryotes and eukaryotes in different symbiotic models].

This paper reviews the Author's contribution to the knowledge of the ultrastructural basis of the prokaryote-eukaryote interactions in different models assessed by an ultrastructural approach. In agreement with the hypothesis of the origin of eukaryotic cells, which are chimeras of several prokaryotes with different morpho-functional specializations, symbiosis had major consequence for evolution of life. In Arthropods, one of the most successful lifestyles, the presence of endosymbiotic prokaryotes, plays an important role in their metabolism. In some cases, genome integration has occurred in
