Acid-base combination principles for preparation of anti-acne dissolving microneedles loaded with azelaic acid and matrine.

To overcome the poor solubility, skin irritation, and low permeability of azelaic acid (AZA) existed on the marketed formulations, a co-drug principle via matrine (MAT) was adopted to prepare anti-acne dissolving microneedles (DMNs). The formula was optimized according to the solubility and antibacterial activity of novel ionic salt. The results indicated solubilization of AZA could be achieved at a molar ratio between AZA and MAT was 1:1. Meanwhile, synergistic antibacterial and anti-irritative properties were acquired. The matrix materials were composed of sodium carboxymethyl cellulose (CMC

pY RNA1-s2: a highly retina-enriched small RNA that selectively binds to Matrin 3 (Matr3).

The purpose of this study was to expand our knowledge of small RNAs, which are known to function within protein complexes to modulate the transcriptional output of the cell. Here we describe two previously unrecognized, small RNAs, termed pY RNA1-s1 and pY RNA1-s2 (processed Y RNA1-stem -1 and -2), thereby expanding the list of known small RNAs. pY RNA1-s1 and pY RNA1-s2 were discovered by RNA sequencing and found to be 20-fold more abundant in the retina than in 14 other rat tissues. Retinal expression of pY RNAs is highly conserved, including expression in the human retina, and occurs in all

Matrine enhances cisplatin sensitivity in oral squamous cell carcinoma through regulation of oxidative stress and apoptosis.

Cisplatin resistance remains a major limitation in the treatment of oral squamous cell carcinoma (OSCC), often associated with altered oxidative stress. This study aimed to evaluate whether matrine can enhance cisplatin sensitivity in OSCC cells and explore its potential underlying mechanisms. Bioinformatics analysis and molecular docking were used to predict potential targets and pathways. In vitro experiments were conducted using CAL27 and cisplatin-resistant CAL27/DDP cells. Cell viability was assessed by CCK-8 assay, and drug interaction was evaluated using combination index (CI). Colony f

Matrine Attenuates Neurological Deficits and Neuroinflammation by Inhibiting the HMGB1/RAGE Axis and Ferroptosis in Intracerebral Hemorrhage Mice.

Intracerebral hemorrhage (ICH) is a life-threatening stroke subtype that lacks effective drug therapy. In the context of ICH, neuroinflammation, and ferroptosis are key contributors to secondary brain injury. In this study, we assessed the neuroprotective effects of matrine (MAT) and its mechanism of action involving the HMGB1/RAGE axis in an ICH mouse model. ICH was induced in mice by autologous blood injection. MAT (1.5 or 20 mg/kg) was administered orally 5 min postinduction. ICH significantly upregulated RAGE and its associated proteins (HMGB1, protein kinase C [PKC], phospholipase C [PLC]

Matrine induces autophagic cell death by triggering ROS/AMPK/mTOR axis and apoptosis in multiple myeloma.

Despite significant advancements in multiple myeloma (MM) treatment in recent years, most patients will eventually develop resistance or experience relapse. Matrine, a primary active compound of traditional Chinese medicinal herb Sophora flavescens Ait, has been found to have anti-tumor properties in various types of malignant tumors. Whether autophagy plays a crucial role in the anti-MM effect of matrine remain unknown. Herein, we found that matrine could trigger apoptosis and cell cycle arrest, and meanwhile induce autophagy in MM cells in vitro. We further ascertained the role of autophagy

Matrine suppresses lipopolysaccharide-induced fibrosis in human peritoneal mesothelial cells by inhibiting the epithelial-mesenchymal transition.

BACKGROUND: Peritoneal fibrosis is the primary reason that patients with end-stage renal disease (ESRD) have to cease peritoneal dialysis. Peritonitis caused by Gram-negative bacteria such as Escherichia coli (E. coli) were on the rise. We had previously shown that matrine inhibited the formation of biofilm by E. coli. However, the role of matrine on the epithelial-mesenchymal transition (EMT) in peritoneal mesothelial cells under chronic inflammatory conditions is still unknown. METHODS: We cultured human peritoneal mesothelial cells (HPMCs) with lipopolysaccharide (LPS) to induce an environ

A novel matrine derivative exerts antitumor effects against hepatocellular carcinoma by inducing oxidative stress and DNA damage.

DNA has long been a major target in the development of anticancer drugs. Natural products, as an important source of antitumor agents, play a key role in cancer prevention and treatment. Matrine is a natural quinoline alkaloid isolated from the medicinal plant Sophora flavescens. In this study, using matrine as a lead compound, we designed and synthesized 25 novel matrine derivatives containing a naphthalimide moiety based on a molecular hybridization strategy. The antiproliferative activities of these derivatives were evaluated against several human cancer cell lines (Hela, A549, HepG2 and MH

Matrine attenuates heterotopic ossification by suppressing TGF-β induced mesenchymal stromal cell migration and osteogenic differentiation.

PURPOSE OF THE STUDY: Heterotopic ossification (HO) is a debilitating disease characterized by extraskeletal bone formation. Active TGF-β recruits mesenchymal stromal cells (MSCs), which contribute to trauma-induced HO. Inhibiting TGF-β induced MSC migration and osteogenic differentiation could be a promising treatment for HO. Matrine is an alkaloid from the genus Sophora that can suppress pancreatic and hepatic fibrosis by regulating TGF-β/Smad signaling. We conducted this study to evaluate the effects of matrine on HO and explore the mechanisms, we carried out this study. MATERIALS AND METH

Novel matrine derivative MD-1 attenuates hepatic fibrosis by inhibiting EGFR activation of hepatic stellate cells.

Matrine (MT), the effective component of Sophora flavescens Ait, has been shown to have anti-inflammation, immune-suppressive, anti-tumor, and anti-hepatic fibrosis activities. However, the pharmacological effects of MT still need to be strengthened due to its relatively low efficacy and short half-life. In the present study, we report a more effective thio derivative of MT, MD-1, and its inhibitory effects on the activation of hepatic stellate cells (HSCs) in both cell culture and animal models. Cytological experiments showed that MD-1 can inhibit the proliferation of HSC-T6 cells with a half
