Co-Digestion of Sugarcane Biorefinery Byproducts in Upflow Anaerobic Sludge Blanket Reactors, in Series, with Application of High Organic Loading Rate.

Anaerobic digestion of products from the sugarcane industry is an interesting option for biogas production. In this work, the anaerobic digestion of vinasse and sugarcane molasses supplemented with filter cake was performed in a two-stage upflow anaerobic sludge blanket (UASB) reactors system (R1 and R2). A volumetric methane production (VMP) of up to 4.3 L CH4 (L d)-1 was obtained for R1 + R2, with the application of an organic loading rate (OLR) in R1 of up to 45.0 g total COD (L d)-1. However, for OLR values higher than those applied, a shock load was observed, and only after its reduction

Coupling Mechanism of Methane Production and Carbon Fixation Regulated by Artificial Humic Acid in Paddy Soils: Insights into Microbial Communities and Metabolic Pathways.

The impact of artificial humic acid (HA) under varying water management conditions on the interactions between carbon metabolic pathways in rice soils remains unclear. Therefore, this study employed incubation experiments using HA-treated soil columns under continuous flooding (CF) and alternating wet-dry (AWD) to explore the relationship between CH4 production and CO2 fixation. The results demonstrate that HA significantly increased CH4 emissions, with a 102.87% rise under CF conditions and a 135.13% increase under AWD. In contrast, CO2 emissions remained unaffected under both conditions. Key

Crystal structure of γ-carbonic anhydrase from the polyextremophilic bacterium Aeribacillus pallidus.

The polyextremophilic bacterium Aeribacillus pallidus produces a thermo- and alkali-stable γ-carbonic anhydrase (γ-apCA), a homotrimeric metalloenzyme containing a zinc ion in its active site that catalyzes the reversible hydration of carbon dioxide (CO2). Here, we present the first crystal structure of γ-apCA at 1.7-Å resolution, revealing 2 trimers in the asymmetric unit. The overall structure is consistent with other γ-CAs, where each monomer adopts a prism-like structure consisting of an N-terminal left-handed β-helix and a C-terminal α-helix. The active site, located at the interface betw

Biogeochemical transformation of mercury driven by microbes involved in anaerobic digestion of municipal wastewater.

Anaerobic digestion (AD) with municipal wastewater contained heavy metal mercury (Hg) highly affects the utilization of activated sludge, and poses severe threat to the health of human beings. However, the biogeochemical transformation of Hg during AD remains unclear. Here, we investigated the biogeochemical transformation and environmental characteristics of Hg and the variations of dominant microbes during AD. The results showed that Hg(II) methylation is dominant in the early stage of AD, while methylmercury (MeHg) demethylation dominates in the later stage. Dissolved total Hg (DTHg) in the

Zero valent iron improved methane production and specifically reduced aminoglycoside and tetracycline resistance genes in anaerobic digestion.

It is unadvisable to discuss the antibiotic resistance genes (ARGs) reduction in anaerobic digestion (AD) system neglecting its main purpose-methane production. The methane production improvement coupling with antibiotic resistance genes (ARGs) reduction in anaerobic digestion (AD) by zero valent iron (ZVI) were simultaneously investigated. Whether the role of ZVI on the ARGs fate was random or specific was clarified through the high-throughput qPCR (HT-qPCR). Results indicated that ZVI improved methane production and ARGs reduction by 23.9% and 25.0%, respectively. The improved methane produc

Fate of mercury and methylmercury in full-scale sludge anaerobic digestion combined with thermal hydrolysis.

Methylmercury (MeHg) is one of the highly toxic and bio-accumulated forms of mercury. Its presence in wastewater treatment processes has been evidenced in recent studies. Considering its enrichment in sewage sludge and the ecological risk associated with its land application, this study investigated the fate of mercury and MeHg in full-scale anaerobic digestion combined with Cambi thermal hydrolysis based on one-year sampling. Results showed that the advanced anaerobic digestion could increase the total mercury (THg) content from 4.35 ± 0.43 mg/kg in raw sludge to 6.37 ± 1.05 mg/kg in digested

The rice field cyanobacteria Anabaena azotica and Anabaena sp. CH1 express vanadium-dependent nitrogenase.

Anabaena azotica FACHB-118 and Anabaena sp. CH1, heterocystous cyanobacteria isolated from Chinese and Taiwanese rice fields, expressed vanadium-containing nitrogenase when under molybdenum deficiency. This is the second direct observation of an alternative nitrogenase in cyanobacteria. The vanadium nitrogenase-specific genes vnfDG are fused and clustered in a phylogenetic tree next to the corresponding genes of Methanosarcina. The expression of vnfH in cells cultured in Mo-free medium and of nifH in Mo-grown cells was shown for the first time by sequencing cDNA derived from cultures of A. azo

Enhanced anaerobic digestion of human feces by ferrous hydroxyl complex (FHC): Stress factors alleviation and microbial resistance improvement.

Anaerobic digestion (AD) offers a reliable strategy for resource recovery from source-separated human feces (HF), but is limited by a disproportionate carbon/nitrogen (C/N) ratio. Ferrous hydroxyl complex (FHC) was first introduced into the HF-AD system to mediate methanogenesis. Mono-digestion of undiluted HF was inhibited by high levels of volatile fatty acids (VFAs), ammonia, and hydrogen sulfide (H2S). FHC addition at optimum dosage (500-1000 mg/L) increased the cumulative methane (CH4) yield by 22.7%, enhanced the peak value of daily CH4 production by 60.5%, and shortened the lag phase by

Characterization of LIC11207, a novel leptospiral protein that is recognized by human convalescent sera and prevents apoptosis of polymorphonuclear leukocytes.

We report the study of a predicted outer-membrane leptospiral protein encoded by the gene lic11207. This protein is conserved in several pathogenic leptospiral strains but is absent in the saprophyte Leptospira biflexa. This putative outer-membrane protein has a domain of unknown function (DUF) 1565 found in several phylogenetically diverse bacteria and in the archaeon Methanosarcina acetivorans. The gene was cloned and expressed in Escherichia coli BL21 (SI) strain using the expression vector pDEST17. The 34 kDa recombinant protein was tagged with N-terminal hexahistidine and purified by meta

Temperature-dependent oligomerization in M-crystallin: lead or lag toward cataract, an NMR perspective.

The oligomerization and/or aggregation of proteins is of critical importance in a wide variety of biomedical situations, ranging from abnormal disease states like Alzheimer's and Parkinson's disease to the production of inclusion bodies, stability, and delivery of protein drugs. In the case of eye-lens proteins, oligomerization is implicated in cataract formation. In the present study, we have investigated the temperature driven oligomerization of M-crystallin, a close homologue of eye-lens proteins, using NMR spectroscopy and dynamic-light scattering (DLS). The NMR data primarily included R(1

Unveiling the regulation of biochar and ferrihydrite on organic carbon stabilization, CH4 emission and microbial community in paddy soil.

Biochar (BC), an accessible and economic soil improvement, has influenced the biogeochemical cycling of iron and organic carbon (OC) in paddy soil. However, little is known about the effect of BC on iron-organic carbon (Fe-OC) associations, as well as how they regulate the associated microbial community in paddy soil. This study employed anoxic soil microcosms with distinct treatments including BC alone, ferrihydrite (Fh) alone, and BC combined with Fh. Results demonstrated that Fh alone significantly reduced CH₄ emission by suppressing key methanogens and stabilizing humic-like dissolved orga

Virus-integrated laboratory models for habitability studies of Enceladus, Europa, and Titan.

Biogeochemical models have become critical for answering questions about the habitability of ocean worlds, especially that of early Earth. However, when extended to icy moons, experimental biogeochemical systems become especially challenging. Due to less constrained physicochemical parameters extrapolated from former flybys, the most comprehensive approaches could combine experimental and computational models of proposed ocean conditions with microbes present. This minireview makes the case for needing to also include viral/phage interactions with microbial models as a mechanism for maintainin

Enhancement of Activated Carbon on Anaerobic Fermentation of Heavy-Metal-Contaminated Plants: Insights into Microbial Responses.

Anaerobic fermentation is a potentially cost-effective approach to disposing of metal-contaminated biowaste collected during phytoremediation. However, the compound heavy metals contained in the biowaste may limit the efficiency of anaerobic fermentation. In this study, anaerobic fermentation with alfalfa harvested from an iron tailing as the feedstock was set up and further enhanced by granular activated carbon (AC). The results showed that adding AC improved the cumulative biogas yields of alfalfa contaminated with metals (AM) by 2.26 times. At the biogas peak stage, plenty of microbes were

Phenotypic properties and microbial diversity of methanogenic granules from a full-scale upflow anaerobic sludge bed reactor treating brewery wastewater.

Methanogenic granules from an anaerobic bioreactor that treated wastewater of a beer brewery consisted of different morphological types of granules. In this study, the microbial compositions of the different granules were analyzed by molecular microbiological techniques: cloning, denaturing gradient gel electrophoresis and fluorescent in situ hybridization (FISH), and scanning and transmission electron microscopy. We propose here that the different types of granules reflect the different stages in the life cycle of granules. Young granules were small, black, and compact and harbored active cel

The potential role of iron-carbon micro-electrolysis materials in curtailing lag-phase stimulates kitchen waste anaerobic digestion at different solid contents: Performance, synergistic effect and microbial response.

High-solid anaerobic digestion (HSAD) of kitchen waste was generally faced to the common problems such as systemic acidification, prolonged lag-phase time and low methane production. Iron-carbon micro-electrolysis (ICME) materials exhibited advantages that porous structure, large specific surface area and excellent conductivity. It was beneficial for organic compounds to hydrolysis. Moreover, ICME materials could establish direct interspecies electron transfer (DIET) pathway between bacteria and methanogens. ICME materials were commonly used to enhance the AD of wastewater, but they were rarel

Characteristics of archaea and bacteria in rice rhizosphere along a mercury gradient.

Several strains of archaea have the ability to methylate or resist mercury (Hg), and the paddy field is regarded to be conducive to Hg methylation. However, our knowledge of Hg-methylating or Hg-resistant archaea in paddy soils is very limited so far. Therefore, the distribution of archaea and bacteria in the rhizosphere (RS) and bulk soil (BS) of the rice growing in Xiushan Hg-mining area of southwest China was investigated. Bacterial and archaeal 16S rRNA gene amplicon sequencing of the rice rhizosphere along the Hg gradient was conducted. THg concentrations in RS were significantly higher t

Biotic and abiotic processes contribute to successful anaerobic degradation of cyanide by UASB reactor biomass treating brewery waste water.

In contrast to the general aerobic detoxification of industrial effluents containing cyanide, anaerobic cyanide degradation is not well understood, including the microbial communities involved. To address this knowledge gap, this study measured anaerobic cyanide degradation and the rearrangements in bacterial and archaeal microbial communities in an upflow anaerobic sludge blanket (UASB) reactor biomass treating brewery waste water using bio-methane potential assays, molecular profiling, sequencing and microarray approaches. Successful biogas formation and cyanide removal without inhibition we

Magnetite Alters the Metabolic Interaction between Methanogens and Sulfate-Reducing Bacteria.

It is known that the presence of sulfate decreases the methane yield in the anaerobic digestion systems. Sulfate-reducing bacteria can convert sulfate to hydrogen sulfide competing with methanogens for substrates such as H2 and acetate. The present work aims to elucidate the microbial interactions in biogas production and assess the effectiveness of electron-conductive materials in restoring methane production after exposure to high sulfate concentrations. The addition of magnetite led to a higher methane content in the biogas and a sharp decrease in the level of hydrogen sulfide, indicating i

Depth-dependent microbial communities potentially mediating mercury methylation and various geochemical processes in anthropogenically affected sediments.

Metal contamination and other geochemical alterations affect microbial composition and functional activities, disturbing natural biogeochemical cycles. Therefore, it is essential to understand the influences of multi-metal and geochemical interactions on microbial communities. This work investigated the distributions of total mercury (THg), methylmercury (MeHg), and trace metals in the anthropogenically affected sediment. The microbial communities and functional genes profiles were further determined to explore their association with Hg-methylation and geochemical features. The levels of THg a
