Environmental conditions are key factors in the progression of plant disease epidemics. Light affects the outbreak of plant diseases, but the underlying molecular mechanisms are not well understood. Here, we report that the light-harvesting complex II protein, LHCB5, from rice is subject to light-induced phosphorylation during infection by the rice blast fungus Magnaporthe oryzae We demonstrate that single-nucleotide polymorphisms (SNPs) in the LHCB5 promoter control the expression of LHCB5, which in turn correlates with the phosphorylation of LHCB5. LHCB5 phosphorylation enhances broad-spectr

Fungal diseases cause enormous crop losses, but defining the nutrient conditions encountered by the pathogen remains elusive. Here, we generated a mutant strain of the devastating rice pathogen Magnaporthe oryzae impaired for de novo methionine biosynthesis. The resulting methionine-requiring strain grew strongly on synthetic minimal media supplemented with methionine, aspartate or complex mixtures of partially digested proteins, but could not establish disease in rice leaves. Live-cell-imaging showed the mutant could produce normal appressoria and enter host cells but failed to develop, indic

A new benzophenanthridine alkaloid glycoside (1), named nitrotyrasanguinarinoside, together with twelve known compounds (2 -13), was isolated from the dichloromethane extract of the aerial parts of Argemone mexicana Linn. Their structures were elucidated using IR, HR-ESI-MS, 1D and 2D NMR, and ECD spectroscopy, as well as by comparison with the published data. The dichloromethane extract exhibited in vivo antifungal activity against Colletotrichum coccodes with 80% inhibition at a concentration of 3000 µg/mL. Five benzophenanthridine alkaloids (1 - 5) inhibited four phytopathogenic fungi in vi

Fatty acids (FAs) have been implicated in signaling roles in plant defense responses. We previously reported that mutation or RNAi-knockdown (OsSSI2-kd) of the rice OsSSI2 gene, encoding a stearoyl acyl carrier protein FA desaturase (SACPD), remarkably enhanced resistance to blast fungus Magnaporthe oryzae and the leaf-blight bacterium Xanthomonas oryzae pv. oryzae (Xoo). Transcriptomic analysis identified six AAA-ATPase family genes (hereafter OsAAA-ATPase1-6) upregulated in the OsSSI2-kd plants, in addition to other well-known defense-related genes. Here, we report the functional analysis of

Plant pathogenic fungi pose a significant threat to global food security. Hence, discovering targeted (bio)chemical methods to combat plant diseases is a critical research avenue. Recent work has implicated copper radical oxidases (CROs) from Auxiliary Activity Family 5 (AA5) in fungal morphogenesis and pathogenesis, including in appressorium penetration by foliar phytopathogens in the genera Colletotrichum and Magnaporthe. We identified orthologous AA5-encoding genes in Verticillium species; however, it was not clear that the functions of the corresponding CROs would be conserved in these vas

The development of nanopesticides with minimized carriers (NMCs) through prodrug design and molecular self-assembly represents a transformative strategy for enhancing pesticide utilization efficiency and addressing global food security challenges. As a proof of concept, we developed A12@DES, a water-based nanodelivery system that combines a target-specific pesticide with a deep eutectic solvent (DES). The system utilizes a laccase inhibitor (A12, structurally optimized from lead compound PMDD-5Y) to self-assemble into monodisperse nanospheres (∼160 nm diameter) with an alkaline phosphatase (AL

The emerging resistance of crop pathogens to fungicides poses a challenge to food security and compels discovery of new antifungal compounds. Here, we show that mono-alkyl lipophilic cations (MALCs) inhibit oxidative phosphorylation by affecting NADH oxidation in the plant pathogens Zymoseptoria tritici, Ustilago maydis and Magnaporthe oryzae. One of these MALCs, consisting of a dimethylsulfonium moiety and a long alkyl chain (C18-SMe2+), also induces production of reactive oxygen species at the level of respiratory complex I, thus triggering fungal apoptosis. In addition, C18-SMe2+ activates

In pursuit of environmentally friendly agrochemicals, we employed the natural quinine alkaloid scaffold as a lead for the design and synthesis of three series of structurally diverse quinine derivatives and systematically evaluated their in vitro antibacterial and antifungal activities. Among them, compounds Ka9 and Ka10 exhibited good antibacterial activity, with MIC values of 6.25 μg/mL against both Xoo and Xac, while compound Kc1 exhibited excellent activity against Magnaporthe oryzae (EC50 = 2.56 μg/mL) and effectively inhibited succinate dehydrogenase (SDH). Furthermore, molecular docking

Rice blast, caused by the fungal pathogen Magnaporthe grisea, is one of the most serious diseases of rice. Here we describe the isolation and characterization of Pib, one of the rice blast resistance genes. The Pib gene was isolated by a map-based cloning strategy. The deduced amino acid sequence of the Pib gene product contains a nucleotide binding site (NBS) and leucine-rich repeats (LRRs); thus, Pib is a member of the NBS-LRR class of plant disease resistance genes. Interestingly, a duplication of the kinase 1a, 2 and 3a motifs of the NBS region was found in the N-terminal half of the Pib p

In plant-pathogen interaction, plants trigger reactive oxygen species (ROS) bursts to defend against pathogen invasion, while pathogens evolved antioxidant systems to scavenge ROS for infection. Our study reveals a regulatory mechanism of ROS adaptation and scavenging in the rice blast fungus Magnaporthe oryzae. We found that KatG1, a bifunctional catalase-peroxidase, acts as a ROS-responsive regulatory enzyme. During infection, the deacetylase Hos2 mediates the decrotonylation of KatG1 at K422, initiating the ROS clearance process. Decrotonylated KatG1 depolymerizes into monomers, interacts w

Resistance to single-target fungicides necessitates structurally innovative antifungal agents with broadened modes of action. Herein, a series of pyrazole-oxadiazole-linked 1,2,4-triazole derivatives (L1-L39) were designed and synthesized via molecular hybridization. Compound L22 exhibited exceptional activity against Magnaporthe grisea (EC50 = 5.96 μg/mL), outperforming the commercial fungicide isoprothiolane, and potently inhibited Botrytis cinerea and Verticillium dahliae. In vivo assays confirmed its excellent protective and curative efficacies against rice blast. Mechanistic investigation

Improving multistress resilience in crops is essential for sustainable agriculture, yet the genetic mechanisms coordinating abiotic and biotic stress responses remain poorly understood. This study identifies CTS1 (Cold Tolerance at the Seedling stage1), encoding the transcription factor OsWRKY74, as a key pleiotropic regulator that mediates both cold tolerance and blast resistance in rice. Through genome-wide association study, we found that CTS1 negatively regulates cold tolerance at the seedling stage. The natural allele CTS1Hap1 confers greater cold tolerance than CTS1Hap2 by mildly repress

BACKGROUND: By 2050, the world population will increase to 10 billion which urged global demand for food production to double. Plant disease and land drought will make the situation more dire, and safer and environment-friendly materials are thus considered as a new countermeasure. The rice blast fungus, Magnaporthe oryzae, causes one of the most destructive diseases of cultivated rice worldwide that seriously threatens rice production. Unfortunately, traditional breeding nor chemical approaches along control it well. Nowadays, nanotechnology stands as a new weapon against these mounting chall

Rice blast, caused by the fungus Magnaporthe oryzae, is one of the three major diseases affecting rice production and quality; it reduces rice grain yield by nearly 30%. In the early stage of this study, a strain of Bacillus velezensis with strong inhibition of M. oryzae was isolated and named ZW10. In vitro assays indicated prolonged germination time of conidia of M. oryzae treated with the antifungal substances of ZW10, 78% of the conidia could not form appressorium, and the conidial tubes expanded to form vacuolar structure and then shrank. The results of FDA-PI composite dyes showed that t

Although the antagonistic effects of host resistance against biotrophic and necrotrophic pathogens have been documented in various plants, the underlying mechanisms are unknown. Here, we investigated the antagonistic resistance mediated by the transcription factor ETHYLENE-INSENSITIVE3-LIKE 3 (OsEIL3) in rice. The Oseil3 mutant confers enhanced resistance to the necrotroph Rhizoctonia solani but greater susceptibility to the hemibiotroph Magnaporthe oryzae and biotroph Xanthomonas oryzae pv. oryzae. OsEIL3 directly activates OsERF040 transcription while repressing OsWRKY28 transcription. The i

Rice is one of the most important crops worldwide and is widely used as a model plant for molecular studies of monocotyledonous species. The plant hormone jasmonic acid (JA) is involved in rice-pathogen interactions. In addition, volatile compounds, including terpenes, whose production is induced by JA, are known to be involved in the rice defense system. In this study, we analyzed the JA-induced terpene synthase OsTPS24 in rice. We found that OsTPS24 was localized in chloroplasts and produced a monoterpene, γ-terpinene. The amount of γ-terpinene increased after JA treatment. γ-Terpinene had s

The nucleolus is a central hub for nuclear functions, including, but not limited to, ribosome biogenesis in mammals and budding yeast. However, how the nucleolus is regulated and contributes to fungal development and pathogenesis is poorly understood. Here we report that a JmjC domain-containing histone demethylase MoJMJD2 is required for rDNA transcription and nucleolar integrity in the rice blast fungus, Magnaporthe oryzae. MoJMJD2 generates two alternatively spliced isoforms with distinct subcellular localization patterns, suggesting coordinated roles in gene regulation in the nucleolus and

Fungal diseases seriously threaten global crop production, highlighting the need for fungicides with novel modes of action. Here, we show that shikonin exerts antifungal activity by targeting dihydroorotase (DHOase). Shikonin significantly inhibited mycelial growth, spore germination, and appressorium formation in representative phytopathogenic fungi in vitro. Biophysical analyses showed that shikonin directly binds to the DHOase of Magnaporthe oryzae (MoPyr4), with dissociation constants of 3.084, 1.22, and 1.26 μM determined by MST, SPR, and ITC, respectively. Rice blast, caused by Magnaporthe oryzae, remains a primary threat to global food production, making the discovery of novel antifungal targets imperative. The Target of Rapamycin (TOR) signaling network is a prime target for therapeutic intervention, but the function of its central components in M. oryzae is not fully understood. In this study, we found that MoLst8 is indispensable for fungal growth, development, and virulence in M. oryzae. Genetic deletion of MoLST8 impairs fungal growth, conidiation, and virulence by disrupting TOR signaling, triggering excessive autophagy, and damaging plas
