Ultrastructure of submandibular salivary glands of mouse: TEM and HRSEM observations.

The fine structure of submandibular glands of mouse were analyzed using light microscopy (LM), high resolution scanning electron microscopy (HRSEM), and transmission electron microscopy (TEM) methods. For LM, the specimens were embedded in Spurr resin, stained by toluidin blue solutions. For TEM, the tissues of submandibular salivary glands were fixed with modified Karnovsky solution and postfixed with osmium tetroxide. For HRSEM, the tissues were fixed with 2% osmium tetroxide solution in 1/15M sodium phosphate buffer (pH 7.4). The samples were immersed successively in dymethylsulphoxide and

The microanatomy of the palatine tonsils of the one-humped camel (Camelus dromedarius).

Tonsils form a first line of defense against foreign antigens and are also a route of entry and a replication site for some pathogens. The palatine tonsils are the largest of all the tonsils. Despite their general importance, little is known about the microanatomy of the palatine tonsils of the one-humped camel. Palatine tonsils of 10 clinically healthy male camels were obtained directly after slaughtering for human consumption. The tonsils were examined macroscopically and by light, scanning, and transmission electron microscopy. Palatine tonsils had the unique form of several spherical macro

Fe(0) nanoparticles for nitrate reduction: stability, reactivity, and transformation.

The pyrophoric character of zerovalent iron nanoparticles and cumbersome handling of this material has been a drawback in practical applications, despite the expectation of an enhanced reactivity. We have been interested in how the iron nanoparticles can gain stability in air without significantly sacrificing reactivity. The freshly synthesized iron nanoparticles ignited spontaneously upon exposure to air. However, when exposed slowly to air, an approximately 5 nm coating of iron oxide was formed on the surface of particles. The oxide shell did not thicken for at least two months, indicating n

Ultrastructure of aortic elastic fibers in copper-deficient Sika deer (Cervus nippon Temminck).

Light microscopic and transmission and scanning electron microscopic observations were performed on the aortas of two 4- and 6-year-old deer affected with cervine ataxia and two 6-month- and 4-year-old healthy deer. Examination of the aortas from affected deer by transmission electron microscopy revealed the absence of distinct elastic laminae in the internal elastic lamina and tunica media, but discontinuous and irregular clumps of elastin were present. Scanning electron microscopy disclosed immature architecture of elastic fibers in the aortas from the copper-deficient deer, and the architec

Sample Preparation Method of Scanning and Transmission Electron Microscope for the Appendages of Woodboring Beetle.

This report described sample preparation methods that scanning and transmission electron microscope observations, demonstrated by preparing appendages of the woodboring beetle, Chlorophorus caragana Xie & Wang (2012), for both types of electron microscopy. The scanning electron microscopy (SEM) sample preparation protocol was based on sample chemical fixation, dehydration in a series of ethanol baths, drying, and sputter-coating. By adding Tween 20 (Polyoxyethylene sorbitan laurate) to the fixative and the wash solution, the insect body surface of woodboring beetle was washed more cleanly in S

Morphology and ultrastructure of Dufour's and venom glands in the ant Camponotus japonicus Mayr (Hymenoptera: Formicidae).

The Dufour's and venom glands are two important exocrine glands in ants. In this study, the morphology and fine structure of these two glands are described in the ant Camponotus japonicus Mayr. The Dufour glands have a characteristic bilobed shape and show a difference in size and color between the female castes (minor and major workers, alate and dealate queens). The external surface of Dufour's gland shows different features among the female castes. It appears more hypertrophied in major workers than in the other castes, indicating for a more pronounced function. The cells of the glandular e

Using human neural crest-derived progenitor cells to investigate osteogenesis: an in vitro study.

Human tooth contains a distinct population of neural crest-derived progenitor cells (dNC-PCs) which are known to give rise to specialized daughter cells of an osteogenic lineage. We hypothesised that dNC-PCs could develop into neural crest-derived bone in a self-propagating and extracorporal culture system. Thus, we examined the three-dimensional structure obtained from osteogenic-stimulated dNC-PCs by morphological, biochemical and spectroscopic methods. After the onset of stimulation, cells formed a multilayer with outer cells covering the surface and inner cells secreting a hyaline matrix.

Whipple's disease: a continuous challenge.

Whipple's disease is a rare disease with protean clinical manifestations, often mimicking those of other pathological conditions. We describe two new cases, one admitted to hospital only after Giardia lamblia infestation had drawn attention to gut symptoms, and the other who was treated for a long time with steroids for suspected Horton's arteritis. Once again, we stress the importance of bearing this polymorph disease in mind, especially in older people.

Itraconazole in pityriasis versicolor: ultrastructural changes in Malassezia furfur produced during treatment.

Twenty-eight patients with pityriasis versicolor were treated orally with 200 mg of itraconazole per day in an open, randomized comparison of five-day and seven-day treatment regimens. The morphologic changes in Malassezia furfur produced by treatment were confirmed by studies with transmission and scanning electron microscopy. The clinical results showed that seven days of treatment were more effective than five days. Transmission electron microscopy revealed cytopathic changes in the fungus at the end of treatment, intracellular necrosis being complete within seven to 28 days. Cell-surface a

Structure and diversity of abdominal exocrine glands in larvae of Leiodidae (Insecta: Coleoptera: Staphylinoidea).

Epidermal glands are well-known in adult beetles, but their presence and function are less studied in larvae. This study reveals, for the first time for the family Leiodidae, the morphology and ultrastructure of epidermal, paired glands of abdominal segments present in larvae. They are documented in all larval stages of the scavenger beetle Sciodrepoides watsoni watsoni (using a multimodal microscopic approach: light and scanning electron, and transmission electron microscopy). Overview of these structures is also documented for other representatives of Leiodidae and Agyrtidae, basal lineages

Artifact-free preparation of biological samples for SEM by optimized water freeze-drying.

Water freeze-drying has traditionally been considered an inappropriate drying method for biological samples for scanning electron microscopy (SEM) because ice crystals can potentially form and damage cell structures. Chemical fixation is also considered essential for the SEM observation of soft cells, such as protozoa and animal cells. We succeeded in preparing specimens of freshwater protists for SEM using simple water freeze-drying without chemical fixation. Specifically, a suspension of live cells in water was frozen by contact with a − 80 °C copper block using an outer cylinder that ensure

Ultrastructure of lymphatic stomata in the tunica vaginalis of humans.

Lymphatic stomata are small openings of lymphatic capillaries on the surface of the mesothelium that lines the serous cavity and have the function of active absorption. They play an important role in physiological and pathological conditions. The cavity of the tunica vaginalis is a typical serous cavity of the testis, but the lymphatic stomata of the tunica vaginalis of humans have never been reported. Here, we studied their ultrastructure by scanning and transmission electron microscopy. The submesothelial connective tissue with foramina was investigated after the mesothelial cells were diges

Ultrastructure of spermatogenesis in the testis of Paragonimus heterotremus.

Lung fluke, Paragonimus heterotremus, is a flatworm causing pulmonary paragonimiasis in cats, dogs, and humans in Southeast Asia. We examined the ultrastructure of the testis of adult P. heterotremus with special attention to spermatogenesis and spermiogenesis using scanning and transmission electron microscopy. The full sequence of spermatogenesis and spermiogenesis, from the capsular basal lamina to the luminal surface, was demonstrated. The sequence comprises spermatogonia, spermatocytes with obvious nuclear synaptonemal complexes, spermatids, and eventual spermatozoa. Moreover, full steps

The morphology of the squirrel spermatozoon: a highly complex male gamete with a massive acrosome.

This study describes the morphology of the spermatozoon from the cauda epididymidis of representative members of two squirrel subfamilies, the Sciurinae and Callosciurinae, as determined by fluorescent, scanning, and transmission electron microscopy. All species examined possess a massive apical segment of the sperm acrosome. It varied markedly in the extent of its caudal flexion but was always much larger, and more complex, than that of the spermatozoon of most other rodents so far documented, although somewhat similar to that of some hystricomorph species. Because this sperm form appears to

Morphology of epithelial cells lining the digestive tract of the giant keyhole limpet, Megathura crenulata (Mollusca; Vetigastropoda).

To understand the digestive functions in the giant keyhole limpet, it is important to know the types of cells present in each region of the gut and their roles in the secretion of digestive enzymes and absorption of nutrients. This study describes the morphology of cells lining the entire gut and identifies sites that may be secreting materials to aid digestion. Previous studies involving electron microscopy and enzyme analysis have focused on the salivary and digestive glands of several gastropods. Studies on the rest of the gut tract typically include only histological descriptions of the ep

High resolution transmission electron microscopy of developing enamel in the Australian lungfish, Neoceratodus forsteri (Osteichthyes: Dipnoi).

The permanent tooth plates of the Australian lungfish, Neoceratodus forsteri, are covered by enamel that develops initially in a similar manner to that of other vertebrates. As the enamel layer matures, it acquires several unusual characteristics. It has radially oriented protoprismatic structures with the long axes of the protoprisms perpendicular to the enamel surface. Protoprisms can be defined as aggregations of hydroxyapatite crystals that lack the highly ordered arrangement of the rods of mammalian enamel but are not without a specific structure of their own. The protoprisms are arranged

Female genital system of the folding-trapdoor spider Antrodiaetus unicolor (Hentz, 1842) (Antrodiaetidae, Araneae): ultrastructural study of form and function with notes on reproductive biology of spiders.

The genitalia of the female folding-trapdoor spider Antrodiaetus unicolor are characterized by two pairs of spermathecae that are arranged in a single row and connected to the roof of the bursa copulatrix. Each single spermatheca is divided into three main parts: stalk, bowl, and bulb, which are surrounded by the spermathecal gland. The epithelium of the spermathecal gland is underlain by a muscle meshwork and consists of different types of cells partly belonging to glandular cell units (Class 3 gland cells) that extend into pores in the cuticle of the stalk and bowl. Interestingly, the bulb l

Ovary Organization and a Putative Germ Plasm in Tubuliferan and Terebrantian Thrips (Thysanoptera).

Comparison of the detailed morphology of ovaries allows for an evolutionary conclusion in insects. The current study characterized the ovary/ovariole organization in both thysanopteran subtaxa (Terebrantia and Tubulifera) and confirmed the presence of a (putative) germ plasm in the oocytes. Using transmission and scanning electron microscopy as well as histological methods, we show that the morphology of ovaries in the studied terebrantians and tubuliferans differs in the organization of the anterior ovary/ovariole region, that is, the germaria and terminal filaments. Whereas in tubuliferan, H

Ultrastructural characterization of Amyloodinium ocellatum (Dinoflagellata): Insights into trophont morphology and parasitic mode.

Amyloodinium ocellatum is a parasitic dinoflagellate that infects marine fish in tropical and subtropical regions, causing significant economic losses in aquaculture. This study used light microscopy (LM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and electron tomography (ET) to comprehensively characterize the morphology, ultrastructure, and specialized attachment apparatus of A. ocellatum trophonts. The trophonts primarily parasitized the gill filaments of infected fish, reaching 89.8 ± 10.3 μm after 48 h of infection. Under LM, trophonts appeared as dark, s
