The primary motor cortex (M1) is one of the main cortical areas involved in motor learning. However, little is known about its differential role in the learning of the relative and absolute dimensions of motor skills. We investigated the role of M1 in the learning of the dimensions of a complex motor skill. Forty-eight participants practiced golf putting and were stimulated for 20 minutes with real or sham bihemispheric tDCS before acquisition. tDCS improved global performance from pre- to post-test. Only those with worse initial performance who were stimulated by tDCS showed a significant imp

This report conveys the results of an immunocytochemical and ultrastructural study of the motor cortices of six patients with clinically and pathologically-diagnosed lower motor neuron disease (LMND) such as progressive spinal muscular atrophy, progressive bulbar palsy, or both. These patients showed neither upper motor neuron signs nor upper motor neuron system involvement including the corticospinal tract in postmortem tissues after conventional stainings. Specimens from 12 age-matched normal individuals served as controls. All patients showed loss of brainstem motor neurons and anterior hor

The primary motor cortex (M1) is broadly acknowledged for its crucial role in executing voluntary movements. Yet, its contributions to cognitive and sensory functions remain largely unexplored. Transcranial direct current stimulation (tDCS) is a noninvasive neurostimulation method that can modify brain activity, thereby enabling the establishment of a causal link between M1 activity and behavior. This study aimed to investigate the online effects of tDCS over M1 on cognitive-motor and sensory-motor functions. Sixty-four healthy participants underwent either anodal or sham tDCS while concurrent

The ability to manipulate objects is a fundamental human skill that relies primarily on the motor system. However, effective object manipulation would not be possible without the continuous information provided by the somatosensory system. Cutaneous tactile feedback is particularly important when a movement must be adjusted while performing an action. Efficient interactions between the tactile and motor systems are therefore paramount for fine motor behaviour, as clearly demonstrated by the profound impairments observed following lesions to sensorimotor brain regions. However, somatosensory de

Motor imagery (MI) has attracted increased interest for motor rehabilitation as many studies have shown that MI shares the same neural networks as motor execution (ME). Nevertheless, MI in terms of facial movement has not been studied extensively; thus, in the present study, we investigated shared neural networks between facial motor imagery (FMI) and facial motor execution (FME). In addition, FMI somatotopy within-face was investigated between the forehead and the mouth. Functional MRI was used to examine 34 healthy individuals with ME and MI paradigms for the forehead and the mouth. The gene

Many recent models of schizophrenia have attempted to explain the so-called first-rank symptoms in terms of a breakdown in the self-monitoring of thoughts and behaviours. These models have focused on the most common symptom of schizophrenia auditory hallucinations-suggesting that they may represent disordered self-monitoring of internal speech. As such, much attention has been given to the role of the temporal and frontal cortices in the clinical presentation of patients with schizophrenia. In this review, we examine the role of the posterior parietal cortex (PPC) in schizophrenia within the c

We investigated the basal ganglia, motor cortex area 4, and supplementary motor area (SMA) using functional magnetic resonance imaging (fMRI) and five motor tasks: switching between finger and toe movements, writing, finger tapping, pronation/supination, and saccadic eye movements. We found reliable activation in the caudate nucleus and putamen in single subjects without the need for inter-subject averaging. Percent signal changes in basal ganglia were smaller by a factor of three than those in SMA or motor cortex (1% vs. 2.5-3%). There was a definite foot-dorsal, hand-ventral basal ganglia so

OBJECTIVES: The ipsilateral silent period (iSP) is used to study interhemispheric control of voluntary motor output. It involves a brief suppression of electromyographic (EMG) activity in a muscle during isometric contraction, triggered by Transcranial Magnetic Stimulation (TMS) over the ipsilateral primary motor cortex (M1). The aim of this study was to investigate cortical activity associated with iSP.
METHODS: The study involved 28 healthy right-handed volunteers who performed maximal contractions of the left hand (unimanual) or both hands (bimanual) across conditions with and without TMS.

Previous research has demonstrated that hypomania is associated with approach motivation and activity in the left prefrontal cortex (PFC). Other research has linked left motor cortex excitability to approach motivation, suggesting the existence of connections between the motor cortex and PFC. The present research extends this work using unilateral hand contractions to manipulate contralateral cortical activity, and examining the relationship between motor cortex and PFC inter-electrode EEG coherence and hypomania. Within the right-hand contraction condition, hypomania related to greater connec

AIM OF THE STUDY: Transcranial magnetic stimulation (TMS) is used to measure corticospinal excitability (CSE) from the primary motor cortex (M1) in humans through motor-evoked potentials (MEPs). The variability of CSE responses to transcranial direct current stimulation (tDCS) protocols is high and needs to be reproduced in the healthy population. The M1 and posterior parietal cortex (PPC) are anatomically and functionally connected and could play a role in understanding the variability in CSE responses. We tested the individual MEPs following a common cathodal (ctDCS) protocol over the M1 and

In common with other secondary motor areas, the macaque ventral premotor cortex (PMv) gives rise to corticospinal projections; it also makes numerous reciprocal corticocortical connections with the primary motor cortex (M1). Repetitive intracortical microstimulation (rICMS) of the PMv gives rise to movements of the hand and digits. To investigate whether these motor effects are dependent on the corticocortical interactions with M1, the effect of reversible inactivation of the M1 hand area was tested in three macaque monkeys with chronically implanted intracortical electrodes in the hand repres

Objective. Enable neural control of individual prosthetic fingers for participants with upper-limb paralysis.Approach. Two tetraplegic participants were each implanted with a 96-channel array in the left posterior parietal cortex (PPC). One of the participants was additionally implanted with a 96-channel array near the hand knob of the left motor cortex (MC). Across tens of sessions, we recorded neural activity while the participants attempted to move individual fingers of the right hand. Offline, we classified attempted finger movements from neural firing rates using linear discriminant analy

UNLABELLED: Background/Goal: Parkinson's disease (PD) disrupts dopaminergic transmission, leading to motor deficits and altered activity in the primary motor cortex (M1). While M1 modulation is critical for motor control, its response to dopaminergic degeneration and treatment remains unclear. This study aimed to characterize M1 neuronal activity and motor behavior in hemiparkinsonian rats using in vivo calcium imaging across naïve, lesioned, and levodopa-treated states.
METHODS: Thirteen Sprague Dawley rats were injected with GCaMP6f in the M1 and implanted with a GRIN lens and guide cannula

Parkinson's Disease (PD) is the second most prevalent neurodegenerative disease worldwide due to loss of dopaminergic neurons projecting from the basal ganglia (BG). It is associated with various motor symptoms that are grouped into subtypes, each with different clinical presentations and disease progressions. Neuroimaging biomarkers focusing on regions a part of motor circuits projecting from the BG can distinguish and improve overall subtyping. The supplementary motor cortex (SMC) is well established in PD neuropathology and associated with freezing of gait and bradykinesia, but has not been

While motor recovery following mild stroke has been extensively studied with neuroimaging, mechanisms of recovery after moderate to severe strokes of the types that are often the focus for novel restorative therapies remain obscure. We used fMRI to: 1) characterize reorganization occurring after moderate to severe subacute stroke, 2) identify brain regions associated with motor recovery and 3) to test whether brain activity associated with passive movement measured in the subacute period could predict motor outcome six months later. Because many patients with large strokes involving sensorimot

Sensory-motor integration is crucial in the processing of chronic pain. The primary motor cortex (M1) is emerging as a promising target for chronic pain treatment. However, it remains elusive how nociceptive sensory inputs influence M1 activity and how rectifying M1 defects, in turn, regulates pain processing at cellular and network levels. We show that injury/inflammation leads to hypoactivity of M1Glu pyramidal neurons by excitation-inhibition imbalance between the primary somatosensory cortex (S1) and the M1. The impaired M1 output further weakens inputs to excitatory parvalbumin neurons of

The dynamical state of cortical neural activity constrains the complexity of functions it can perform. A marginally stable dynamical state - called criticality - is thought to be beneficial for brain functions that require multiple time scales, broad dynamic range, and large information storage and transmission. A growing body of evidence suggests that criticality is a feature of healthy brain dynamics, but breaks down in certain brain disorders. Here we ask whether Parkinson's disease incurs deviation from criticality compared to healthy controls. We analyze human resting state EEG activity i

BACKGROUND: Essential tremor (ET) is one of the most prevalent movement disorders. However, the complete understanding of ET pathophysiology remains elusive.
OBJECTIVE: To explore the pathophysiological role of primary motor cortex (M1) in ET, specifically exploring its neurophysiological changes and their correlation with voluntary motor abnormalities.
METHODS: We recruited 30 ET patients and 18 healthy controls (HC). Evaluations were conducted on patients using clinical scales. Transcranial magnetic stimulation (TMS) was used to assess M1 excitability, including motor thresholds and motor

In patients with Parkinson's disease, beta (β) and gamma (γ) oscillations are altered in the basal ganglia, and this abnormality contributes to the pathophysiology of bradykinesia. However, it is unclear whether β and γ rhythms at the primary motor cortex (M1) level influence bradykinesia. Transcranial alternating current stimulation (tACS) can modulate cortical rhythms by entraining endogenous oscillations. We tested whether β- and γ-tACS on M1 modulate bradykinesia in patients with Parkinson's disease by analysing the kinematic features of repetitive finger tapping, including movement amplit
