The emergence of drug resistance presents a major challenge for the treatment of tuberculosis (TB). Reporter phage can provide an effective method for drug susceptibility testing (DST) of Mycobacterium tuberculosis (Mtb), but limited for parts of antitubercular drugs and the process of operation is usually time-consuming. Herein, we developed a new, sensitive, reporter phage with optimized method to detect drug susceptibility of Mtb clinical isolates for all first-line antitubercular drugs. The P furAma promoter and nanoluciferase (Nluc) reporter sequences were integrated into the genome of th

A cell-free biosensor for the detection of transcription induction by specific small-molecule ligands is presented. As model systems, tetracycline and mercury-inducible promoters were used containing firefly luciferase as reporter gene. Escherichia coli S30 extract was prepared and used for coupled transcription-translation reactions. By using purified Tet repressor and MerR regulatory proteins, we could study repressor-operator interactions for optimizing the relative concentrations of each component. Previously, detection of tetracycline and mercury using similar transcriptional regulation i

To facilitate the detection of the tick-borne encephalitis virus (TBEV), the causative agent of one of the most severe human neuroinfections, we have developed an immunoassay based on bioluminescent hybrid protein 14D5a-Rm7 as a detection probe. The protein containing Renilla luciferase as a reporter and a single-chain variable fragment (scFv) of murine immunoglobulin to TBEV as a recognition element was constructed, produced by bacterial expression, purified, and tested. Both domains were shown to reveal their specific biological properties-affinity to the target antigen and bioluminescent ac

BACKGROUND: Tuberculosis (TB) continues to be a major global infectious disease, and the ability of Mycobacterium tuberculosis (Mtb) to persist within macrophages is a major barrier to eradication. Autophagy is a critical host defense mechanism, yet the epitranscriptomic regulation of this process is poorly understood.
METHODS: PBMCs isolated from individuals with TB and THP-1 macrophages infected with Mtb H37Ra were analyzed for METTL14 expression. Functional modulation experiments were conducted to investigate autophagy, m6A modification, and bacterial survival. Techniques included qPCR, We

Engineered bacterial biotherapeutics have shown increasingly promising potential for cancer treatment, yet they are hindered by unresolved challenges regarding off-target toxicity and suboptimal therapeutic efficacy. In this study, we report the first systematic screening of strong constitutive anaerobic promoters in the probiotic Escherichia coli Nissle 1917 (EcN). Via RNA-seq analysis, we initially identified forty candidate promoters from EcN. Subsequent functional characterization using a firefly luciferase reporter assay yielded six robust constitutive anaerobic promoters, whose activity

Saliva is a complex heterogeneous biofluid whose matrix effects can substantially influence biosensor performance. Here, we show that control-referenced bacterial NAD(P)H:FMN oxidoreductase-luciferase kinetics provides a compact functional readout of these matrix effects and reveals a structured salivary response phenotype in breast disease. We analyzed 200 clinically annotated saliva samples representing healthy controls, benign breast disease, early-stage breast cancer and late-stage breast cancer. Three descriptors were extracted from paired sample-control curves: residual luminescence (LI%

Black rockfish (Sebastes schlegelii) is one of the most important mariculture fish in the Western North Pacific. Although the crucial roles of CXCL12 in immune responses against bacterial infection are well-known, the specific mechanisms underlying its involvement in fish immunity remain poorly understood. In this study, we systematically characterized CXCL12a and CXCL12b in Sebastes schlegelii (SsCXCL12a and SsCXCL12b), Firstly, SsCXCL12a/b were ubiquitously expressed across all seven tissues. Both SsCXCL12a/b were significantly differentially expressed in gill, kidney, liver, and spleen afte

Quorum sensing (QS) is a means of bacterial communication accomplished by microbe-produced signals and sensory systems. QS systems regulate important population-wide behaviors in bacteria, including secondary metabolite production, swarming motility, and bioluminescence. The human pathogen Streptococcus pyogenes (group A Streptococcus [GAS]) utilizes Rgg-SHP QS systems to regulate biofilm formation, protease production, and activation of cryptic competence pathways. Given their reliance on small-molecule signals, QS systems are attractive targets for small-molecule modulators that would then a

The role of nucleotide-binding oligomerization domain-1 (NOD1) and nucleotide-binding oligomerization domain-2 (NOD2), cytoplasmic receptors which detect bacterial cell wall molecules, in pulmonary innate immune responses is poorly understood. We determined that both NOD1 and NOD2 detect heat-killed Legionella and stimulate NF-κb and IFN-β promoter activity using an in vitro luciferase reporter system. We next infected NOD1- and NOD2-deficient animals with aerosolized Legionella pneumophila. At 3 days post infection, Nod1(-/-) mice had impaired bacterial clearance compared to WT controls. In a

Neonates have increased vulnerability to life-threatening infections due to the distinct immune landscape. Interleukin (IL)-27 is a key component of this immune profile that we have previously shown to be elevated in both newborn humans and mice. IL-27 continues to increase in the serum and tissues consistent with poor outcomes during gram-negative neonatal bacterial sepsis. Presently, we dissected the IL-27 producer profile at a single-cell level using IL-27p28eGFP reporter mice in our previously established model of neonatal sepsis with luciferase-expressing K1-encapsulated Escherichia coli.

INTRODUCTION: Incorporating minimal antibody regions into engineered proteins is an effective strategy for creating targeted therapeutics and diagnostics. However, a major barrier to clinical translation is the lack of efficient methods to track their performance in vivo. Bioluminescence imaging using targeted reporter proteins offers a promising solution to this challenge. The aim of this study was to develop a novel in vivo imaging strategy using computationally engineered targeted synthetic proteins and to establish a validated pipeline for their design, construction, and testing.

MicroRNAs (miRNAs), a subclass of non-coding small RNA molecules, exert pivotal functions in regulating the immune responses of fish during pathogen infections. The Japanese pufferfish (Takifugu rubripes), a prominent commercial fish species in China, has experienced substantial economic losses due to infections caused by Vibrio harveyi. However, research on the immune modulatory effects of miRNAs in conferring resistance to Vibrio infections in the Japanese pufferfish remains scarce. In this study, high-throughput sequencing was applied to characterize the miRNA profile within the spleen of J

Electrospun nanofibers offer a highly promising platform for the delivery of vaginal lactobacilli, providing an innovative approach to preventing and treating vaginal infections. To advance the application of nanofibers for the delivery of lactobacilli, tools for studying their safety and efficacy in vitro need to be established. In this study, fluorescent (mCherry and GFP) and luminescent (NanoLuc luciferase) proteins were expressed in three vaginal lactobacilli (Lactobacillus crispatus, Lactobacillus gasseri and Lactobacillus jensenii) and a control Lactiplantibacillus plantarum with the aim

Bacillus anthracis, the causative agent of anthrax, is considered a high-priority agent that may be used in a food-related terrorist attack because it can be contracted by ingestion and it also forms spores with heat and chemical resistance. Thus, novel surveillance methodologies to detect B. anthracis on adulterated foods are important for bioterrorism preparedness. We describe the development of a phage-based bioluminescence assay for the detection of B. anthracis on deliberately contaminated foods. We previously engineered the B. anthracis phage Wβ with genes encoding bacterial luciferase (

NahR, a transcriptional regulator for naphthalene degradation in response to salicylate, is a central element in the microbial biosensor for detection of naphthalene and salicylate. To maximize the sensitivity of the biosensor, we have chosen a rational design of highly-sensitive microbial biosensors by introducing site directed mutagenesis to nahR gene. Eight single mutants (N169A, N169C, N169K, N169S, R248H, R248M, R248Q, and R248Y) were made at residues 169 and 248 known as the central inducer-recognition and the C-terminal multimerization domain. The effects of these mutations were examine

Infection with Japanese encephalitis virus (JEV), a mosquito-borne flavivirus, may cause acute encephalitis in humans and induce severe cytopathic effects in various types of cultured cells. We observed that JEV replication rendered infected baby hamster kidney (BHK-21) cells sensitive to the translational inhibitor hygromycin B or alpha-sarcine, to which mock-infected cells were insensitive. However, little is known about whether any JEV nonstructural (NS) proteins contribute to virus-induced changes in membrane permeability. Using an inducible Escherichia coli system, we investigated which p

There is an urgent need to develop novel antibiotics since antibiotic resistance is an increasingly serious threat to global public health. Whole-cell biosensors are one of the promising strategies for new antibiotic discovery. The peptidoglycan (PG) of the bacterial cell wall is one of the most important targets for antibiotics. However, the biosensors for the detection of PG-targeting antibiotics in Gram-negative bacteria have not been developed, mainly because of the lack of the regulatory systems that sense and respond to PG stress. Recently, we identified a novel two-component signal tran
