Topical delivery of cultivated limbal stem cells via porous photocurable hydrogel: A noninvasive, convenient, and efficient stem cells delivery strategy for limbal stem cell deficiency.

Topical administration of stem cells is one of the simplest approaches to stem cell-based therapy for limbal stem cell deficiency (LSCD). Without timely supplementation of deficient stem cells, LSCD may worsen, carrying a high risk of blindness in severe cases. However, challenges such as stem cell loss, poor cell attachment, short retention time, and phenotypic transformation of stem cells limit the efficiency of direct stem cell delivery. To address these problems, this study conducted in vitro and in vivo screenings of photocurable hydrogels and identified a porous Gelatin Methacryloyl (PG)

Therapeutic effects of adipose mesenchymal stem cell-derived exosomes on limbal stem cells deficiency in cats.

Corneal transparency is maintained by the limbus and its stem cells, and damage or deficiency in this region leads to conjunctivalization and subsequent vision impairment. This study aims to evaluate the therapeutic effects of feline adipose mesenchymal stem cell-derived exosomes (fAD-MSCExo) in the treatment of limbal stem cell deficiency in cats. For this purpose, 30 animals presenting with severe conjunctivalization were included in the study. Cats were randomly assigned to two groups: Group 1 (n = 15), serving as the control group and receiving steroid treatment, Group 2 (n = 15), receivin

A novel case of limbal stem cell deficiency in a patient with acrodermatitis enteropathica.

An 18-year-old male diagnosed with acrodermatitis enteropathica (AE) since early childhood presented with worsening of dermatitis along with photophobia and watering in both eyes. Systemic evaluation by dermatology and gastroenterology specialists confirmed a diagnosis of acute exacerbation of AE, and oral zinc supplements were initiated. A best-corrected visual acuity of 20/20 was documented in both eyes. Slit-lamp examination revealed bilateral subepithelial corneal opacities in a radial fan-like pattern extending from the superior limbus toward the center. A whorled appearance of fluorescei

Treatment of vernal keratoconjunctivitis simulating total limbal stem cell deficiency: a case report.

BACKGROUND: Severe vernal keratoconjunctivitis (VKC) can lead to total limbal stem cell deficiency (LSCD), a condition characterized by conjunctivalization of the cornea and neovascularization that often results in severe vision loss and typically requires surgical intervention. This report presents a novel case where a condition clinically simulating total LSCD, induced by chronic inflammation from VKC, was reversed using only anti-inflammatory therapy. CASE PRESENTATION: A 15-year-old male with a 7-year history of inadequately managed VKC presented with typical signs of clinically apparent t

Stability of limbal stem cell deficiency after mechanical and thermal injuries in mice.

We studied the reproducibility and stability of limbal stem cell deficiency (LSCD) in mice following controlled injuries to the corneal and limbal epithelia. In one method, corneal and limbal epithelia were entirely removed with a 0.5 mm metal burr. In the other, limbus to limbus epithelial removal with the burr was followed by thermal injury to the limbus. These two methods were compared with a previously published one. Unwounded corneas were used as control. The corneas were examined monthly for three months by slit lamp with fluorescein staining. Immunofluorescence staining for cytokeratin

Phenotypic alteration of limbal niche-associated limbal epithelial stem cell deficiency by ultraviolet-B exposure-induced phototoxicity in mice.

Good vision requires a healthy cornea, and a healthy cornea needs healthy stem cells. Limbal epithelial stem cells (LESCs) are a traditional source of corneal epithelial cells and are recruited for the continuous production of epithelium without seizing throughout an animal's life, which maintains corneal transparency. Like the maintenance of other adult somatic stem cells, the maintenance of LESCs depends on the specific microenvironmental niche in which they reside. The purpose of this study was to determine the microenvironmental damage associated with LESCs fate due to ultraviolet (UV)-B e
