Computed tomography of granulomatous pneumonia with oxalosis in an American alligator (Alligator mississippiensis) associated with Metarhizium anisopliae var anisopliae.

An 18-yr-old, male, albino, American alligator (Alligator mississippiensis) was evaluated for decreased appetite and abnormal buoyancy. Computed tomography (CT) of the coelomic cavity showed multifocal mineral and soft tissue attenuating pulmonary masses consistent with pulmonary fungal granulomas. Additionally, multifocal areas of generalized, severe emphysema and pulmonary and pleural thickening were identified. The alligator was euthanized and necropsy revealed severe fungal pneumonia associated with oxalosis. Metarhizium anisopliae var. anisopliae was cultured from lung tissue and exhibite

A New Strain of Metarhizium robertsii Isolated from Loess Plateau and Its Virulence and Pathological Characteristics against Monochamus alternatus.

Monochamus alternatus is a serious trunk-boring pest. The isolation and utilization of entomopathogenic fungi to manage M. alternatus is important. Here, a new strain GQH6 of Metarhizium robertsii, isolated from the Loess Plateau, was identified morphologically and molecularly. The virulence of the strain GQH6 against the third-instar larvae of M. alternatus was studied. Then, the pathological process, including symptom observation and histopathological observation, was also researched. The corrected mortality was 100% at 109 and 108 conidia/mL, and 88.89 ± 5.88% at 107 conidia/mL. The LC50 wa

Increased pathogenicity against coffee berry borer, Hypothenemus hampei (Coleoptera: Curculionidae) by Metarhizium anisopliae expressing the scorpion toxin (AaIT) gene.

Coffee berry borer (CBB) is the Worlds most devastating coffee pest causing an estimated US$500 million worth of losses annually through damage and control costs. Beauveria bassiana and Metarhizium anisopliae have been employed to control this pest but their low virulence (slow kill and large inoculums) is an important factor constraining their use. M. anisopliae (AaIT-Ma549) has been modified to express the scorpion toxin (AaIT) in insect hemolymph and this greatly increased pathogenicity against Manduca sexta and Aedes aegypti. Here, we demonstrate that AaIT-Ma549 was also dramatically more

Acyl-CoA Oxidase-Mediated β-Oxidation Regulates the Lifestyle Transition of Metarhizium rileyi from Insect Pathogen to Plant Mutualist.

Metarhizium rileyi as a widely distributed insect pathogenic fungus, has a promising application prospect in pest control. This study investigates M. rileyi strain XSBN200920 following its endophytic colonization of Nicotiana benthamiana. The fungus establishes systemic endophytic relationships via foliar spray and root drench, significantly improving plant height, stem thickness, leaf area, biomass, and chlorophyll content. Colonized plants showed more than 50% reduction in Spodoptera litura damage and approximately 70% decrease in Alternaria alternata disease severity. Multiomics analyses re

Heterologous expression of the Metarhizium anisopliae barc gene cluster reveals a diphenyl ether-to-xanthene transformation in barceloneic acid biosynthesis.

Filamentous fungi generate chemical diversity through several evolutionary strategies, including the elaboration of conserved biosynthetic intermediates into structurally distinct secondary metabolites. Emodin and its corresponding anthrone are central intermediates in fungal aromatic polyketide metabolism, yet the enzymatic logic that remodels these precursors into more complex scaffolds remains incompletely understood. To investigate this process, we examined an infection-responsive non-reducing polyketide synthase biosynthetic gene cluster (barc) from the entomopathogenic fungus Metarhizium

Metarhizium anisopliae infection reduces Trypanosoma congolense reproduction in Glossina fuscipes fuscipes and its ability to acquire or transmit the parasite.

BACKGROUND: Tsetse fly-borne trypanosomiasis remains a significant problem in Africa despite years of interventions and research. The need for new strategies to control and possibly eliminate trypanosomiasis cannot be over-emphasized. Entomopathogenic fungi (EPF) infect their hosts through the cuticle and proliferate within the body of the host causing death in about 3-14 days depending on the concentration. During the infection process, EPF can reduce blood feeding abilities in hematophagous arthropods such as mosquitoes, tsetse flies and ticks, which may subsequently impact the development a

Susceptibility of adult and larval stages of the horn fly, Haematobia irritans, to the entomopathogenic fungus Metarhizium anisopliae under field conditions.

The efficacy of M. anisopliae strain E9 as a biological insecticide for the adult and larval stages of H. irritans was assessed under field conditions. To assess larvicidal activity, nine heifers were randomly divided into three groups, which were maintained separated from each other. The first group ingested fungal spores encapsulated in alginate pellets. The second group ingested in natura spores that were grown on sterilized rice. In both groups, each animal received three meals a day, with each meal containing 2 x 10(10)conidia. The third group received no treatment and was used as a contr

Commercial Product Based on Metarhizium Anisopliae Affects Survival, Midgut Morphology, and Behavior in the Stingless Bee Scaptotrigona depilis (Hymenoptera: Apidae).

Brazilian social stingless bees are important pollinators and producers of honey, wax, royal jelly, propolis, and other products. These species are experiencing a population decline, attributed to different factors, such as the excessive use of pesticides. Biological control has been proposed as an alternative to chemical control, relying on the use of natural enemies to manage pests, including microorganisms such as entomopathogenic fungi. However, little is known about the effects of these agents on stingless bees. Therefore, this research evaluated the effect of the commercial bioproduct Me

Biocontrol effect of entomopathogenic fungi Metarhizium anisopliae ethyl acetate-derived chemical molecules: An eco-friendly anti-malarial drug and insecticide.

Insect pests represent a major threat to human health and agricultural production. With a current over-dependence on chemical insecticides in the control of insect pests, leading to increased chemical resistance in target organisms, as well as side effects on nontarget organisms, the wider environment, and human health, finding alternative solutions is paramount. The employment of entomopathogenic fungi is one such potential avenue in the pursuit of greener, more target-specific methods of insect pest control. To this end, the present study tested the chemical constituents of Metarhizium aniso

Attachment of Metarhizium anisopliae to the southern green stink bug Nezara viridula cuticle and fungistatic effect of cuticular lipids and aldehydes.

In this paper we examined the conidial attachment of Metarhizium anisopliae on the southern green stink bug, Nezara viridula, using the exuvia and nymphal stage of the host as a substrate for M. anisopliae conidiospores. Initial studies using fluorescein isothiocyanate (FITC)-labeled conidia examined the differential binding of conidia to various sites on the cuticle. Both the topography and the chemistry of the cuticle affected conidial adhesion. Conidia were trapped in areas containing large numbers of setae (e.g., antennal tips, apical portions of tibia and tarsi). Chemical treatments to re

Novel nanoparticle formulations improve the tolerance of Metarhizium brunneum to ultraviolet radiation and biocontrol efficacy against Frankliniella fusca (Thysanoptera: Thripidae).

Oil-in-water Pickering emulsions stabilized by titania (TiO2) nanoparticles have shown potential to increase ultraviolet (UV) protection in the entomopathogenic fungus Metarhizium brunneum. In this study, we expand the findings by comparing several nanoparticle-based formulations for UV tolerance, and investigating the impact of protective formulations in a biocontrol setting. The fungal viability in colony forming units (CFUs) and virulence to the 2nd instar tobacco thrips, Frankliniella fusca, were assessed. Under UV-A (380-nm), a titania-based formulation was effective in protecting the fun

Asphyxiation of Metarhizium robertsii during mycelial growth produces conidia with increased stress tolerance via increased expression of stress-related genes.

Little is known about the impact of hypoxia and anoxia during mycelial growth on tolerance to different stress conditions of developing fungal conidia. Conidia of the insect-pathogenic fungus Metarhizium robertsii were produced on potato dextrose agar (PDA) medium under normoxia (control = normal oxygen concentrations), continuous hypoxia, and transient anoxia, as well as minimal medium under normoxia. The tolerance of the conidia produced under these different conditions was evaluated in relation to wet heat (heat stress), menadione (oxidative stress), potassium chloride (osmotic stress), UV

Characterization of Metarhizium viride Mycosis in Veiled Chameleons (Chamaeleo calyptratus), Panther Chameleons (Furcifer pardalis), and Inland Bearded Dragons (Pogona vitticeps).

Metarhizium viride has been associated with fatal systemic mycoses in chameleons, but subsequent data on mycoses caused by this fungus in reptiles are lacking. The aim of this investigation was therefore to obtain information on the presence of M. viride in reptiles kept as pets in captivity and its association with clinical signs and pathological findings as well as improvement of diagnostic procedures. Beside 18S ribosomal DNA (rDNA) (small subunit [SSU]) and internal transcribed spacer region 1 (ITS-1), a fragment of the large subunit (LSU) of 28S rDNA, including domain 1 (D1) and D2, was s

Divergent immune strategies of Colorado potato beetle larvae against the entomopathogenic fungi Metarhizium robertsii and Beauveria bassiana: A comparative transcriptomic analysis.

The entomopathogenic fungi Beauveria bassiana and Metarhizium robertsii are natural pathogens of the Colorado potato beetle (CPB) and hold promise for biocontrol. However, the insect's immune responses to fungal infections remain poorly understood. In this study, we performed transcriptomic analyses of haemocytes and fat body tissue from CPB larvae topically infected with B. bassiana and M. robertsii at the stage of pathogen encapsulation. Quantitative PCR validation of gene expression, along with complementary physiological and biochemical assays, was also conducted. Reference gene annotation

Two Iron(II), α-Ketoglutarate-Dependent Enzymes Encoded by the PPZ Gene Cluster of Metarhizium majus Enable Production of 8-Hydroxyperamine.

Entomopathogenic fungus Metarhizium majus contains the nine-gene PPZ cluster, with ppzA, encoding a peramine-producing nonribosomal peptide synthetase, as the central component. In this work, the roles of two α-ketoglutarate, iron-dependent oxygenases encoded by the PPZ genes ppzC and ppzD were elucidated. PpzD was found to produce both trans-4-hydroxy-l-proline and trans-3-hydroxy-l-proline in a 13.1:1 ratio, yielding a key precursor for peramine biosynthesis. PpzC was found to act directly on peramine, yielding the novel analogue 8-hydroxyperamine.
