Proximity labeling reveals non-centrosomal microtubule-organizing center components required for microtubule growth and localization.

Microtubules are polarized intracellular polymers that play key roles in the cell, including in transport, polarity, and cell division. Across eukaryotic cell types, microtubules adopt diverse intracellular organization to accommodate these distinct functions coordinated by specific cellular sites called microtubule-organizing centers (MTOCs). Over 50 years of research on MTOC biology has focused mainly on the centrosome; however, most differentiated cells employ non-centrosomal MTOCs (ncMTOCs) to organize their microtubules into diverse arrays, which are critical to cell function. To identify

Increased acetylation of microtubules rescues human tau-induced microtubule defects and neuromuscular junction abnormalities in Drosophila.

Tau normally associates with and stabilizes microtubules (MTs), but is hyperphosphorylated and aggregated into neurofibrillary tangles in Alzheimer's disease and related neurodegenerative diseases, which are collectively known as tauopathies. MTs are regulated by different forms of post-translational modification, including acetylation; acetylated MTs represent a more stable microtubule population. In our previous study, we showed that inhibition of histone deacetylase 6 (HDAC6), which deacetylates tubulin at lysine 40, rescues defects in MTs and in neuromuscular junction growth caused by tau

Microtubule inner proteins in apicomplexan parasites.

Microtubule inner proteins (MIPs) are integral components within the microtubule lumen of various organisms, contributing to microtubule structural integrity and functionality. Apicomplexan parasites, including Plasmodium spp. and Toxoplasma gondii, exhibit a range of specialized tubulin structures, such as axonemal microtubules, subpellicular microtubules (SPMTs), and conoid fibers, playing critical roles in cellular morphology and motility. Yet, in contrast with model organisms, only a few MIPs have been characterized in apicomplexans so far. Recent advances in cryo-electron tomography and s

Mitochondrial and microtubule defects in Exfoliation Glaucoma.

Exfoliation Syndrome is an age-related systemic condition characterized by large aggregated fibrillar material deposition in the anterior eye tissues. This aggregate formation and deposition on the aqueous humor outflow pathway are significant risk factors for developing Exfoliation Glaucoma (XFG). XFG is a multifactorial late-onset disease that shares common features of neurodegenerative diseases, such as increased protein aggregation, impaired protein degradation, and oxidative and cellular stress. XFG patients display decreased mitochondrial membrane potential and mitochondrial DNA deletion

Microtubule-based transport in filamentous fungi.

Defects in microtubule-based transport are implicated in many neuropathologies. The filamentous fungi Aspergillus nidulans and Ustilago maydis are valuable models for studying transport owing to their yeast-like genetic and biochemical tractability and metazoan-like dependence on microtubule-based transport for cellular trafficking. In these organisms the role of microtubules in nuclear positioning is well studied, but recent work has expanded the range of cargos to include endosomes, messenger RNA, secretory vesicles, peroxisomes, and nuclear pore complexes, reflecting the diversity of metazo

Association of adenovirus with the microtubule organizing center.

Adenoviruses (Ad) must deliver their genomes to the nucleus of the target cell to initiate an infection. Following entry into the cell and escape from the endosome, Ad traffics along the microtubule cytoskeleton toward the nucleus. In the final step in Ad trafficking, Ad must leave the microtubule and establish an association with the nuclear envelope. We hypothesized that in cells lacking a nucleus, the capsid moves to and associates with the microtubule organizing center (MTOC). To test this hypothesis, we established an experimental system to examine Ad trafficking in enucleated cells compa

Influence of monoclonal antibodies on microtubule assembly.

The influence on microtubule assembly in vitro of monoclonal antibodies against microtubule-associated proteins (MAPs) was studied. Light scattering was used for measuring net polymer formation and electron microscopy for determining the influence of antibodies on microtubule morphology. Control experiments showed that nonimmune mouse IgG had no effect on either the assembly or appearance of microtubules. The same was true for monoclonal antibodies against MAP1. At low levels, antibodies against MAP2 caused the aggregation of microtubules into bundles, an effect that did not occur with antibod

Microtubules and nuclear pores position centromeres and genes in neural stem cells.

Chromatin's spatial organization is important for cell division, epigenetic memory, and gene regulation. Chromatin architecture is often reflected in nonrandom centromere positioning, which changes during development and disease. However, the mechanisms and functions of centromere positioning remain largely unknown. Here, we report that Drosophila neural stem cells cluster centromeres near the nuclear envelope, adjacent to the apical centrosome. This asymmetric centromere positioning persists throughout interphase but is lost in differentiated progeny. Asymmetric centromere positioning is regu

The cortical microtubules of Toxoplasma gondii underlie the helicity of parasite movement.

Motility is essential for apicomplexan parasites to infect their hosts. In a three-dimensional (3-D) environment, the apicomplexan parasite Toxoplasma gondii moves along a helical path. The cortical microtubules, which are ultra-stable and spirally arranged, have been considered to be a structure that guides the long-distance movement of the parasite. Here we address the role of the cortical microtubules in parasite motility, invasion, and egress by utilizing a previously generated mutant (dubbed "TKO") in which these microtubules are destabilized in mature parasites. We found that the cortica

NDP52 tunes cortical actin interaction with astral microtubules for accurate spindle orientation.

Oriented cell divisions are controlled by a conserved molecular cascade involving Gαi, LGN, and NuMA. Here, we show that NDP52 regulates spindle orientation via remodeling the polar cortical actin cytoskeleton. siRNA-mediated NDP52 suppression surprisingly revealed a ring-like compact subcortical F-actin architecture surrounding the spindle in prophase/prometaphase cells, which resulted in severe defects of astral microtubule growth and an aberrant spindle orientation. Remarkably, NDP52 recruited the actin assembly factor N-WASP and regulated the dynamics of the subcortical F-actin ring in mit

Crowding and Pausing Strongly Affect Dynamics of Kinesin-1 Motors along Microtubules.

Molecular motors of the kinesin-1 family move in a directed and processive fashion along microtubules. It is generally accepted that steric hindrance of motors leads to crowding effects; however, little is known about the specific interactions involved. We employ an agent-based lattice gas model to study the impact of interactions that enhance the detachment of motors from crowded filaments on their collective dynamics. The predictions of our model quantitatively agree with the experimentally observed concentration dependence of key motor characteristics including their run length, dwell time,

Drosophila spichthyin inhibits BMP signaling and regulates synaptic growth and axonal microtubules.

To understand the functions of NIPA1, mutated in the neurodegenerative disease hereditary spastic paraplegia, and of ichthyin, mutated in autosomal recessive congenital ichthyosis, we have studied their Drosophila melanogaster ortholog, spichthyin (Spict). Spict is found on early endosomes. Loss of Spict leads to upregulation of bone morphogenetic protein (BMP) signaling and expansion of the neuromuscular junction. BMP signaling is also necessary for a normal microtubule cytoskeleton and axonal transport; analysis of loss- and gain-of-function phenotypes indicate that Spict may antagonize this

Discodermolide, a cytotoxic marine agent that stabilizes microtubules more potently than taxol.

Computer-assisted structure analysis indicated (+)-discodermolide, a polyhydroxylated alkatetraene lactone marine natural product, was an antimitotic compound, and we confirmed this prediction. Previous work had shown an accumulation of discodermolide-treated cells in the G2/M portion of the cell cycle, and we have now found that discodermolide arrests Burkitt lymphoma cells in mitosis. Discodermolide-treated breast carcinoma cells displayed spectacular rearrangement of the microtubule cytoskeleton, including extensive microtubule bundling. Microtubule rearrangement that occurred with 10 nM di

Dynamic properties of nucleated microtubules: GTP utilisation in the subcritical concentration regime.

Microtubule assembly kinetics have been studied quantitatively under solution conditions supporting microtubule dynamic instability. Purified GTP-tubulin (Tu-GTP) and covalently cross-linked short microtubule seeds (EGS-seeds; Koshland et al. (1988) Nature 331, 499) were used with and without biotinylation. Under sub-critical concentration conditions ([Tu-GTP] < 5.3 microM), significant microtubule growth of limited length was observed on a proportion of the EGS-seeds by immuno-electron microscopy. A sensitive fluorescence assay for microtubule GDP production was developed for parallel assessm

Occurrence of fasciculated microtubules at nodes of Ranvier in rat spinal roots.

Fascicles of microtubules, previously considered to be unique to the initial segments of myelinated axons, were found at nodes of Ranvier of sensory and motor axons in rat spinal roots, though their occurrence was limited to the proximal portion of the axons. No fasciculated microtubules were noticed in the internodes of myelinated axons or in unmyelinated axons. In transverse sections, microtubules in fascicles were characteristically cross-linked by short filamentous strands at a centre-to-centre distance of 36-40 nm. In sensory axons, the density of fasciculated microtubules at the node of

Supernumerary microtubules in the cilia of two siblings causing "immotile cilia syndrome".

The function and ultrastructure of nasal cilia in 2 siblings with unexplained chronic sinusitis and bronchitis were investigated. Both patients lacked nasal mucociliary clearance (greater than 30 min). On electron microscopy, the ultrastructure of nasal cilia showed an extra doublet or supernumerary microtubules with disorganized microtubular network. Other axonemal components were normally present. It is conceivable that an "extra" component of the axoneme may be added to those "defects" already recognized as a cause of the "immotile-cilia syndrome".

Stability of neuronal microtubules to high pressure in vivo and in vitro.

Neuronal microtubules in a variety of nerve cell types are unaffected by high hydrostatic pressures over a range of 1400-10,000 pounds/inch(2) and periods of 10-45 min. Similarly, purified tubulin polymerized to form microtubules in vitro were not depolymerized by the same range of pressures. The depolymerization of microtubules in several types of non-neuronal cells, which has been reported, may have been over-generalized with regard to the direct action of pressure on microtubule stability.

FAP53 facilitates formation of microtubule doublets.

The formation of microtubule doublets (MTDs) is a foundational step in cilia biogenesis, yet how B-tubule nucleation is initiated at the molecular level remains elusive. Here, we identify FAP53 as a factor that facilitates B-tubule assembly in a scaffold-dependent manner. In vitro reconstitution demonstrated that recombinant FAP53 enhances MTD formation in the presence of preassembled microtubule scaffolds, but is insufficient to drive de novo assembly from free tubulin alone. In cultured HeLa cells, coexpression of CFAP53 and CFAP20-an inner junction protein-induced ectopic MTD-like structure
