Retrolental Opacity: Atypical Manifestation of Lens-Induced Uveitis.

The crystalline lens can cause intraocular inflammation when its capsule is ruptured or allows leakage of lens proteins into the anterior chamber, a condition known as lens-induced uveitis (LIU). LIU is a rare form of uveitis and can be classified based on the integrity of the lens capsule. When the capsule is ruptured, the inflammation is called phacoantigenic or phacoanaphylactic uveitis, whereas if the capsule remains intact but leaks proteins, it is termed phacolytic uveitis. Common causes include ocular trauma, previous surgery, or spontaneous rupture in hypermature cataracts. Initial tre

Controlling unwanted sources of threshold change in disability glare studies: a prototype apparatus and procedure.

One of the main purposes of cataract surgery is to reduce the debilitating effects of light scatter by the lens. An important effect of this intraocular forward scatter is to produce a veiling retinal illuminance from a glare source. The retinal contrast of the stimulus is thus reduced, and a quantifiable "disability glare" effect may be measured. A complication is introduced by the fact that intraocular scatter arises from other sources in addition to the lens. All present tests of glare disability fail to eliminate or control for these factors, nor do they control for the light adaptation ef

Risk factors for age-related cortical, nuclear, and posterior subcapsular cataracts. The Italian-American Cataract Study Group.

A total of 1,008 cases and 469 controls, aged 45-79 years, were included in a clinic-based case-control study of age-related cataract in Parma, Italy, from 1987 to 1989. Associations were examined between specific types of cataract and physiologic, behavioral, environmental, and biochemical factors. Cases included subjects with pure cortical (n = 489), pure nuclear (n = 110), pure posterior subcapsular (n = 28), and mixed (n = 381) cataracts. With polychotomous logistic regression, an increased risk of cataract was found for females (cortical; odds ratio (OR) = 2.20) and persons with less than

Structural study of the G57W mutant of human gamma-S-crystallin, associated with congenital cataract.

PURPOSE: Human γS-crystallin (CrygS) is an important component of the human eye lens nucleus and cortex. The mutation G57W in the molecule is reported to be associated with congenital cataract in children. We compare the conformational features and aggregation properties of the mutant protein G57W with the wild-type CrygS to understand how the structural changes in the mutant are related to the mechanism of opacification. METHODS: Wild-type and mutant proteins were cloned, expressed, and purified, and their structural properties were studied in solution. Conformational features and the struct

Drug-induced lenticular opacity and accumulation of cholesterol-related substances in the lens cortex of dogs.

TP0446131, developed as an antidepressant agent, was found to cause lenticular opacity in a 13-week repeated-dose study in dogs. Histopathologically, the lenticular opacity was observed as a degeneration of the lens fibers, characterized by irregularity in the ordered arrangement of the fibers which is necessary to maintain the transparency of the lens, and was considered to manifest clinically as cataract. To evaluate the development mechanism of the lenticular opacity, the chemical constituents of the lens, which is known to be associated with the development of cataract, were examined. The

Location and severity of cortical opacities in different regions of the lens in age-related cataract.

PURPOSE: To examine the distribution of cortical opacities across the lens in the Italian-American Natural History Study of Age-Related Cataract and to study the association between an index of sunlight exposure and the location of cortical cataract within the lens. METHODS: Lens photographs of one eye of 731 persons with cortical opacities (503 with pure and 228 with mixed types of opacity) were included in the analysis. A radial grid superimposed on the photographs was used to assess presence, location, and severity of wedge-shaped cortical opacities. RESULTS: Both the prevalence and the e

Immunohistochemical localization of aldose reductase. II. Rat eye and kidney.

Aldose reductase (AR) was purified from rat seminal vesicles. Specific antibodies to this enzyme were prepared in rabbits and were used in the unlabeled antibody-enzyme (PAP) technique to localize AR in a number of tissues known to be sites of diabetic lesions. AR was localized in the following structures in the eye: lens epithelial lining and cortical lenticular fibers; corneal endothelium; the inner, nonpigmented layer of ciliary body epithelium and its extension as the posterior surface of the iris; and neuroglial (Müller) cells in the retina. Retinal capillary endothelium did not contain i

Alzheimer's disease amyloid-β pathology in the lens of the eye.

Neuropathological hallmarks of Alzheimer's disease (AD) include pathogenic accumulation of amyloid-β (Aβ) peptides and age-dependent formation of amyloid plaques in the brain. AD-associated Aβ neuropathology begins decades before onset of cognitive symptoms and slowly progresses over the course of the disease. We previously reported discovery of Aβ deposition, β-amyloidopathy, and co-localizing supranuclear cataracts (SNC) in lenses from people with AD, but not other neurodegenerative disorders or normal aging. We confirmed AD-associated Aβ molecular pathology in the lens by immunohistopatholo

The mutation V42M distorts the compact packing of the human gamma-S-crystallin molecule, resulting in congenital cataract.

BACKGROUND: Human γS-crystallin is an important component of the human eye lens nucleus and cortex. The mutation V42M in the molecule causes severe congenital cataract in children. We compare the structure of the mutant protein with that of the wild type in order to understand how structural changes in the mutant relate to the mechanism of opacification. METHODS: Both proteins were made using conventional cloning and expression procedures. Secondary and tertiary structural features of the proteins were analyzed using spectral methods. Structural stabilities of the proteins were analyzed using

(1) Human crystalline lens and vision In order to investigate the hue changes in eyes with UV-absorbing intraocular lenses (IOLs) and tinted IOLs, we simulated the changes in the chromaticity coordinates of the 16 colors of the Farnsworth dichotomous test-panel D-15 (panel D-15), considering the ratio of the spectral transmittance of the IOL and the human crystalline lens, and the results were plotted on a CIE chromaticity diagram. The chromaticity coordinates of each color for UV-absorbing IOLs shifted to close to the origin of coordinates while retaining their hue circle. However, the chroma

Adaptive regulation of amino acid transport across the cell membrane in avian and mammalian tissues.

The regulation of amino acid transport across the cell membrane by adaptive mechanisms has been studied in a variety of mesenchymal and epithelial cells and tissues of avian and mammalian origin. Changes in transport activity as a function of time under various in vitro conditions (amino acid dependence, active and inhibited protein synthesis) have been evaluated by measurements of initial entry rates with representative amino acids. Results and conclusions based on the adopted experimental approach include the following. (1) An adaptive control mechanism for the transport of neutral amino aci
