Inactivation of Sonic Hedgehog Signaling and Polydactyly in Limbs of Hereditary Multiple Malformation, a Novel Type of Talpid Mutant.

Hereditary Multiple Malformation (HMM) is a naturally occurring, autosomal recessive, homozygous lethal mutation found in Japanese quail. Homozygote embryos (hmm-/-) show polydactyly similar to talpid2 and talpid3 mutants. Here we characterize the molecular profile of the hmm-/- limb bud and identify the cellular mechanisms that cause its polydactyly. The hmm-/- limb bud shows a severe lack of sonic hedgehog (SHH) signaling, and the autopod has 4 to 11 unidentifiable digits with syn-, poly-, and brachydactyly. The Zone of Polarizing Activity (ZPA) of the hmm-/- limb bud does not show polarizin

Suppressor of Fused Is Required for Determining Digit Number and Identity via Gli3/Fgfs/Gremlin.

The anterior-posterior patterning of the vertebrate limb bud requires closely coordinated signaling interactions, including Sonic Hedgehog (Shh)-mediated counteraction of the Gli3 transcription factor in the distal and posterior mesenchyme of the limb bud. Suppressor of Fused (Sufu), an intracellular negative regulator of Shh signaling via Gli2 and Gli3, is implicated in early development of the mouse limb bud. However, how Sufu is involved in the genetic regulation of limb bud patterning still remains elusive. In this study, we show that the conditional deletion of Sufu in the mesenchyme of t

A duplicated zone of polarizing activity in polydactylous mouse mutants.

The positional signaling along the anteroposterior axis of the developing vertebrate limb is provided by the zone of polarizing activity (ZPA) located at the posterior margin. Recently, it was established that the Sonic hedgehog (Shh) mediates ZPA activity. Here we report that a new mouse mutant, Recombination induced mutant 4 (Rim4), and two old mutants, Hemimelic extra toes (Hx) and Extra toes (Xt), exhibit mirror-image duplications of the skeletal pattern of the digits. In situ hybridization of the embryos of these mutants revealed ectopic expression of Shh and fibroblast growth factor-4 (F

DLX5, FGF8 and the Pin1 isomerase control ΔNp63α protein stability during limb development: a regulatory loop at the basis of the SHFM and EEC congenital malformations.

Ectrodactyly, or Split-Hand/Foot Malformation (SHFM), is a congenital condition characterized by the loss of central rays of hands and feet. The p63 and the DLX5;DLX6 transcription factors, expressed in the embryonic limb buds and ectoderm, are disease genes for these conditions. Mutations of p63 also cause the ectodermal dysplasia-ectrodactyly-cleft lip/palate (EEC) syndrome, comprising SHFM. Ectrodactyly is linked to defects of the apical ectodermal ridge (AER) of the developing limb buds. FGF8 is the key signaling molecule in this process, able to direct proximo-distal growth and patterning

KIF3B promotes a PI3K signaling gradient causing changes in a Shh protein gradient and suppressing polydactyly in mice.

Digit determination in limb buds is driven by a posteriorizing Sonic hedgehog (Shh) protein gradient; however, the mechanism regulating this is unclear. Here, we propose a diffusion-and-trapping hypothesis for Shh gradient formation based on data from the preaxial polydactyly phenotype of KIF3B motor hypomorphic mice. In the limb buds of these mice, a distal-to-proximal gradient of fibroblast growth factor (FGF) and phosphatidylinositol 3-kinase (PI3K) signaling and a posterior-to-anterior gradient of Shh were disorganized. This phenotype was reproduced by transplanting FGF8b-soaked beads. At

Skeleton pattern and joint formation in chorioallantoic grafts containing the distal parts of the chick wing bud.

Skeleton pattern formation was examined in chick wing bud grafts using the chorioallantoic grafting method. The distal parts of the wing bud were excised from the donor wing and transplanted onto the chorioallantoic membrane (the experimental groups). Transplants with intact limb bud material served as the control group. The skeleton pattern formation in the grafts depended on the amount of transplanted material and donor's limb bud stage. The younger the donor's stage and the bigger the piece of the transplanted material the more proximal parts grafts had, more retarded growth and abnormal sk

MicroRNA-34a is dispensable for p53 function as teratogenesis inducer.

The tumor suppressor protein p53 is a powerful regulator of the embryo's susceptibility to diverse teratogenic stimuli, functioning both as a teratogenesis inducer and suppressor. However, the targets that p53 engages to fulfill its functions remain largely undefined. We asked whether the microRNA (miRNA) miR-34 family, identified as one of the main targets of p53, mediates its function as a teratogenesis inducer. For this, pregnant ICR-, p53- and miR-34a-deficient mice, as well as rats, were exposed to 5-aza-2'-deoxycytidine (5-aza), a teratogen inducing limb reduction anomalies (LRA) of the

Evidence for genetic control of Sonic hedgehog by Gli3 in mouse limb development.

Sonic hedgehog (Shh) expression in the developing limb is associated with the zone of polarising activity (ZPA), and both are restricted to the posterior part of the limb bud. We show that the expression patterns of Shh and Gli3, a member of the Gli-family believed to function in transcriptional control, appear to be mutually exclusive in limb buds of mouse embryos. In the polydactyly mouse mutant extra toes (Xt), possessing a null mutation of Gli3, Shh is additionally expressed in the anterior region of the limb bud. The transcript of Ptc, the putative receptor for Shh protein, can be detecte

Polydactyly and ectopic ZPA formation in Alx-4 mutant mice.

Correct development of the limb is dependent on coordination between three distinct signaling centers. Recently, fibroblast growth factor-4 has been identified as a crucial determinant of AER function, which directs limb bud outgrowth, and Sonic hedgehog has been identified as a signaling molecule that mediates ZPA function, which specifies anterior-posterior patterning in the developing limb bud. In addition, Shh and FGF-4 reciprocally reinforce each other's expression via a positive feedback loop, providing a molecular basis for the coordination of limb bud outgrowth and anterior-posterior p

Analysis of a new allele of limb deformity (ld) reveals tissue- and age-specific transcriptional effects of the Ld Global Control Region.

The mouse limb deformity (ld) phenotype is characterized by developmental failure of distal limb structures often associated with renal anomalies. It is caused by loss of the BMP-antagonist Gremlin in the limb buds, either through mutation of Grem1, or by loss of a transcriptional global control region (GCR) located in the neighboring Fmn1 gene. In this report, we describe a new allele of ld due to complete deletion of Fmn1, including its GCR. Unlike many other ld strains, these mice are viable and fertile as homozygotes. As expected, this genomic deletion causes loss of Gremlin in the develop

The role of cell death in limb development of rats manifesting Lx allele on different genetic backgrounds.

Control of vertebrate digital pattern is a phylogenetically old mechanism. Animal strains with abnormal digital counts are a useful model system to study tissue, cell and molecular factors involved in limb patterning. The aim of this study was to investigate rat limb morphogenesis on gestation days 13 to 16 in normodactylous, polydactylous and oligodactylous fetuses where the deviation from the normal pentadactylous phenotype is caused by interaction of mutant Lx allele with different genetic backgrounds. General development was assessed by measurements of crown-rump length, and limb morphogen

Teratogenic effects of cholinergic insecticides in chick embryos--IV. The role of tryptophan in protecting against limb deformities.

The mechanism by which organophosphate (OP) insecticides cause micromelia in embryonic chick limbs was examined using a tissue culture approach. Limb bud cells in micromass culture were assayed for their proliferative and chondrogenic activities, [3H]thymidine and 35SO4 = incorporation, respectively, into the trichloroacetic acid-insoluble constituents of the cell masses and/or the accumulation of 35S-labeled soluble macromolecular products in the culture medium. There was no obvious correlation between either the teratogenicity or toxicity of the insecticide in ovo and the inhibition of proli

Role of the brachial somites in the development of the appendicular musculature in rat embryos.

DiI, a fluorescent lipophilic dye, was micro-injected into the brachial somites of 10.5 day rat embryos to determine whether these somites can contribute cells to the development of the fore-limb bud. The injected embryos were cultured and harvested at the 20-25-somite stage. The dye did not interfere with somitogenesis because, at the injection site, the DiI-labelled somites were able to differentiate into dermomyotome and sclerotome. We have analyzed cryo-sections of 20-21-somite stage embryos and were unable detect the presence of DiI-labelled cells in the fore-limb buds. However, at the 22

Molecular analysis of Aldh1a2 conditional inactivation in developing limbs reveals mechanisms of retinoic-acid mediated interdigital apoptosis.

INTRODUCTION: Retinoic acid (RA) plays a plethora of functions during embryonic development as well as post-natal life. In particular, RA function during limb development is associated with interdigital cell death. However, the molecular mechanism(s) through which RA signaling contributes to interdigital cell death remains poorly understood. METHODS: To gain insights into the mechanism(s) involved, we used a conditional approach to inactivate the RA catalyzing enzyme Aldh1a2 thereby abrogating RA signaling in the distal limb. RESULTS AND DISCUSSION: Our data first show that Aldh1a2 mutant li

[Regulator of expression of the brachypodism-H gene in mice].

Extracts were prepared from the whole limb buds of the 11 day-old mouse embryos and from the postaxial or preaxial parts of the 12 day-old embryo limb buds. Effects of these extracts on the growth of the 13 day-old bpH/bpH embryo tibia rudiments were studied in vitro. It was shown that the limb buds of the 11- and 12 day-old mouse embryos contain a factor regulating bpH gene expression in differentiating cartilage cells. This factor is present in the postaxial part of the limb bud and is absent in its preaxial one. When the extract from the limb bud postaxial parts of the 12 day-old +/+ embryo

Twist plays an essential role in FGF and SHH signal transduction during mouse limb development.

Loss of Twist gene function arrests the growth of the limb bud shortly after its formation. In the Twist(-/-) forelimb bud, Fgf10 expression is reduced, Fgf4 is not expressed, and the domain of Fgf8 and Fgfr2 expression is altered. This is accompanied by disruption of the expression of genes (Shh, Gli1, Gli2, Gli3, and Ptch) associated with SHH signalling in the limb bud mesenchyme, the down-regulation of Bmp4 in the apical ectoderm, the absence of Alx3, Alx4, Pax1, and Pax3 activity in the mesenchyme, and a reduced potency of the limb bud tissues to differentiate into osteogenic and myogenic

Regionalized Twist1 activity in the forelimb bud drives the morphogenesis of the proximal and preaxial skeleton.

Development of the mouse forelimb bud depends on normal Twist1 activity. Global loss of Twist1 function before limb bud formation stops limb development and loss of Twist1 throughout the mesenchyme after limb bud initiation leads to polydactyly, the ulnarization or loss of the radius and malformations and reductions of the shoulder girdle. Here we show that conditional deletion of Twist1 by Mesp1-Cre in the mesoderm that migrates into the anterior-proximal part of the forelimb bud results in the development of supernumerary digits and carpals, the acquisition of ulna-like characteristics by th

The chick oligozeugodactyly (ozd) mutant lacks sonic hedgehog function in the limb.

We have analyzed a new limb mutant in the chicken that we name oligozeugodactyly (ozd). The limbs of this mutant have a longitudinal postaxial defect, lacking the posterior element in the zeugopod (ulna/fibula) and all digits except digit 1 in the leg. Classical recombination experiments show that the limb mesoderm is the defective tissue layer in ozd limb buds. Molecular analysis revealed that the ozd limbs develop in the absence of Shh expression, while all other organs express Shh and develop normally. Neither Ptc1 nor Gli1 are detectable in mutant limb buds. However, Bmp2 and dHAND are exp
