Metabolomics in angiotensin II-induced cardiac hypertrophy.

Angiotensin II (Ang II) induces mitochondrial dysfunction. We tested whether Ang II alters the "metabolomic" profile. We harvested hearts from 8-week-old double transgenic rats harboring human renin and angiotensinogen genes (dTGRs) and controls (Sprague-Dawley), all with or without Ang II type 1 receptor (valsartan) blockade. We used gas chromatography coupled with time-of-flight mass spectrometry to detect 247 intermediary metabolites. We used a partial least-squares discriminate analysis and identified 112 metabolites that differed significantly after corrections (false discovery rate q <0.

Metabolomics: a complementary tool in renal transplantation.

Renal transplant success is closely tied to the ability to monitor transplant recipients and responsively change their medications. However, transplant monitoring still depends on relatively dated technologies - serum creatinine levels, urine output, and histopathology of biopsy samples. These techniques do not offer sufficient specificity, sensitivity, or accuracy for appropriate and timely interventions. As a result, more specific diagnostic techniques, based on proteomics, genomics and metabolomics are being sought. Metabolomics (the high-throughput measurement and analysis of metabolites)

Metabolomics Based Biomarkers of Green Beans Intake.

While there is considerable interest in legumes as a food source, there are no validated biomarkers for green beans (Phaseolus vulgaris, L.) intake. The objective of the current work is to identify and assess the dose-response of food intake biomarkers of green beans. In an acute intervention study, 27 participants consumed green beans in the study center and urine samples were collected pre- and post-consumption. Metabolites were measured by nuclear magnetic resonance (1H-NMR) and liquid chromatography-mass spectrometry (LC-MS). Statistical analysis determined features that changed post-consu

Levothyroxine Treatment During Pregnancy: A Metabolomics Study.

Levothyroxine is one of the most prescribed drugs during pregnancy. Our aim was to determine levothyroxine treatment-associated changes in the metabolite profiles and link these to the health of the newborn. Metabolite profiles of 118 levothyroxine-treated and 118 healthy control pregnancies were determined with nuclear magnetic resonance-based metabolomics from (1) umbilical cord blood samples, (2) samples collected during the first trimester and (3) during delivery from the pregnant women. There was a modest negative correlation between cord blood thyroid stimulating hormone (CBTSH) concentr

Applications of metabolomics in cow health assessment.

BACKGROUND: The dairy cow is an integral part of the global agricultural economy and plays a vital role in human nutrition. Compromised health in dairy cows leads to substantial economic losses and adverse environmental impacts. Understanding cow physiology and the etiology of common diseases is essential for developing effective strategies to improve animal health and mitigate negative consequences. Over the past two decades, metabolomics has emerged as a powerful approach not only for assessing and monitoring the health status of dairy cows but also for predicting and diagnosing diseases. A

Kick-start for metabolomics in liver disease.

It is not complicated for the clinician to diagnose a patient with advanced fibrosis or liver cirrhosis when he has already presented some decompensation of his liver disease. However, it is in the earliest stages when the patient's prognosis can be modified the most. Since liver disease is generally asymptomatic, not invasive markers are of great relevance. In the era of omics, it is time for metabolomics to accompany genomics and proteomics, which are more established in the diagnostics and prognostics clinical toolbox. Metabolomics, understood as the comprehensive evaluation of the metaboli

Decoding aging clocks: New insights from metabolomics.

Chronological age is a crucial risk factor for diseases and disabilities among older adults. However, individuals of the same chronological age often exhibit divergent biological aging states, resulting in distinct individual risk profiles. Chronological age estimators based on omics data and machine learning techniques, known as aging clocks, provide a valuable framework for interpreting molecular-level biological aging. Metabolomics is an intriguing and rapidly growing field of study, involving the comprehensive profiling of small molecules within the body and providing the ultimate genome-e

In Situ Metabolomics of Cortisol-Producing Adenomas.

BACKGROUND: Recent advances in omics techniques have allowed detailed genetic characterization of cortisol-producing adrenal adenoma (CPA). In contrast, the pathophysiology of CPAs has not been elucidated in detail on the level of tumor metabolic alterations. METHODS: The current study conducted a comprehensive mass spectrometry imaging (MSI) map of CPAs in relation to clinical phenotypes and immunohistochemical profiles of steroidogenic enzymes. The study cohort comprised 46 patients with adrenal tumors including CPAs (n 35) and nonfunctional adenomas (n 11). RESULTS: Severity of cortisol h

Metabolic and Metabolomics Insights into Dilated Cardiomyopathy.

BACKGROUND: Dilated cardiomyopathy (DCM) is the most common form of heart muscle disease characterized by progressive dilatation and ventricular dysfunction. Metabolomics is an emerging and powerful discipline that provides a global information on the phenotype of mammalian systems via the study of endogenous and exogenous metabolites in cells, tissues, and biofluids. These studies aid in the identification of biomarkers to prevent diseases in later life or help to early detect onset of diseases as well as aiding in the elucidation of disease mechanisms. SUMMARY: Metabolomics provides a uniqu

Application of data augmentation techniques towards metabolomics.

Niemann-Pick Class 1 (NPC1) disease is a rare and debilitating neurodegenerative lysosomal storage disease (LSD). Metabolomics datasets of NPC1 patients available to perform this type of analysis are often limited in the number of samples and severely unbalanced. In order to improve the predictive capability and identify new biomarkers in an NPC1 disease urinary dataset, data augmentation (DA) techniques based on computational intelligence have been employed to create synthetic samples, i.e. the addition of noise, oversampling techniques and conditional generative adversarial networks. These t

CSF Metabolomics of Tuberculous Meningitis: A Review.

From the World Health Organization's global TB report for 2020, it is estimated that in 2019 at least 80,000 children (a particularly vulnerable population) developed tuberculous meningitis (TBM)-an invariably fatal disease if untreated-although this is likely an underestimate. As our latest technologies have evolved-with the unprecedented development of the various "omics" disciplines-a mountain of new data on infectious diseases have been created. However, our knowledge and understanding of infectious diseases are still trying to keep pace. Metabolites offer much biological information, but

Narrative review of metabolomics in cardiovascular disease.

Cardiovascular diseases are accompanied by disorders in the cardiac metabolism. Furthermore, comorbidities often associated with cardiovascular disease can alter systemic and myocardial metabolism contributing to worsening of cardiac performance and health status. Biomarkers such as natriuretic peptides or troponins already support diagnosis, prognosis and treatment of patients with cardiovascular diseases and are represented in international guidelines. However, as cardiovascular diseases affect various pathophysiological pathways, a single biomarker approach cannot be regarded as ideal to re

NMR-Based Metabolomics in Gallbladder Cancer Research.

Gallbladder cancer (GBC) is a common form of malignancy, which has high incidence rates in the northern parts of India, South America, Japan, and in Native American populations. A few metabolomic studies have revealed changes in the composition of biofluids, tissues, and gallstones by nuclear magnetic resonance (NMR), which may prove useful in understanding GBC. In this chapter, we focus on the use of NMR in unveiling the metabolomics of GBC.

In situ metabolomics of aldosterone-producing adenomas.

Recent genetic examinations and multisteroid profiles have provided the basis for subclassification of aldosterone-producing adenomas (APAs). The objective of the current study was to produce a comprehensive, high-resolution mass spectrometry imaging (MSI) map of APAs in relation to morphometry, immunohistochemical profiles, mutational status, and clinical outcome. The study cohort comprised 136 patients with unilateral primary aldosteronism. Matrix-assisted laser desorption/ionization-Fourier transform-ion cyclotron resonance MSI was conducted, and metabolite profiles were analyzed with genot

Metabolomics-A Promising Approach to Pituitary Adenomas.

Background: Metabolomics-the novel science that evaluates the multitude of low-molecular-weight metabolites in a biological system, provides new data on pathogenic mechanisms of diseases, including endocrine tumors. Although development of metabolomic profiling in pituitary disorders is at an early stage, it seems to be a promising approach in the near future in identifying specific disease biomarkers and understanding cellular signaling networks. Objectives: To review the metabolomic profile and the contributions of metabolomics in pituitary adenomas (PA). Methods: A systematic review was con

Global serum metabolomics profiling of colorectal cancer.

Accurate diagnosis of colorectal cancer (CRC) relies on the use of invasive tools such as colonoscopy and sigmoidoscopy. Non-invasive tools are less sensitive in detecting the disease, particularly in the early stage. A number of researchers have used metabolomics analyses on serum/plasma samples of patients with CRC compared with normal healthy individuals in an effort to identify biomarkers for CRC. The aim of the present review is to compare reported serum metabolomics profiles of CRC and to identify common metabolites affected among these studies. A literature search was performed to inclu

Metabolomics of mammalian brain reveals regional differences.

BACKGROUND: The mammalian brain is organized into regions with specific biological functions and properties. These regions have distinct transcriptomes, but little is known whether they may also differ in their metabolome. The metabolome, a collection of small molecules or metabolites, is at the intersection of the genetic background of a given cell or tissue and the environmental influences that affect it. Thus, the metabolome directly reflects information about the physiologic state of a biological system under a particular condition. The objective of this study was to investigate whether va

Mass spectrometry-based metabolomics for tuberculosis meningitis.

Tuberculosis meningitis (TBM) is a prevalent form of extra-pulmonary tuberculosis that causes substantial morbidity and mortality. Diagnosis of TBM is difficult because of the limited sensitivity of existing laboratory techniques. A metabolomics approach can be used to investigate the sets of metabolites of both bacteria and host, and has been used to clarify the mechanisms underlying disease development, and identify metabolic changes, leadings to improved methods for diagnosis, treatment, and prognostication. Mass spectrometry (MS) is a major analysis platform used in metabolomics, and MS-ba
