Lumpy skin disease outbreaks in Greece during 2015-16, implementation of emergency immunization and genetic differentiation between field isolates and vaccine virus strains.

The objective of this study is to present epizootiological data from the lumpy skin disease (LSD) outbreaks in Greece during 2015-16, following the implementation of emergency vaccination and total stamping-out, along with laboratory data regarding the genetic differentiation between field isolates and live attenuated vaccine virus strains. Descriptive geographical chronology analysis was conducted to present the progressive shift of the outbreaks westwards, and at the same time, the absence of further outbreaks in previously affected regional units where high vaccination coverage was achieved

A lumpy skin disease virus deficient of an IL-10 gene homologue provides protective immunity against virulent capripoxvirus challenge in sheep and goats.

Sheep and goat pox continue to be important livestock diseases that pose a major threat to the livestock industry in many regions in Africa and Asia. Currently, several live attenuated vaccines are available and used in endemic countries to control these diseases. One of these is a partially attenuated strain of lumpy skin disease virus (LSDV), KS-1, which provides cross-protection against both sheep pox and goat pox. However, when used in highly stressed dairy cattle to protect against lumpy skin disease (LSD) the vaccine can cause clinical disease. In order to develop safer vaccines effectiv

Windborne Dispersal of Arthropod Vectors as a Pathway for Lumpy Skin Disease Virus Introduction into South Korea: A Multi-Species, Source-Attributed Modelling Study.

Lumpy skin disease (LSD) is a rapidly expanding transboundary vector-borne disease threatening livestock systems across Asia, with increasing risk of introduction into previously unaffected regions such as South Korea. This study developed an integrated, spatially explicit framework to assess the plausibility, timing and source-region structure of LSD virus (LSDV) introduction via windborne dispersal of infected arthropod vectors. Maximum Entropy models based on climatic, environmental, and topographic variables were used to determine habitat suitability for four major vector families (Culex s

Targeting of the cellular ATM rather than ATR kinase exhibits therapeutic potential during the lumpy skin disease virus infection in vivo and in vitro.

Due to the lack of timely vaccine prevention and effective drug treatment, lumpy skin disease is increasingly becoming a global epidemic, including in China. There is an urgent need to explore the pathogenic mechanism of lumpy skin disease virus (LSDV) and develop practical therapeutic approaches. The present study provides concrete evidence for the simultaneous induction and activation of nuclear ATM-mediated double-strand break and ATR kinase-dependent single-strand break signaling cascades during LSDV replication in the cytoplasm. Specific drug-inhibitory experiments targeting ATM and ATR k

Molecular diagnosis of three outbreaks during three successive years (2018, 2019, and 2020) of Lumpy skin disease virus in cattle in Sharkia Governorate, Egypt.

BACKGROUND: Lumpy skin disease (LSD) is endemic in Egypt despite the Egyptian authorities' annual mass vaccination of cattle with sheeppox vaccine (Veterinary Serum and Vaccine Research Institute, Egypt), and the LSD virus (LSDV) continues to thrive practically every summer. The disease has a huge economic impact on the trade of the animal and its by-product. AIM: This paper study the molecular characterization of LSDV strains that have been circulating in Sharkia Governorate, Egypt, for three successive years (2018, 2019, and 2020). METHODS: A total of 61 specimens (26 skin nodules and 35 o

Detection of lumpy skin disease virus in cattle using real-time polymerase chain reaction and serological diagnostic assays in different governorates in Egypt in 2017.

BACKGROUND AND AIM: Lumpy skin disease (LSD), is a highly infectious viral disease of cattle, caused by LSD virus (LSDV) which belongs to the genus Capripoxvirus of family Poxviridae. In the summer of 2017, skin lesions suggestive of LSD were observed in cattle at several governorates in Egypt. This study aimed to detect LSDV in cattle specimens using rapid serological and molecular diagnostic assays. MATERIALS AND METHODS: A total of 46 skin biopsies and uncoagulated blood samples were collected from cattle with LSD suggestive clinical signs, as well as 290 coagulated whole blood samples fro

Development of a Synthesized Gene Unique to Lumpy Skin Disease Virus and Its Application in Serological Differentiation of Naturally Infected from Vaccinated Cattle with Attenuated Goat Pox Vaccine.

Lumpy skin disease (LSD) is an important infectious disease caused by lumpy skin disease virus (LSDV) in bovine. LSDV, sheep pox virus (SPPV), and goat pox virus (GTPV) from the same genus Capripoxvirus (CaPV) of the Poxviridae family exhibit a nucleotide sequence similarity of up to 97%. Therefore, attenuated vaccines of GTPV and SPPV are often used to vaccinate cattle against LSD. However, available serological testing methods cannot accurately differentiate cattle vaccinated with GTPV from those infected with LSDV, posing a significant risk for disease spread. In this study, we developed a

Development and Validation of a New DIVA Real-Time PCR Allowing to Differentiate Wild-Type Lumpy Skin Disease Virus Strains, Including the Asian Recombinant Strains, from Neethling-Based Vaccine Strains.

The current epidemic in Asia, driven by LSDV recombinants, poses difficulties to existing DIVA PCR tests, as these do not differentiate between homologous vaccine strains and the recombinant strains. We, therefore, developed and validated a new duplex real-time PCR capable of differentiating Neethling-based vaccine strains from classical and recombinant wild-type strains that are currently circulating in Asia. The DIVA potential of this new assay, seen in the in silico evaluation, was confirmed on samples from LSDV infected and vaccinated animals and on isolates of LSDV recombinants (n = 12),

Lumpy skin disease: A growing threat to the global livestock industry.

Lumpy Skin Disease (LSD) is an infectious disease in cattle caused by the Lumpy Skin Disease Virus by the Poxviridae family. Historically, LSD was first documented in Northern Rhodesia (now Zambia) in 1929 as a symptom of bug bite poisoning or hypersensitivity, described as pseudo-urticaria. This disease, which causes widespread malaise and persistent weakness, is a serious, economically devastating, and reportable condition that lowers cattle output. The illness starts as a biphasic fever. After fever, emaciation, ocular discharge, and agalactia, the clinical signs of a minor infection develo

Approaches to Next-Generation Capripoxvirus and Monkeypox Virus Vaccines.

Globally, there are two major poxvirus outbreaks: mpox, caused by the monkeypox virus, and lumpy skin disease, caused by the lumpy skin disease virus. While vaccines for both diseases exist, there is a need for improved vaccines. The original vaccines used to eradicate smallpox, which also protect from the disease now known as mpox, are no longer acceptable. This is mainly due to the risk of serious adverse events, particularly in HIV-positive people. The next-generation vaccine for mpox prevention is modified vaccinia Ankara, which does not complete the viral replication cycle in humans and,

A study of the pathology of lumpy skin disease in cattle.

Microscopic lesions in cattle infected with the virus of the Neethling form of lumpy skin disease comprised a granulomatous reaction in the dermis and hypodermis which extended to the surrounding tissue. During the early stages of the lesions a vasculitis and lymphangitis with concomitant thrombosis and infarction resulted in necrosis and oedema. A hallmark of the acute to subacute stages of the lesions was the presence of intracytoplasmic eosinophilic inclusions in various cell types. The inclusions consisted of the viroplasm which was identified as aggregates of electron-dense, finely granul

Cellular infiltration, cytokines, and histopathology of skin lesions associated with different clinical forms and stages of naturally occurring lumpy skin disease in cattle.

Lumpy skin disease (LSD) caused by the Capripoxvirus LSD virus which infects cattle, leading to a serious disease characterized by fever and the eruption of skin nodules all over the surface of the body. Our understanding of the pathogenesis of this disease is still incomplete, particularly the immunopathological alterations occurring in the skin nodules of infected animals. Therefore, we collected skin nodules from naturally infected cattle with different forms of the disease, both in the early stage of clinical infection and after disease progression. The skin samples were examined both hist

Potential of Using Capripoxvirus Vectored Vaccines Against Arboviruses in Sheep, Goats, and Cattle.

The genus capripoxvirus consists of sheeppox virus, goatpox virus, and lumpy skin disease virus, which affect sheep, goats, and cattle, respectively. Together capripoxviruses cause significant economic losses to the sheep, goat, and cattle industry where these diseases are present. These diseases have spread into previously free bordering regions most recently demonstrated with the spread of lumpy skin disease virus into the Middle East, some Eastern European countries, and Russia. This recent spread has highlighted the transboundary nature of these diseases. To control lumpy skin disease viru

Global expansion of lumpy skin disease: transmission trends, economic consequences, and preventive measures.

Lumpy Skin Disease (LSD) is an emerging transboundary poxviral infection affecting cattle and buffaloes, caused by the Lumpy Skin Disease Virus (LSDV), a member of the Capripoxvirus genus. Characterized by fever, nodular dermatitis, lymphadenopathy, and production loss, the disease has evolved from a regionally confined threat in Africa to a global epizootic. While mortality remains below 5%, morbidity can reach 100% in affected herds, leading to severe economic disruption in livestock-dependent economies. Between 2005 and 2020, over 41,000 outbreaks were reported globally, nearly 30,000 in Af
