Fungi from Malus in Qujing, China: two new species, three new records, and insights into potential host jumping and lifestyle switching.

Apple trees [Malus domestica Borkh. (Rosaceae)] are one of the important temperate fruit crops in China. In comparison to other temperate fruits, such as grapes and pears, fungal studies (in Yunnan) associated with M. domestica are fewer in number. In the present study, we investigated fungi associated with M. domestica in Qujing City, Yunnan Province, China. Samples were collected from apple gardens in different locations. Single spore isolation was carried out to isolate saprobic fungi, while the surface sterilization method was carried out to isolate endophytic fungi. Molecular analyses wer

NO treatment of embryos isolated from artificially aged apple (Malus domestica Borkh.) seeds modifies the pattern of nitrated and carbonylated proteins.

Accelerated ageing of orthodox seeds of apple (Malus domestica Borkh.), resulting in loss of vigour, is associated with the overaccumulation of reactive oxygen species (ROS) and the depletion of reactive nitrogen species (RNS). Nitric oxide (NO) mitigates the detrimental effects of seed ageing. The aim of this work was to study the effects of NO treatment on the profile of proteins modified by ROS (carbonylated) and RNS (nitrated), in embryonic axes isolated from apple seeds artificially aged for 7, 14, and 21 days. We linked ageing-related deterioration in seeds with the activity of the prote

Diminishing returns in the scaling of lamina and petiole biomass and its relation to lamina geometric occupancy: evidence from 12 Malus cultivars.

BACKGROUND AND AIMS: Leaf biomass allocation involves scaling relationships between photosynthetic surface area and supporting structures. However, whether these relationships remain consistent among closely related cultivars and whether they are associated with leaf geometry remains unclear. This study examined the scaling relationship between lamina fresh mass (ML) and petiole fresh mass (MP), the relationship between lamina area (A) and ML, and the association between lamina geometric occupancy and these scaling patterns across 12 Malus cultivars. METHODS: Lamina geometric occupancy was qu

Genome-wide identification of WOX transcription factors and functional characterization of WOX4 and WOX13 involved in cold stress response in Malus baccata.

INTRODUCTION: Cold stress is a major abiotic threat to apple production. Malus baccata has exceptional cold hardiness and is widely used as a superior cold-resistant rootstock. The WUSCHEL-related homeobox (WOX) transcription factor family regulates plant growth, development and stress adaptation, whereas the functions of WOX genes in cold tolerance of M. baccata remain elusive. METHODS: In the present work, 19 MbWOX family members were identified and characterized at the genome-wide level. Evolutionary analysis, cis-element prediction, transcriptome profiling and real-time quantitative PCR (

MdUGT88F1 enhances plant resistance to Fusarium proliferatum f.sp. malus domestica MR5 via root exudate-mediated assembly of disease-suppressive rhizosphere microbiota.

BACKGROUND: Apple replant disease (ARD) is a major threat to the sustainable development of China's apple industry. It is primarily caused by the accumulation of phloridzin and the pathogen Fusarium proliferatum f.sp. malus domestica MR5 (Fpmd MR5). MdUGT88F1-mediated phloridzin biosynthesis is known to enhance disease resistance, but its role in shaping the rhizosphere microbiome and conferring resistance against Fpmd MR5 remains unclear. In this study, we used wild-type (WT) and MdUGT88F1 transgenic apple lines to systematically investigate the mechanism by which MdUGT88F1 regulates the rhiz

Identification of Malus halliana R2R3-MYB gene family under iron deficiency stress and functional characteristics of MhR2R3-MYB4 in Arabidopsis thaliana.

Iron (Fe) is an essential element for plant growth and development. Fe deficiency can trigger leaf chlorosis and reduce fruit yield. Therefore, it is necessary to explore transcription factors in response to Fe deficiency stress. A total of 29 MhR2R3-MYB transcription factors were identified based on the transcriptome of Malus halliana under Fe deficiency stress. A comprehensive analysis of physical and chemical properties, gene structures, conserved motif composition, evolutionary relationship and chromosome distribution was performed. Subsequently, based on the transcriptome, 14 genes with t

Apple Extract (Malus sp.) and Rutin as Photochemopreventive Agents: Evaluation of Ultraviolet B-Induced Alterations on Skin Biopsies and Tissue-Engineered Skin.

The skin is exposed to the solar ultraviolet B (UVB) radiation, which leads to the formation of several types of skin damage responsible for cancer initiation and aging. Malus sp. is a genus of apples, which are a good source of polyphenolic compounds. Malus sp. and more precisely one of its components, rutin, have preventive effects on many diseases caused by reactive oxygen species. In addition, previous studies have suggested the topical usage of the extract as a cosmetic product to prevent skin damage caused by oxidative stress. In this study, we evaluated the efficacy of two topical formu

H2S Enhanced the Tolerance of Malus hupehensis to Alkaline Salt Stress through the Expression of Genes Related to Sulfur-Containing Compounds and the Cell Wall in Roots.

Malus is an economically important plant that is widely cultivated worldwide, but it often encounters saline-alkali stress. The composition of saline-alkali land is a variety of salt and alkali mixed with the formation of alkaline salt. Hydrogen sulfide (H2S) has been reported to have positive effects on plant responses to abiotic stresses. Our previous study showed that H2S pretreatment alleviated the damage caused by alkaline salt stress to Malus hupehensis Rehd. var. pingyiensis Jiang (Pingyi Tiancha, PYTC) roots by regulating Na+/K+ homeostasis and oxidative stress. In this study, transcri

Comprehensive genomic analysis of the TYROSINE AMINOTRANSFERASE (TAT) genes in apple (Malus domestica) allows the identification of MdTAT2 conferring tolerance to drought and osmotic stresses in plants.

Tyrosine aminotransferase (TAT, EC 2.6.1.5) is the first key enzyme that catalyzes the reversible interconversion of tyrosine and 4-hydroxyphenylpyruvate in the tyrosine-derived pathway for syntheses of important secondary metabolites and compounds. Although plant TAT genes have been proposed to be important in response to abiotic stress, there is little information about TAT genes in woody perennial tree species, especially in economic fruit trees. Based on TAT domain searching, sequence homology screening and phylogenetic analysis, we identified four TATs in apple genome. Then, we carried ou

Pulp Callus Extract From Malus domestica var. Mela Rosa Marchigiana Preserves Intestinal Barrier Integrity and Confers Neuroprotection in a Gut-Brain Axis Co-Culture Model.

The gut-brain axis is a key target in neuroinflammatory disorders. We investigated the protective effects of Mela Rosa Marchigiana pulp callus extract (MRME), a phytocomplex with a unique triterpenic profile. Using a validated transwell co-culture model of the intestinal-neural interface, differentiated Caco-2 cells formed a polarized epithelial barrier (apical), while BV2 microglia or SH-SY5Y neurons were seeded in the basolateral compartment. Apical MRME pretreatment preserved Caco-2 barrier integrity against lipopolysaccharide or dextran sodium sulfate-induced damage. MRME maintained occlud

Phyllobilomics: a comprehensive analysis of phyllobilins in apple (Malus × domestica Borkh.) leaves after fungal infections suggests a general role of the PaO/PB pathway in chlorotic infections.

INTRODUCTION: Chlorophyll (Chl) degradation via the pheophorbide a oxygenase/phyllobilin (PaO/PB) pathway produces linear tetrapyrrolic phyllobilins (PBs) during leaf senescence and infection by certain pathogens, such as phytoplasmas. Since leaf yellowing, or chlorosis, is a common symptom of fungal disease in apple (Malus × domestica Borkh.) and other plants, PB formation was analyzed during four fungal diseases to evaluate the general relevance of the PaO/PB pathway during infection.
