Burn injury sensitizes rat Kupffer cells via mechanisms dependent on gut-derived endotoxin.

BACKGROUND: Bacterial translocation occurs after thermal injury in association with intestinal barrier loss. Recently, we found that sensitization of Kupffer cells involved gut-derived endotoxin; therefore, the purpose of this work was to study the mechanisms of sensitization of Kupffer cells in burn injury. METHODS: Rats received a 30% body surface area full-thickness steam burn 24 h before experiments. Serum alanine aminotransferase (ALT) was measured to assess liver damage, and plasma endotoxin in the portal vein were measured. Kupffer cells were isolated 24 h after the burn. Intracellular

Kupffer cells inhibition prevents hepatic lipid peroxidation and damage induced by carbon tetrachloride.

The aim of this work was to determine if the action mechanism of gadolinium on CCl(4)-induced liver damage is by preventing lipid peroxidation (that may be induced by Kupffer cells) and its effects on liver carbohydrate metabolism. Four groups of rats were treated with CCl(4), CCl(4)+GdCl(3), GdCl(3), and vehicles. CCl(4) was given orally (0.4 g 100 g(-1) body wt.) and GdCl(3) (0.20 g 100 g(-1) body wt.) was administered i.p. All the animals were killed 24 h after treatment with CCl(4) or vehicle. Glycogen and lipid peroxidation were measured in liver. Alkaline phosphatase, gamma-glutamyl tran

Secretion of prostaglandins elicited by lipopolysaccharide and ethanol in cultured rat Kupffer cells.

Prostaglandins (PGs) released by cultured rat Kupffer cells in response to stimulation with lipopolysaccharide (LPS) or ethanol were extracted from culture media, separated by HPLC and measured by radioimmunoassay. LPS (0.5-5 micrograms/ml) enhanced, after a 3-4 hrs lag period, the production of PGE2 (7-10 fold by 24 hrs), thromboxane B2 (2-3 fold) and PGD2. PG 6-keto-F1 alpha, PGF2 alpha (20-50% each). This effect was not inhibited by 30 microM aspirin but was reduced by dexamethasone. Ethanol (25-85 mM) gradually increased the release of PGE2 (40-90% by 24 hrs) and other PGs (10-30%), with 3

Oxygen radical production by peritoneal macrophages and Kupffer cells elicited with Lactobacillus casei.

BALB/c mice were injected intraperitoneally (i.p.) or intravenously (i.v.) with Lactobacillus casei YIT9018 (LC 9018). The i.p. injected LC 9018 augmented oxygen radical (OR) production by peritoneal macrophages (PM) and suppressed the production of prostaglandin E2 by PM. The growth of i.p. inoculated Meth A fibrosarcoma was also inhibited by an i.p. injection of LC 9018. i.v. injection of LC 9018 stimulated OR production by fixed macrophages and inhibited the growth of Listeria monocytogenes in the liver. Furthermore, glucose-6-phosphate dehydrogenase activity in the liver was significantly

Protocol to isolate and co-culture primary mouse hepatocytes, hepatic stellate cells, and Kupffer cells under physiological conditions.

With the rising incidence of metabolic diseases, the ability to isolate and co-culture primary hepatic cells has become increasingly critical for investigating liver pathophysiology. We present a protocol to isolate and co-culture primary mouse hepatocytes, hepatic stellate cells, and Kupffer cells. We describe steps for perfusing the liver without a pump, digesting it with collagenase, and separating cells by density centrifugation. We then detail procedures for culturing hepatocytes in a defined, serum-free, plasma-like medium to mimic in vivo conditions.

Apoptotic cells attenuate fulminant hepatitis by priming Kupffer cells to produce interleukin-10 through membrane-bound TGF-β.

UNLABELLED: The liver, a unique tolerogenic organ, is regarded as the site to trap and destroy aging erythrocytes and activated T cells. However, to date, the mechanisms for why the liver is tolerogenic and whether liver Kupffer cells (KC) are critical phagocytes for apoptotic cells (AC) contributing to the liver immunosuppression remain unclear. Here we report that KC is the main phagocyte for AC in the liver. Contact of AC inhibits proinflammatory cytokine but enhances anti-inflammatory cytokine production of KC in response to lipopolysaccharide (LPS) stimulation. Membrane-bound transforming

Physiological role of sinusoidal endothelial cells and Kupffer cells and their implication in the pathogenesis of liver injury.

We reviewed the morphological characteristics and physiological functions of hepatic sinusoidal endothelial cells (SECs) and Kupffer cells (KCs), both of which are major components of the hepatic sinusoid, and we showed the implication of these hepatic sinusoidal lining cells in the pathophysiology of the liver, based on our experimental studies. The most outstanding feature of SECs is that they are provided with numerous fenestrae, thereby allowing direct communication between the sinusoidal lumen and the space of Disse. Physiologically, SECs play a role in filtration function, endocytic func

Fatty acid synthesis is indispensable for Kupffer cells to eliminate bacteria in ALD progression.

BACKGROUND: Dysregulated fatty acid metabolism is closely linked to the development of alcohol-associated liver disease (ALD). KCs, which are resident macrophages in the liver, play a critical role in ALD pathogenesis. However, the effect of alcohol on fatty acid metabolism in KCs remains poorly understood. The current study aims to investigate fatty acid metabolism in KCs and its potential effect on ALD development. METHODS: Wild-type C57BL/6 mice were fed a Lieber-DeCarli ethanol liquid diet for 3 days. Then, the liver injury and levels of intrahepatic bacteria were assessed. Next, we inves

Kupffer cells abrogate homing and repopulation of allogeneic hepatic progenitors in injured liver site.

BACKGROUND: Allogeneic hepatocyte transplantation is an emerging approach to treat acute liver defects. However, durable engraftment of the transplanted cells remains a daunting task, as they are actively cleared by the recipient's immune system. Therefore, a detailed understanding of the innate or adaptive immune cells-derived responses against allogeneic transplanted hepatic cells is the key to rationalize cell-based therapies. METHODS: Here, we induced an acute inflammatory regenerative niche (3-96 h) on the surface of the liver by the application of cryo-injury (CI) to systematically eval

Reactive oxygen species regulation by NCF1 governs ferroptosis susceptibility of Kupffer cells to MASH.

Impaired self-renewal of Kupffer cells (KCs) leads to inflammation in metabolic dysfunction-associated steatohepatitis (MASH). Here, we identify neutrophil cytosolic factor 1 (NCF1) as a critical regulator of iron homeostasis in KCs. NCF1 is upregulated in liver macrophages and dendritic cells in humans with metabolic dysfunction-associated steatotic liver disease and in MASH mice. Macrophage NCF1, but not dendritic cell NCF1, triggers KC iron overload, ferroptosis, and monocyte-derived macrophage infiltration, thus aggravating MASH progression. Mechanistically, elevated oxidized phospholipids

Kupffer Cells and Blood Monocytes Orchestrate the Clearance of Iron-Carbohydrate Nanoparticles from Serum.

Intravenous (IV) iron nanoparticle preparations are widely used to treat iron deficiency. The mechanism of mononuclear phagocyte system-mediated clearance of IV iron nanoparticles is unknown. The early uptake and homeostasis of iron after injection of ferric carboxymaltose (FCM) in mice was studied. An increase in serum iron was observed at 2.5 h followed by a return to baseline by 24 h. An increase in circulating monocytes was observed, particularly Ly6Chi and Ly6Clow. FCM was also associated with a time-dependent decrease in liver Kupffer cells (KCs) and increase in liver monocytes. The incr

TNFα-mediated liver destruction by Kupffer cells and Ly6Chi monocytes during Entamoeba histolytica infection.

Amebic liver abscess (ALA) is a focal destruction of liver tissue due to infection by the protozoan parasite Entamoeba histolytica (E. histolytica). Host tissue damage is attributed mainly to parasite pathogenicity factors, but massive early accumulation of mononuclear cells, including neutrophils, inflammatory monocytes and macrophages, at the site of infection raises the question of whether these cells also contribute to tissue damage. Using highly selective depletion strategies and cell-specific knockout mice, the relative contribution of innate immune cell populations to liver destruction

Kupffer cell and interleukin-12-dependent loss of natural killer T cells in hepatosteatosis.

UNLABELLED: Hepatosteatosis is associated with increased expression of tumor necrosis factor alpha (TNF-alpha) and interleukin (IL)-12, major T helper (Th) 1 cytokines, and reduced hepatic natural killer T (NKT) cell numbers. The relationship between lipid accumulation, cytokine expression, and hepatic NKT cells is not known. This study was conducted to assess the role of IL-12 in the development of hepatic steatosis and its potential impact on liver NKT cells. Male C57Bl/6 wildtype (WT) and IL-12-deficient (IL-12(-/-)) mice were fed a choline-deficient diet (CDD) for 0, 10, or 20 weeks. CDD l

CD14 expression on Kupffer cells during the course of carbon tetrachloride-mediated liver injury.

OBJECTIVE: To investigate the expression of CD14 on Kupffer cells during the course of carbon tetrachloride (CCl(4))-mediated liver injury and its role in the activation of Kupffer cells. METHODS: Rats were administered CCl(4) twice weekly for up to 8 weeks. Kupffer cells were isolated from normal and CCl(4)-treated rats by the combined 'collagenase-pronase' perfusion method, discontinuous density gradient centrifugation. On the day after isolation, the cells were incubated with RPMI-1640 containing varying doses of lipopolysaccharide (LPS) for 6 h. Supernatants were then collected for measur

Immunogenic role of Kupffer cells in a rat model of acute liver allograft rejection.

Kupffer cells (KCs) are of bone-marrow origin. After liver transplantation, recipient KCs are supposed to replace donor KCs. On the other hand, KCs are currently hypothesized to play a major immunogenic role in acute liver allograft rejection. In the present study, we investigated the immunogenic role of KCs in acute rat liver allograft rejection. For this purpose, we depleted the donor KCs using intravenous injection of liposome-encapsulated dichloromethylene diphosphonate (DMDP) in the fully allogenic ACI-to-LEW rat liver transplantation model. Kupffer cell depletion was confirmed using mono

Effector function of hepatocytes and Kupffer cells in the resolution of systemic bacterial infections.

It has been suggested that mononuclear phagocytes serve as the principal site of replication for a number of intracellular pathogens including Listeria monocytogenes. To determine the role of the tissue macrophages (Kupffer cells) in the proliferation of Listeria taken up in the liver, the hepatic cell populations were purified and the associated bacteria were quantified at periodic intervals postinfection. Here we report that the bulk of Listeria injected intravenously into nonimmune mice replicated within hepatocytes rather than Kupffer cells. Whereas a 200-fold increase in the number of hep

Exercise reduces hepatic pre-metastatic niche formation via TREM-1 signaling in kupffer cells.

BACKGROUND: Tumor-derived exosomes contribute to the formation of a hepatic pre-metastatic niche (PMN) by activating Kupffer cells (KCs) and hepatic stellate cells (HSCs), thereby promoting metastatic seeding. Whether exercise modulates exosome-driven PMN formation and subsequent liver metastasis remains unclear. METHODS: Eight-week-old male mice were randomized to sedentary (SED) and exercise training (ExT) groups. Mice were pre-educated with cancer-derived exosomes to induce a hepatic PMN and subsequently subjected to experimental liver metastasis model. Kupffer cells were isolated for tran

Targeting NLRP3 palmitoylation: Celastrol alleviates inflammasome activation induced by immunometabolism dysregulation in Kupffer cells.

Protein palmitoylation, the only reversible lipid-linked post-translational modification, acts as a critical regulatory mechanism for modulating protein function and subcellular localization. However, its specific roles and underlying mechanisms in the inflammatory response of liver macrophages (Kupffer cells) remain largely undefined. This study aimed to elucidate the precise mechanism by which palmitoylation regulates inflammation in Kupffer cells and to explore potential therapeutic interventions targeting this modification. Through metabolomic and membrane proteomic analyses, we demonstrat

Molecular Mechanism of POSTN Mediating M2 Polarization of Kupffer Cells to Promote Hepatic Fibrosis.

Background/Objectives: Liver diseases cause more than 2 million annual deaths globally, accounting for 4% of the total global mortality rate. Hepatic fibrosis (HF) acts as an indispensable pathological mediator in the progressive deterioration of chronic liver diseases. Thus, the identification of effective anti-fibrotic targets and rational development of corresponding therapeutic agents are expected to deliver profound clinical value for patients suffering from chronic liver disorders. Methods: An in vivo HF model was established to detect Kupffer cell (KC) polarization and periostin (POSTN)

Infiltrating macrophages replace Kupffer cells and play diverse roles in severe alcohol-associated hepatitis.

Patients with alcohol-associated cirrhosis (AC) may develop severe alcohol-associated hepatitis (sAH), a disease with high short-term mortality. Our previous studies demonstrated that sAH, but not AC livers, are infiltrated with a high number of self-sustaining IL-8+ neutrophils that likely drive the transition from AC to sAH. Monocyte-derived macrophages (MoMFs) also infiltrate the liver in sAH, but their roles remain largely obscure. In the present study, we characterized liver macrophages in human liver explants from sAH and AC patients. Our data revealed a marked reduction in Kupffer cells
