Post-tetanic potentiation, habituation and facilitation of synaptic potentials in reticulospinal neurones of lamprey.

1. Synaptic potentials evoked by electrical stimulation of cranial nerves were recorded in giant reticulospinal neurones (Müller cells) of lamprey. A variety of patterns of stimulation was employed to explore further the functional properties of the pathways intervening between the cranial nerve fibres and Müller cells.2. Simultaneous low intensity stimulation of two different cranial nerves produced excitatory short-latency synaptic potentials whose amplitudes summed linearly.3. Tetanic (10/sec) stimulation of a cranial nerve depressed the evoked short-latency synaptic response, but following

An invariant VLR-like receptor refines the T-like cell framework in lamprey.

Jawless vertebrates employ variable lymphocyte receptors (VLRs) to mount adaptive immunity. Although six VLRs (VLRA-F) and T/B-like lymphocytes are identified in lampreys, it remains unclear how VLR diversity is organized at the cellular level and whether individual lymphocytes may integrate multiple VLRs. Using single-cell transcriptomics, we identify a germline-encoded, mature, non-diversified VLR-like receptor (VLR-like) in lamprey Lethenteron reissneri. VLR-like retains structural features of canonical VLRs, and is a GPI-anchored receptor expressed by T-like cells. We generate a VLR-like m

A Cftr-independent, Ano1-rich seawater-adaptive ionocyte in sea lamprey gills.

All ionoregulating marine fishes examined to date utilize seawater-type ionocytes expressing the apical Cl- channel, cystic fibrosis transmembrane conductance regulator (Cftr) to secrete Cl-. We performed transcriptomic, molecular and functional studies to identify Cl- transporters in the seawater-type ionocytes of sea lamprey (Petromyzon marinus). Gill cftr expression was minimal or undetectable in larvae and post-metamorphic juveniles. We identified other Cl- transporters highly expressed in the gills and/or upregulated following metamorphosis and further investigated two candidates that sto

Genetic and Functional Characterization of Novel Brown-Like Adipocytes Around the Lamprey Brain.

During the process of vertebrate evolution, many thermogenic organs and mechanisms have appeared. Mammalian brown adipose tissue (BAT) generates heat through the uncoupling oxidative phosphorylation of mitochondria, acts as a natural defense against hypothermia and inhibits the development of obesity. Although the existence, cellular origin and molecular identity of BAT in humans have been well studied, the genetic and functional characteristics of BAT from lampreys remain unknown. Here, we identified and characterized a novel, naturally existing brown-like adipocytes at the lamprey brain peri

Insights into the evolutionary origin of the pineal color discrimination mechanism from the river lamprey.

BACKGROUND: Pineal-related organs in cyclostomes, teleosts, amphibians, and reptiles exhibit color opponency, generating antagonistic neural responses to different wavelengths of light and thereby sensory information about its "color". Our previous studies suggested that in zebrafish and iguana pineal-related organs, a single photoreceptor cell expressing both UV-sensitive parapinopsin and green-sensitive parietopsin generates color opponency in a "one-cell system." However, it remains unknown to what degree these opsins and the single cell-based mechanism in the pineal color opponency are con

Protein synthetic machinery and mRNA in regenerating tips of spinal cord axons in lamprey.

Polyribosomes, mRNA, and other elements of translational machinery have been reported in peripheral nerves and in elongating injured axons of sensory neurons in vitro, primarily in growth cones. Evidence for involvement of local protein synthesis in regenerating central nervous system (CNS) axons is less extensive. We monitored regeneration of back-labeled lamprey spinal axons after spinal cord transection and detected mRNA in axon tips by in situ hybridization and microaspiration of their axoplasm. Poly(A)+mRNA was present in the axon tips, and was more abundant in actively regenerating tips

A single sea lamprey attack causes acute anemia and mortality in lake sturgeon.

The effects of sea lamprey Petromyzon marinus parasitism on hematological variables have not been quantified for lake sturgeon Acipenser fulvescens. Our study objectives were to (1) assess changes in lake sturgeon hematology immediately after a single sea lamprey attack and after a 2-week recovery period and (2) assess changes in the histological condition of major hematopoietic organs. Lake sturgeon from four size-groups (470-570, 570-650, 650-760, and 950-1,500 mm fork length) were individually subjected to a sea lamprey attack in a series of 55 experimental trials. Survival of lake sturgeon

Developmental increases in expression of neurofilament mRNA selectively in projection neurons of the lamprey CNS.

Neurofilaments of the sea lamprey are unique in being homopolymers of a single subunit (NF-180). Digoxigenin-labeled RNA probes complementary to NF-180 were used to determine the distribution and timing of expression of neurofilament message in the brain and spinal cord of the lamprey. In the brainstem, detection of NF-180 mRNA was restricted to neurons with axons projecting to the spinal cord or the periphery. The majority of brainstem neurons, whose axons project locally, did not express NF-180 within the detection limits of this technique. NF-180-positive neurons included cells with a wide

Crystals in supporting cells of pineal organ in larval lamprey not likely to be pure guanine.

Spectrophotometry of larval pineal organs, digested in either 1 M HCl or 1 M NaOH, revealed peak absorbances at 253 nm and 268 nm, respectively. This result is not consistent with the view that the crystals found in lamprey pineals are chemically made of guanine.

Features of entrainment of spinal pattern generators for locomotor activity in the lamprey spinal cord.

The in vitro lamprey spinal cord contains a "central pattern generator" (CPG) that can generate locomotor activity with excitatory amino acids added to the bath. The motor pattern can be entrained by imposed rhythmic bending of either the caudal or rostral end of the notochord/spinal cord. In the present study, the quantitative and mechanistic features of entrainment were investigated. Increasing the amplitude of the imposed movement increased the range of frequencies over which entrainment occurred. Brief, pulsed, imposed movements could reset the locomotor rhythm. During entrainment at diffe

The morphology and histology of the endocrine pancreas of the southern hemisphere lamprey, Geotria australis gray.

The location and arrangement of the pancreatic endocrine tissue in larval and adult Geotria australis (Geotriidae) differ markedly from those exhibited by the comparable stages of Northern Hemisphere lampreys (Petromyzontidae). In larval Geotria australis, the main zones of islet proliferation are located laterally between the oesophagus and the inner edge of the two large intestinal diverticula unique to this species rather than dorsal and ventral to the oesophagus. In adult Geotria australis, the islet follicles are closely packed into a single discrete capsule which could be easily removed

Using stable isotopes and C:N ratios to examine the life-history strategies and nutritional sources of larval lampreys.

Natural abundance stable-isotope analysis (δ(13)C and δ(15)N) and C:N ratios were used to study the ammocoete phase of two common non-parasitic lamprey species (least brook lamprey Lampetra aepyptera and American brook lamprey Lethenteron appendix) in two tributaries of the Ohio River (U.S.A.). The C:N ratios suggest that each species employs different lipid accumulation strategies to support its metamorphosis and recruitment into an adult animal. Ammocoete δ(13)C values generally increased with increasing C:N values. In contrast to δ(13)C, ammocoete δ(15)N values were weakly related to the to

Reproductive and morphological traits distinguish early- and late-maturing Pacific lamprey (Entosphenus tridentatus).

Pacific lamprey, an anadromous fish native to the North Pacific Ocean, are imperiled in parts of their range. Many aspects of their biology are not fully understood, including the physiology of their spawning migration. Pacific lamprey exhibit two divergent reproductive life histories: early-maturing and late-maturing. Interior Columbia River early-maturing lamprey complete the final stages of gonadal maturation ~ 10 months after entering freshwater. From the same migrating cohort, the late-maturing lamprey wait ~ 22 months to complete maturation. This study sought to characterize the reproduc

Liver metabolomic profiles of sea lamprey (Petromyzon marinus) are influenced by sex and maturation stages.

INTRODUCTION: Sea lamprey (Petromyzon marinus) is a unique vertebrate model to examine how liver metabolomes support different reproductive functions. Juvenile sea lamprey prey on other fish species by attaching to their body and feeding on their blood and body fluids. Once reaching adulthood, they cease feeding, migrate to spawning streams and begin their final sexual maturation. During these processes, the male livers produce large quantities of bile acid pheromone precursors to be modified and released via gills, whereas the female livers synthesize vast amounts of vitellogenin (yolk lipoph

Molecular characterization of the sea lamprey retina illuminates the evolutionary origin of retinal cell types.

The lamprey, a primitive jawless vertebrate whose ancestors diverged from all other vertebrates over 500 million years ago, offers a unique window into the ancient formation of the retina. Using single-cell RNA-sequencing, we characterize retinal cell types in the lamprey and compare them to those in mouse, chicken, and zebrafish. We find six cell classes and 74 distinct cell types, many shared with other vertebrate species. The conservation of cell types indicates their emergence early in vertebrate evolution, highlighting primordial designs of retinal circuits for the rod pathway, ON-OFF dis

Restoration of the Topological Organization of the Trigeminal System Following Trigeminal Nerve Root Injury in the Lamprey.

Two issues were examined regarding the trigeminal system in larval lampreys: (1) for normal animals, double labeling was used to confirm that the trigeminal system has a topological organization; (2) following trigeminal nerve root transections, double labeling was used to test whether the topological organization of the trigeminal system is restored. First, for normal animals, Alexa 488 dextran amine applied to the medial oral hood (anterior head) labeled trigeminal motoneurons (MNs) in the ventromedial part of the trigeminal motor nuclei (nVm) and axons of trigeminal sensory neurons (SNs) in

Molecular cloning and pharmacological characterization of two novel GnRH receptors in the lamprey (Petromyzon marinus).

This paper reports the identification, expression, binding kinetics, and functional studies of two novel type III lamprey GnRH receptors (lGnRH-R-2 and lGnRH-R-3) in the sea lamprey, a basal vertebrate. These novel GnRH receptors share the structural features and amino acid motifs common to other known gnathostome GnRH receptors. The ligand specificity and activation of intracellular signaling studies showed ligands lGnRH-II and -III induced an inositol phosphate (IP) response at lGnRH-R-2 and lGnRH-R-3, whereas the ligand lGnRH-I did not stimulate an IP response. lGnRH-II was a more potent ac
