Aboveground herbivory induced jasmonates disproportionately reduce plant reproductive potential by facilitating root nematode infestation.

Different plant feeders, including insects and parasitic nematodes, can influence each other by triggering systemic changes in their shared host plants. In most cases, however, the underlying mechanisms are unclear, and the consequences for plant fitness are not well understood. We studied the interaction between leaf feeding Manduca sexta caterpillars and root parasitic nematodes in Nicotiana attenuata. Simulated M. sexta attack increased the abundance of root parasitic nematodes in the field and facilitated Meloidogyne incognita reproduction in the glasshouse. Intact jasmonate biosynthesis w

Identification of ecdysis-triggering hormone from an epitracheal endocrine system.

Developing insects repeatedly shed their cuticle by means of a stereotyped behavior called ecdysis, thought to be initiated by the brain peptide eclosion hormone. Here an ecdysis-triggering hormone, Mas-ETH, is described from the tobacco hornworm Manduca sexta. Mas-ETH contains 26 amino acids and is produced by a segmentally distributed endocrine system of epitracheal glands (EGs). The EGs undergo a marked reduction in volume, appearance, and immunohistochemical staining during ecdysis, at which time Mas-ETH is found in the hemolymph. Injection of EGs extract or synthetic Mas-ETH into pharate

Structural and chemical properties of insect's chitin-containing extracellular matrices.

Insect's body barriers rely on specialized extracellular matrices that protect against harmful environmental influences. The outer barrier is the cuticle, which is composed of chitin, cuticle proteins and lipids. The peritrophic matrix (PM) serves as an inner barrier lining the midgut epithelium. It is composed of chitin fibers that are organized by PM proteins. While cuticle and PM proteins have received considerable attention in the past, supramolecular organization and physicochemical properties of the chitin component - particularly of the PM - remain poorly understood. Here, we combine sy

LC and LD50 values of Bacillus thuringiensis Serovar japonensis strain buibui toxin to Oriental beetle and northern masked chafer larvae (Coleoptera: Scarabaeidae).

Bacillus thuringiensis serovar japonensis strain Buibui has the potential to be an important control agent for pest scarabs. Bioassays were designed to test B. t. japonensis against two of the major turf and ornamental scarab pests infesting turfgrasses and ornamentals and to serve as a basis for further tests against other scarab pests. LC and LD50 values of B. t. serovarjaponensis strain Buibui toxin and spores were determined by four different bioassays for the oriental beetle, Anomala orientalis (Waterhouse), and northern masked chafer, Cyclocephala borealis Arrow. Oriental beetle larvae w

Caterpillar Climbing: Robust, Tension-Based Omni-Directional Locomotion.

Animals that must transition from horizontal to inclined or vertical surfaces typically change their locomotion strategy to compensate for the relative shift in gravitational forces. The species that have been studied have stiff articulated skeletons that allow them to redistribute ground reaction forces (GRFs) to control traction. Most also change their stepping patterns to maintain stability as they climb. In contrast, caterpillars, most of which are highly scansorial, soft-bodied, and lack rigid support or joints, can move with the same general kinematics in all orientations. In this study,

The neural bases of host plant selection in a Neuroecology framework.

Understanding how animals make use of environmental information to guide behavior is a fundamental problem in the field of neuroscience. Similarly, the field of ecology seeks to understand the role of behavior in shaping interactions between organisms at various levels of organization, including population-, community- and even ecosystem-level scales. Together, the newly emerged field of "Neuroecology" seeks to unravel this fundamental question by studying both the function of neurons at many levels of the sensory pathway and the interactions between organisms and their natural environment. EPR Studies of V-ATPase with Spin-Labeled Inhibitors DCC and Archazolid: Interaction Dynamics with Proton Translocating Subunit c.

Vacuolar-type H(+) -ATPases (V-ATPases) have gained recent attention as highly promising anticancer drug targets, and therefore detailed structural analyses and studies of inhibitor interactions are very important research objectives. Spin labeling of the V-ATPase holoenzyme from the tobacco hornworm Manduca sexta and V-ATPase in isolated yeast (Saccharomyces cerevisiae) vacuoles was accomplished by two novel methods involving the covalent binding of a (2,2,6,6-tetramethylpiperidin-1-yl)oxyl (TEMPO) derivative of N,N'-dicyclohexylcarbodiimide (DCC) to the essential glutamate residue in the act

BmSerpin-7 negatively regulates prophenoloxidase activation and antimicrobial peptide synthesis in Bombyx mori.

The prophenoloxidase (PPO) activation pathway and the Toll-mediated antimicrobial peptide (AMP) synthesis pathway constitute critical immune responses within the insect immune system. The activation of these pathways is driven by extracellular serine protease cascades, a process counterbalanced by the negative regulation of serpins. In this study, we identified a typical serpin, BmSerpin-7, and characterized its function in the silkworm, Bombyx mori. BmSerpin-7 expression was up-regulated in the hemocytes but down-regulated in the fat body after Micrococcus luteus and Yersinia pseudotuberculos

In vitro analyses of interactions between olfactory receptor growth cones and glial cells that mediate axon sorting and glomerulus formation.

During development, the axons of olfactory receptor neurons project to the CNS and converge on glomerular targets. For vertebrate and invertebrate olfactory systems, neuron-glia interactions have been hypothesized to regulate the sorting and targeting of olfactory receptor axons and the development of glomeruli. In the moth Manduca sexta, glial reduction experiments have directly implicated two types of central olfactory glia, the sorting zone- and neuropil-associated glia, in key events in olfactory development, including axon sorting and glomerulus stabilization. By using cocultures containi

Behavioral responses to host foodplants of two populations of the insect parasitoid Cotesia congregata (Say).

To test the hypothesis that natural enemy populations differ in their behavioral responses to plants or to plant allelochemicals, we compared two populations of the gregarious larval endoparasitoid, Cotesia congregata (Say) (Hymenoptera: Braconidae) that differed in their historical and present exposure to tobacco. The major hosts for both populations were Manduca sexta L. and M. quinquemaculata (Haworth) (Lepidoptera: Sphingidae), but these hosts were typically encountered on tobacco by parasitoids in one population (Upper Marlboro) and on tomato by parasitoids in another population (Wye). Polyamines modulate multiple protein phosphorylation pathways in the insect prothoracic gland.

Multiple endogenous substrates phosphorylated by four distinct protein kinases were identified in particulate and cytosolic fractions from the larval prothoracic gland of the tobacco hornworm, Manduca sexta. Three prominent particulate-associated phosphoprotein substrates (19, 21, and 34 kDa) were of particular interest. The in vitro phosphorylation of the 19 and 21 kDa peptides was markedly enhanced by cAMP, Ca2+/calmodulin, as well as Ca2+/phospholipids, presumably via cAMP-dependent protein kinase (cAMP-PK), Ca2+/calmodulin-dependent protein kinase (Ca2+/CaM-PK), and protein kinase C (PKC),

Integration of endocrine signals that regulate insect ecdysis.

The extremely large number of insects and members of allied groups alive today suggests that molting--shedding of an old cuticle--may be one of the most commonly performed behaviors on our planet. Removal of an old cuticle in insects is associated with stereotyped, species-specific patterns of behavior referred to as ecdysis. It has been recognized for decades that the initiation of ecdysis is under hormonal control, but until recently many of the key peptides that regulate ecdysis were unknown. The report in 1996 of a new ecdysis-triggering hormone (ETH) sparked an era of significant advances

Insect cuticle, an in vivo model of protein trafficking.

In the course of this study more than 20 proteins have been isolated from the larval cuticle of Manduca sexta. Synthesis, secretion, transport and accumulation of four particular proteins, representative members of four characteristic groups, were followed during metamorphosis by immunoblot and immuncytochemical methods and are described in detail in this paper. We established that only some of the proteins of the soft cuticle of Lepidopteran larvae are synthesized in epidermal cells at the beginning of the larval stages and are digested during the moulting period (MsCP29). Other proteins (MsC

Proteomic analysis of dimorphic sperm in the cabbage white butterfly, Pieris rapae.

Sperm dimorphism, the production of two distinct sperm morphs by a single male, is a widespread but enigmatic reproductive phenomenon. In Lepidoptera, fertilizing eupyrene sperm coexist with anucleate apyrene sperm, which cannot fertilize eggs but are nevertheless required for successful reproduction. Despite the prevalence and presumed adaptive significance of sperm dimorphism, the molecular basis of this trait remains limited. Here, we characterize the proteome of dimorphic sperm in the Cabbage White butterfly, Pieris rapae, an emerging model for sexual selection and postcopulatory interacti

Herbivory rapidly activates MAPK signaling in attacked and unattacked leaf regions but not between leaves of Nicotiana attenuata.

Mitogen-activated protein kinase (MAPK) signaling plays a central role in transducing extracellular stimuli into intracellular responses, but its role in mediating plant responses to herbivore attack remains largely unexplored. When Manduca sexta larvae attack their host plant, Nicotiana attenuata, the plant's wound response is reconfigured at transcriptional, phytohormonal, and defensive levels due to the introduction of oral secretions (OS) into wounds during feeding. We show that OS dramatically amplify wound-induced MAPK activity and that fatty acid-amino acid conjugates in M. sexta OS are

Increases in cyclic 3', 5'-guanosine monophosphate (cGMP) occur at ecdysis in an evolutionarily conserved crustacean cardioactive peptide-immunoreactive insect neuronal network.

At the end of each instar, insects shed their old cuticle by performing the stereotyped ecdysis behavior. In the month, Manduca sexta, larval ecdysis is accompanied by increases in intracellular cyclic 3', 5'-guanosine monophosphate (cGMP) in a small network of 50 peptidergic neurons within the ventral central nervous system (CNS). Studies on a variety of insects show that this cGMP response has been associated with ecdysis throughout most of insect evolution. In the mealbeetle (Tenebrio, Coleoptera) and the mosquito (Aedes, Diptera), all 50 neurons showed increases in cGMP immunoreactivity (-

A herbivore-induced salicylic acid carboxyl methyl transferase produces methyl salicylate in tomato to mediate defense signaling and deter pests.

Methyl salicylate (MeSA) is a component of the herbivore-induced volatile blend of tomato (Solanum lycopersicum), and an intermediate in long distance salicylic acid (SA) signaling. To investigate the role of MeSA in interactions between tomato and the lepidopteran pests Helicoverpa zea and Manduca sexta, we used transgenic tomato lines that were antisense (AS) or overexpressors (OE) of the tomato SA carboxyl methyl transferase SlSAMT1, which under- or over-produce MeSA, respectively. Transgenic AS plants produced less MeSA and were more susceptible to both insects compared to wild-type plants

Bacillus thuringiensis: a natural endophytic bacterium found in wild plants.

Despite the fact that Bacillus thuringiensis is the most widely used bacterium in biological pest control, its ecology has been notoriously neglected. Its role in nature is uncertain, and a defined habitat and niche are under discussion. In this report, wild-type strains were isolated from the inner plant tissues as natural endophytic bacteria in wild plants. Once a reliable superficial sterilization technique was standardized, leaf samples from 110 wildlife plant species within 52 families were processed to obtain their endophytic microflora, which were able to grow in artificial media. Biotin-containing proteins of the insect nervous system, a potential source of interference with immunocytochemical localization procedures.

When the biotinylated Manduca sexta adipokinetic hormone gene was used as a probe for in situ hybridization, the intrinsic neurosecretory cells were stained with a biotin detection system that contained streptavidin or avidin. Further experiments showed that the DNA probe was not necessary for staining these cells by streptavidin-alkaline phosphatase, and that they were not stained by alkaline phosphatase alone. Similarly, the intrinsic neurosecretory cells were stained directly by streptavidin conjugated to a fluorescent dye. Other parts of the central nervous system could also be stained wit
