A methylene blue-mediated enzyme electrode for the determination of trace mercury(II), mercury(I), methylmercury, and mercury-glutathione complex.

A methylene blue-mediated enzyme biosensor has been developed for the detection of inhibitors including mercury(II), mercury(I), methylmercury, and mercury-glutathione complex. The inhibition to horseradish peroxidase was apparently reversible and noncompetitive in the presence of HgCl2 in less than 8 s and irreversibly inactivated when incubated with different concentrations of HgCl2 for 1-8 min. The binding site of horseradish peroxidase with HgCl2 probably was a cysteine residue SH. Mercury compounds can be assayed amperometrically with the detection limits 0.1 ng ml(-1) Hg for HgCl2 and me

Apoptotic process of cerebellar degeneration in experimental methylmercury intoxication of rats.

This report deals with the mechanism involved in the cerebellar degeneration following experimental methylmercury poisoning of male Wistar rats. The cerebellar granule cells of animals that exhibited typical hind leg paresis were shrunken and displayed marked nuclear pyknosis. At the ultrastructural level, the nuclei of these cells were condensed and fragmented, features which are characteristic of apoptosis. In situ staining for DNA strand breaks revealed that the pyknotic nuclei were positively labeled. DNA fragmentation was confirmed by agarose gel electrophoresis; a ladder pattern of multi

Organic mercury: an environmental threat to the health of dietary-exposed societies?

As a natural element, mercury is ubiquitous in the environment. The largest amount of mercury, amounting to approximately 100,000 tons per year, originates from the degassing of the earth's crust. To this amount, such anthropogenic activities as combustion of fossil fuels and releases from industrial activities add approximately 20,000 tons of mercury every year. The emitted mercury, both natural and anthropogenic, is in an inorganic form, predominantly as the metallic vapor (Hgzero). In aquatic environments, however, inorganic mercury is microbiologically transformed into the lipophilic organ

Environmental challenges for the nervous system and the brain in Japan.

Japan provides many lessons for the Environmental Neurology's issues. Drama and disasters have paved the recent history of Japan. The Japanese people have been intoxicated by chemical compounds (methylmercury, sulfur dioxide, cadmium, PCBs and other dioxin-related compounds) and were the victims of several dramatic disasters (atomic bombing, nuclear disaster, sarin gas attack). They are still exposed to air pollution. Prion diseases including dura-graft-associated CJD are still an issue. In addition, continuously spreading chronic wasting disease is a worldwide challenge .

Methylmercury Resistance in Desulfovibrio desulfuricans Strains in Relation to Methylmercury Degradation.

Two strains of Desulfovibrio desulfuricans, one known to synthesize monomethylmercury from ionic mercury, were grown to determine methylmercury toxicity and for comparison with an anaerobic strain of Clostridium pasteurianum, a H(2) producer, and with the broad-spectrum mercury-resistant Pseudomonas putida strain FB-1, capable of degrading 1 mug of methylmercury to methane and elemental mercury in 2 h. The CH(3)HgCl resistance of D. desulfuricans strains was 10 times that of P. putida FB-1 and 100 times that of C. pasteurianum. The methylmercury resistance of D. desulfuricans was related to th

Mercury and monomethylmercury: present and future concerns.

Global atmospheric changes carry the potential to disrupt the normal cycling of mercury and its compounds. Acid rain may increase methylmercury levels in freshwater fish. Global warming and increased ultraviolet radiation may affect the global budget of methylmercury, including its formation and degradation in both biotic and abiotic environments. In this article we review current knowledge on mercury and monomethylmercury with regard to their environmental fate and the potential for human health effects. Recent findings indicate that atmospheric Hg deposition readily accounts for the total ma

Mercury in human hair as an indicator of the fish consumption.

OBJECTIVES: Mercury and most of its compounds are extremely toxic and should be handled with care. It can be inhaled and absorbed through the skin and mucous membranes. The most toxic forms of mercury are its organic compounds such as dimethylmercury and methylmercury. Fish have a natural tendency to accumulate mercury. Methylmercury is produced by microbial methylation of inorganic mercury in water sediment then it infiltrates the food chain and it consequently accumulates in fish. Fish are the main source of methylmercury in human food. Mercury is transferred into a hair; and this can be tha

Methylation of mercury and tin by estuarine microbial mats.

After tin and mercury salts were added to estuarine microbial mats increasing amounts of methyltin and methylmercury, respectively, were formed over a 30 to 100 hour time period. Inhibition of the methylation by molybdate, a metabolic inhibitor of sulfate reduction, stimulation by pyruvate addition and lack of methylation by sterilized mats, were evidence that sulfate reducing bacteria within the mats were responsible for the tin and mercury methylation. Methyl mercury was formed from mercuric chloride and mercuric cysteine, but not from mercuric sulfide. We suggest that mercury bound to organ

The Impact of the Major Baltic Inflow of December 2014 on the Mercury Species Distribution in the Baltic Sea.

The Baltic Sea is a marginal sea characterized by stagnation periods of several years. Oxygen consumption in its deep waters leads to the buildup of sulfide from sulfate reduction. Some of the microorganisms responsible for these processes also transform reactive ionic mercury to neurotoxic methylmercury. Episodic inflows of oxygenated saline water from the North Sea temporally re-establish oxic life in deep waters of the Baltic Sea. Thus, this sea is an especially important region to better understand mercury species distributions in connection with variable redox conditions. Mercury species

Mercury and human genotoxicity: critical considerations and possible molecular mechanisms.

Mercury compounds versatility explains their numerous applications in diverse areas of industry. The growing use of this metal has resulted in a significant increase of environment contamination and episodes of human intoxication, arousing the concern of international organisms. Meanwhile, consequences of these intoxication outbreaks are still not fully understood, especially if we consider long-term effects of chronic exposure to relatively low levels of mercury compounds. In the present manuscript, studies about the genotoxicity of mercury compounds, performed in vitro, in vivo, and/or inclu

Mercury methylation in aquatic systems affected by acid deposition.

Recently, it has been noted that fish in acidified lakes may contain elevated levels of mercury. While there is correlation among lakes between depressed pH and high mercury concentrations in fish, the cause of this problem is unknown. A number of hypotheses have been advanced in explanation, including increased mercury deposition, changes in mercury mobility due to acidification, pH dependent changes in mercury uptake by biota, and alterations in population size and/or structure which result in increased bioaccumulation in fish. Because fish accumulate mercury mainly in an organic form, methy

Morphine protects against methylmercury intoxication: a role for opioid receptors in oxidative stress?

Mercury is an extremely dangerous environmental contaminant responsible for episodes of human intoxication throughout the world. Methylmercury, the most toxic compound of this metal, mainly targets the central nervous system, accumulating preferentially in cells of glial origin and causing oxidative stress. Despite studies demonstrating the current exposure of human populations, the consequences of mercury intoxication and concomitant use of drugs targeting the central nervous system (especially drugs used in long-term treatments, such as analgesics) are completely unknown. Morphine is a major

Dynamic Protein Adduct Formation by Methylmercury: Molecular Targets, Ligand Exchange, and Toxicological Consequences.

Methylmercury (MeHg) is a highly toxic environmental contaminant that preferentially interacts with sulfur- and selenium-containing biomolecules. Although MeHg exposure is associated with disruption of redox homeostasis, calcium signaling, mitochondrial function, and neuronal processes, the molecular mechanisms linking MeHg chemistry to these diverse biological effects remain incompletely understood. A central feature of MeHg chemistry is its high affinity for thiol and selenol groups and its ability to undergo ligand exchange reactions with low-molecular-weight and protein thiols and selenols

A Novel Diselenide-Probucol-Analogue Protects Against Methylmercury-Induced Toxicity in HT22 Cells by Upregulating Peroxide Detoxification Systems: a Comparison with Diphenyl Diselenide.

Methylmercury (MeHg) is a ubiquitous environmental neurotoxicant whose mechanisms of action involve oxidation of endogenous nucleophilic groups (mainly thiols and selenols), depletion of antioxidant defenses, and disruption of neurotransmitter homeostasis. Diphenyl diselenide-(PhSe)2-a model diaryl diselenide, has been reported to display significant protective effects against MeHg-induced neurotoxicity under both in vitro and in vivo experimental conditions. In this study, we compared the protective effects of (PhSe)2 with those of RC513 (4,4'-diselanediylbis(2,6-di-tert-butylphenol), a novel

Production of Methylmercury in Peatlands Following Permafrost Thaw Increases along a Trophic Gradient.

Permafrost thaw in peatlands risks increasing the production and mobilization of methylmercury (MeHg), a bioaccumulative neurotoxin that poses a health hazard to humans. We studied 12 peatlands on a trophic gradient in northwestern Canada, including permafrost peat plateaus and thawed bogs and fens, to determine the effects of thaw on MeHg production from measures of soil and porewater MeHg and in situ methylation assays. The production of MeHg was greater in thawed peatlands, especially rich fens, as indicated by higher potential rates of microbial methylation of inorganic mercury (Hg) to MeH

Assessing comparability and uncertainty of analytical methods for methylated mercury species in seawater.

BACKGROUND: The relative distribution and importance of monomethylmercury (MMHg) and dimethylmercury (DMHg) in seawater is still under debate. A lack of comparability between measurements at sub-picomolar levels hampered the further understanding of the biogeochemical Hg cycle. To overcome this, we assessed the relative standard measurement uncertainties (Uex,r) for direct measurements of MMHg and DMHg by species-specific isotope dilution ICP-MS and cryo-focusing GC-ICP-MS at femtomolar concentrations. Furthermore, Uex,r was determined for the indirect determination of DMHg (DMHgcalc = MeHg -

Methylmercury Determination in Fish by Direct Mercury Analyzer.

BACKGROUND: There are two methods for quantifying methylmercury (MeHg) in fish using GC-electron capture detection (ECD): AOAC INTERNATIONAL Official Method 988.11 and the Korean Food Code (KFC) method. Both of these methods consume a large amount of chemicals and require long pretreatment times because of several complicated MeHg extraction steps. OBJECTIVE: In this study, a new method for the simple and rapid determination of MeHg in fish has been developed. The method is based on the investigation of oxygen combustion-gold amalgamation using a direct mercury analyzer (DMA) after the comple

Chelating agents for the sequestration of mercury(II) and monomethyl mercury(II).

Both mercury(II) and monomethyl mercury(II) poisonings are of great concern for several reasons. As it happens for other metals, chelation therapy is the most indicated treatment for poisoned patients. The efficacy of the therapy and the reduction of side-effects can be sensibly enhanced by an accurate knowledge of all the physiological mechanisms involved in metal uptake, transport within and between various tissues, and (possibly) clearance. All these aspects, however, are strictly dependent on the chemical speciation (i.e., the distribution of the chemical species of a component in a given

Spatial characteristics of net methylmercury production hot spots in peatlands.

Many wetlands are sources of methylmercury (MeHg) to surface waters, yet little information exists about the distribution of MeHg within wetlands. Total mercury (THg) and MeHg in peat pore waters were studied in four peatlands in spring, summer, and fall 2005. Marked spatial variability in the distribution of MeHg, and %MeHg as a proxy for net MeHg production, was observed, with highest values occurring in discrete zones. We denote these zones "MeHg hot spots", defined as an area where the pore water %MeHg exceeded the 90th percentile of the data set (n=463) or >22% of THg as MeHg. MeHg hot sp
