Peritoneal T lymphocyte regulation by macrophages.

The T cell composition of the peritoneal cavity (PerC) in naïve BALB/c, C57BL/6, DBA/2J, and B-1 B cell-defective BALB.xid mice was investigated. The BALB.xid PerC T cell pool had a high CD4:CD8 T cell ratio relative to the other strains whose ratios were similar to those found in their lymph node and spleen. All mice had significant representation of T cells with an activated (CD25(+), GITR(hi), CD44(hi), CD45RB(lo), CD62L(lo)) phenotype and low numbers of Foxp3(+) T(reg) cells in their PerC. Despite a phenotype indicative of activation, peritoneal T cell responses to CD3 ligation were very l

Monocytes/macrophages in renal allograft rejection.

Monocytes/macrophages (MO) have long been recognized to be involved in renal allograft rejection. Monocytes/macrophages have been detected in the glomerular, vascular, and tubulointerstitial compartments during rejection. The recent demonstration that peritubular capillary deposition of complement split factor C4d, a marker for antibody-mediated rejection, is associated with relatively marked MO infiltration of the allograft during acute rejection is a significant development in our understanding of the role of the MO in rejection. High levels of MO in rejecting allografts have been associated

Salbutamol increases tristetraprolin expression in macrophages.

Tristetraprolin (TTP) is a tandem zinc finger protein that can bind to AU-rich elements (AREs) in the 3'-untranslated regions (3'-UTR) in mRNAs of transiently expressed genes, e.g. tumor necrosis factor-alpha (TNF-alpha) and granulocyte macrophage colony-stimulating factor (GM-CSF). TTP increases the turnover rate of the target mRNAs, thereby reducing, for example, the expression of TNF-alpha and GM-CSF. We examined the role of beta(2)-agonists, cAMP analogs, and forskolin (an activator of adenylate cyclase) on TTP mRNA and protein expression by quantitative real-time RT-PCR and Western blotti

Oxidative stress-inducible proteins in macrophages.

Macrophages produce reactive oxygen species such as O2-, H2O2 and *OH that contribute to the pathogenesis of diseases such as inflammation and atherosclerosis. The cells have multiple defense systems against those reactive oxygen species, and we describe here such an oxidative stress-inducible defense system. Upon exposure to reactive oxygen species and electrophilic agents, murine peritoneal macrophages induce stress proteins to protect themselves. Using differential screening, we cloned two novel proteins designated MSP23 and A170 that are induced in the cells by low levels of reactive oxyge

Renal interstitial foam cells are macrophages.

Immunohistochemical studies on renal biopsies from eight patients with various types of glomerulonephritis showed that the interstitial foam cells belonged to the monocyte-macrophage lineage. There was a strong association between hypercholesterolaemia and the presence of renal interstitial foam cells.

Monocytes/Macrophages Stimulate Hairy-Cell Proliferation.

During a study of the effect of B-cell growth factors on the growth of hairy cells (HCs), proliferation of a sub-population of HCs was observed in the absence of added growth factors in 1 (WB) of 9 patients with hairy-cell leukaemia (HCL). Double staining analysis using monoclonal antibodies detecting the Ki-67 nuclear proliferation antigen and a range of surface/cytoplasmic antigens, confirmed that the HCs were proliferating and that these cells broadly resembled the majority non-proliferative population. The HC proliferation of patient WB was shown to be dependent on the presence of autologo

Lysosomal gold accumulations in pulmonary macrophages.

Although soluble gold has been widely used in the treatment of rheumatoid arthritis, little is known about the distribution of gold deposits in extra-articular tissues. In the synovial lining cells and articular cartilage, the morphology of lysosomes containing gold, the aurosomes, is well documented. Because gold may cause pulmonary injury, the morphology and distribution of gold deposits in pulmonary tissue should be recognized. We found morphologically typical aurosomes with electron-dense membranes and granules, giving the spectrum of gold in electron microprobe analysis, in the interstiti

Atherosclerosis nanotheranostics: a perspective from macrophages.

The mortality rate of cardiovascular diseases (CVDs) has ranked number one in the world over the years. Atherosclerosis (AS), a chronic inflammatory disease, is a key pathological factor in the occurrence and development of CVDs, characterized by lipid accumulation and plaque formation in arteries. Macrophages is the critical contributor to the progression of AS, and macrophage-related pathological processes have received considerable attention as an extremely important target for the diagnosis and treatment of AS. Biomaterial-based nanotechnology offers the possibility of selective and effici

Peristalsis-Induced Mechanical Activation of Macrophages.

Inflammation is a complex process that drives both acute and chronic diseases. It is modulated by cascading biochemical and biophysical cues that result in immune cell activation. Macrophages are a primary immune cell type that are involved in multiple inflammatory conditions, including cancer, fibrosis, and autoimmune diseases. Traditional methods of investigating macrophage activation involve biochemical stimulation with defined cytokine or chemokine cues, activating macrophages into the edges of a pro- or anti-inflammatory state. Emerging evidence suggests that biophysical cues also play a

Role of macrophages in neuroimmune regulation.

Macrophages, as essential components of the immune system, play pivotal roles in regulating neuroimmune interactions. These cells exist in two primary forms: tissue-resident macrophages and monocyte-derived macrophages (MDMs), each exhibiting distinct functions in neuroimmune responses. Tissue-resident macrophages maintain tissue homeostasis and act as the first line of defense against pathogens, while MDMs are recruited during inflammation, performing both phagocytic and immunomodulatory functions. Despite the recognized roles of these macrophage populations in systemic immunity, their specif

Tumour-associated macrophages in human meningiomas.

Tumour-associated macrophages (TAMs) are regarded as potential therapeutic targets due to "pro-tumoral" and "anti-tumoral" phenotypes. Human meningiomas contain considerable number of TAMs, but their clinical impact is sparsely investigated in these tumours. The aim of this study was therefore to investigate the presence, morphology, and distribution of TAMs in human meningiomas, and relate these findings to histopathology, meningioma subtypes, World Health Organization (WHO) grade, risk of recurrence, and overall survival. In this study, 147 WHO grade 1 and 2 primary meningiomas prepared as t

Macrophages and Age-Related Macular Degeneration.

Age-related macular degeneration (AMD) is characterised by the dysfunction of the retinal pigment epithelium (RPE), deposition of drusen and degeneration of the overlying photoreceptors, leading to the loss of central vision. AMD is a complex and multifactorial disease, with the greatest risk factor being age. As we age, changes occur to our immune system, a phenomenon known as inflammageing. There is mounting evidence indicating a role for the immune system in the development, progression, pathology and potentially, treatment of AMD. However, the specific contribution played by immune cells r

Macrophages in the male genital tract.

Macrophages play essential roles in maintaining immune defence and tissue homeostasis within the male genital tract, which is essential for reproductive health. Since their initial discovery in the testes, macrophages have been established as highly adaptable immune cells that perform diverse functions ranging from immune surveillance to tissue repair. In the testes, macrophages play a crucial role in maintaining immune privilege and tissue homeostasis by regulating inflammatory responses and supporting steroidogenesis and spermatogenesis via intricate crosstalk with Leydig and Sertoli cells.

Functional In Vivo Imaging of Macrophages.

Resident tissue macrophages (RTMs) are specialized phagocytes that are widely distributed throughout the body and are responsible for maintaining homeostasis. Recent advances in experimental techniques have enabled us to gain a greater insight into the actual in vivo biology of RTMs by observing their spatiotemporal dynamics directly in their native environment. Here, we detail a method for live tracking macrophages in a prototypical stromal tissue with high spatial and temporal resolution and great experimental versatility. Our approach builds on a custom intravital imaging platform and strai

Peripheral and central macrophages in obesity.

Obesity is associated with chronic, low-grade inflammation. Excessive nutrient intake causes adipose tissue expansion, which may in turn cause cellular stress that triggers infiltration of pro-inflammatory immune cells from the circulation as well as activation of cells that are residing in the adipose tissue. In particular, the adipose tissue macrophages (ATMs) are important in the pathogenesis of obesity. A pro-inflammatory activation is also found in other organs which are important for energy metabolism, such as the liver, muscle and the pancreas, which may stimulate the development of obe

Effects of synovial macrophages in osteoarthritis.

Osteoarthritis (OA) is a common degenerative disease in mammals. However, its pathogenesis remains unclear. Studies indicate that OA is not only an aging process that but also an inflammation-related disease. Synovitis is closely related to the progression of OA, and synovial macrophages are crucial participants in synovitis. Instead of being a homogeneous population, macrophages are polarized into M1 or M2 subtypes in OA synovial tissues. Polarization is highly associated with OA severity. However, the M1/M2 ratio cannot be the only factor in OA prognosis because intermediate stages of macrop
