Gelatin-methacryloyl hydrogel stiffness influences epithelial-mesenchymal transition in MCF7 but not MDA-MB-231 breast cancer cells in 3D culture.

This study focuses on advancing the understanding of breast cancer through 3D in vitro models, which provide biomimetic environments superior to many 2D cultures and animal models. Ex vivo analyses show that malignant breast tissues exhibit increased stiffness with higher tumour grade. Tumour stiffening is associated with altered cell phenotype, promoting progression, invasion, and metastasis. This research aims to design 3D models that mimic the evolving tumour microenvironment to study how matrix stiffness affects breast cancer cell behaviour. Using gelatin-methacryloyl (GelMA) hydrogels, we

Evaluation of the anticancer effects of ellagic acid and cisplatin in cisplatin-sensitive and -resistant MDA-MB-231 breast cancer cells.

Ellagic acid (EA) is a natural polyphenol noted for its antiproliferative and pro-apoptotic effects. This study investigates the impact of EA, alone or combined with cisplatin (CIS), on the expression of angiogenesis, apoptosis, metastasis, and chemoresistance-related genes and proteins in cisplatin-sensitive and -resistant MDA-MB-231 breast cancer cells. Cell viability was evaluated by cytotoxicity assay, while gene and protein expression levels were analyzed via qPCR and immunocytochemistry. Molecular docking was used to assess EA's binding affinity to target proteins. The IC₅₀ values of EA

A distinctive and proficient fluorescent switch for ratiometric recognition of the menacing cyanide ion: biological studies on MDA-MB-231 cells.

A new fluorescent ratiometric switch (BOHB) was developed for swift and selective detection of cyanide ions in aqueous media without any interference from other competitive anions. Upon gradual addition of cyanide ions into the probe solution, a prominent fluorescence color change from yellow to cyan was observed under a UV chamber. The fluorescence changes thus observed were ratiometric, and the detection limit of this new probe was found to be (22.1 ± 0.89) μM, suggesting that the efficiency of BOHB for the detection of cyanide ions is brilliant even at a minute level. The blue shift in fluo

Insights into the Behavior of Triple-Negative MDA-MB-231 Breast Carcinoma Cells Following the Treatment with 17β-Ethinylestradiol and Levonorgestrel.

Oral contraceptives (OCs) are widely used due to their efficiency in preventing unplanned pregnancies and treating several human illnesses. Despite their medical value, the toxicity of OCs remains a public concern. Previous studies indicate the carcinogenic potential of synthetic sex hormones and their link to the development and progression of hormone-dependent malignancies such as breast cancer. However, little is known about their influence on the evolution of triple-negative breast carcinoma (TNBC), a malignancy defined by the absence of estrogen, progesterone, and HER2 receptors. This stu

MDA-MB-231 breast cancer cells fuel osteoclast metabolism and activity: A new rationale for the pathogenesis of osteolytic bone metastases.

Recent progress in dissecting the molecular paracrine circuits of cancer and stromal cells in bone metastases (BM) are offering new options to improve current merely palliative approach. The study of tumor-stroma metabolic interplay may further ameliorate this scenario. In this context, we demonstrated that highly glycolytic MDA-MB-231 cancer cells, that form osteolytic BM in vivo, release a large amount of lactate at a significantly higher level than MCF7 cells. Thus, we speculated that lactate released from carcinoma cells is uptaken and metabolically used by osteoclasts, the key players of

Coordination of centrosome homeostasis and DNA repair is intact in MCF-7 and disrupted in MDA-MB 231 breast cancer cells.

When cells encounter substantial DNA damage, critical cell cycle events are halted while DNA repair mechanisms are activated to restore genome integrity. Genomic integrity also depends on proper assembly and function of the bipolar mitotic spindle, which is required for equal chromosome segregation. Failure to execute either of these processes leads to genomic instability, aging, and cancer. Here, we show that following DNA damage in the breast cancer cell line MCF-7, the centrosome protein centrin2 moves from the cytoplasm and accumulates in the nucleus in a xeroderma pigmentosum complementat

Radiosensitizing Effect of Gold Nanoparticles Stabilized with Sodium Citrate and Thioglycolic Acid on MDA-MB-231 and MCF7 Breast Cancer Cells.

Gold nanoparticles (AuNP) have been increasingly investigated as potential radiosensitizing agents for the treatment of cancer. In the present study, AuNP stabilized with sodium citrate and thioglycolic acid (AuNP-Cit-TGA) were synthesized to assess their radiosensitizing effects on MDA-MB-231 and MCF7 breast cancer cells after X-ray exposure. The AuNP-Cit-TGA displayed an average diameter of 22 nm, showcasing extremely low hemolytic activity in vitro, along with a moderate rate of cellular uptake. AuNP-Cit-TGA notably decreased cell viability in both cell lines 48 h after exposure to a radiat

Targeted delivery of chrysin and 5-fluorouracil on MDA-MB-231 cancer cells by a peptide-functionalized L-DOPA-imprinted polymer.

Triple-negative breast cancer (TNBC) is a very aggressive and deadly form of breast cancer for which chemotherapy is the only systemic treatment option. Therefore, novel and more effective targeted or combined therapies, such as specific drug delivery systems that selectively target cancer cells, have received much attention. This research aimed to investigate the effect of targeted delivery of chrysin (CH) and 5-fluorouracil (5FU) using polymer nanoparticles on MDA-MB-231 cells. In this regard, CH and 5FU were individually used as the template to polymerize L-DOPA on the surface of silica nan

Whey Protein Concentrate Renders MDA-MB-231 Cells Sensitive to Rapamycin by Altering Cellular Redox State and Activating GSK3β/mTOR Signaling.

Whey protein concentrate (WPC) is an amino acid-rich supplement that has been shown to increase cellular antioxidant capacity. Mammalian target of rapamycin (mTOR) is a crucial regulator of signaling in mammalian cells, and serves as a therapeutic target for triple-negative breast cancer (TNBC). This study was designed to investigate the effect of combining WPC with rapamycin on MDA-MB-231 human breast cancer cells. These cells were found to be insensitive to rapamycin and exhibited higher glutathione (GSH) and reactive oxygen species levels than non-tumorigenic MCF-10A cells. However, for MDA

Cytotoxicity and cell cycle arrest induced by andrographolide lead to programmed cell death of MDA-MB-231 breast cancer cell line.

BACKGROUND: Breast cancer is considered as an increasing major life-threatening concern among the malignancies encountered globally in females. Traditional therapy is far from satisfactory due to drug resistance and various side effects, thus a search for complementary/alternative medicines from natural sources with lesser side effects is being emphasized. Andrographis paniculata, an oriental, traditional medicinal herb commonly available in Asian countries, has a long history of treating a variety of diseases, such as respiratory infection, fever, bacterial dysentery, diarrhea, inflammation e

Combination of Berberine and NVP-BEZ235 inhibits metastasis of triple-negative breast cancer MDA-MB-231 cell line.

Triple Negative Breast Cancer (TNBC), responsible for 20 % of breast cancers cases, is the most lethal and aggressive type. Its resistance to hormonal therapy and frequent development of chemotherapy resistance render conventional treatments inadequate for TNBC. The phosphatidylinositol 3-kinase (PI3K)/AKT/mTOR pathway, commonly activated in breast cancer, is critical in tumor growth and metastasis. Inhibitors for this pathway have shown to have therapeutic potential and their combinations has led to their active investigation in preclinical and clinical studies for TNBC treatment. Berberine i

Gonadotropin-releasing hormone gene expression in MDA-MB-231 and ZR-75-1 breast carcinoma cell lines.

The presence of gonadotropin-releasing hormone (GnRH)-binding sites in human breast carcinomas and breast tumor cell lines as well as the demonstration of the inhibitory effects of GnRH analogues on the growth of these cells raised the possibility that GnRH is produced locally by breast tumor cells themselves. Immunoreactive GnRH was shown to be present in acetic acid extracts of cultured MDA-MB-231 and ZR-75-1 breast carcinoma cells. These extracts were separated by high-performance liquid chromatography and were analyzed by means of region-specific antisera with differing GnRH sequence speci

MDA-MB-231 cell morphology influences chemotactic sensing of CXCL12 gradients in type 1 bovine collagen matrix.

Chemotaxis plays a critical role in the metastatic progression of breast cancer. The chemokine CXCL12 is well recognized as an essential component of chemotactic migration in triple-negative breast cancer (TNBC) cells in vivo. The purpose of this study is to determine how the highly metastatic TNBC cell line, MDA-MB-231, migrates in response to well-defined CXCL12 gradients in vitro. Traditional 2D transwell migration assays were optimized to gauge the MDA-MB-231 cells' responsiveness to various CXCL12 concentrations. The optimum chemoattractant concentrations were applied to a commercially av

Screening of kinase inhibitors in the triple negative KRAS G13D-mutated MDA-MB-231 breast cancer cell line.

Triple negative breast cancer (TNBC) is more chemoresistant and has a poorer prognosis than other breast cancer types. A subset of TNBCs carries KRAS mutations or amplifications. MDA-MB-231 cells, which have a KRAS G13D mutation and express HER2-103, were previously shown to be sensitive to the KRAS G12D inhibitor MRTX1133 and ERBB2-targeting drugs. A kinase inhibitor library of 157 compounds was screened against MDA-MB-231 cells. Effective compounds and ERBB2-directed drugs were tested for synergistic antiproliferative effects in combination with two KRAS inhibitors and for their impact on ce
