Interventional Microbubble Enhanced Sonothrombolysis on Left Ventricular Assist Devices.

The left ventricular assist device (LVAD) is often used in the treatment of heart failure. However, 4% to 9% implanted LVAD will have thrombosis problem in one year, which is fatal to the patient's life. In this work, an interventional sonothrombolysis (IST) method is developed to realize the thrombolysis on LVAD. A pair of ultrasound transducer rings is installed on the shell of LVAD, and drug-loaded microbubbles are injected into the LVAD through the interventional method. The microbubbles are adhere on the thrombus with the coated thrombus-targeted drugs, and the thrombolytic drugs carried

Intravascular forward-looking ultrasound transducers for microbubble-mediated sonothrombolysis.

Effective removal or dissolution of large blood clots remains a challenge in clinical treatment of acute thrombo-occlusive diseases. Here we report the development of an intravascular microbubble-mediated sonothrombolysis device for improving thrombolytic rate and thus minimizing the required dose of thrombolytic drugs. We hypothesize that a sub-megahertz, forward-looking ultrasound transducer with an integrated microbubble injection tube is more advantageous for efficient thrombolysis by enhancing cavitation-induced microstreaming than the conventional high-frequency, side-looking, catheter-m

Microbubble Contrast Agents for Molecular Ultrasound Imaging of Tumor Vasculature.

Molecular ultrasound imaging was proposed more than two decades ago. The ultrasound contrast agents used for selective targeting of vascular biomarkers of disease are formulated as gas microbubbles surrounded by a thin shell.Microbubbles are decorated by the ligands that specifically bind to the biomarkers of disease overexpressed on the surface of vascular endothelium. The microbubble gas core is most often a poorly soluble perfluorinated gas (preferably C4F10 as the least soluble), to assure circulation lifetime for at least several minutes. The gas from circulating bubbles is exhaled via th

Enhancing the role of paediatric ultrasound with microbubbles: a review of intravenous applications.

Contrast-enhanced ultrasound (CEUS) represents a complementary technique to greyscale and colour Doppler ultrasonography which allows for real-time visualization and characterization of tissue perfusion. Its inherent advantages in the child makes ultrasonography an ideal imaging modality; repeatability and good tolerance along with the avoidance of CT, a source of ionizing radiation, renders ultrasonography imaging desirable. Although currently paediatric CEUS is principally used in an "off-label" manner, ultrasonography contrast agents have received regulatory approval for assessment of paedi

Microbubbles at the bedside: intracavitary CEUS to confirm chest-drain position and loculations.

BACKGROUND: CEUS is increasingly used beyond hepatobiliary imaging; its pleuro-pulmonary applications remain limited, especially for device assessment. CASE DESCRIPTION: We describe a postoperative patient with moderate right pleural effusion and associated middle-basal atelectasis causing hypoxemic respiratory failure. After ultrasound-guided insertion of a 12-Fr pigtail drain, the catheter trajectory and tip could not be confidently visualized due to limited patient mobilization and small-bore caliber. A second-generation UCA (sulfur hexafluoride) diluted 1 mL in 10 mL saline was instilled

Safety of contrast-enhanced ultrasound using microbubbles in human pregnancy: A scoping review.

Successful placentation is crucial for fetal development and maintaining a healthy pregnancy. Placental insufficiency can cause a variety of obstetric complications. Despite the many efforts to enhance diagnosing placental insufficiency, no imaging technique has proven satisfactory. A promising imaging technique is contrast-enhanced ultrasound (CEUS) using microbubbles which has proven capable of (micro)vascular imaging. Its use for placental vascularization assessment in human pregnancies remains constrained by limited evidence and safety concerns. This scoping review aims to demonstrate the

Modulating Nonlinear Acoustic Response of Phospholipid-Coated Microbubbles with pH for Ultrasound Imaging.

Activatable microbubble contrast agents for contrast-enhanced ultrasound have a potential role for measuring physiologic and pathologic states in deep tissues, including tumor acidosis. In this study, we describe a novel observation of increased harmonic oscillation of phosphatidylcholine microbubbles (PC-MBs) in response to lower ambient pH using a clinical ultrasound scanner. MB echogenicity and nonlinear echoes were monitored at neutral and acidic pH using B-mode and Cadence contrast pulse sequencing (CPS), a harmonic imaging technique at 7.0 and 1.5 MHz. A 3-fold increase in harmonic signa

CO2 Microbubbles in Silicone Oil (Part I: Study of the Frequency of Formation).

The objective of this work is the study of gas microbubble dissolution in a carrier liquid. To achieve this, we will analyze, using microfluidic techniques, the formation and evolution of carbon dioxide (CO2) gas microbubbles in silicone oil, monitoring the size, position, and distance between the formed bubbles as they advance through a microchannel. This work consists of two parts (Part I and Part II): in Part I, we analyze the mechanisms determining the variation in the spatial frequency of bubbles as they move through the microchannel, while Part II examines the evolution of their size and

The involvement of transient receptor potential channels in mast cell activation by microbubbles.

This study was to explore the activation of mast cells by microbubbles, with the focus on transient receptor potential (TRP) channels mediated degranulation and calcium influx. Bone marrow-derived mast cells (BMMCs) were primarily obtained from femurs in mice and induced differentiation for 4 weeks. After the purity identification, BMMCs were contacted by homogeneous microbubbles with the diameter of 1 mm for 1 h. β-hexosaminidase and histamine levels in supernatants were assessed by enzyme-linked immunosorbent assay (ELISA) and the CD63 expression was tested by flow cytometry. Microbubbles Remove Listeria monocytogenes from the Surface of Stainless Steel, Cucumber, and Avocado.

Fresh produce may be contaminated by bacterial pathogens, including Listeria monocytogenes, during harvesting, packaging, or transporting. A low-intensity cavitation process with air being injected into water was studied to determine the microbubbles' efficiency when detaching L. monocytogenes from stainless steel and the surface of fresh cucumber and avocado. Stainless steel coupons (1″ × 2″), cucumber, and avocado surfaces were inoculated with L. monocytogenes (LCDC strain). After 1, 24 or 48 h, loosely attached cells were washed off, and inoculated areas were targeted by microbubbles (~0.1-

Targeting of VEGF-mediated angiogenesis to rat myocardium using ultrasonic destruction of microbubbles.

Myocardial angiogenesis mediated by human vascular endothelial growth factor 165 (hVEGF165) cDNA was promoted in rat myocardium using an in vivo-targeted gene delivery system known as ultrasound-targeted microbubble destruction (UTMD). Microbubbles carrying plasmids encoding hVEGF165, or control solutions were infused intravenously during ultrasonic destruction of the microbubbles within the myocardium. Biochemical and histological assessment of gene expression and angiogenesis were performed 5, 10, and 30 days after UTMD. UTMD-treated myocardium contained hVEGF165 protein and mRNA. The myocar

Acoustic control of nonspherical shape oscillations in microbubbles using a Koopman linear quadratic regulator.

Large-amplitude oscillations of spherical bubbles are known to be destabilizing and to induce nonspherical shape dynamics. Acoustic driving of microbubbles can often reach this threshold for excitation, whether the bubbles are in extended free fluid media or near boundaries. In many medical applications, such as diagnostic ultrasound and therapeutic drug delivery, nonspherical shape oscillations produce important secondary effects, such as microstreaming, jetting, or acoustic backscatter. The active control of these shape oscillations is therefore an important open problem. This work continues

Toward a Theranostic Approach for the Brain Tumor Toxicity Profile of Polymer-Shelled Microbubbles.

The establishment of theranostic devices by combining multimodal real-time intraoperative imaging for brain tumor surgery with targeted drug delivery may provide therapeutic advantages for patients with malignant gliomas. Our group has recently developed a new generation of novel microbubbles (MBs), with an air core and a crosslinked poly(vinyl alcohol) shell, called PVA MBs. The PVA MB surface was engineered to support near-infrared (NIR) imaging with a fluorescence probe (C790) for the surgical microscope. The attachment to a cyclic pentapeptide containing the RGD sequence promotes active ad

Contrast-enhanced ultrasonography combined with microbubbles for uterine disorders: Current trends and future perspectives.

Uterine disorders, including fibroids, polyps, and carcinomas, represent significant health concerns for women. Conventional diagnostic methods, including ultrasound and magnetic resonance imaging, often lack the sensitivity required to accurately delineate and evaluate these lesions. Consequently, there is a growing need for advanced imaging techniques to enhance both diagnostic precision and therapeutic decision-making. Standard imaging modalities typically provide low resolution, limiting their effectiveness in evaluating uterine pathology and determining optimal treatment strategies. In th

Ultrasound-Triggered Microbubbles: Novel Targeted Core-Shell for the Treatment of Myocardial Infarction Disease.

Myocardial infarction (MI) is known as a main cardiovascular disease that leads to extensive cell death by destroying vasculature in the affected cardiac muscle. The development of ultrasound-mediated microbubble destruction has inspired extensive interest in myocardial infarction therapeutics, targeted delivery of drugs, and biomedical imaging. In this work, we describe a novel therapeutic ultrasound system for the targeted delivery of biocompatible microstructures containing basic fibroblast growth factor (bFGF) to the MI region. The microspheres were fabricated using poly(lactic-co-glycolic

Improving acute cardiac transplantation rejection therapy using ultrasound-targeted FK506-loaded microbubbles in rats.

Targeted delivery of immunosuppressants to allografts can increase the concentrations of drugs in pathological tissues, improve therapeutic effects and reduce unfavorable side effects. Therefore, we synthesized FK506-loaded microbubbles (FK506-MBs) for site-specific release of FK506 into transplanted hearts by the ultrasound-targeted microbubble destruction (UTMD) technique. The average particle size of FK506-MBs was 1.65 ± 0.32 μm and they had high drug loading and encapsulation efficiency. The in vivo drug concentration in transplanted hearts that were treated with FK506-MBs plus UTMD was ab

Image-Guided Hydrogen Gas Delivery for Protection from Myocardial Ischemia-Reperfusion Injury via Microbubbles.

Cardiomyocyte death induced by ischemia-reperfusion is a major cause of morbidity and mortality worldwide. Hydrogen (H2), as an antioxidant, has been shown to have great potential in preventive and therapeutic applications against lethal injury that occurs from ischemia-reperfusion. However, H2 is sparingly soluble in water, resulting in its poor bioavailability in blood and damaged tissues. Here, we have developed an ultrasound-visible H2 delivery system by loading H2 inside microbubbles (H2-MBs) to prevent myocardial ischemia-reperfusion injury. Using this system, the concentration of H2 in
