Posttraining administration of pentylenetetrazol dissociates gabapentin effects on memory consolidation from that on memory retrieval process in mice.

Gabapentin (GBP), an anticonvulsant drug, 10 mg/kg, i.p., but not 100 mg/kg, i.p., enhanced retention of an inhibitory avoidance task when given 20 min after training, as indicated by retention performance 48 h later. The immediate post-training administration of pentylenetetrazol (PTZ, 45 mg/kg, i.p.) impaired retention performance. The amnesic effects of the convulsant drug PTZ were not influenced by GBP at any level of doses. However, GBP 100 mg/kg, but not 10 mg/kg, delayed the latency to first clonic body seizures and decreased the duration of convulsion induced by PTZ. The enhancing effe

Short-term and long-term memory deficit following intracarotid Amytal injection: further support for the memory consolidation hypothesis.

To test the three main hypotheses of the human amnesic syndrome (encoding, consolidation, retrieval), we designed an original protocol for memory assessment under Amytal that included, in addition to a retrograde memory measure, both short-term and long-term anterograde memory measures. Twenty epileptic patients with SEEG-confirmed unilateral temporal lobe foci were given right and left injections on successive days. Only the long-term memory measure for material presented under Amytal was significantly related to the presence of a temporal contralateral epileptogenic focus, even if it was ass

Wdr5 Modulates Synaptic Development and Memory Consolidation in the Hippocampus and Cerebral Cortex.

WD-repeat domain 5 (Wdr5), a core component of the SET1/MLL family of histone methyltransferases, is widely involved in the regulation of development, epigenetic control, transcription, and cell division. Wdr5 has been linked to neurodevelopmental disorders characterized by intellectual disability and developmental delay-yet its regulatory roles in neurons remain poorly understood. Here, we report that postnatal deletion of Wdr5 in excitatory forebrain neurons impairs learning and memory in male Wdr5fl/fl; CaMKIIα-iCre conditional knockout (Wdr5 cKO) mice. We showed that the conditional Wdr5 k

Anterograde Amnesia Induced by Disruption of Consolidation or Reconsolidation of Long-Term Memory.

Mechanisms of amnesia caused by impairment of consolidation or reconsolidation of conditioned food aversion memory with protein synthesis inhibitor cycloheximide were studied in Helix lucorum. Cycloheximide injection during training or memory reconsolidation in trained snails produced amnesia. In both cases, repeated training 10 days after amnesia induction led to short-term memory formation, while long-term memory was not formed, despite the fact that the number of conditioned and reinforcing stimuli combinations was higher than during initial training. The possibility of formation of short-t

Distributed Population Coding and Memory Consolidation in the Cerebellar Cortex and Dentate Nucleus.

The cerebellum contributes to associative learning, yet how cortical and nuclear populations interact during memory formation remains unclear. We recorded single neurons from Purkinje cells (PCs) in Crus I/II and from the dentate nucleus (DN) in monkeys learning a visuomotor association. Both populations encoded the association in a temporally distributed manner spanning stimulus onset to movement. Once learning was consolidated, DN population activity preceded that of PCs, suggesting a progressive transfer of information from the cerebellar cortex to the nuclei, where long-term memory traces

Memory Impairing Effect of Propranolol on Consolidation and Reconsolidation for Various Learning Tasks.

Newly acquired memory traces have been thought to become stable and resistant to interruption after they are stored in long-term memory. However, according to a recent research drugs such as beta-adrenergic receptor antagonists enable memories to be updated and rewritten when administered during consolidation and reconsolidation. Propranolol is a widely used beta-adrenergic receptor antagonist that disrupts the consolidation and reconsolidation processes of memory formation as it inhibits protein synthesis in the central nervous system. This review aims to discuss the memory impairing effect o

Effect of pristine fullerene on acquisition, consolidation and retrieval memory in wistar rats.

The present study evaluated the effect of fullerene (C60) under in vitro conditions, in hippocampus homogenates from rats and on the induction of behavioral disabilities. Exposure to in vitro C60 led to an increase in the concentration of reactive oxygen species (ROS) and lipid peroxidation (LPO) of hippocampus treated with of fullerene and suspension. These results indicate that the oxidative stress caused by the exposure to C60 was in part related to an absence of an antioxidant response. In this sense, one-trial inhibitory avoidance task were performed and results showed that fullerene at 0

Cocaine, nicotine, and their conditioned contexts enhance consolidation of object memory in rats.

To test the hypothesis that drugs of abuse and their conditioned stimuli (CSs) enhance memory consolidation, the effects of post-training exposure to cocaine and nicotine were compared to the effects of post-training exposure to contextual stimuli that were paired with the effects of these drugs. Using the object recognition (OR) task, it was first demonstrated that both 10 and 20 mg/kg cocaine, and 0.2 and 0.4 mg/kg nicotine, enhanced recognition memory when administered immediately after, but not 6 h after the sample phase. To establish the drug CSs, rats were confined for 2 h in a chamber (

Memory Integration as a Challenge to the Consolidation/Reconsolidation Hypothesis: Similarities, Differences and Perspectives.

We recently proposed that retrograde amnesia does not result from a disruption of the consolidation/reconsolidation processes but rather to the integration of the internal state induced by the amnesic treatment within the initial memory. Accordingly, the performance disruption induced by an amnesic agent does not result from a disruption of the memory fixation process, but from a difference in the internal state present during the learning phase (or reactivation) and at the later retention test: a case of state-dependency. In the present article, we will review similarities and differences the

Generalization of perceptual and motor learning: a causal link with memory encoding and consolidation?

In both perceptual and motor learning, numerous studies have shown specificity of learning to the trained eye or hand and to the physical features of the task. However, generalization of learning is possible in both perceptual and motor domains. Here, I review evidence for perceptual and motor learning generalization, suggesting that generalization patterns are affected by the way in which the original memory is encoded and consolidated. Generalization may be facilitated during fast learning, with possible engagement of higher-order brain areas recurrently interacting with the primary visual o

Hippocampal replay in the awake state: a potential substrate for memory consolidation and retrieval.

The hippocampus is required for the encoding, consolidation and retrieval of event memories. Although the neural mechanisms that underlie these processes are only partially understood, a series of recent papers point to awake memory replay as a potential contributor to both consolidation and retrieval. Replay is the sequential reactivation of hippocampal place cells that represent previously experienced behavioral trajectories and occurs frequently in the awake state, particularly during periods of relative immobility. Awake replay may reflect trajectories through either the current environmen

On the requirement of nitric oxide signaling in the amygdala for consolidation of inhibitory avoidance memory.

Evidence suggests that the NO/sGC/PKG pathway plays a key role in memory processing but the actual participation of this signaling cascade in the amygdala during memory consolidation remains unknown. Here, we show that when infused in the amygdala immediately after inhibitory avoidance training, but not later, the NO synthase inhibitor L-NNA hindered long-term memory retention without affecting locomotion, exploratory behavior, anxiety state or retrieval of the avoidance response. The amnesic effect of L-NNA was not state-dependent and was mimicked by the soluble guanylyl cyclase inhibitor LY8

Combining Stimulation Protocols to Probe Memory Consolidation-Commentary on Hausdorf et al. 2025.

A recent study by Hausdorf et al. reported that combining closed-loop acoustic stimulation with transcranial direct current stimulation during sleep does not improve memory consolidation. This contrasts with past studies that reported a beneficial effect of each method individually. In this commentary, I discuss the relevance of the study in the current literature and offer potential explanations for this absence of effect.

Transcranial alternating current stimulation of the posterior parietal cortex enhances episodic memory consolidation.

Transcranial alternating current stimulation (tACS) has been reported to improve episodic memory (EM) by entraining neural oscillations. However, the effects of tACS delivered over the posterior parietal cortex (PPC) immediately after learning remain unclear. We aimed to determine whether tACS over the PPC, administered immediately after learning, enhances EM retention. Thirty-six healthy adults were randomly assigned to the tACS or sham-control groups. On day 1, participants completed three sessions consisting of a word-learning task followed by a recognition test. Subsequently, 40 Hz tACS (1

Exposure to sounds during sleep impairs hippocampal sharp wave ripples and memory consolidation.

Sleep is critical for the consolidation of recent experiences into long-term memories. As a key underlying neuronal mechanism, hippocampal sharp-wave ripples (SWRs) occurring during sleep define periods of hippocampal reactivation of recent experiences and have been causally linked with memory consolidation. Hippocampal SWR-dependent memory consolidation during sleep is often referred to as occurring during an "offline" state, dedicated to processing internally generated neural activity patterns rather than external stimuli. However, the brain is not fully disconnected from the environment dur

Reduced sleep spindles and spindle coherence in schizophrenia: mechanisms of impaired memory consolidation?

BACKGROUND: Sleep spindles are thought to induce synaptic changes and thereby contribute to memory consolidation during sleep. Patients with schizophrenia show dramatic reductions of both spindles and sleep-dependent memory consolidation, which may be causally related. METHODS: To examine the relations of sleep spindle activity to sleep-dependent consolidation of motor procedural memory, 21 chronic, medicated schizophrenia outpatients and 17 healthy volunteers underwent polysomnography on two consecutive nights. On the second night, participants were trained on the finger-tapping motor sequen

The intra-hippocampal leucine administration impairs memory consolidation and LTP generation in rats.

Leucine accumulates in fluids and tissues of patients affected by maple syrup urine disease, an inherited metabolic disorder, predominantly characterized by neurological dysfunction. Although, a variable degree of cognition/psychomotor delay/mental retardation is found in a considerable number of individuals affected by this deficiency, the mechanisms underlying the neuropathology of these alterations are still not defined. Therefore, the aim of this study was to investigate the effect of acute intra-hippocampal leucine administration in the step-down test in rats. In addition, the leucine eff

Single-cell resolution spatial transcriptomic signature of the retrosplenial cortex during memory consolidation.

The retrosplenial cortex (RSC) is a critical brain region that is activated during spatial memory tasks and plays a crucial role in the consolidation of long-term memory. Various classes of RSC excitatory neurons across different laminar layers serve as the central hub for neuronal connections between the RSC and other brain regions, such as the hippocampus. Despite the established role of the RSC in spatial memory, the transcriptomic signature of the neuronal subtypes in the RSC during spatial memory consolidation remained elusive. Here, we used unbiased and targeted spatial transcriptomics t

A visual paired associate learning (vPAL) paradigm to study memory consolidation during sleep.

Sleep improves the consolidation and long-term stability of newly formed memories and associations. Most research on human declarative memory and its consolidation during sleep uses word-pair associations requiring exhaustive learning. In the present study, we present the visual paired association learning (vPAL) paradigm, in which participants learn new associations between images of celebrities and animals. The vPAL is based on a one-shot exposure that resembles learning in natural conditions. We tested if vPAL can reveal a role for sleep in memory consolidation by assessing the specificity
