Autoantibodies to glutamate receptor GluRepsilon2 in a patient with limbic encephalitis associated with relapsing polychondritis.

Limbic encephalitis is a rare central nervous system (CNS) manifestation of relapsing polychondritis (RP). Vasculitis is assumed to be the cause of CNS involvement in RP. Several studies, however, have described CNS involvement in RP with no evidence of vasculitis but with a more nonspecific inflammatory picture. We report a patient with limbic encephalitis associated with RP who presented with anti-glutamate receptor (GluR) epsilon2 (NR2B) autoantibodies in his cerebrospinal fluid and sera. Brain MRI showed a high signal intensity lesion in the medial temporal lobe and progressive atrophy wit

18 F-FDG PET and MRI Evaluation of Paraneoplastic Limbic Encephalitis.

Autoimmune limbic encephalitis (paraneoplastic or nonparaneoplastic) is a rare condition involving autoantibodies against intracellular or cell-surface proteins of neurons. Patients typically present with acute and progressive neuropsychiatric symptoms. Although the criterion standard for diagnosis remains detection of autoantibodies in cerebrospinal fluid, there are characteristic imaging features that can aid in diagnosis, notably abnormalities in the bilateral medial temporal lobes on imaging, particularly with nuclear imaging. Here, we present 18 F-FDG PET findings of paraneoplastic limbic

Molecular, Structural, and Functional Neuroimaging in Major Depression.

Structural and functional imaging have been applied to understand the pathophysiological alterations of brain in major depressive disorder (MDD) for several decades. The interaction between gene and brain in the pathophysiology of MDD can be explored by molecular imaging. Structural imaging studies support the crucial role of fronto-limbic model. Gray matter alterations may include frontal cortex, anterior cingulate cortex, amygdala, hippocampus, and parietal lobe. White matter alterations may include superior longitudinal fasciculus, uncinate fasculus, and cingulum. Structural connectome comb

Age, BMI and diabetes as independent predictors of brain hypoperfusion.

BACKGROUND: Cerebral blood flow abnormalities are supposed to be potential risk factors for developing cognitive dysfunction in the general population. Aging, obesity and type 2 diabetes mellitus are associated with perfusion abnormalities leading to cognitive impairment, neurodegeneration and future development of dementia. In our study, we aimed at identifying independent factors that contribute to the appearance of regional brain perfusion changes besides those that are already known. MATERIAL AND METHODS: Forty-three type 2 diabetic and twenty-six obese patients were enrolled. After the i

Architecture and connectivity of the human angular gyrus and of its homolog region in the macaque brain.

The angular gyrus roughly corresponds to Brodmann's area 39, which is a multimodal association brain region located in the posterior apex of the human inferior parietal lobe, at its interface with the temporal and occipital lobes. It encompasses two cyto- and receptor architectonically distinct areas: caudal PGp and rostral PGa. The macaque brain does not present an angular gyrus in the strict sense, and the establishment of homologies was further hindered by the fact that Brodmann defined a single cytoarchitectonic area covering the entire guenon inferior parietal lobule in the monkey brain,

Aberrant brain network topology in the frontoparietal-limbic circuit in bipolar disorder: a graph-theory study.

Characterizing the properties of brain networks across mood states seen in bipolar disorder (BP) can provide a deeper insight into the mechanisms involved in this type of affective disorder. In this study, graph theoretical methods were used to examine global, modular and nodal brain network topology in the resting state using functional magnetic resonance imaging data acquired from 95 participants, including those with bipolar depression (BPD; n = 30) and bipolar mania (BPM; n = 39) and healthy control (HC) subjects (n = 26). The threshold value of the individual subjects' connectivity matrix

Inter-network functional connectivity increases by beta-amyloid and may facilitate the early stage of tau accumulation.

Alzheimer's disease (AD) is pathologically marked by tau tangles and beta-amyloid (Aβ) plaques. It has been hypothesized that Aβ facilitates spread of tau outside of the medial temporal lobe (MTL), but exact mechanism of this facilitation remains unclear. We aimed to test the hypothesis that abnormal Aβ induces an increase in inter-network functional connectivity, which in turn induces early-stage tau elevation in limbic network. Our study used 18F-Florbetaben Aβ positron emission tomography (PET), 18F-MK6240 tau-PET, and resting-state functional magnetic resonance imaging (rs-fMRI) from 489 h

Anti-glutamic acid decarboxylase antibody paraneoplastic limbic encephalitis associated with Acute Myeloid Leukemia.

A 75-year-old woman developed a frontal lobe disorder a few days after the diagnostic of an Acute Myeloid Leukemia secondary to a myelodysplastic syndrome. The patterns on the cerebral Magnetic Resonance Imaging and positron emission tomography and the find of antiglutamic acid decarboxylase antibody on cerebral spinal fluid were in favor of a paraneoplastic limbic encephalitis. In cerebral spinal fluid, there were no micro-organisms nor leukemic cells. We found no sign of cancer on full body computerized tomography-scan, on full-body PET and on rectosigmoidoscopy. The patient was treated by c

Inter-subject variability in brain connectivity predicts nicotine dependence severity and differentiates smokers via machine learning.

BACKGROUND: Tobacco use disorder (TUD) is a major public health issue with significant individual differences. A deeper understanding of its neurobiology and reliable biomarkers is needed. This study investigated whether inter-subject variability in resting-state brain functional connectivity (IVFC) could serve as such a marker for TUD. METHODS: Resting-state fMRI data from 123 male TUD patients and 123 healthy controls (HCs) were collected and analyzed. IVFC was computed within seven major brain lobes. Five machine learning models (random forest, gradient boosting, extra trees, multi-layer p

White matter abnormalities in major depression: a tract-based spatial statistics and rumination study.

Increasing evidence indicates that major depressive disorder (MDD) is usually accompanied by altered white matter in the prefrontal cortex, the parietal lobe and the limbic system. As a behavioral abnormity of MDD, rumination has been believed to be a substantial indicator of the mental state of the depressive state. So far, however, no report that we are aware of has evaluated the relationship between white matter alterations and the ruminative state. In this study, we first explored the altered white matter using a tract-based spatial statistics (TBSS) method based on diffusion tensor imagin

Promising Neuroimaging Biomarkers in Depression.

The neuroimaging has been applied in the study of pathophysiology in major depressive disorder (MDD). In this review article, several kinds of methodologies of neuroimaging would be discussed to summarize the promising biomarkers in MDD. For the magnetic resonance imaging (MRI) and magnetoencephalography field, the literature review showed the potentially promising roles of frontal lobes, such as anterior cingulate cortex (ACC), dorsolateral prefrontal cortex (DLPFC) and orbitofrontal cortex (OFC). In addition, the limbic regions, such as hippocampus and amygdala, might be the potentially prom

Acute amnesia and seizures in a young female.

Limbic encephalitis is a condition characterised by an acute or sub-acute onset of memory disorder, associated with seizures and psychiatric manifestations. Investigations such as brain MRI usually reveal a high intensity signal in the medial temporal lobe and cerebrospinal fluid analysis shows mild pleocytosis and oligoclonal bands. It may occur in association with cancer, infection, or as an isolated clinical condition, often accompanying autoimmune disorders. Immune-mediated limbic encephalitis is now subclassified according to the presence and type of autoantibodies, which has significant

Adenylate kinase 5 (AK5) autoimmune encephalitis: Clinical presentations and outcomes in three new patients.

Adenylate kinase 5 (AK5) antibodies are biomarkers of a poorly responsive to immunotherapy, non-paraneoplastic, autoimmune limbic encephalitis. We detected 6 patients (all female, median age: 72 years [49-80]) with identical CSF antibody staining by indirect immunofluorescence on mouse tissues. We identified AK5 as the antigen and confirmed with standardized assays. Three patients with clinical information had limbic encephalitis, inflammatory CSF and mesiotemporal lobe T2 hyperintensities that evolved to atrophy on brain MRI. One patient had burning smell sensation with no evidence of seizure
