Microcystin-LR induces lung injury in mice through the NF-κB/NLRP3 pathway.

Microcystin-LR (MC-LR) a cyclic toxin produced by cyanobacterial species is known to exert detrimental effects on various organs, including lung. Several investigators demonstrated that MC-LR exerts pulmonary toxicity, but the underlying mechanisms remain unclear. This study aimed to investigate whether exposure to MC-LR-induced lung inflammation and examine the underlying mechanisms. Thirty specific pathogen-free (SPF) male mice were allocated into control and MC-LR treatment groups. Mice were intraperitoneally injected with physiological saline or MC-LR (20 μg/kg) daily for a total of 21 day

Short-term ingestion of sublethal microcystin levels disrupts stress response in male mice.

Cyanobacterial harmful algal blooms (cHABs) produce various cyanotoxins, primarily microcystin leucine arginine (MC-LR). Studies demonstrated that intraperitoneal delivery of high-dose MC-LR affects the rat stress response, which is regulated by the hypothalamus-pituitary-adrenal (HPA) axis. In general, during stress parvocellular neurons in the hypothalamic paraventricular nucleus (PVN) secrete corticotropin-releasing hormone (CRH), triggering adrenocorticotropic hormone (ACTH) release from the anterior pituitary, which leads to increased cortisol in humans and corticosterone in rats and mice

Microcystin-LR activates serine/threonine kinases and alters the phosphoproteome in human HepaRG cells.

Microcystin-LR (MCLR) exposure has been associated with development of hepatocellular carcinoma (HCC). Many of the carcinogenic mechanisms for MCLR have been attributed to the induction of cell survival and proliferation through altered protein phosphorylation pathways by inhibition of protein phosphatases 1 (PP1) and PP2A. The current study determined MCLR effects on the phosphoproteome in human HepaRG cells. Differentiated HepaRG cells were treated with either vehicle or MCLR followed by phosphoproteomic analysis and Western blotting of MAPK-activated proteins. MCLR decreased cell viability

Co-exposure to polystyrene microplastics and microcystin-LR aggravated male reproductive toxicity in mice.

Microplastics (MPs) are plastic pollutants with a diameter of less than 5 mm and microcystins (MCs) are natural toxins produced by cyanobacteria. In recent years, the pollution of MPs and MCs attracted widespread attention. However, our understanding about the toxic effects of co-exposure of MPs and MCs on male reproduction is limited. Mice were continuously exposed to 0.04mg/(kg*bw) microcystin-leucine-arginine (MC-LR) or 45 mg/(kg*bw) polystyrene microplastics (PS-MPs) or a mixed solution of 0.04mg/(kg*bw) MC-LR and 45 mg/(kg*bw) PS-MPs by gavage for 28 days in this study. The results showed

Unraveling the intestinal epithelial barrier in cyanotoxin microcystin-treated Caco-2 cell monolayers.

Microcystin is a widespread cyanobacterial toxin that affects the intestine to produce diarrheal symptoms after ingestion of freshwater blue-green algae. Our study aimed to characterize the mechanism by which the toxin leads to diarrhea via epithelial barrier dysfunction in a small intestine Caco-2 cell model. Microcystin-treated human Caco-2 epithelial monolayers were functionally and molecularly analyzed for barrier dysfunction. Tight junctions (TJs) and cell damage were analyzed in relation to transepithelial electrical resistance (TER) changes. TER of microcystin-treated Caco-2 cells was r

Microcystin-LR aerosol induces inflammatory responses in healthy human primary airway epithelium.

Harmful algal blooms plague bodies of freshwater globally. These blooms are often composed of outgrowths of cyanobacteria capable of producing the heptapeptide Microcystin-LR (MC-LR) which is a well-known hepatotoxin. Recently, MC-LR has been detected in aerosols generated from lake water. However, the risk for human health effects due to MC-LR inhalation exposure have not been extensively investigated. In this study, we exposed a fully differentiated 3D human airway epithelium derived from 14 healthy donors to MC-LR-containing aerosol once a day for 3 days. Concentrations of MC-LR ranged from

Chronic Microcystin-LR Exposure Induces Hepatocarcinogenesis via Increased Gankyrin in Vitro and in Vivo.

BACKGROUND/AIMS: Our recent study indicated that the serum microcystin-LR (MC-LR) level is positively linked to the risk of human hepatocellular carcinoma (HCC). Gankyrin is over-expressed in cancers and mediates oncogenesis; however, whether MC-LR induces tumor formation and the role of gankyrin in this process is unclear. METHODS: We induced malignant transformation of L02 liver cells via 35 passages with exposure to 1, 10, or 100 nM MC-LR. Wound healing, plate and soft agar colony counts, and nude mice tumor formation were used to evaluate the tumorigenic phenotype of MC-LR-treated cells.

A Method to Prepare Magnetic Nanosilicate Platelets for Effective Removal of Microcystis aeruginosa and Microcystin-LR.

Algal toxin is a unique type of toxin generated with harmful algal blooms in water bodies. This phenomenon is worsened by eutrophication caused by excessive discharge of nutrients into surface water bodies. Since algal toxins are hard to remove after they enter the water treatment processes, an efficient method is required to inhibit the growth of algal cells, to settle the cells at the bottom of the water body and to removes the toxin from the water. We report an efficient way to prepare a novel nanohybrid material, i.e., magnetic nanosilicate platelet (MNSP), and its effects on the removal o

The ionic strength effect on microcystin and natural organic matter surrogate adsorption onto PAC.

This work aims to contribute to a better understanding of the ionic strength effect on microcystin and natural organic matter (NOM) surrogate adsorption by analyzing the importance of adsorbate molecular size, and surface concentration. Adsorption kinetics and/or isotherms were performed on PAC Norit SA-UF for four microcystin variants (MC-LR, MC-LY, MC-LW, MC-LF), and three NOM surrogates (salicylic acid (SA), tannic acid (TA), Aldrich humic acid (AHA)) at different solution ionic strengths. Results showed that the ionic strength effect depends upon the adsorbate surface concentration, cation

Effect of process variables and natural organic matter on removal of microcystin-LR by PAC-UF.

The release of cyanobacterial toxins, such as microcystin-LR, in drinking water supplies is of increasing concern. In this study, we investigated the use of ultrafiltration (UF) combined with adsorption on powdered activated carbon (PAC) for the removal of microcystin-LR from drinking water. Process variables examined included PAC type, PAC dosage, membrane characteristics (material and pore size), and the presence of natural organic matter (NOM). Due to greater mesopore volume, wood-based activated carbon was up to 4-times more effective at removing microcystin-LR than coconut-based carbon, d

Effects of microcystin-LR on actin and the actin-associated proteins alpha-actinin and talin in hepatocytes.

Microcystin-LR (MCLR) is a commonly encountered blue-green algal hepatotoxin and a known inhibitor of cellular protein phosphatase types 1 and 2A. The toxin causes alterations in, and redistribution of, intermediate filaments, microtubules, and actin microfilaments (MFs) in affected cells. In this study, the effect of MCLR on the sequence of alterations in MFs and actin-associated proteins (AAPs) of isolated hepatocytes was examined in an effort to determine whether morphologic changes induced in MFs by microcystins are a result of prior dislocation of AAPs. We studied the effects of MCLR expo

Broad Cross-Reactivity of a Microcystin ELISA Confirmed Using 19 Quantitative Reference Materials.

Microcystins (MCs) are toxins produced by cyanobacteria and are found worldwide. Exposure may be acute or chronic through contaminated drinking water, food (such as vegetables), dietary supplements, and recreational activities. Acute exposure can cause severe liver damage due to the hepatotoxicity of MCs. More than 300 variants of MCs have been reported, making analysis complex, especially since standards are available for only a few MCs. Therefore, a rapid, cost-effective method capable of detecting all MCs in one analysis is essential to protecting public health and livestock. An antibody wa

Nitrogen pulse drives a prolonged microcystin event by increasing the proportion of toxigenic Microcystis genotypes.

Microcystins (MCs) pose a major risk to drinking water safety, yet their production drivers remain poorly understood. We conducted a year-long, high-frequency monitoring program in a large subtropical reservoir to identify environmental triggers and regulatory mechanisms of Microcystis blooms and toxin production. Microcystis blooms occurred in May and July, followed by a six-month MC contamination event from August to January. An extreme nitrogen pulse was detected on August 1 and was followed by a sharp increase in MC concentrations that peaked in October, suggesting that the N pulse was the

Synergistic reproductive toxicity of microcystin-LR and polystyrene micro/nano-plastics in male zebrafish.

Microcystins (MCs) and micro- or nano-plastics (MPs or NPs) coexist in aquatic environment. The combined toxic effects and underlying mechanisms of MCs and MPs or NPs in male reproductive toxicology remain poorly studied. To investigate potential male reproductive interference, male zebrafish were exposed to microcystin-LR (MCLR; 0, 5, 25 μg/L), polystyrene micro-plastics (PSMPs; 5 μm, 1 mg/L), polystyrene nano-plastics (PSNPs; 80 nm, 1 mg/L), and MCLR + PSMPs or PSNPs for 45 days. UCHL3-mediated proteasomal degradation of GOPC drives microcystin-LR-induced acrosomal dysgenesis.

Growing evidence links environmental exposure to microcystin-leucine-arginine (MC-LR) with declining male fertility, yet its molecular pathogenesis remains poorly understood. In this study, we observed that MC-LR disrupted acrosome biogenesis, a critical process for sperm function. Mechanistically, MC-LR induced proteasomal degradation of Golgi-associated PDZ- and coiled-coil motif-containing protein (GOPC), a key acrosome assembly protein, by disrupting its ubiquitin homeostasis. This degradation resulted from dual inhibition of MC-LR on the deubiquitinase ubiquitin carboxyl terminal hydrolas

Microcystin-LR-Exposure-Induced Kidney Damage by Inhibiting MKK6-Mediated Mitophagy in Mice.

Previous studies have reported that microcystin-LR (MC-LR) levels are highly correlated with abnormal renal function indicators, suggesting that MC-LR is an independent risk factor for kidney damage. However, the evidence for the exact regulation mechanism of MC-LR on kidney damage is still limited, and further in-depth exploration is needed. In addition, the mitochondria-related mechanism of MC-LR leading to kidney damage has not been elucidated. To this end, the present study aimed to further explore the mechanism of mitophagy related to kidney damage induced by MC-LR through in vitro and in

Measurement of Microcystin Activity in Human Plasma Using Immunocapture and Protein Phosphatase Inhibition Assay.

Microcystins are toxic chemicals generated by certain freshwater cyanobacteria. These chemicals can accumulate to dangerous levels during harmful algal blooms. When exposed to microcystins, humans are at risk of hepatic injury, including liver failure. Here, we describe a method to detect microcystins in human plasma by using immunocapture followed by a protein phosphatase inhibition assay. At least 279 microcystins have been identified, and most of these compounds share a common amino acid, the Adda side chain. We targeted this Adda side chain using a commercial antibody and extracted microcy

Recovery of reproductive function of female zebrafish from the toxic effects of microcystin-LR exposure.

Fish has a strong resistance to microcystins (MCs), cyclic heptapeptide cyanotoxins, known as endocrine disrupting chemicals (EDCs) which are released during cyanobacterial blooms and many laboratory and field studies have found the hepatic recovery of fish from the MCs exposure. The aim of the present study was to investigate the recovery mechanisms of reproductive function of adult zebrafish (Danio rerio) from microcystin-LR (MC-LR) exposure. Therefore, adult female zebrafish were exposed to 0, 1 or 50 μg/L of MC-LR for 21days and transferred to MC free water for another 21 days to investiga
