Akt regulates the fertility of Coridius chinensis by insulin signaling pathway.

Akt (also known as protein kinase B) belongs to the multifunctional serine/threonine kinase family and is an important component of the insulin signaling pathway that plays a key role in many biological processes such as cell growth, proliferation, and survival. However, few studies have reported the effect of Akt on reproduction in Hemiptera. In this study, we cloned and characterized the Akt gene from Coridius chinensis (CcAkt). The open reading frame of CcAkt has a length of 1,563 bp and encodes 520 amino acids. It has a conserved pleckstrin homology domain (PH), catalytic domain of serine/

RXR Ligands Negatively Regulate Thrombosis and Hemostasis.

OBJECTIVE: Platelets have been found to express intracellular nuclear receptors including the retinoid X receptors (RXRα and RXRβ). Treatment of platelets with ligands of RXR has been shown to inhibit platelet responses to ADP and thromboxane A2; however, the effects on responses to other platelet agonists and the underlying mechanism have not been fully characterized. APPROACH AND RESULTS: The effect of 9-cis-retinoic acid, docosahexaenoic acid and methoprene acid on collagen receptor (glycoprotein VI [GPVI]) agonists and thrombin-stimulated platelet function; including aggregation, granule

Efficacy of Broadline® in cats against induced infections with developing fourth-stage larval and adult Ancylostoma ceylanicum hookworms.

Ancylostoma ceylanicum hookworms parasitize dogs and cats in subtropical and tropical South East Asia and Pacific region, the Arab Peninsula, southern Africa and South America but cause patent infections in humans too. To evaluate the efficacy of Broadline®, a combination product comprising eprinomectin, fipronil, (S)-methoprene and praziquantel, against developing fourth-larval stage (L4) and adult A. ceylanicum in cats, a blinded, induced infection study was conducted. Following oral inoculation of 24 purpose bred cats with ˜300 infectious A. ceylanicum larvae each, cats were allocated to on

Larviciding Culex spp. (Diptera: Culicidae) Populations in Catch Basins and Its Impact on West Nile Virus Transmission in Urban Parks in Atlanta, GA.

In urban environments, road-side catch basins are common larval habitats of Culex spp. (Diptera: Culicidae) mosquitoes and important targets of larval control in areas subject to West Nile virus (WNv) transmission. We quantified the impact of larviciding basins on Culex spp. populations and WNv infection prevalence by treating basins in and around urban parks in Atlanta, GA, using Mosquito Dunks and Bits (active ingredient, a.i., Bacillus thuringiensis subsp. isrealensis, Dunks-10.31%, Bits-2.86%) and Altosid 30-Day Briquets (a.i., S-methoprene 8.62%) in two separate seasons. A field evaluation of four larval mosquito control methods in urban catch basins.

Effective control of mosquitoes in rural Midwestern communities that lack organized mosquito control districts would be aided by baseline data on optimal breeding sites, interannual effects of climate on population emergence and abundance, and efficacy of various control options under field conditions. During 19 surveillance weeks in the summer of 2005, we sampled 100 catch basins each week that were distributed among 10 study zones. Catch basins within each study zone were subjected to 1 of 4 different mosquito control methods or were left untreated. Of the 10 study zones, 5 were in high-inte

Hormonal regulation of microRNA expression dynamics in the gut of the yellow fever mosquito Aedes aegypti.

The yellow fever mosquito Aedes aegypti is an obligatory blood feeder and a major arboviral disease vector, evoking severe public health concerns worldwide. In adult female mosquitoes, the gut is critical for blood digestion and pathogen entry. We aimed for a systematic exploration of microRNA expression dynamics in the gut during the gonadotrophic cycle. Small RNA libraries were constructed from female mosquito gut tissues at five time points. Unsupervised hierarchical clustering revealed three expression clusters (early, mid and late) peaking at sequential time points - 24, 48 and 72 h poste

Two juvenile hormone suppressible storage proteins may play different roles in Hyphantria cunea Drury.

We isolated and sequenced cDNA clones corresponding to two storage proteins (HcSP-1 and HcSP-2) from fall webworm, Hyphantria cunea. The cDNAs for HcSP-1 (2,337 bp) and HcSP-2 (2,572 bp) code for 753 and 747 residue proteins with predicted molecular masses of 88.3 and 88.5 kDa, respectively. The calculated isoelectric points are pI = 8.4 (HcSP-1) and 7.6 (HcSP-2). Multiple alignment analysis of the amino acid sequence revealed that HcSP-1 is most similar to SL-1 from S. litura (73.8% identity) and other methionine-rich hexamers, whereas HcSP-2 is most similar to the SL-2 alpha subunit from S.

Differential Juvenile Hormone Variations in Scale Insect Extreme Sexual Dimorphism.

Scale insects have evolved extreme sexual dimorphism, as demonstrated by sedentary juvenile-like females and ephemeral winged males. This dimorphism is established during the post-embryonic development; however, the underlying regulatory mechanisms have not yet been examined. We herein assessed the role of juvenile hormone (JH) on the diverging developmental pathways occurring in the male and female Japanese mealybug Planococcus kraunhiae (Kuwana). We provide, for the first time, detailed gene expression profiles related to JH signaling in scale insects. Prior to adult emergence, the transcrip

Membrane-localized MET engages PVR to mediate extranuclear juvenile hormone signaling in Aedes aegypti.

Although Methoprene-tolerant (MET) is well established as the intracellular receptor mediating the genomic actions of juvenile hormone (JH) in insects, the identity of the receptor responsible for initiating extranuclear JH responses has remained elusive. In the yellow fever mosquito Aedes aegypti, we identify the platelet-derived growth factor- and vascular endothelial growth factor-receptor related (PVR) protein as a key mediator of membrane-initiated JH signaling. JH treatment induces robust PVR phosphorylation in the fat body of adult female mosquitoes. Disruption of PVR function suppresse

Transcriptional regulation of juvenile hormone-mediated induction of Krüppel homolog 1, a repressor of insect metamorphosis.

The Krüppel homolog 1 gene (Kr-h1) has been proposed to play a key role in the repression of insect metamorphosis. Kr-h1 is assumed to be induced by juvenile hormone (JH) via a JH receptor, methoprene-tolerant (Met), but the mechanism of induction is unclear. To elucidate the molecular mechanism of Kr-h1 induction, we first cloned cDNAs encoding Kr-h1 (BmKr-h1) and Met (BmMet1 and BmMet2) homologs from Bombyx mori. In a B. mori cell line, BmKr-h1 was rapidly induced by subnanomolar levels of natural JHs. Reporter assays identified a JH response element (kJHRE), comprising 141 nucleotides, loca

Juvenile hormone receptor relies on conserved domain interfaces for dimerization-dependent activity.

Juvenile hormone (JH) is vital to insects. In Drosophila, JH signals through a basic helix-loop-helix-PER-ARNT-SIM (bHLH-PAS) protein methoprene-tolerant (MET) and its paralog germ cell-expressed (GCE). Binding of JH to the PAS-B domain of MET or GCE triggers the assembly of a DNA-bound, transcriptionally active JH receptor (JHR) heterodimer comprising either MET or GCE and their partner taiman (TAI). Molecular mechanisms of the JHR complex activation remain unclear. Here, we use the Drosophila GCE protein to address the presumably critical but as yet unexplored function of the other PAS domai

Juvenile Hormone Involved in the Defensive Behaviors of Soldiers in Termite Reticulitermes aculabialis.

Eusocial insects have evolved specific defensive strategies to protect their colonies. In termite colonies, soldiers perform a colony-level defense by displaying mechanical biting, head-banging and mandible opening-closing behaviors. However, few studies have been reported on the factors modulating defensive behaviors in termites. Owing to JH (juvenile hormone) being involved in soldier differentiation, JH was speculated to affect defensive behaviors in termite soldiers. To determine the effect of JH on the defensive behaviors of termite soldiers, we performed a JHA-feeding and RaSsp1-silencin

Lungworms and gastrointestinal parasites of domestic cats: a European perspective.

With the exception of Aelurostrongylus abstrusus, feline lungworms have been poorly studied. Information on their distribution is patchy and mostly limited to case reports. In this study, the occurrence of feline lungworms and co-infecting gastrointestinal parasites has been investigated in 12 European countries (i.e. Austria, Belgium, Bulgaria, France, Greece, Hungary, Italy, Portugal, Romania, Spain, Switzerland and the United Kingdom). An average of 10 domestic cats, with regular outdoor access, was sampled each month for 12months, and freshly passed faeces were collected. Stools were proce

Stage- and cell-specific expression of ecdysone receptors and ecdysone-induced transcription factors during midgut remodeling in the yellow fever mosquito, Aedes aegypti.

In insects, especially in mosquitoes that are adult blood feeders, midgut remodeling is an important event during metamorphosis. It involves two processes viz., programmed cell death (PCD) of larval cells, and proliferation and differentiation of imaginal cells to form pupal/adult midgut. These processes are regulated by 20-hydroxyecdysone (20E) and juvenile hormone (JH), but the signaling mechanisms, which trigger specific changes remain poorly understood. Here, we report stage- and cell-specific expression of ecydone receptor (EcR), ultraspiracle (USP), broad (Br), E75B and hormone receptor

Juvenile hormone regulation of microRNAs is mediated by E75 in the Dengue vector mosquito Aedes aegypti.

MicroRNAs (miRNAs) are small noncoding RNAs that play critical roles in controlling posttranscriptional gene regulation and have a profound effect on mosquito reproduction and metabolism. Juvenile hormone (JH) is critical for achieving reproductive competence in the main vector of human arboviral diseases, Aedes aegypti We report a JH-mediated mechanism governing miRNA expression. Using a transcription factor screen with multiple primary miRNA (pri-miRNA) promoters, we identified that the Ecdysone-induced protein E75 (E75) isoform (E75-RD) induced miRNA gene promoter activity. Field-Recovered Polystyrene Exposure Alters Developmental Endocrine Signaling and Cholinergic Function in the Plastivorous Insect Zophobas morio.

The use of insect larvae for plastic waste biodegradation has attracted increasing attention as a potential strategy to mitigate environmental plastic pollution. However, the toxicological consequences of ingesting plastics recovered from the environment, particularly those acting as vectors for sorbed contaminants, remain poorly understood. In this study, we investigated the endocrine and neurotoxic effects of field-recovered (environmental) polystyrene (ePS) in the plastivorous insect Zophobas morio. Chitin synthase A: a novel epidermal development regulation gene in the larvae of Bombyx mori.

Chitin synthase is the key regulatory enzyme for chitin synthesis and excretion in insects, as well as a specific target of insecticides. The chitin synthase A gene (BmChsA) cloned from Bombyx mori, the model species of lepidopteran, is an epidermis-specific expressed gene during the molting stage. Knockdown BmChsA gene in 3rd instar larvae increased the number of non-molting and abnormal molting larvae. Exposure to nikkomycin Z, a chitin synthase inhibitor downregulated the expression of BmChsA and decreased the amount of epidermis chitin during the molting process. The thickness of the new e

Lysine Triggers Acute Oviposition by Activating the 20E-ETH-JH Signaling Cascade in Drosophila melanogaster.

Reproductive performance is closely linked to nutrient availability, yet the specific nutrient-derived cues governing oviposition decision-making remain inadequately defined. Chemically defined holidic media used in Drosophila melanogaster studies provide precise control over dietary components; however, the mechanisms by which individual nutrients exert acute effects on inter-organ endocrine signaling to trigger oviposition behavior are not well understood. In a two-choice oviposition assay, where substrates are presented within the same chamber, we observed that HM induces a rapid increase i
