Homotypic fusion of ER membranes requires the dynamin-like GTPase atlastin.

Establishment and maintenance of proper architecture is essential for endoplasmic reticulum (ER) function. Homotypic membrane fusion is required for ER biogenesis and maintenance, and has been shown to depend on GTP hydrolysis. Here we demonstrate that Drosophila Atlastin--the fly homologue of the mammalian GTPase atlastin 1 involved in hereditary spastic paraplegia--localizes on ER membranes and that its loss causes ER fragmentation. Drosophila Atlastin embedded in distinct membranes has the ability to form trans-oligomeric complexes and its overexpression induces enlargement of ER profiles,

In vitro fusion of plant Golgi membranes can be influenced by divalent cations.

The fusogenic activity of plant Golgi membranes was studied in a cell-free system by assaying lipid mixing and content leakages of fluorescence probes. Golgi membranes from mung bean (Vigna radiata L.) hypocotyl cells fused to liposomes in the absence of any cytosolic proteins and nucleotides. It was demonstrated that the fusion was mediated by integral membrane protein(s), and was influenced by divalent cations (mm). Mg(2+), Ca(2+), and Mn(2+) ions enhanced the lipid mixing by reducing repulsive forces between membranes. In the content leakage assay, Mg(2+) ions also showed a stimulative effe

OmpA fusion proteins for presentation of foreign antigens on the bacterial outer membrane.

The ompA genes of Escherichia coli and Shigella dysenteriae have been used to construct a group of enterobacterial surface expression vectors for foreign genes. Linker oligonucleotides were inserted into the sequence corresponding to the third or fourth outer domain to allow in-frame sandwich fusion of foreign genes or epitopes into ompA. Influenza haemagglutinin was inserted without its leader peptide and anchor sequences and shown to be transferred as an ompA fusion protein to the bacterial surface in large amounts. The stability of this system depends on the stem structure (i.e. the bottom

Regulating the membrane-fusion effect of liposomes via the protonation of tertiary amines.

Membrane fusion, a fundamental biological process enabling precise exchange of biomolecules within and between cells, has inspired the development of various fusogenic model systems for applications in drug delivery and beyond. However, conventional systems typically operate in an "always-on" state, lacking spatiotemporal control. Although cleavable polyethylene glycol coatings can confer temporal regulation through stimuli-responsive activation, such strategies are often unidirectional and synthetically challenging. Here, we report a simple yet reversible strategy to regulate liposomal membra

Increased efficiency of charge-mediated fusion in polymer/lipid hybrid membranes.

Due to their augmented properties, biomimetic polymer/lipid hybrid compartments are a promising substitute for natural liposomes in multiple applications, but the protein-free fusion of those semisynthetic membranes is unexplored to date. Here, we study the charge-mediated fusion of hybrid vesicles composed of poly(dimethylsiloxane)-graft-poly(ethylene oxide) and different lipids and analyze the process by size distribution and the mixing of membrane species at μm and nano scales. Remarkably, the membrane mixing of oppositely charged hybrids surpasses by far the degree in liposomes, which we c

Aggregation and fusion of vesicles composed of N-palmitoyl derivatives of membrane phospholipids.

N-Acylphosphatidylethanolamines and N-acylphosphatidylserines have been isolated from mammalian cells and have been associated with some tissue degenerative changes, although the relationship between their synthesis and the uncontrolled sequence of events that ends in irreversible tissue damage is not completely established. Our results show that monovalent and divalent cations induce aggregation and fusion of liposomes constituted by N-palmitoylphosphatidylethanolamine (NPPE) and N-palmitoylphosphatidylserine (NPPS). The effectiveness of cations to induce the aggregation of NPPE and NPPS lipo

Association of human hepatocellular membrane fusions with non-A, non-B hepatitis.

Liver biopsies from patients with alcoholic hepatitis, chemical hepatitis, or viral hepatitis types A, B, or non-A, non-B were examined by electron microscopy. Circular, fused, cytoplasmic membranes were observed in hepatocytes of 17% of patients with hepatitis type B and 92% of patients with hepatitis type non-A, non-B. The membrane alterations were not observed in hepatocytes of patients with the other types of hepatitis. The greater frequency of altered cytoplasmic membranes in hepatocytes of patients with non-A, non-B hepatitis was shown to be statistically significant (p less than 0.05) w

The membrane-proximal domain of vesicular stomatitis virus G protein functions as a membrane fusion potentiator and can induce hemifusion.

Recently we showed that the membrane-proximal stem region of the vesicular stomatitis virus (VSV) G protein ectodomain (G stem [GS]), together with the transmembrane and cytoplasmic domains, was sufficient to mediate efficient VSV budding (C. S. Robison and M. A. Whitt, J. Virol. 74:2239-2246, 2000). Here, we show that GS can also potentiate the membrane fusion activity of heterologous viral fusion proteins when GS is coexpressed with those proteins. For some fusion proteins, there was as much as a 40-fold increase in syncytium formation when GS was coexpressed compared to that seen when the f

Bovine coronavirus enters PBIECs via membrane fusion and clathrin-, caveolin-mediated endocytosis, macropinocytosis.

Bovine coronavirus (BCoV) is an important pathogen that exhibits dual tropism for the respiratory and intestinal tracts, causing winter dysentery in adult cattle, diarrhea and respiratory infections in calves, thus imposing considerable economic losses on the global cattle industry. Our previous studies demonstrated that BCoV gains entry into susceptible HRT-18 cells through membrane fusion and clathrin-mediated endocytosis (CME). However, the precise mechanisms by which BCoV enters host cells remain incompletely elucidated, particularly in primary bovine intestinal epithelial cells (PBIECs).

Stiffness-Gated Cytoplasmic mRNA Delivery Through Engineered Membrane Fusion for Breast Cancer Immunotherapy.

Traditional lipid nanoparticles (LNPs) suffer from low lysosomal escape efficiency (<4%) and off-target toxicity, hindering mRNA therapy translation. While membrane fusion carriers bypass endosomal entrapment, their lack of cell specificity induces non-target cytotoxicity. To overcome these limitations, we developed a stiffness-gated mRNA delivery platform (PGC@FM). This system features a PLGA core loaded with G0-C14 dendrimer/mRNA complexes and coated with an engineered, low-stiffness tumor cell fusion membrane (FM). The membrane stiffness is strategically reduced via unsaturated fatty acid e

Erythrocyte Membrane Incorporation Modulates the Stability and Fusion-Associated Behavior of Biomimetic Liposomes.

This study investigates how membrane composition governs the stability and fusogenic behavior of biomimetic hybrid liposomes composed of phosphatidylcholine (PC) and red blood cell (RBC) membrane fragments. While prior work on RBC-derived hybrid systems has largely focused on biological performance metrics such as circulation time, cellular uptake, and immune evasion, the biophysical and thermodynamic consequences of membrane hybridization at the molecular level remain poorly explored. Here, we address this gap through a systematic, multitechnique physicochemical characterization of a series o

Engineering of liposomes containing cholesterol analogues to enhance melanoma therapy via membrane fusion.

A key challenge in disease treatment and prevention is the efficient delivery of therapeutics to the cells. To address this, a primary strategy is the use of liposomes to protect the payload and enhance cellular uptake. However, the delivery efficiency of traditional liposomes remains limited, as cellular internalization primarily occurs through endocytosis. Here, we present a strategy to engineer liposomes with cholesterol analogues to enhance delivery efficiency via membrane fusion. Liposomes containing cholesterol analogues including β-sitosterol (Sito/Lip) and stigmasterol (Stig/Lip) show

Promoting plant protoplast fusion via membrane decoration with cell-penetrating peptide-conjugated lipids.

Protoplast fusion is a powerful plant cell engineering approach that enables the formation of new hybrids and facilitates genetic exchange across species barriers. However, conventional fusion techniques, such as polyethylene glycol (PEG)-induced fusion and electrofusion, often have low efficiency and poor reproducibility. In this study, we used Tat peptide-conjugated PEG-lipids (Tat-PEG-lipids) with different alkyl chain lengths (C9, C12, and C14) to examine the effect of PEG-lipid conjugates on fusion efficiency in rice (Oryza sativa L. cv. Nipponbare) protoplasts. Initially, we used rhodami

A Broad-Spectrum Antiviral Phase-Transformable Liposome with Membrane Fusion-Mediated Virucidal Capability.

Viral infection is a significant threat to global public health. Available antiviral drugs are effective only for specific viruses. It is imperative to develop broad-spectrum antiviral drugs. The heparin sulfate proteoglycan (HSPG) biomimetic strategy holds great promise to design broad-spectrum antiviral materials. However, the reversible binding of HSPG-mimicking nanoparticles with virions presents a potential risk of reinfection. Herein, a sulfonated liposome (SC_Lip) that can trigger phase transition below pH 6.5 is screened to combat viral infection at all stages. SC_Lip can realize pH-in

Membrane Fusion-Mediated Loading of Therapeutic siRNA into Exosome for Tissue-Specific Application.

Tissue-specific delivery of oligonucleotide therapeutics beyond the liver remains a key challenge in nucleic acid drug development. To address this issue, exploiting exosomes as a novel carrier has emerged as a promising approach for efficient nucleic acid drug delivery. However, current exosome-based delivery systems still face multiple hurdles in their clinical applications. Herein, this work presents a strategy for constructing a hybrid exosome vehicle (HEV) through a DNA zipper-mediated membrane fusion approach for tissue-specific siRNA delivery. As a proof-of-concept, this work successful

Direct cytosolic delivery of siRNA via cell membrane fusion using cholesterol-enriched exosomes.

Efficient cytosolic delivery is a significant hurdle when using short interfering RNA (siRNA) in therapeutic applications. Here we show that cholesterol-rich exosomes are prone to entering cancer cells through membrane fusion, achieving direct cytosolic delivery of siRNA. Molecular dynamics simulations suggest that deformation and increased contact with the target cell membrane facilitate membrane fusion. In vitro we show that cholesterol-enriched milk-derived exosomes (MEs) achieve a significantly higher gene silencing effect of siRNA, inducing superior cancer cell apoptosis compared with the

The Ebola virus VP40 matrix layer undergoes endosomal disassembly essential for membrane fusion.

Ebola viruses (EBOVs) assemble into filamentous virions, whose shape and stability are determined by the matrix viral protein 40 (VP40). Virus entry into host cells occurs via membrane fusion in late endosomes; however, the mechanism of how the remarkably long virions undergo uncoating, including virion disassembly and nucleocapsid release into the cytosol, remains unknown. Here, we investigate the structural architecture of EBOVs entering host cells and discover that the VP40 matrix disassembles prior to membrane fusion. We reveal that VP40 disassembly is caused by the weakening of VP40-lipid

Outer nuclear membrane fusion of adjacent nuclei in varicella-zoster virus-induced syncytia.

Syncytia formation has been considered important for cell-to-cell spread and pathogenesis of many viruses. As a syncytium forms, individual nuclei often congregate together, allowing close contact of nuclear membranes and possibly fusion to occur. However, there is currently no reported evidence of nuclear membrane fusion between adjacent nuclei in wild-type virus-induced syncytia. Varicella-zoster virus (VZV) is one typical syncytia-inducing virus that causes chickenpox and shingles in humans. Here, we report, for the first time, an interesting observation of apparent fusion of the outer nucl
