Cyclodextrin and Meglumine-Based Microemulsions as a Poorly Water-Soluble Drug Delivery System.

Cyclodextrins (CDs) and meglumine (MEG) are pharmaceutical excipients widely used to improve solubility of poorly water-soluble drugs. The purpose of this work was to study the effect of CDs or MEG on the internal microstructure of soya oil-based O/W microemulsions (MEs) and on the modulation of the solubility and release rate of Class II model hydrophobic drugs, sulfamerazine and indomethacin. The pseudoternary phase diagrams revealed that higher proportions of oil phase, as well as the presence of β-cyclodextrin (ßCD), methyl-ßCD, and MEG, favored the incorporation of the drugs. The conducti

Experimental evaluation of iosefamate meglumine and its derivatives as hepatobiliary CT contrast agents.

Iosefamate meglumine has attracted attention as a possible hepatobiliary contrast agent for CT scanning. However, its limited hepatic opacification has prevented clinical acceptance. To find a more efficient agent, the efficacy and toxicity of six derivatives of iosefamate were compared with those of the parent compound in dogs. Twenty dogs received intravenous doses ranging from 150 to 600 mg I/kg of one of these seven water-soluble, ionic, dimeric agents. Three control animals received equivalent amounts of physiologic saline. The CT densities of liver, biliary tract, kidneys, and blood were

Comparative double-blind investigation of meglumine metrizoate, metrizamide, and iohexol in carotid angiography.

The new nonionic contrast medium iohexol (Omnipaque) was compared with its predecessor metrizamide (Amipaque) and with the conventional ionic medium meglumine metrizoate (Isopaque Cerebral) in carotid angiography using a double-blind crossover technique. The results indicated that iohexol and metrizamide caused less discomfort than the ionic medium. The circulatory effects of the three media were generally mild, and the diagnostic effectiveness was comparable when the iodine concentration was kept in the range of 280-300 mg I/ml.

Synthesis of 14C-meglumine salicylate and its disposition in humans after oral administration.

The synthesis of 14C-meglumine salicylate was accomplished by heating 14C-meglumine with salicylic acid, in equimolar ratios, in 2-propanol. The average radiochemical yield was 97.5%. Ten healthy adult male volunteers were given 1.2 g of the compound orally. Five took 1.2 g of 1-deoxy-1-[14C]-methylamino-D-glucitol salicylate (containing about 47 micronCi), and five others took 1.2 g of 1-deoxy-1-methylamino-D-[U-14 micronCi), and five others took 1.2 g of 1-deoxy-1-methylamino-D-[U-14C]-glucitol salicylate (containing about 45 micronCi). Detection of flunixin meglumine in pork based on magnetic solid-phase extraction technology.

This study established two methods for detecting flunixin meglumine (FM) in pork. The MSPE-LC-MS/MS method utilizes the accuracy and quantifiability of liquid chromatography-tandem mass spectrometry. FM in pork can be accurately analyzed after optimizing the pretreatment, liquid phase conditions, and mass spectrometry parameters. It shows a good linear relationship (correlation coefficient > 0.999), with a detection limit of 0.2 μg/kg, limit of quantification of 0.6 μg/kg, spiked recovery rates of 90.6 % - 97.4 %, matrix effects of 96.7 % - 107.2 %, intra-day precision of 1.4 % - 5.6 %, and in

Immuno-pharmacokinetics of Meglumine Antimoniate in Patients With Cutaneous Leishmaniasis Caused by Leishmania (Viannia).

BACKGROUND: Control of cutaneous leishmaniasis (CL) relies on chemotherapy, yet gaps in our understanding of the determinants of therapeutic outcome impede optimization of antileishmanial drug regimens. Pharmacodynamic (PD) parameters of antimicrobials are based on the relationship between drug concentrations/exposure and microbial kill. However, viable Leishmania persist in a high proportion of individuals despite clinical resolution, indicating that determinants other than parasite clearance are involved in drug efficacy. METHODS: In this study, the profiles of expression of neutrophils, mo
