Induction of apoptosis by mercury compounds depends on maturation and is not associated with microglial activation.

The earliest sign of neurotoxicity observed after exposure of three-dimensional brain cell cultures to low concentrations of mercury compounds is a microglial reaction. We hypothesized that an induction of apoptosis by mercury compounds could be an activating signal of the microglial reaction. Aggregating brain cell cultures of fetal rat telencephalon were treated for 10 days with either mercury chloride or monomethylmercury chloride at noncytotoxic concentrations during two developmental periods: from day 5 to 15, corresponding to an immature stage, and from day 25 to 35 corresponding to a ma

[The comparative toxicity of inorganic mercury compounds for a cell culture and the whole organism].

In experiments with two tissue culture lines it was shown, that cytotoxicity of mercury (1) derivatives (measured by CL50) is lesser two times, than it was for mercury (2) derivates. No significant differences in cytotoxicity were noted by the cell morphology and monolayer mitosis activity coefficient studies. This demonstrates the equal toxicity and dangerous to both derivates.

The action of organic mercury compounds on the function of isolated mammalian heart muscle.

The effects of four organic mercury compounds (methylmercuric chloride; bromomercurihydroypropane, BMHP; chlormerodrin; p-chloromercuribenzoic acid, PCMB) on mechanical and electrical functions of guinea-pig papillary muscles were investigated. An initial decline in contraction force was followed by a transient positive inotropic response. The first was accompanied by a shortening of the action-potential duration and by a reduction of the depolarization velocity and the duration of the Ca2+-dependent slow response. The latter was characterized by an indirect component (release of noradrenaline

Genetic variation in the susceptibility to mercury and other metal compounds in Drosophila melanogaster.

The tolerance of Drosophila melanogaster to heavy metal compounds was investigated with special emphasis on methylmercury. A pronounced variation in tolerance to CH3HgOH, HgCl2, (C2H5)3PbCl, (CH3)3SnCl, and CdCl2 was recorded between 12 wild-type strains. After ranking the tolerance of the strains with respect to the five compounds rank correlations for experiments within and between compounds were calculated. The results showed a high degree of correlation within compounds but no unequivocal indication of a correlation between compounds, indicating that different mechanisms of genetic control

Isotope fractionation of mercury during its photochemical reduction by low-molecular-weight organic compounds.

Photochemical reduction of Hg(II) by various low-molecular-weight organic compounds (LMWOC) was investigated to evaluate the effect of specific functional groups that are typically encountered in natural dissolved organic matters (DOM) on the photoreactivity and isotope fractionation of Hg. LMWOC with reduced sulfur functional groups (e.g., cysteine, glutathione) resulted in slower photochemical reduction of Hg(II) than those without reduced sulfur groups (e.g., serine, oxalic acid). Reduction rate constants were specifically determined for two contrasting LMWOC: dl-serine (0.640 h(-1)) and l-

Differential effects of mercury compounds on mutagenicity, genotoxicity and repair of UV-DNA damage.

Mercury (Hg) is a global contaminant that is present in human diet as methylmercury (MeHg). Recent studies linked MeHg exposure with high risks of skin cancers. It is unknown whether MeHg is directly genotoxic in skin cells or able to enhance mutagenic effects of UV radiation. We examined mutagenicity and genotoxicity of MeHg and its metabolite inorganic Hg2 + (iHg) and their impact on processing of carcinogenic UV-DNA damage. We found that iHg and MeHg were both nonmutagenic in the Hprt assay in rodent CHO cells. iHg but not MeHg strongly enhanced mutagenicity of UV-B without changes in repai

Series of Lanthanide-Mercury Compounds with Three-Dimensional Structures: Rational Preparation, Structures and Properties.

Three novel Ln-Hg complexes [Ln(H2O)2(μ-IA)3Hg3Br6] n (Ln = Pr (1), Nd (2), and Er (3); HIA is isonicotinic acid) are synthesized and characterized. They feature three-dimensional (3-D) motifs. Solid-state UV/vis diffuse reflectance spectroscopy found that their band gaps are 4.91, 4.59, and 2.68 eV. It is found that lanthanide ions could adjust the band structures of semiconductors. Their photoluminescence comes from their characteristic emissions of 1D2 → 3H4 of Pr3+, 7F7/2 → 4S3/2 and 4F3/2 → 4I9/2 of Nd3+, and 4I15/2 → 4F7/2 and 4I15/2 → 4S3/2 of Er3+. Adsorptive behaviour of mercury on algal biomass: competition with divalent cations and organic compounds.

Biosorption processes constitute an effective technique for mercury elimination. Sorption properties of native and acid-treated Sargassum muticum have been studied. Effect of pH, initial mercury concentration and contact time studies provided fundamental information about the sorption process. This information was used as the reference values to analyse mercury sorption under competition conditions. Saline effect has shown little influence in sorption, when only electrostatic modifications took place upon salt addition. On the contrary, if mercury speciation dramatically changed owing to the a

Distinct genotoxicity of phenylmercury acetate in human lymphocytes as compared with other mercury compounds.

In the present study, the frequency of sister chromatid exchanges (SCEs) was assayed to evaluate the genotoxic effects of mercury nitrate (Hg2+), methylmercury chloride (CH3HgCl and phenylmercury acetate (PMA) on human lymphocytes. The free radical scavengers, catalase (CA) and superoxide dismutase (SOD) were tested for their antigenotoxic effects toward PMA. PMA (1-30 microM) increased SCE frequency in a concentration-dependent manner. However, CH3HgCl significantly increased SCE frequency only at a concentration of 20 microM, and all concentrations treated with Hg2+ did not induce a positive

[Effects of mercury compounds on growth, cell composition and glucose uptake in Tetrahymena pyriformis].

The effects of the mercuric compounds methylmercuric chloride (MMC) and mercuric chloride (MC) on growth, cell composition, and glucose uptake were studied in the ciliate protozoan Tetrahymena pyriformis GL. MMC was the most effective growth inhibitor of Tetrahymena among mercuric compounds and sulfhydryl inhibitors; it completely inhibited growth of Tetrahymena at concentrations of 1/30-1/3000 of the others. The growth inhibition of Tetrahymena was inversely proportional to concentrations of chemicals in medium. Particularly in the case of MMC and MC the relationship between the concentration

Mercury compounds induce a rapid increase in procoagulant activity of monocyte-like U937 cells.

When monocytic leukaemia line U937 cells were incubated in the presence of HgCl2 there was a rapid increase in tissue factor (TF)-dependent procoagulant activity, reaching a maximum (equivalent to the total TF activity observed when cells had been subjected to a freeze/thaw cycle) after 15 min at 50 microM HgCl2 and after 30 min at 10 microM HgCl2. Two other heavy metal compounds, AgNO3 and phenylmercuric acetate, caused a similar increase in TF activity. The increase was independent of protein synthesis. Other reagents tested, CdCl2, ZnCl2, NiCl2, ADP, FMLP and monocyte chemotactic factor (MC

Comparison of selenoneine found in marine organisms with selenite in the interaction with mercury compounds in vitro.

Selenium (Se) is an essential micronutrient because it forms the active center of selenoenzymes/selenoproteins in the form of selenocysteine. Another biological significance of Se is that it detoxifies inorganic mercury (iHg) by directly interacting with it. Recently, a novel selenometabolite, selenoneine (2-selenyl-N,N,N-trimethyl-L-histidine), was identified in several marine animals. However, its biological significance is still unclear. In this study, the ability of selenoneine to form a complex with iHg and methyl Hg (MeHg) was evaluated in vitro. Whereas selenite serving as the positive

[The aggregative stability of human oxyhemoglobin in aqueous media in the presence of mercury(II) compounds].

An analytical review of studies on human oxyhemoglobin coagulation has been performed by the author jointly with V. S. Koniaeva and L. D. Bogdanova within a period from 1985 to 1990. It was shown that the oxyhemoglobin coagulation modified by mercurials proceeded without any essential alteration of native protein conformation. A hypothesis is discussed that the oxyhemoglobin coagulation results from the primary polyaggregation of dimer fragments and that hydrophobic sites which provide for dimer-to-dimer contacts in native tetrameric oxyhemoglobin, participate in this process.

Differential effects of mercurial compounds on the electroolfactogram (EOG) of salmon (Salmo salar L.).

The effects on the salmon (Salmo salar L.) electroolfactogram (EOG) of the two mercurials, mercuric chloride (HgCl2) and methylmercuric chloride (CH3HgCl), were studied. The EOG responses were evoked by stimulating the olfactory epithelium with 340 microM L-alanine for 10 sec every second minute during a 1-hr period. Each EOG response consisted of an initial peak component followed by a sustained component with an amplitude about 40% below the peak value. Three experimental series, each comprising six fish, were carried out. In the first series, the rosette was irrigated solely with artificial

Comments on the article "the toxicology of mercury and its chemical compounds" by Clarkson and Magos (2006).

Clarkson and Magos (2006) provide their perspectives on the toxicology of mercury vapor and dental amalgam. As scientists who are involved in preparing a German federal guideline regarding dental amalgam, we welcome additional scientific data on this issue. However, Clarkson and Magos do not present all the relevant studies in their review. The additional data provided here show that: (a) Dental amalgam is the main source of human total mercury body burden, because individuals with amalgam have 2-12 times more mercury in their body tissues compared to individuals without amalgam; (b) there is

Protective roles of glutathione in the toxicity of mercury and cadmium compounds to C6 glioma cells.

The neurotoxic effects of mercury (II) chloride, methylmercury (MeHg) and cadmium chloride on astrocytes were modelled using C6 glioma cell cultures. All three compounds were cytotoxic to these cells with an order of potency of cadmium > MeHg > mercurychloride. Addition of reduced glutathione (GSH) to the media protected the cells in all three cases, whereas depletion of GSH with l-buthionine-S,R-sulfoximine enhanced the toxicity of cadmium and mercury chloride but not MeHg. The effects of subcytotoxic concentrations of these compounds on intracellular GSH levels were assessed using chlorobima

The assessment of the condition of the liver in patients chronically exposed to mercury compounds in the place of settlement.

Problems with the assessment of organ damage caused by toxic substances in places of residence have recently appeared with increasing frequency. In spite of this there have been so far no uniform, objective research methods which could allow their accurate evaluation. This is why the attempt has been made to assess morphological and functional condition of the liver in patients chronically exposed to mercury compounds in the place of settlement. The research group consisted of 62 patients exposed to metallic mercury at a yearly rate exceeding 24 kg for average duration of 16 years. Patients wi
