Auto-titration technique using intracranial self-stimulation and effects of antianxiety drugs.

In order to study effects of antianxiety drugs on the threshold of intracranial self-stimulation (ICSS), the auto-titration technique was used in the two lever Skinner box. This procedure consisted of conventional ICSS except the brain stimulation current intensity was decreased after every 20 lever press for ICSS, and the animal could press the another reset lever (at any time) to reset the stimulation current intensity to the preseted level. The current intensity at which the animal pressed the reset lever (reset current) was defined as the threshold of ICSS. Since reset currents that the an

First enantioseparation and stereoselective metabolism of carisoprodol in rat liver microsomes.

Carisoprodol, a widely prescribed muscle relaxant, is primarily metabolized into its main active metabolites, meprobamate and hydroxy carisoprodol. Despite its frequent use, the pharmacokinetics of carisoprodol have not been extensively studied, with the majority of research focusing on meprobamate. Moreover, even though carisoprodol has a stereogenic center, no chiral separation of carisoprodol has ever been reported in the literature. The present study describes the first enantioseparation of carisoprodol by a heart-cut bidimensional achiral-chiral chromatographic method in reversed-phase mo

Occurrence and distribution of psychoactive compounds and their metabolites in the urban water cycle of Berlin (Germany).

The occurrence and distribution of six psychoactive compounds (primidone, phenobarbital, oxazepam, diazepam, meprobamate, and pyrithyldione) and a metabolite of primidone (phenylethylmalonamide) were investigated in wastewater treatment plant (WWTP) effluents, surface water, groundwater of a bank filtration site, raw and final drinking water, and in groundwater affected by former sewage irrigation. Primidone and its metabolite phenylethylmalonamide were found to be ubiquitous in environmental water samples in Berlin. Maximum concentrations of 0.87 and 0.42 μg/L, respectively, were encountered

Abuse Potential of Soma: the GABA(A) Receptor as a Target.

Soma(®) (carisoprodol) is an increasingly abused, centrally-acting muscle relaxant. Despite the prevalence of carisoprodol abuse, its mechanism of action remains unclear. Its sedative effects, which contribute to its therapeutic and recreational use, are generally attributed to the actions of its primary metabolite, meprobamate, at GABA(A) receptors (GABA(A)R). Meprobamate is a controlled substance at the federal level; ironically, carisoprodol is not currently classified as such. Using behavioral and molecular pharmacological approaches, we recently demonstrated carisoprodol, itself, is capab

Carisoprodol: abuse potential and withdrawal syndrome.

Carisoprodol (N-isopropyl-2 methyl-2-propyl-1,3-propanediol dicarbamate; N-isopropylmeprobamate) is a centrally acting skeletal muscle relaxant whose primary active metabolite is meprobamate, a substance with well established abuse potential similar to that of benzodiazepines. A number of reports show that carisoprodol has been abused for its sedative and relaxant effects, to augment or alter the effects of other drugs, and by the intentional combination of carisoprodol and other noncontrolled medications because of the relative ease (as compared to controlled substances) of obtaining prescrip

Septal driving of hippocampal theta rhythm: role of gamma-aminobutyrate-benzodiazepine receptor complex in mediating effects of anxiolytics.

In free-moving male rats, when the hippocampal theta rhythm is artificially driven by stimulation in the septum at frequencies between 5 and 10 Hz, the function relating frequency to the threshold current required to drive the theta rhythm has a minimum at 7.7 Hz. This minimum is eliminated by anxiolytic drugs. Dose-response curves for this effect are reported for chlordiazepoxide, diazepam and meprobamate. The effect of meprobamate was reversed by two gamma-aminobutyrateA antagonists, picrotoxin and bicuculline, which have previously been shown to be without effects of their own. The gamma-am

Pesticide-drug interaction in rats.

The possible interactions between a fungicide, thiram, and some drugs acting on the central nervous system were investigated in rats by means of behavioral methods. Potentiation was found after combined treatment with thiram and promethazine, and after thiram and meprobamate. In the case of trihexyphenidyl, an additive interaction was observed. It is presumed that these observations will be of practical toxicological significance.

[The effect of phytin, benzonal and their combination administered prophylactically on the pharmacodynamics of drugs metabolized in the liver in hypokinesia].

Hexenal, meprobamate, amidopyrine and ethylmorphine produced a significantly marked effect in animals under hypokinesia as compared with normal rats. When phytin, benzonal and their combination were used for preventive purposes, impaired pharmacodynamics of the tested drugs metabolizing in the liver disappeared. The investigations demonstrated that the preventive use of phytin in combination with benzonal is the most optimal in correcting the impairments of drug pharmacodynamics in hypokinesia.

SOME ACTIONS OF CENTRALLY ACTIVE AND OTHER DRUGS ON THE TRANSMISSION OF SINGLE NERVE IMPULSES THROUGH THE ISOLATED SUPERIOR CERVICAL GANGLION PREPARATION OF THE RABBIT.

The effect of some centrally-active and other drugs on the transmission of single nerve impulses through the isolated superior cervical ganglion preparation of the rabbit has been studied by recording both preganglionic and postganglionic action potentials. Block of conduction in the axon could be distinguished from block of the synaptic mechanism. The drugs did not appear to exert any one characteristic form of blocking action. A continuous spectrum of drug action linked an agent such as meprobamate which acted predominantly on the synapse to benactyzine which acted mainly by blocking axonal

[Characteristics of the choice of psychotropic drugs in suicide attempts].

UNLABELLED: Review of results of Pecs centre in WHO/EURO Multicentre Study on Suicidal Behaviour. OBJECTIVE: Studies concerning the choice of methods and psychotropics in the suicidal acts have a great importance because the outcome of suicides is decisively determined by the potential lethality of the method. In the sample of patients who attempted suicides, the features of overdoses have been investigated as well as their relations to the age, sex and repetition. METHOD: Within the framework of the WHO/EURO Multicentre Study on Suicidal Behaviour data, 1158 cases with suicide attempts were

Routine analysis of barbiturates and some other hypnotic drugs in the blood plasma as an aid to the diagnosis of acute poisoning.

A gas-liquid chromatographic method has been devised whereby both the qualitative and the quantitative analysis of a number of hypnotic drugs in the blood may be accomplished simultaneously. Small (50-mul) plasma aliquots are required. The technique is rapid and will detect 2 mg/l concentrations of these compounds. The barbiturates glutethimide, methaqualone, and meprobamate are the principal drugs covered by the scheme. After overdose the concentrations that occur in man are at, or above, this minimum level of detection.

Neuropharmacologic specificity of a simple animal model for the behavioral actions of benzodiazepines.

A simple model system for the behavioral actions of benzodiazepines is analyzed for its dose-response predictiveness, using several benzodiazepines, and for its pharmacological specificity, using other non-anxiolytic classes of psychoactive drugs. The model demonstrates an increase in mouse exploratory activity between a brighty-lit open field and a small, dark compartment by clonazepam, diazepam, flurazepam, chlordiazepoxide and meprobamate, but not by the peripheral benzodiazepine R05-4864, nor by clorgyline, butriptyline, or chlorpromazine.

[Voluntary drug poisoning: epidemiology, performance and limits of the emergency laboratory].

The aim of this study is to determine the efficiency ot toxicologic screening (detection of barbiturates, benzodiazepines, tricyclic antidepressants, salicylates, phenothiazines, meprobamate and ethanol assay), during acute drug poisoning. In 1988, 898 patients are enclose in this study. Screenings are negative in 17% of cases; benzodiazepines, alcohol and antidepressants are often found. The recovery is very good for barbiturates and salicylates but it's not perfect for benzodiazepines, particularly flunitrazepam triazolam, loflazepate, oxazepam, and non tricyclic antidepressants. The human distribution of some barbiturate sedatives in combination with miscellaneous CNS-active drugs.

This paper presents 14 cases in which the distribution of barbiturates and the hydroxylated metabolites in combination with miscellaneous CNS-active drugs was studied. In 7 of these cases the other drug present was methadone, and in the remainder dihydrocodeine, morphine, propoxyphene, amitriptyline, meprobamate, cyclizine and dipipanone, diphenhydramine and methaqualine. Amitriptyline, methadone, cyclizine and dipipanone, methaqualone and diphenhydramine appeared to modify the distribution of amylorbarbitone and quinalbarbitone. Likewise, the barbituates seemed to alter the distribution of am

Integrated bioinformatics analysis of chromosomal deletion regions in glioblastoma identifies key genes linked to GABA(A) receptor signaling.

Glioblastoma (GBM) is the most common aggressive brain tumor in adults. The development of therapy resistance in GBM necessitates the identification of novel biomarkers to guide molecularly targeted treatment strategies. This study aims to identify potential tumor suppressor genes in GBM and to evaluate their potential as therapeutic targets. Genes localized on deletion regions in GBM tumors were selected using the BioMart database. Microarray datasets containing GBM tumor and normal brain tissue samples were analyzed with GEO2R. A protein-protein interaction (PPI) network was constructed for

Behavioral effects of nonbenzodiazepine anxiolytic drugs: a comparison of CGS 9896 and zopiclone with chlordiazepoxide.

Zopiclone and CGS 9896 are two nonbenzodiazepine compounds which have been shown to displace benzodiazepines from their binding sites. The present study compared the behavioral effects of these two compounds in rats with those of chlordiazepoxide. The three drugs produced dose-related increases in punished drinking as did pentobarbital and meprobamate but not PK 9084, which also acts at benzodiazepine binding sites, or buspirone. Rates of lever pressing suppressed by punishment were also increased by chlordiazepoxide and zopiclone. CGS 9896 exerted a similar although less marked effect. Lever

Thirty years' war: a battle with insomnia.

A 72-year-old man with a 30-year complaint of intractable insomnia had a positive family history of depression. He first came to psychiatric attention in 1958, after attacking his wife. He was prescribed barbiturates, and later was given meprobamate and nitrazepam, but with no effect on his complaint. The patient tended to increase the dosage of any drug given, of his own accord. EEG sleep recording confirmed the diagnosis of nocturnal myoclonus. It was hoped that at the case conference further treatment stratagems would be suggested.
