Herlyn-Werner-Wunderlich Syndrome: Case Report and Review of the Literature.

Herlyn-Werner-Wunderlich (HWW) syndrome is a very rare congenital malformation of the urogenital tract involving both the müllerian and the wolffian ducts, and it consists of the association of didelphys uterus, ipsilateral obstructed hemivagina, and ipsilateral kidney agenesis. Its etiology is related to the abnormal development of two embryonic structures-mesonephros and paramesonephros-although its precise mechanism is not known, but theories involving signaling molecules and gene expression are studied for potential explanations. Because of its rarity, there is limited literature on this s

Clonal Analysis of Embryonic Hematopoietic Stem Cell Precursors Using Single Cell Index Sorting Combined with Endothelial Cell Niche Co-culture.

The ability to study hematopoietic stem cell (HSC) genesis during embryonic development has been limited by the rarity of HSC precursors in the early embryo and the lack of assays that functionally identify the long-term multilineage engraftment potential of individual putative HSC precursors. Here, we describe methodology that enables the isolation and characterization of functionally validated HSC precursors at the single cell level. First, we utilize index sorting to catalog the precise phenotypic parameter of each individually sorted cell, using a combination of phenotypic markers to enric

Ectopic expression of Ptf1a induces spinal defects, urogenital defects, and anorectal malformations in Danforth's short tail mice.

Danforth's short tail (Sd) is a semidominant mutation on mouse chromosome 2, characterized by spinal defects, urogenital defects, and anorectal malformations. However, the gene responsible for the Sd phenotype was unknown. In this study, we identified the molecular basis of the Sd mutation. By positional cloning, we identified the insertion of an early transposon in the Sd candidate locus approximately 12-kb upstream of Ptf1a. We found that insertion of the transposon caused overexpression of three neighboring genes, Gm13344, Gm13336, and Ptf1a, in Sd mutant embryos and that the Sd phenotype w

Transcriptional activators, repressors, and epigenetic modifiers controlling hematopoietic stem cell development.

Hematopoietic stem cells (HSCs) are pluripotent cells that give rise to all of the circulating blood cell types. Their unique ability to self-renew while generating differentiated daughter cells permits HSCs to sustain blood cell production throughout life. In mammals, the pool of HSCs shifts from early sites in the aorta-gonad-mesonephros region and the placenta to the fetal liver and ultimately bone marrow. During the past decade, a map of transcriptional activators and repressors that regulate gene expression in HSCs, their precursors and their progeny, at distinct stages of development has

[A case of Mayer-Rokitansky syndrome].

The Mayer-Rokitansky syndrome has been generally defined as a congenital absence of the vagina in genotypic and phenotypic female subjects with normal endocrine status. Many authors have reported that urological anomalies associate commonly with this syndrome, but recently a new concept of this syndrome was proposed by Tarry and associates. They considered the embryogenesis of the Müllerian system and recognized a spectrum of Müllerian anomalies without total vaginal agenesis as this syndrome. They also proposed a new classification of this syndrome (Müllerian grade) considering grade of Mülle

Immunopathological study of mesonephric lesions of cervix uteri and vagina.

Seventeen cases of mesonephric cervical remnants, four cases of mesonephric cervical carcinoma and nine vaginal Gardner cysts were studied by histochemical and immunohistochemical methods for the presence of mucin, proteoglucans, glucogen, CEA, cytokeratins, secretory component and vimentin. Mesonephric lesions in contrast to endocervical glandular lesions are negative for mucin, glucogen, CEA, and secretory component and positive for vimentin, and broad spectrum cytokeratins. Mesonephric carcinomas present certain immunopathological characteristics that permit their identification and proper

Segmental aplasia of the left paramesonephric duct in the cow.

Segmental aplasia of the left uterine horn in a multiparous Holstein cow was diagnosed by palpation and ultrasonography. Treatment with prostaglandin was unsuccessful in eliminating the fluid from the distended uterine horn. Segmental aplasia should be included in the list of differential diagnoses for cows with nonresponsive uterine enlargement.

Cytogenetic and histological features of a human embryo with homogeneous chromosome 8 trisomy.

BACKGROUND: Homogeneous and complete trisomy 8 is extremely rare. With one recent neonatal exception, all reported cases have been mosaic, due to mitotic non-disjunction during early zygotic development. We report a case of chromosome 8 trisomy in a human embryo examined at Carnegie stage 11 (25 days post-fertilization). It presented severe cardiovascular and central nervous system malformations. METHODS: The unusual bifid heart in this embryo spurred a detailed histological examination, karyotyping of a chorionic villus sample and subsequent FISH on inter-phase nuclei of intra-embryonic sect

Mammalian twisted gastrulation is essential for skeleto-lymphogenesis.

Dorsoventral patterning depends on the local concentrations of the morphogens. Twisted gastrulation (TSG) regulates the extracellular availability of a mesoderm inducer, bone morphogenetic protein 4 (BMP-4). However, TSG function in vivo is still unclear. We isolated a TSG cDNA as a secreted molecule from the mouse aorta-gonad-mesonephros region. Here we show that TSG-deficient mice were born healthy, but more than half of the neonatal pups showed severe growth retardation shortly after birth and displayed dwarfism with delayed endochondral ossification and lymphopenia, followed by death withi

Steroidogenic enzyme activity and ovarian differentiation in the saltwater crocodile, Crocodylus porosus.

It has been hypothesized that steroid hormone production is involved in sexual differentiation of the gonads in reptiles with temperature-dependent sex determination. We have therefore examined steroidogenic enzymes and ovarian differentiation in Crocodylus porosus embryos incubated at 30 degrees, a temperature producing 100% female hatchlings. delta 5-3 beta-Hydroxysteroid dehydrogenase (3 beta-HSD) activity was detected only in the adrenal gland before, during, and after the period of ovary differentiation. The same pattern was observed during testis differentiation in embryos incubated at 3

How does the boar epididymis regulate the emission of fertile spermatozoa?

The epididymis is responsible for peripheral immune tolerance of maturing spermatozoa even though these have xeno-antigens foreign to the male and female immune system. The epididymis also produces factors required for fertilization and serves as a sperm repository until the time of ejaculation. These reproduction-relevant epididymal functions occur in the mesonephros-derived duct-system that is composed of absorptive and secretory epithelial cells with the capacity for merocrine and apocrine secretion of proteins, antioxidative- and electrolyte/pH-regulating enzymes and small, non-coding RNAs

Expression patterns of genes encoding small GTPases Ras-dva-1 and Ras-dva-2 in the Xenopus laevis tadpoles.

Small GTPases of the recently discovered Ras-dva family are specific to the Vertebrate phylum. In Xenopus laevis, Ras-dva-1 is expressed during gastrulation and neurulation in the anterior ectoderm where it regulates the early development of the forebrain and cranial placodes (Tereshina et al., 2006). In the present work, we studied the expression of Ras-dva-1 at later developmental stages. As a result, the Ras-dva-1 expression was revealed in the eye retina, epiphysis (pineal gland), hypophysis (pituitary), branchial arches, pharynx, oesophagus, stomach and gall bladder of swimming tadpoles.

Mesonephric tubules expressing estrogen and androgen receptors remain in the rete ovarii of adult mice.

The rete ovarii and epoophoron in females are homologous structures of the rete testis and efferent/epididymal duct in males and are derived from the developing rete cells and mesonephric tubules, respectively. Sex steroid hormones play a critical role in reproductive function for both sexes, and we recently reported expression patterns of sex steroid receptors in developing male reproductive tracts. However, their expression patterns in females remain unclear. We, therefore, investigated the three-dimensional structure and expression patterns of sex steroid receptors in the rete ovarii and ep

[Bilateral persistence of the common mesonephric duct in children. Apropos of a case. Review of the literature].

We report a case of bilateral persistence of the common mesonephric duct in a child. This anomaly is an error of implantation of the vas deferens in the distal part of the Wolffian duct. This abnormality is extremely rare (6 cases have been reported in the world literature). The diagnosis is usually incidental during laparotomy or on retrograde cystography (vesico-ureteric and vesico-deferential reflux). It is simple to treat: bilateral deferential ligation in view of the context of multiple malformations, in order to prevent ascending genital infections (prostatitis, epididymitis, testicular

Wolffian duct development.

The Wolffian ducts (WDs) are the progenitors of the epididymis, vas deferens and seminal vesicles. They form initially as nephric ducts that acquire connection to the developing testis as the mesonephros regresses. The development of the WDs is dependent on androgens. Conventionally, the active androgen is believed to be testosterone delivered locally rather than via the systemic circulation. However, recent studies in marsupials show that 5α-reduced steroids are essential and that these can induce virilisation even when they are delivered via the systemic circulation. The development of the W

Rac1 is essential for intraembryonic hematopoiesis and for the initial seeding of fetal liver with definitive hematopoietic progenitor cells.

Definitive hematopoietic stem and progenitor cells (HSCs/Ps) originating from the yolk sac and/or para-aorta-splanchno-pleura/aorta-gonad-mesonephros are hypothesized to colonize the fetal liver, but mechanisms involved are poorly defined. The Rac subfamily of Rho GTPases has been shown to play essential roles in HSC/P localization to the bone marrow following transplantation. Here, we study the role of Rac1 in HSC/P migration during ontogeny and seeding of fetal liver. Using a triple-transgenic approach, we have deleted Rac1 in HSCs/Ps during very early embryonic development. Without Rac1, th

[Unilateral agenesis of structures dependent on the mesonephric duct. Presentation of a case].

The mesonefric duct plays an essential role in the development of the genitourinary tract. From that duct derive structures as the ureter, the vas deferens and the seminal vesicles. Also, the ureteral bud interact with the mesonefric blastema and induce its conversion to renal tissue. The origin of these anomalies of the embrionary development is unknown. In some cases, mutations on the gene of the cystic fibrosis (CFRT) could play a role, assuming that there is a relationship between the congenital absence of the vas deferens and the cystic fibrosis. Usually, the absence of these structures h

A case of endometrioid carcinoma of the fallopian tube mimicking an adnexal tumor of probable Wolffian origin.

We report a very uncommon case of endometrioid adenocarcinoma of the fallopian tube that mimicked, based on histology, a female adnexal tumor of probable Wolffian origin (FATPWO). We present our microscopic and immunohistochemical findings, and a review of the literature concerning these two entities. The differential diagnosis can be of great consequence, owing to the very different prognoses of the two tumors, and is based mainly on macroscopic appearance and immunohistochemical profile: epithelial membrane antigen (EMA) and CA125, generally lacking in FATPWO, are expressed in endometrioid a

Hematopoietic Stem Cell Tropism Mediated Targeting of Breast Cancer Stem Cells via Alteration of Tumor Immune-Microenvironment.

The ontogenic development of hematopoietic stem cells (HSCs) occurs across diverse niches, with HSCs migrating from the aorta-gonad-mesonephros (AGM) to the fetal liver and finally residing in the bone marrow after birth, where adult HSCs replenish the hematopoietic system. The HSC niche critically regulates tropism and proliferation via factors secreted by the microenvironment interaction. Here, we hypothesized that HSCs display tropism toward the aggressive cancer stem cell (CSC) niches of triple-negative breast cancer (TNBC) and MCF-7 cells breast cancer, which exhibit high relapse rates an

Somatic Cells in the Anuran Testis.

BACKGROUND: The anuran testis is a heterogeneous organ composed of germline and somatic cells mainly derived from the coelomic epithelium (Sertoli cells) and mesonephros (Leydig cells), which together regulate spermatogenesis and testicular architecture. SUMMARY: We provide an integrative overview of the developmental origin, structure, and function of somatic cells in evolutionary primitive and advanced anurans, with comparisons to other vertebrates. Initially bipotential gonad differentiates into testes by forming seminiferous cords and tubules composed of gonocytes and Sertoli cells, and a
