[Phage conversion of Clostridium novyi type A (author's transl)].

Clostridium novyi type A, the cause of gas gangrene in man and animal, produces four main toxic components (alpha, gamma, delta, and epsilon). The alpha toxin is a necrotizing toxin produced in lethal amounts by strains of type A. This lethal toxin induces a characteristic subcutaneous colorless gelatineous edema when guinea pigs are infected with C. novyi type A. In this paper we report further evidence on the relationship of bacteriophages to the production of alpha toxin by C. novyi type A. The results showed that five phage-sensitive bacterial strains that did not produce alpha toxin could

The isolation and characterisation of a temperate phage ɸPh_AJ01 against Acinetobacter junii.

BACKGROUND: Acinetobacter junii is an emerging opportunistic human and animal pathogen, fast gaining antimicrobial resistance. As conventional treatment becomes ineffective, alternatives are needed to address this challenge. Bacteriophages offer a promising yet underexplored solution in combating A. junii infections. Here, we report the isolation and characterisation of a temperate phage, ɸPh_AJ01, effective against a carbapenem-resistant A. junii. Moreover, the study adopts a technique for biasing the temperate phage to the lytic life cycle to address the issues of lysogeny. The efficacy of t

[Spontaneous phage production in Pseudomonas pseudomallei and in a range of hosts of melioidosis phages among representatives in the genus Pseudomonas].

A capacity to spontaneous phage production has been studied in 57 melioidosis museum strains. The strains were comparatively analyzed by the levels of frequencies of spontaneous phage production. It is established that 40 of 57 studied strains of Pseudomonas pseudomallei are capable to spontaneously produce phages with frequency from 10(7) to 1. On the indicator lawn of P. mallei phages formed negative colonies of different morphology transparent, turbid and turbid with secondary growth in the centre. No distinct correlation between the frequency of spontaneous phage production and morphology

[Antibiotic properties of cultures of the Streptomyces coelicolor group that suppress the growth of strains of the same species].

Antimicrobial spectrum of 45 cultures of the S. coelicolor group with respect to closely related strains and other streptomycete species was studied. The spectrum of 7 cultures was narrow and they inhibited closely related strains and certain streptomycete species. These cultures produced antibiotic substances of bacteriocin-like nature. The bacteriocin-like substances of the cultures differed in sensitivity to temperature and proteolytic enzymes and capacity for dialysis through cellophane membranes. Biosynthesis of the antibiotic substances increased after exposure to UV light.

Phage influence on the synthesis of extracellular toxins in group A streptococci.

Phage conversion of group A streptococci to produce streptococcal exotoxins was shown to occur more widely than has been previously reported. Toxigenic conversion was found in 19 newly constructed lysogenic and pseudolysogenic strains resulting in synthesis of exotoxin types A and B. Conversion was accomplished by a positive conversion effector, which was a phage characteristic expressed by the prophage and vegetatively reproducing phage. Exotoxin production was determined by the rabbit skin test and by countercurrent immunoelectrophoresis with type-specific antisera. New lysogens and pseudoly

Identification of novel inhibitors of Mycobacterium smegmatis growth through genome-wide overexpression of Cluster P3 mycobacteriophage Xavia genes.

Bacteriophages encode numerous genes with no known function, many of which can affect essential cellular processes when expressed in the bacterial host. For mycobacteriophages, genome-wide overexpression in Mycobacterium smegmatis can be used to identify proteins that impair growth. To evaluate the cytotoxic potential of the Cluster P3 phage Xavia, we constructed a plasmid library containing 71 predicted Xavia genes under the anhydrotetracycline inducible promoter pTet and screened this library in a plate-based cytotoxicity assay to measure impacts on M. smegmatis growth. Two genes prevented t

Isolation and Characterization of New Bacteriophages against Staphylococcal Clinical Isolates from Diabetic Foot Ulcers.

Staphylococcus sp. is the most common bacterial genus in infections related to diabetic foot ulcers (DFUs). The emergence of multidrug-resistant bacteria places a serious burden on public health systems. Phage therapy is an alternative treatment to antibiotics, overcoming the issue of antibiotic resistance. In this study, six phages (SAVM01 to SAVM06) were isolated from effluents and were used against a panel of staphylococcal clinical samples isolated from DFUs. A genomic analysis revealed that the phages belonged to the Herelleviridae family, with sequences similar to those of the Kayvirus g

Coupled induction of prophage and virulence factors during tick transmission of the Lyme disease spirochete.

The alternative sigma factor RpoS plays a central role in the critical host-adaptive response of the Lyme disease spirochete, Borrelia burgdorferi. We previously identified bbd18 as a negative regulator of RpoS but could not inactivate bbd18 in wild-type spirochetes. In the current study we employed an inducible bbd18 gene to demonstrate the essential nature of BBD18 for viability of wild-type spirochetes in vitro and at a unique point in vivo. Transcriptomic analyses of BBD18-depleted cells demonstrated global induction of RpoS-dependent genes prior to lysis, with the absolute requirement for

Genomic characteristics of vB_PpaP_PP74, a T7-like Autographivirinae bacteriophage infecting a potato pathogen of the newly proposed species Pectobacterium parmentieri.

Bacteriophage vB_PpaP_PP74 (PP74) is a novel virulent phage that infects members of the species Pectobacterium parmentieri, a newly established species of soft-rot-causing bacteria in the family Pectobacteriaceae, derived from potato-specific Pectobacterium wasabiae. vB_PpaP_PP74 was identified as a member of the family Podoviridae by transmission electron microscopy. The phage has a 39,790-bp dsDNA genome containing 50 open reading frames (ORFs). Because of the absence of genes encoding toxins or lysogeny factors, PP74 may be considered a candidate phage for pathogen biocontrol applications.

Genome sequence and analysis of a Propionibacterium acnes bacteriophage.

Cutaneous propionibacteria are important commensals of human skin and are implicated in a wide range of opportunistic infections. Propionibacterium acnes is also associated with inflammatory acne vulgaris. Bacteriophage PA6 is the first phage of P. acnes to be sequenced and demonstrates a high degree of similarity to many mycobacteriophages both morphologically and genetically. PA6 possesses an icosahedreal head and long noncontractile tail characteristic of the Siphoviridae. The overall genome organization of PA6 resembled that of the temperate mycobacteriophages, although the genome was much

In Vivo Targeting of Clostridioides difficile Using Phage-Delivered CRISPR-Cas3 Antimicrobials.

Clostridioides difficile is an important nosocomial pathogen that causes approximately 500,000 cases of C. difficile infection (CDI) and 29,000 deaths annually in the United States. Antibiotic use is a major risk factor for CDI because broad-spectrum antimicrobials disrupt the indigenous gut microbiota, decreasing colonization resistance against C. difficile Vancomycin is the standard of care for the treatment of CDI, likely contributing to the high recurrence rates due to the continued disruption of the gut microbiota. Thus, there is an urgent need for the development of novel therapeutics th

The secret life of the anthrax agent Bacillus anthracis: bacteriophage-mediated ecological adaptations.

Ecological and genetic factors that govern the occurrence and persistence of anthrax reservoirs in the environment are obscure. A central tenet, based on limited and often conflicting studies, has long held that growing or vegetative forms of Bacillus anthracis survive poorly outside the mammalian host and must sporulate to survive in the environment. Here, we present evidence of a more dynamic lifecycle, whereby interactions with bacterial viruses, or bacteriophages, elicit phenotypic alterations in B. anthracis and the emergence of infected derivatives, or lysogens, with dramatically altered

A receptor-centered approach identifies Lom as a LamB-bound superinfection exclusion factor in bacteriophage λ.

Bacteriophages face intense competition within bacterial populations. Although bacteria encode diverse anti-phage mechanisms, strategies protecting virions at the host surface remain poorly understood. Here, we develop a receptor-centered discovery approach that captures phage proteins bound to host receptors during infection. Applying this strategy to bacteriophage λ and its outer-membrane receptor LamB, we identify Lom as a phage-encoded outer membrane protein that binds LamB. Structural, biochemical, and functional analyses show that Lom occupies the same LamB surface recognized by the rece

12/111phiA Prophage Domestication Is Associated with Autoaggregation and Increased Ability to Produce Biofilm in Streptococcus agalactiae.

CC17 Streptococcus agalactiae carrying group-A prophages is increasingly responsible for neonatal infections. To investigate the impact of the genetic features of a group-A prophage, we first conducted an in silico analysis of the genome of 12/111phiA, a group-A prophage carried by a strain responsible for a bloodstream infection in a parturient. This revealed a Restriction Modification system, suggesting a prophage maintenance strategy and five ORFs of interest for the host and encoding a type II toxin antitoxin system RelB/YafQ, an endonuclease, an S-adenosylmethionine synthetase MetK, and a

PRISM: A unified platform for phage isolation and characterization from single-droplet microenvironments.

The exploration of bacteriophage diversity remains constrained by reliance on conventional plaque assays, which bias discovery toward phages that form visible plaques. We present PRISM (Phage Recovery and Investigation in Single-droplet Microenvironments), a droplet microfluidics-based, plating-independent platform for high-throughput isolation, quantification, and characterization of bacteriophages. By encapsulating phage-host interactions in water-in-oil microdroplets at single-phage resolution and coupling them with fluorescence-based detection and sorting, we demonstrate PRISM's capabiliti

Isolation, Characterization, and In Vivo Evaluation Efficacy of Lytic Bacteriophage SEP1 Against Salmonella Paratyphi C.

Multidrug-resistant Salmonella Paratyphi poses a severe threat to public health. As conventional antibiotics lose efficacy against emerging resistant strains, the need to develop alternative antimicrobial agents has become increasingly urgent. In this study, we isolated and characterized a novel lytic bacteriophage, designated SEP1, from poultry sewage using the S. Paratyphi C strain QH as the host and systematically evaluated its therapeutic potential in a murine infection model Genomic analysis confirmed that SEP1 belongs to the genus Felixounavirus, with an 85,703-bp genome devoid of lysoge
