Lipoxygenase-catalyzed oxidation of chlorpromazine by hydrogen peroxide at acidic pH.

The hydroperoxidase activity of soybean lipoxygenase, a non-heme protein, oxidizes chlorpromazine using H2O2 at acidic pHs ranging from 3.0 to 4.0. The enzyme is assayed at pH 3.5, at which the half-life is 2 h (lower pHs cause higher inactivation rates). This oxidation is enzymatical since boiled enzyme or even iron ions both with H2O2 failed to produce any increase in absorbance. In addition, the concentration of CPZ radical cation formed and the concomitant enzyme activity directly depends on the enzyme concentration up to 0.23 microM. The Vmax value is 125 mumol/min per mg protein and the

[Distribution of 5-, 12-lipoxygenases in porcine ciliary epithelial cells].

Nonpigmented epithelial cells (NPE) and pigmented epithelial cells (PE) were separated from porcine ciliary processes without protease treatment. The relative capabilities of NPE and PE to synthesize 5-hydroxyeicosatetraenoic acid (5-HETE), 12-HETE, 5,12-diHETE from 1- (14C) arachidonic acid were determined by high pressure liquid chromatography. The capability of NPE to synthesize 5-lipoxygenase and 12-lipoxygenase products was, respectively, 5-fold and 2,5-fold higher than that of PE. The formation of 5-HETE, 12-HETE and 5,12-diHETE was inhibited almost completely by 50 microM nordihydroguai

Prevention of ocular inflammation by matrine, prednisolone, and cyclooxygenase and lipoxygenase inhibitors.

Lens protein-induced ocular inflammation in rabbits was used to study the action mechanism of some anti-inflammatory agents. Indomethacin, a cyclooxygenase inhibitor, markedly reduced PGE2 and PGF2 alpha in the iris and ciliary body at 2 h but PGE2 only at 4 h. REV 5901, a lipoxygenase inhibitor, only significantly reduced PGE2 levels in the ciliary body at 4 h. PGF2 alpha levels were not affected by REV 5901. When indomethacin and REV 5901 were combined, both PGE2 and PGF2 alpha were suppressed at 2 and 4 h in both iris and ciliary body. Neither matrine nor prednisolone produced significant e

Role of lipoxygenase metabolism in platelet function: effect of aspirin and salicylate.

Aspirin inhibits thromboxane A2 (TxA2) production whereas its salicylate moiety inhibits 12-hydroxy-eicosatetraenoic acid (12-HETE) production in the platelet. The significance of the latter effect on platelet function is unclear. We examined the effects of aspirin and salicylate on (i) platelet/collagen adhesion using 3H-adenine-labelled human platelets and collagen-coated discs, (ii) platelet aggregation induced by thrombin, collagen, ADP and arachidonic acid, and (iii) platelet TxA2 and 12-HETE synthesis as measured by radioimmunoassay and high pressure liquid chromatography respectively. Extraction, purification and characterization of lipoxygenase from foxtail millet (Setaria italica).

This study established a method for extracting and purifying foxtail millet lipoxygenase (LOX) by combining ammonium sulfate precipitation and ion exchange chromatography, and investigated its enzymatic properties. The purified LOX showed high specific activity (448.82 ± 13.26 U/mg) and a molecular weight of 96.6 kDa. It achieved maximum catalytic activity at 30 °C and pH 7.0, with good stability within the pH range of 5.0-6.0. Circular dichroism and ultraviolet spectral analyses demonstrated that the heat-induced reduction in LOX enzymatic activity derived from alterations in the protein's sp

Light-Controlled Modulation of 15-Lipoxygenase-1 Regulates Intestinal Inflammatory Signaling.

Photopharmacology offers powerful opportunities for the spatiotemporal control of biological processes, yet the rational design of photoswitchable enzyme inhibitors remains challenging. Here, we report a target-guided strategy for the development of diazo-based photoswitchable inhibitors of human 15-lipoxygenase-1 (15-LOX-1), a key enzyme in inflammatory signaling, ferroptosis, and cancer. Guided by the structural features of known ligands, we developed three complementary photoswitch classes: reversible azobenzenes (ABs), azo-heteroarenes (HAs), and covalent azo-bis-alkynes (BAs). These compo

Inhibition of 5-Lipoxygenase in Hepatic Stellate Cells Alleviates Liver Fibrosis.

Background and Purpose: Activation of hepatic stellate cells (HSC) is a central driver of liver fibrosis. 5-lipoxygenase (5-LO) is the key enzyme that catalyzes arachidonic acid into leukotrienes. In this study, we examined the role of 5-LO in HSC activation and liver fibrosis. Main Methods: Culture medium was collected from quiescent and activated HSC for target metabolomics analysis. Exogenous leukotrienes were added to culture medium to explore their effect in activating HSC. Genetic ablation of 5-LO in mice was used to study its role in liver fibrosis induced by CCl4 and a methionine-choli

Eosinophils promote corneal wound healing via the 12/15-lipoxygenase pathway.

Lipid mediators play important roles in regulating inflammatory responses and tissue homeostasis. Since 12/15-lipoxygenase (12/15-LOX)-derived lipid mediators such as lipoxin A4 (LXA4 ) and protectin D1 (PD1) protect against corneal epithelial cell damage, the major cell types that express 12/15-LOX and contribute to the corneal wound healing process are of particular interest. Here, we found that eosinophils were the major cell type expressing 12/15-LOX during the corneal wound healing process. Eosinophils were recruited into the conjunctiva after corneal epithelium wounding, and eosinophil-d

12/15-Lipoxygenase choreographs the resolution of IgG-mediated skin inflammation.

Autoimmune diseases are defined by an immune response against a specific autoantigen, driven by antigen-specific T cells or antibodies. While the mechanisms resolving brief episodes of acute inflammation elicited by microbial components or tissue injury are well understood, the mechanisms resolving tissue inflammation in autoimmune diseases are still largely elusive. We have, therefore, addressed the mechanisms of resolution in IgG-mediated autoimmune diseases using a mouse model of the pemphigoid disease "bullous pemphigoid-like epidermolysis bullosa acquisita" (BP-like EBA) as prototypical e

Inhibition of lipoxygenase pathway reduces blood pressure in renovascular hypertensive rats.

To assess the potential role of the lipoxygenase (LO) pathway in the vasculature in an angiotensin II (ANG II)-dependent model of hypertension, we investigated the effect of LO pathway inhibition on blood pressure in the two-kidney, one-clip (2K,1C) Goldblatt hypertensive rat. The development of renovascular hypertension in 2K,1C rats was attenuated by oral administration of phenidone (Phe, 60 mg.kg-1.day-1), a nonselective LO inhibitor, throughout the 3 wk of observation after renal artery constriction. In contrast, the same treatment protocol had no effect on the evolution of hypertension in

Structural analyses of oxygen channels of animal, soybean and manganese lipoxygenases.

Lipoxygenases (LOX) are found in many organisms and oxidize polyunsaturated fatty acids to hydroperoxides. The first tertiary structures predicted already 30 years ago a dedicated oxygen channel to the non-heme iron metal center, which has been a topic of discussion ever since. Analysis of more than fifteen lipoxygenases revealed (mostly) conserved channels from the iron metal center to the surface. They were lined by residues of α-helices 9 and 11 and characteristically passed through a loop between two β-strands below the surface. The channel orifices were generally open and the other ends w

Activation of the 15-lipoxygenase pathway in aspirin-exacerbated respiratory disease.

BACKGROUND: Aspirin-exacerbated respiratory disease (AERD) is characterized by asthma, chronic rhinosinusitis with nasal polyps (CRSwNP), and an intolerance of medications that inhibit cyclooxygenase-1. Patients with AERD have more severe upper and lower respiratory tract disease than do aspirin-tolerant patients with CRSwNP. A dysregulation in arachidonic acid metabolism is thought to contribute to the enhanced sinonasal inflammation in AERD. OBJECTIVE: Our aim was to utilize an unbiased approach investigating arachidonic acid metabolic pathways in AERD. METHODS: Single-cell RNA sequencing

Identification and Characterization of a New Protein Isoform of Human 5-Lipoxygenase.

Leukotrienes (LTs) are inflammatory mediators that play a pivotal role in many diseases like asthma bronchiale, atherosclerosis and in various types of cancer. The key enzyme for generation of LTs is the 5-lipoxygenase (5-LO). Here, we present a novel putative protein isoform of human 5-LO that lacks exon 4, termed 5-LOΔ4, identified in cells of lymphoid origin, namely the Burkitt lymphoma cell lines Raji and BL41 as well as primary B and T cells. Deletion of exon 4 does not shift the reading frame and therefore the mRNA is not subjected to non-mediated mRNA decay (NMD). By eliminating exon 4,

The 12/15-lipoxygenase as an emerging therapeutic target for Alzheimer's disease.

Alzheimer's disease (AD) is a chronic neurodegenerative condition characterized by progressive memory loss. Mutations in genes involved in the production of amyloid-β (Aβ) are linked to the early-onset variant of AD. However, the most common form, sporadic AD, is considered to be the result of an interaction between environmental risk factors and various genes. Among them, recent work has highlighted the potential role that the 12/15-lipoxygenase (12/15LO) pathway may play in AD pathogenesis. 12/15LO is widely distributed in the central nervous system, and its levels are upregulated in patient

Involvement of 5-lipoxygenase in Alzheimer's disease: a role for DNA methylation.

Lipoxygenases play a major role in the neuropathology of Alzheimer's disease (AD), even though the underlying mechanisms are as yet poorly understood. Here, we studied the epigenetic regulation of 5-lipoxygenase (5-LOX) in peripheral blood mononuclear cells of subjects with late-onset AD and age-matched controls. We found a significant increase in 5-LOX gene expression in AD subjects compared to healthy controls, paralleled by increased 5-LOX protein and leukotriene B4, the 5-LOX product. In addition, a consistent reduction in DNA methylation at 5-LOX gene promoter was documented in AD versus

Role of arachidonic acid, lipoxygenase, and mitochondrial depolarization in reperfusion arrhythmias.

We have sought evidence that arachidonic acid (AA) induces mitochondrial depolarization in isolated myocytes by a lipoxygenase (LOX)-dependent mechanism and that such depolarization might contribute to arrhythmogenesis following ischemia-reperfusion injury. A method was developed for measuring mitochondrial depolarization in isolated adult rat myocytes in suspension, using tetramethylrhodamine ethyl ester. The addition of AA to myocytes resulted in mitochondrial depolarization that was inhibited by the LOX inhibitor baicalein, by the reactive oxygen species (ROS) scavenger mercaptoproprionylgl

Regulation of the activity of 5-lipoxygenase, a key enzyme in leukotriene biosynthesis.

5-Lipoxygenase (5LO) catalyzes two steps in the biosynthesis of leukotrienes (LTs), lipid mediators of inflammation derived from arachidonic acid. LTs function in normal host defense, and have pathophysiological roles in chronic inflammatory diseases as asthma and atherosclerosis. Also, possible effects of 5LO products in relation to tumorigenesis have been described. Thus, insight regarding the biochemistry of 5LO is relevant for better understanding of normal physiology, and for development of therapy.

Cyclooxygenases, lipoxygenases, and epoxygenases in CNS: their role and involvement in neurological disorders.

Three enzyme systems, cyclooxygenases that generate prostaglandins, lipoxygenases that form hydroxy derivatives and leukotrienes, and epoxygenases that give rise to epoxyeicosatrienoic products, metabolize arachidonic acid after its release from neural membrane phospholipids by the action of phospholipase A(2). Lysophospholipids, the other products of phospholipase A(2) reactions, are either reacylated or metabolized to platelet-activating factor. Under normal conditions, these metabolites play important roles in synaptic function, cerebral blood flow regulation, apoptosis, angiogenesis, and g
