Endocrine-metabolic effects of kisspeptin in mammals.

Kisspeptins (Kp) play crucial roles in regulating reproductive functions. In recent decades, new effects of Kp on other systems, such as the cardiovascular, respiratory, and musculoskeletal systems, and its possible therapeutic effects on gestational hypothyroidism, diabetes mellitus, and neoplasms have been identified. Additionally, Kp signaling has been investigated in endocrine-metabolic regulation; nonetheless, there are few reviews on this subject. This study aims to provide a comprehensive overview of the effects of Kp on endocrine-metabolic physiology in mammals. Kp modulates the releas

Multiscale Regulation of Meiotic Recombination in Mammals.

Meiotic recombination is a multilayered process that shapes genetic diversity while safeguarding genome stability. In mammals, the final distribution of crossovers and non-crossovers emerges from the integration of sequence-specific hotspot specification, chromosome architecture, replication timing, and crossover patterning mechanisms. PRDM9 directs most double-strand break formation to short (typically 1-2 kb), rapidly evolving hotspots by depositing histone marks and remodeling chromatin, whereas in PRDM9-independent contexts, breaks are redirected toward accessible promoters and CpG islands

Molecular genetics of meiotic initiation in mammals.

Mammalian fertility is dependent upon meiosis, a specialized cell division by which diploid progenitors undergo one round of DNA replication followed by two rounds of chromosomal segregation to produce haploid gametes. The germ line in the testis and ovary undergoes several rounds of mitotic divisions before ultimately transitioning to the meiotic cell cycle. This transition is achieved by replacing the mitotic cell cycle program with the meiotic one. Here, we discuss the molecular players that regulate the transition from mitosis to meiosis. In spermatogenesis, MEIOC, YTHDC2, and RBM46 form a

Histoplasma capsulatum in wild mammals from Ecuador.

BACKGROUND: Histoplasma capsulatum is a globally distributed dimorphic fungal pathogen endemic to the Americas, Africa, and Asia. It causes histoplasmosis, a disease acquired via inhalation of spores from contaminated environments. It thrives in nitrogen-rich soils and is disseminated by avian and chiropteran reservoirs. Histoplasma capsulatum has been found in wild mammals such as rodents, marsupials, felines, and xenarthrans, suggesting diverse reservoirs that may influence its maintenance and transmission in endemic areas. This study aimed to detect H. capsulatum in tissues of wild small ma

Hallmarks of uterine receptivity predate placental mammals.

Embryo implantation requires tightly coordinated signaling between the blastocyst and the endometrium, and is crucial for the establishment of a uteroplacental unit that persists until term in eutherian mammals. In contrast, marsupials, with a unique life cycle and short gestation, make only brief fetal-maternal contact and lack implantation. To better understand the evolutionary link between eutherian implantation and its ancestral equivalent in marsupials, we compare single-cell transcriptomes from the receptive and non-receptive endometrium of the mouse and guinea pig with that of the oposs

The global H5N1 influenza panzootic in mammals.

Influenza A viruses have caused more documented global pandemics in human history than any other pathogen1,2. High pathogenicity avian influenza viruses belonging to the H5N1 subtype are a leading pandemic risk. Two decades after H5N1 'bird flu' became established in poultry in Southeast Asia, its descendants have resurged3, setting off a H5N1 panzootic in wild birds that is fuelled by: (1) rapid intercontinental spread, reaching South America and Antarctica for the first time4,5; (2) fast evolution via genomic reassortment6; and (3) frequent spillover into terrestrial7,8 and marine mammals9.

The molecular evolution of spermatogenesis across mammals.

The testis produces gametes through spermatogenesis and evolves rapidly at both the morphological and molecular level in mammals1-6, probably owing to the evolutionary pressure on males to be reproductively successful7. However, the molecular evolution of individual spermatogenic cell types across mammals remains largely uncharacterized. Here we report evolutionary analyses of single-nucleus transcriptome data for testes from 11 species that cover the three main mammalian lineages (eutherians, marsupials and monotremes) and birds (the evolutionary outgroup), and include seven primates. We find

Molecular evolutionary insights from PRLR in mammals.

Prolactin (PRL) is a pleiotropic neurohormone secreted by the mammalian pituitary gland into the blood, thus reaching many tissues and organs beyond the brain. PRL binds to its receptor, PRLR, eliciting a molecular signaling cascade. This system modulates essential mammalian behaviors and promotes notable modifications in the reproductive female tissues and organs. Here, we explore how the intracellular domain of PRLR (PRLR-ICD) modulates the expression of the PRLR gene. Despite differences in the reproductive strategies between eutherian and metatherian mammals, there is no clear distinction

Exercise-Induced Adult Cardiomyocyte Proliferation in Mammals.

Loss of cardiomyocytes is a vital manifestation and predisposing factor of many cardiovascular diseases and will eventually lead to heart failure (HF). On the other hand, adult mammalian cardiomyocytes have a very limited regenerative capacity and cannot achieve self-repair of the myocardium after injury. Therefore, it is necessary to promote regeneration and repair of the myocardium through effective intervention means. Exercise plays an important role in the prevention and rehabilitation of cardiovascular diseases. Exercise can improve ischemia-reperfusion injury, reduce the size of the infa

Embryo-Maternal Interactions Underlying Reproduction in Mammals.

This Special Issue, "Embryo-Maternal Interactions Underlying Reproduction in Mammals", gathers a collection of 23 articles, 16 original research articles and 7 up-to-date reviews, providing new findings or summarizing current knowledge on embryo-maternal interactions in seven different mammalian species including humans. Considering the different players involved in these embryo-maternal interactions, articles are mainly focused on one of these different players: the oviduct, the uterus, the embryo or the emergent extracellular vesicles. Additionally, a few articles bring up the impact of repr

Amino acid dependent formaldehyde metabolism in mammals.

Aldehyde dehydrogenase class 3, encoded by ADH5 in humans, catalyzes the glutathione dependent detoxification of formaldehyde. Here we show that ADH5 deficient cells turn over formaldehyde using alternative pathways starting from the reaction of formaldehyde with free amino acids. When mammalian cells are exposed to formaldehyde, the levels of the reaction products of formaldehyde with the amino acids cysteine and histidine - timonacic and spinacine - are increased. These reactions take place spontaneously and the formation of timonacic is reversible. The levels of timonacic are higher in the

Diet and Hydrogen Sulfide Production in Mammals.

Significance: In recent times, it has emerged that some dietary sulfur compounds can act on mammalian cell signaling systems via their propensity to release hydrogen sulfide (H2S). H2S plays important biochemical and physiological roles in the heart, gastrointestinal tract, brain, kidney, and immune systems of mammals. Reduced levels of H2S in cells and tissues correlate with a spectrum of pathophysiological conditions, including heart disease, diabetes, obesity, and altered immune function. Recent Advances: In the last decade, researchers have now begun to explore the mechanisms by which diet

Hemozoin produced by mammals confers heme tolerance.

Free heme is cytotoxic as exemplified by hemolytic diseases and genetic deficiencies in heme recycling and detoxifying pathways. Thus, intracellular accumulation of heme has not been observed in mammalian cells to date. Here we show that mice deficient for the heme transporter SLC48A1 (also known as HRG1) accumulate over ten-fold excess heme in reticuloendothelial macrophage lysosomes that are 10 to 100 times larger than normal. Macrophages tolerate these high concentrations of heme by crystallizing them into hemozoin, which heretofore has only been found in blood-feeding organisms. SLC48A1 de

Research advances on embryonic diapause in mammals.

Embryonic diapause occurs in over 130 species of mammals. In embryonic diapause, there is a reversible cessation phase in embryo development and subsequent reactivation and continuation of development with no adverse effects. This allows animals to prolong gestation and give birth to offspring at a more favorable time from a survival perspective. Embryonic diapause is divided into two types: facultative diapause which is induced by physiological conditions and obligate diapause which is induced by season of the year. The process of embryonic diapause includes three phases: onset, maintenance,

Diaphragm: A vital respiratory muscle in mammals.

The diaphragm is a respiratory muscle that is primarily responsible for the respiratory function in normal individuals. In mammals, the diaphragm muscle has been studied from the early days of zoology, comparative and experimental anatomy, physiology, medicine, physics, and philosophy. However, even with these early advances in knowledge pertaining to the diaphragm, comprehensive morphological data on the diaphragm are still incomplete. In this review, we summarize the beginnings of the morphological description of the diaphragm, and we describe the current status of the known morphological an

Circannual Testis Changes in Seasonally Breeding Mammals.

In the non-equatorial zones of the Earth, species concentrate their reproductive effort in the more favorable season. A consequence of seasonal breeding is seasonal testis regression, which implies the depletion of the germinative epithelium, permeation of the blood-testis barrier, and reduced androgenic function. This process has been studied in a number of vertebrates, but the mechanisms controlling it are not yet well understood. Apoptosis was assumed for years to be an important effector of seasonal germ cell depletion in all vertebrates, including mammals, but an alternative mechanism has

Highly athletic terrestrial mammals: horses and dogs.

Evolutionary forces drive beneficial adaptations in response to a complex array of environmental conditions. In contrast, over several millennia, humans have been so enamored by the running/athletic prowess of horses and dogs that they have sculpted their anatomy and physiology based solely upon running speed. Thus, through hundreds of generations, those structural and functional traits crucial for running fast have been optimized. Central among these traits is the capacity to uptake, transport and utilize oxygen at spectacular rates. Moreover, the coupling of the key systems--pulmonary-cardio

[Micro RNA and viral infections in mammals].

RNA silencing plays an important role in development through the action of micro (mi) RNAs that fine tune the expression of a large portion of the genome. But, in plants and insects, it is also a very important player in innate immune responses, especially in antiviral defense. It is now well established that the RNA silencing machinery targets plant as well as insect viruses. While the genetic basis underlying this defense mechanism in these organisms starts being elucidated, much less is known about the possible antiviral role of RNA silencing in mammals. In order to identify siRNAs coming f

Male contraceptive technology for nonhuman male mammals.

Contraceptive techniques applied to males have potential to mitigate diverse instances of overpopulation in human and animal populations. Different situations involving different species dictate that there is no ideal male contraceptive, and emphasizes the value of varying approaches to reducing male fertility. A majority of work in this field has focused on non-surgically destroying the testes or obstructing the epididymis, and suppressing gonadotropin secretion or inducing immune responses to reproductive hormones or sperm-specific antigens.
