Mechanistic Insights into Ultrafast Excitation Energy Transfer in Hybrid Light-Harvesting System of BODIPY Derivatives and LH2 from Rhodobacter sphaeroides 2.4.1 Co-assembled in Lipid Bilayers.

To further explore the intrinsic mechanism of excitation energy transfer (EET) in a hybrid light-harvesting system composed of a natural antenna complex and fluorophores, the peripheral light-harvesting complex LH2 from Rhodobacter (Rba.) sphaeroides and a bistyryl-BODIPY derivative (B6) were co-assembled into phospholipid bilayers, followed by experimental and theoretical investigations. The singlet EET efficiency from B6 to B850 BChls of LH2 was determined by fluorescence excitation profile, exceeding 60%. The ultrafast dynamics study reveals that EET from optimally aligned B6 monomers to B8

In Vitro Hydrolysis of Zinc Chlorophyllide a Homologues by a BciC Enzyme.

Chlorosomes in green photosynthetic bacteria are the largest and most efficient light-harvesting antenna systems of all phototrophs. The core part of chlorosomes consists of bacteriochlorophyll c, d, or e molecules. In their biosynthetic pathway, a BciC enzyme catalyzes the removal of the C132-methoxycarbonyl group of chlorophyllide a. In this study, the in vitro enzymatic reactions of chlorophyllide a analogues, C132-methylene- and ethylene-inserted zinc complexes, were examined using a BciC protein from Chlorobaculum tepidum. As the products, their hydrolyzed free carboxylic acids were obser

Expression of xanthophyll biosynthetic genes during light-dependent chloroplast differentiation.

In higher plants, etioplast to chloroplast differentiation is characterized by dramatic ultrastructural changes of the plastid and a concomitant increase in chlorophylls and carotenoids. Whereas the formation and function of carotenes and their oxygenated derivatives, the xanthophylls, have been well studied, little is known about the regulation of the genes involved in xanthophyll biosynthesis. Here, we analyze the expression of three xanthophyll biosynthetic genes (i.e. beta-carotene hydroxylase [bhy], zeaxanthin epoxidase [zep], and violaxanthin de-epoxidase [vde]) during de-etiolation of s

Using Rayleigh-Mie scattered light and polarised multidimensional fluorescence emission in combination for protein quantification in a model clarified bioreactor harvest.

BACKGROUND: Accurate monitoring and control of bioprocesses require the measurement of critical parameters, such as product and interfering protein concentrations, particularly in the bioreactor harvest prior to during downstream purification. One promising approach is to combine polarised multidimensional fluorescence measurements of intrinsic emission with light scattering measurements. This offers a non-invasive and sensitive method for protein quantification that can be implemented using either polarised Excitation Emission Matrix (pEEM) or Total Synchronous Fluorescence (pTSFS) measuremen

Improving photosynthesis and crop productivity by accelerating recovery from photoprotection.

Crop leaves in full sunlight dissipate damaging excess absorbed light energy as heat. When sunlit leaves are shaded by clouds or other leaves, this protective dissipation continues for many minutes and reduces photosynthesis. Calculations have shown that this could cost field crops up to 20% of their potential yield. Here, we describe the bioengineering of an accelerated response to natural shading events in Nicotiana (tobacco), resulting in increased leaf carbon dioxide uptake and plant dry matter productivity by about 15% in fluctuating light. Because the photoprotective mechanism that has b

The influence of hydrogen bonds on the electronic structure of light-harvesting complexes from photosynthetic bacteria.

The influence of hydrogen bonds on the electronic structure of the light-harvesting I complex from Rhodobacter sphaeroides has been examined by site-directed mutagenesis, steady-state optical spectroscopy, and Fourier-transform resonance Raman spectroscopy. Shifts of 4-23 nm in the Q(y) absorption band were observed in seven mutants with single or double changes at Leu alpha44, Trp alpha43, and Trp beta48. Resonance Raman spectra were consistent with the loss of a hydrogen bond with the alteration of either Trp alpha43 or Trp beta48 to Phe. However, when the Trp alpha43 to Phe alteration is co

Molecular interactions of the redox-active accessory chlorophyll on the electron-donor side of photosystem II as studied by Fourier transform infrared spectroscopy.

A Fourier transform infrared (FTIR) difference spectrum upon photooxidation of the accessory chlorophyll (Chlz) of photosystem II (PS II) was obtained at 210 K with Mn-depleted PS II membranes in the presence of fericyanide and silicomolybdate. The observed Chlz+/Chlz spectrum showed two differential bands at 1747/1736 and 1714/1684 cm-1. The former was assigned to the free carbomethoxy C = 0 and the latter to the keto C = 0 that is hydrogen-bonded or in a highly polar environment. Also, the negative 1614 cm-1 band assignable to the macrocycle mode indicated 5-coordination of the central Mg. T

Characterization of Vibrational Coherence in Monomeric Bacteriochlorophyll a by Two-Dimensional Electronic Spectroscopy.

Bacteriochlorophyll a (BChla) is the most abundant pigment found in the Bacterial Reaction Center (BRC) and light-harvesting proteins of photosynthetic purple and green bacteria. Recent two-dimensional electronic spectroscopy (2DES) studies of photosynthetic pigment-protein complexes including the BRC and the Fenna-Matthews-Olson (FMO) complex have shown oscillatory signals, or coherences, whose physical origin has been hotly debated. To better understand the observations of coherence in larger photosynthetic systems, it is important to carefully characterize the spectroscopic signatures of th

nblS, a gene involved in controlling photosynthesis-related gene expression during high light and nutrient stress in Synechococcus elongatus PCC 7942.

The HliA protein of the cyanobacterium Synechococcus elongatus PCC 7942 is a small, thylakoid-associated protein that appears to play a role in photoprotection; its transcript rapidly accumulates in response to high-intensity light (HL) and the hli gene family is required for survival of cells in high light. In order to discover regulatory factors involved in HL acclimation in cyanobacteria, a screen was performed for chemically generated mutants unable to properly control expression of the hliA gene in response to HL. One such mutant was identified, and complementation analysis led to the ide

TaCAP1 Interacts with TaLHCB1s and Positively Regulates Wheat Resistance Against Stripe Rust.

Actin filaments and their associated actin-binding proteins play key roles in plant innate immune signaling. CAP1, or cyclase-associated protein 1, is an important regulatory factor of the actin cytoskeleton-associated signaling network and was hypothesized here to be involved in resistance against wheat stripe rust because TaCAP1 expression was upregulated in response to Puccinia striiformis f. sp. tritici (Pst). Downregulation of TaCAP1 expression led to decreased resistance against Pst, in contrast to increased resistance upon TaCAP1 overexpressing, as demonstrated by the changes of phenoty

Development and use of domain-specific antibodies in a characterization of the large subunits of soybean photosystem 1.

The molecular architecture of the soybean photosystem 1 reaction center complex was examined using a combination of surface labeling and immunological methodology on isolated thylakoid membranes. Synthetic peptides (12 to 14 amino acids in length) were prepared which correspond to the N-terminal regions of the 83 and 82.4 kDa subunits of photosystem 1 (the PsaA and PsaB proteins, respectively). Similarly, a synthetic peptide was prepared corresponding to the C-terminal region of the PsaB subunit. These peptides were conjugated to a carrier protein, and were used for the production of polyclona

The out of phase 1 mutant defines a role for PHYB in circadian phase control in Arabidopsis.

Arabidopsis displays circadian rhythms in stomatal aperture, stomatal conductance, and CO(2) assimilation, each of which peaks around the middle of the day. The rhythmic opening and closing of stomata confers a rhythm in sensitivity and resistance, respectively, to the toxic gas sulfur dioxide. Using this physiological assay as a basis for a mutant screen, we isolated mutants with defects in circadian timing. Here, we characterize one mutant, out of phase 1 (oop1), with the circadian phenotype of altered phase. That is, the timing of the peak (acrophase) of multiple circadian rhythms (leaf mov

Structural and functional changes in photosynthetic apparatus of wheat under exposure to sulfur dioxide fumes.

Changes in photosynthesis of wheat (pigment content and chlorophyll fluorescence) have been measured to study effect of gaseous sulfur dioxide on wheat cenosis. Several biochemical and biophysical techniques have been estimated to evaluate their ability for early detection of destructive effect of SO2. A mathematical model of toxic gas absorption in a closed system with plants has been proposed.

Chlorophyll-deficient mutants of Chlamydomonas reinhardtii that accumulate magnesium protoporphyrin IX.

Two Chlamydomonas reinhardtii mutants defective in CHLM encoding Mg-protoporphyrin IX methyltransferase (MgPMT) were identified. The mutants, one with a missense mutation (chlM-1) and a second mutant with a splicing defect (chlM-2), do not accumulate chlorophyll, are yellow in the dark and dim light, and their growth is inhibited at higher light intensities. They accumulate Mg-protoporphyrin IX (MgProto), the substrate of MgPMT and this may be the cause for their light sensitivity. In the dark, both mutants showed a drastic reduction in the amounts of core proteins of photosystems I and II and

Coexistence of plant and algal energy dissipation mechanisms in the moss Physcomitrella patens.

Although light is the source of energy for photosynthetic organisms, it causes oxidative stress when in excess. Plants and algae prevent reactive oxygen species (ROS) formation by activation of nonphotochemical quenching (NPQ), which dissipates excess excitation energy as heat. Although NPQ is found in both algae and plants, these organisms rely on two different proteins for its activation, Light harvesting complex stress-related (LHCSR) and Photosystem II subunit S (PSBS). In the moss Physcomitrella patens, both proteins are present and active. Several P. patens lines depleted in or over-expr

[Effects of magnesium deficiency on senescence of Dimocarpus longana leaves].

180 days old Dimocarpus longana seedlings were grown in nutrition solution with three magnesium concentrations as 4 mmol.L-1 Mg2+ (sufficient supply), 0.4 mmol.L-1 Mg2+ (deficient supply), and Mg-free (without supply). Leaves were sampled 120 and 150 days later after treatment. With an increase in severity of Mg-deficient, the chlorophyll content, PS II activities, photosynthetic rate, protein, nucleic acid (DNA, RNA), and ZRs contents declined progressively. Whereas the content of H2O2, the rate of O2-. generation as well as the content of malondialdehyde (MDA) increased. The results suggeste

Ameliorative role of nitric oxide on H2O2 toxicity to a chlorophycean alga Scenedesmus obliquus.

A concentration-dependent toxicity of hydrogen peroxide (H(2)O(2)) was observed on growth yield, chlorophyll a content and chlorophyll fluorescence characteristics of the green microalga Scenedesmus obliquus under laboratory batch culture conditions. The addition of sodium nitroprusside, a nitric oxide (NO) donor, in combination with H(2)O(2) prevented chlorophyll losses, and the inhibition level of growth yield, maximum quantum yield of photosystem II (PSII) and the light-adapted quantum yield of PSII were significantly reduced. Anticandidal Efficacy of Oral Probiont Limosilactobacillus fermentum Against Dental Caries Pathogens in Children, Tamil Nadu, India.

Functional proteomics of membrane proteins is an important tool for the understanding of protein networks in biological membranes but structural studies on this part of the proteome are limited. In this study we undertook such an approach to analyse photosynthetic thylakoid membranes isolated from wild-type and mutant strains of Chlamydomonas reinhardtii. Thylakoid membrane proteins were separated by high-resolution two-dimensional gel electrophoresis (2-DE) and analysed by immuno-blotting and mass spectrometry for the presence of membrane-spanning proteins. Our data show that light-harvesting

Light-driven dinitrogen reduction catalyzed by a CdS:nitrogenase MoFe protein biohybrid.

The splitting of dinitrogen (N2) and reduction to ammonia (NH3) is a kinetically complex and energetically challenging multistep reaction. In the Haber-Bosch process, N2 reduction is accomplished at high temperature and pressure, whereas N2 fixation by the enzyme nitrogenase occurs under ambient conditions using chemical energy from adenosine 5'-triphosphate (ATP) hydrolysis. We show that cadmium sulfide (CdS) nanocrystals can be used to photosensitize the nitrogenase molybdenum-iron (MoFe) protein, where light harvesting replaces ATP hydrolysis to drive the enzymatic reduction of N2 into NH3

Bio serves nano: biological light-harvesting complex as energy donor for semiconductor quantum dots.

Light-harvesting complex (LHCII) of the photosynthetic apparatus in plants is attached to type-II core-shell CdTe/CdSe/ZnS nanocrystals (quantum dots, QD) exhibiting an absorption band at 710 nm and carrying a dihydrolipoic acid coating for water solubility. LHCII stays functional upon binding to the QD surface and enhances the light utilization of the QDs significantly, similar to its light-harvesting function in photosynthesis. Electronic excitation energy transfer of about 50% efficiency is shown by donor (LHCII) fluorescence quenching as well as sensitized acceptor (QD) emission and corrob
