Medetomidine, an alpha 2-agonist, alleviates post-thoracotomy pain in dogs.

Twelve laboratory beagles underwent a routine left thoracotomy to insert permanent instrumentation. Every second dog was given 10 micrograms/kg of medetomidine, an alpha 2-agonist sedative. The rest of the animals were treated with 20 micrograms/kg of buprenorphine, an opioid agonist-antagonist, which is regularly used to treat postoperative pain in laboratory animals. The drugs were given at the end of operation (0) and 4, 8, 20, and 24 h postoperatively. Blood samples for catecholamines (adrenaline and noradrenaline) and blood gases (pCO2 and pO2) and pH were drawn immediately before any dru

A Case of Maternal and Neonatal Withdrawal After Exposure to Fentanyl Adulterated With Medetomidine.

The emergence of medetomidine, a potent veterinary alpha-2 adrenergic receptor agonist, as a common fentanyl adulterant in many areas of the United States poses new threats to pregnant women who use illicit opioids and their neonates. We present a case of severe withdrawal seen in a postpartum woman and her neonate after exposure to fentanyl adulterated with medetomidine up to the time of delivery. The case demonstrates the need for obstetric and pediatric providers to be aware of this emerging trend to allow for early recognition and prompt treatment of the potentially life-threatening sympto

Medetomidine-Associated Opioid Overdoses in Chicago, Illinois: A Report of 3 Cases.

BACKGROUND: Medetomidine has recently emerged as an adulterant in street-level opioids and stimulants in North America, complicating opioid overdose presentations and resisting reversal with naloxone. Medetomidine is an alpha-2 adrenergic receptor agonist used in veterinary medicine as an anesthetic, analgesic, anxiolytic, and muscle relaxant and is not approved for human use by the Food and Drug Administration. Medetomidine's pharmacological profile resembles xylazine, another medication of the same class, but demonstrates higher potency and longer sedative effects in canine models. CASE SUM

In sheep undergoing general anaesthesia does inclusion of medetomidine result in hypoxaemia?

PICO QUESTION: In healthy adult female non-pregnant sheep undergoing general anaesthesia for research studies does the inclusion of intravenous medetomidine as part of the anaesthetic protocol cause hypoxaemia? CATEGORY OF RESEARCH: Treatment. NUMBER AND TYPE OF STUDY DESIGNS REVIEWED: Four papers were available for critical appraisal. The quality of the evidence is weak. There were four experimental studies; three of which were cross-over studies and one study which was run in parallel with primary orthopaedic research. None of the studies were specifically focussed on the potential hypoxae

PROTOCOL FOR HUMAN EXPOSURE TO OPIOIDS AND CONCENTRATED MEDETOMIDINE USED IN FIELD APPLICATIONS.

Wildlife professionals routinely use potent sedatives and anesthetics when chemically immobilizing wildlife and zoo species in remote environments. Accidental exposure to these prescription veterinary drugs is rare but could be rapidly fatal. Commonly used agents include opioids and α2 adrenoreceptor agonists. These drugs can be reversed with specific antagonists; however, they are often not approved for human use. The protocol created here can be used by wildlife health professionals in a field setting with basic human emergency medical response training in coordination with local Emergency M

Ketamine-butorphanol-medetomidine for the immobilisation of free-living hyenas (Crocuta crocuta).

Free-ranging spotted hyenas (Crocuta crocuta) are immobilised for a variety of purposes, including wildlife-human conflict mitigation, research, and veterinary treatment. Combinations of tiletamine-zolazepam (Zoletil) and medetomidine are commonly used for immobilisation of hyenas, however, recovery times are long. In this descriptive study, a total of 20 adult or subadult free-ranging hyenas were immobilised near Skukuza in the Kruger National Park using ketamine, butorphanol, and medetomidine. The goal of the study was to evaluate a suitable dose and measure cardiorespiratory effects of this

Emergence of Medetomidine in the Unregulated Drug Supply and Its Association With Hallucinogenic Effects.

INTRODUCTION: The unregulated drug supply in the United States is rapidly evolving, and veterinary tranquillisers have emerged as adulterants of concern, especially in illicitly-manufactured fentanyl. Following the proliferation of xylazine, medetomidine, a more potent sedative, has recently appeared in multiple US states. This study describes the characteristics of medetomidine samples from a national mail-based drug checking program and aims to determine whether medetomidine is associated with hallucinogenic effects. METHODS: We conducted a retrospective analysis of 11,363 drug samples betw

Evaluation of a mixture of medetomidine, alfaxalone and butorphanol as an alternative drug for euthanasia in mice.

Euthanasia agents should induce a rapid and painless loss of consciousness, followed by cardiopulmonary arrest and subsequent brain death. Injectable drugs such as pentobarbital sodium are commonly used for laboratory rodents due to their quick and smooth action. However, the discontinuation of pharmaceutical-grade pentobarbital sodium and secobarbital sodium in Japan, along with a global shortage of pentobarbital in late 2020, has increased the demand for new injectable euthanasia drugs. In Japan, the combination of medetomidine, midazolam, and butorphanol (MMB), as well as a newer formulatio

Recommended doses of medetomidine-midazolam-butorphanol with atipamezole for preventing hypothermia in mice.

A non-narcotic anesthetic combination (Me/Mi/Bu) of medetomidine (Me), midazolam (Mi), and butorphanol (Bu) has been recommended as the injectable anesthesia in mice. An original dose of Me/Mi/Bu (0.3/4.0/5.0 mg/kg) has provided sufficient anesthetic duration of 40-50 min in mice. In addition, atipamezole is available for reversal of Me/Mi/Bu anesthesia. As an adverse effect of Me/Mi/Bu anesthesia, however, severe hypothermia has been also observed in mice. In the present study, we investigated 1) the main agent in Me/Mi/Bu to cause of hypothermia, 2) the effects of the differential doses of a
